N:P Ratio 조절에 의한 단기 영양염 첨가 바이오에세이(NEBs) 및 1차 생산력(엽록소-a)의 반응성 테스트 (Short-term Nutrient Enrichment Bioassays (NEBs) by Manipulation of TN:TP Ratios and the Response of Primary Productivity (as Chlorophyll-a))

  • 본 연구는 식물성 플랑크톤의 1차 생산력에 대한 N:P ratio의 영향을 분석하기 위해 "영양염 첨가실험(NEBs)"을 실시하였다. 영양염 첨가실험(NEBs)에 의한 N:P Ratio의 영향은 대청호에서 측정된 수질데이터와 비교분석하였다. 단기 영양염 첨가실험 결과, 인 (P)을 첨가한 처리군들 (N:P Ratio=5, 15, 20, 30)에서의 1차 생산력의 반응이 대조군 (Control)과 인(P)을 첨가한 처리군 (N:P Ratio=80, $T_V$), 질소(N)를 첨가한 처리군(N:P Ratio=150, $T_{VI}$)에서보다 높았다. 또한 질소 (N)를 처리한 처리군에서는 대조군과 모든 처리군에서보다 1차 생산력의 반응이 유의하게 작았다. 영양염 첨가실험의 결과, 식물성 플랑크톤의 성장에 인이 제한영양염으로 작용하고 있었으며, 질소첨가 (Spiking N)는 식물성 플랑크톤의 성장을 억제한 것으로 사료된다. 대청호의 영양염 변이 분석 결과, 최소 N:P Ratio에서 엽록소-a의 최대농도가 나타났고, N:P Ratio는 식물성 플랑크톤의 성장에 대한 핵심 조절자로 사료되었다. 본 연구결과를 종합해 볼 때, N:P Ratio가 식물성 플랑크톤의 성장을 조절하는 핵심 인자로 작용 할 것으로 사료된다.

Production of Lipase-catalyzed Structured Lipid from Olive Oil with Omega-3 Polyunsaturated Fatty Acids

  • Acidolysis of olive oil with omega-3 (n-3) polyunsaturated fatty acids (PUFAs) was carried out to produce a structured lipid. Novozym $435^{(R)}$ from Candida antarctica was used as the biocatalyst. Response surface methodology (RSM) was used to determine optimum conditions for lipase-catalyzed enrichment of olive oil. Three factors, 5 levels, central composite design was used. The effects of incubation time, temperature, and substrate mole ratio on incorporation ratio (n-3 fatty acids/total fatty acids, %) were investigated. From the evaluation of response surface graphs, the optimal conditions for incorporation of long chain n-3 PUFAs into olive oil were $40-60^{\circ}C$ for temperature, 30-45 hr for reaction time, and 3:1-5:1 (n-3 fatty acids/olive oil) for substrate mole ratio. Experiments conducted under optimized conditions predicted by the model equation obtained from RSM yielded structured lipids with 50.8% n-3 PUFAs. This value agreed well with that predicted by the model. Oxidative stability tests showed that the product was more susceptible to oxidation than unmodified olive oil. Antioxidant addition improved the oxidative stability of the product.

Lead(II)-selective Polymeric Electrode Using a Schiff Base Complex of N,N'-Bis-thiophene-2-ylmethylene-ethane-1,2-diamine as an Ion Carrier

  • We prepared lead ion-selective PVC membranes that were based on N,N'-bis-thiophene-2-ylmethylene-ethane-1,2-diamine as a membrane carrier. The membrane electrode has a linear dynamic range between 1.0 ${\times}$ $10^{-5}$ and 1.0 ${\times}$ $10^{-1}$ M with a Nernstian slope of 29.79 mV per decade, and its detection limit was 2.04 ${\times}$ $10^{-6}$ M at room temperature. The potentiometric response is independent of the pH of the solution in the pH range of 5-7. The proposed electrode revealed good selectivity and response for $Pb^{2+}$ over a wide variety of other metal ions in pH 5.0 buffer solutions, and there was good reproducibility of the base line on the subsequent measurements. The membrane electrode has a relatively fast response time, satisfactory reproducibility and a relatively long life time.

반응표면분석을 이용한 PoN 블록체인 시스템 합의품질 개선 (Improvement of Consensus Quality for PoN Blockchain System Using Response Surface Methodology)

  • Purpose: The purpose of this study was to suggest an improved version of Proof-of-Nonce (PoN) algorithm, which is a distributed consensus algorithm used for block chain system. Methods: First, we used response surface method for design of experiment that is to generate experimental points considering non-linear relationship among variables. Then, we employed overlapped contour plots for visualizing the impact of control variables to performance target. Results: First, we modified the consensus procedure of the existing PoN algorithm by diminishing the content of the exchanged message. Then, we verified the performance improvement of the new PoN algorithm by performing a numerical experiment and paired t-test. Finally, we established new regression models for consensus time and Transactions per second (TPS) and proposed a method for optimizing control variables for obtaining performance target. Conclusion: We could improve the performance of the previous version of PoN algorithm by modifying the content of the exchanged message during 4-steps of consensus procedure, which might be a stepping stone for designing an efficient and effective consensus algorithm for blockchain system with dynamic operation environment.

과도운전시 가솔린기관의 성능평가 (Evaluation of Transient Performance of Carburettered Gasoline Engine)

  • An experimental study was carried out to evaluate the characteristics of transient performance of carburettered gasoline engine under rapid accelerating transient driving conditions. In order to evaluate the characteristics of transient performance quantitatively, the concept of dead time $t_d$ response delay time $t_r$ are introduced. Performance parameters such as air mass fiowrate Gat, engine speed N, manifold boost pressure Pb, and output torque T are measured simultaneously during the rapid opening of the throttle valve by the stepping motor. During the rapid opening of the throttle valve, air mass fiowrate Gat is increased immediately without delay time, but response of engine revolution N, and output torque T are delayed. Therefore hesitation, and stumble phenomena are occurred. Dead time $t_d$ and response delay time $t_r$ of engine revolution N, which is extremely delayed comparing to other performance parameters, are respectively 0.2-0.3sec., 3.0-4.6sec., and dead time rate $t_d/{\Delta}t$ and response delay time rate $t_r/{\Delta}t$ are linearly increased with the throttle valve opening rate ${\theta}$ during the acceleration from 12 degree to 20 degree at 1250rpm.

Backbone Assignment of the N-terminal Domain of Human Replication Protein A 70 kDa

  • Replication Protein A (RPA) is the eukaryotic single-stranded DNA binding protein. It involves in DNA replication, repair, and damage response. Among three subunits, RPA70 has a protein-protein binding domain (RPA70N) at the N-terminal. It has known that the domain recruits several damage response proteins to the damaged site. Also, it is suggested that there are more candidates that interact with RPA70N. Even though several studies performed on the structural aspects of RPA70N and its ligand binding, the backbone assignments of RPA70N is not available in public. In this study, we present the backbone assignments of RPA70N.

Using harmonic class loading for damage identification of plates by wavelet transformation approach

  • In this paper, the harmonic displacement response of a damaged square plate with all-over part-through damage parallel to one edge is utilized as the input signal function in wavelet analysis. The method requires the properties of the damaged plate, i.e., no information about the original undamaged structure is required. The location of damage is identified by sudden changes in the spatial variation of transformed response. The incurred damage causes a change in the stiffness or mass of the plate. This causes a localized singularity which can be identified by a wavelet analysis of the displacement response. In this study via numerical examples shown by using harmonic response is more versatile and effective compared with the static deflection response, specially in the presence of noise. In the light of the obtained results, suggestions for future work are presented and discussed.

Dynamic bending response of SWCNT reinforced composite plates subjected to hygro-thermo-mechanical loading

  • The dynamic bending response of single walled carbon nanotube reinforced composite (SWCNTRC) plates subjected to hygro-thermo-mechanical loading are investigated in this paper. The mechanical load is considered as wind pressure for dynamic bending responses of SWCNTRC plate. The dynamic version of the High Order shear deformation Theory (HSDT) for a composite plate with Matrix and SWCNTRC plate is first formulated. Distribution of fibers through the thickness of the SWCNTRC plate could be uniform or functionally graded (FG). The dynamic displacement response is predicted by using Nemarck integration method. The effective material properties of SWCNTRC are estimated by using micromechanics based modeling approach. The effect of different environmental condition, volume fraction of SWCNT, Width-to-thickness ratio, wind pressure, different SWCNTRC-FG plates, boundary condition, E1/E2 ratio, different temperature on dynamic displacement response is investigated. The dynamic displacement response is compared with the available literature and it shows good agreement.

A Humidity Sensor Using an Electrochemically Prepared Poly(1,5-Diaminonaphthalene)Film

  • An electrochemical humidity sensor was fabricated with poly(1,5-diaminonaphthalene) film coated on a gap of two splitted gold electrodes, which were made by vacuum deposition. Response currents according to humidity were measured by the potential sweep method and chronoamperometry. The stability of the polymer film was improved by double step chronoamperometry using the applied voltage of ${\pm}0.5$ Vdc. The response time determined by the pulse technique was about ${\sim}50$ msec and the relative standard deviation of current response was within ${\pm}5.0%$. The response current of the film was intrinsically humidity dependent. The film exhibited a non-linear but reproducible response in ordinary range of relative humidity. The linear equations were $I(nA)=0.28{\times}%RH-1.01$ between 10 to 70 %RH and $I(nA)=6.05{\times}%RH-403.21$ between 70 to 90 %RH.

  • The polymeric membrane electrodes based on N,N'-bis-pyridin-2-ylmethylene-naphthalene-1,8-diamine as an ion carrier were prepared and tested for the copper-ion selective electrode. The membrane has a linear dynamic range between $10^{-6}$ and $10^{-2}$ M with a Nernstian slope of 29.6 mV per decade, and its detection limit was $10^{-5.62}$M. The potentiometric response is independent of the pH range of 3-5. The proposed electrode showed good selectivity and response for $Cu^{2+}$ over a wide variety of other metal ions in pH 4.0 buffer solutions.
