Influence of the Composted Liquid Manure on the Turf Growth of Zoysiagrass (Zoysiagrass japonica) and Soil Properties

  • This study was conducted to investigate the effects of composted liquid manure (CLM) on soil properties and turf quality of zoysiagrass. The CLMs were produced at 5 facilities for livestock excretions treatments located in Cheongwon, Gunwi, Iksan, Nonsan, and Yeoju in Korea, respectively. Field experiments were conducted at 5 golf courses and a sod farm located near each facility for livestock excretion treatments. Chemical fertilizer (CF) with N at $12g\;m^{-2}year^{-1}$ and CLMs were applied four times, respectively. The constituents of the different CLMs were variable based on when and where the fertilizer was produced. Soil K content significantly increased when the soil was treated with CLMs. The soil treated with CF showed a higher content of total P than that treated with CLM. CF and CLMs treatments significantly increased the turf color index compared with control. Tiller density and shoot dry weight of fertilized plots were also higher than those of non-fertilized plots. However, there was no significant difference in turf color index and tiller density among plots treated by CLMs or CF. The results of this study demonstrated that CLMs could be a substitute for CF.

  • Mucolipidosis (ML) II/III are autosomal recessive diseases caused by deficiency of post-translational modification of lysosomal enzymes. The mannose-6-phosphate (M6P) residue in lysosomal enzymes synthesized by N-acetylglucosamine 1-phosphotransferase (GlcNAc-phosphotransferase) serves as recognition marker for trafficking in lysosomes. GlcNAc-phosphotransferase is encoded by GNPTAB and GNPTG. Mutations in GNPTAB cause severe ML II alpha/beta and the attenuated ML III alpha/beta. Whereas mutations in GNPTG cause the ML III gamma, the attenuated type of ML III variant. For the diagnostic approaches, increased urinary oligosaccharides excretion could be a screening test in clinically suspicious patients. To confirm the diagnosis, instead of measuring the activity of GlcNAc phosphotransferase, measuring the enzymatic activities of different lysosomal hydrolases are useful for diagnosis. The activities of several lysosomal hydrolases are decreased in fibroblasts but increased in serum of the patients. In addition, the sequence analysis of causative gene is warranted. Therefore, the confirmatory diagnosis requires a combination of clinical evaluation, biochemical and molecular genetic testing. ML II/III show complex disease manifestations with lysosomal storage as the prime cellular defect that initiates consequential organic dysfunctions. As there are no specific therapy for ML to date, understanding the molecular pathogenesis can contribute to develop new therapeutic approaches ultimately.

Near Infrared Spectroscopy for Measuring Purine Derivatives in Urine and Estimation of Microbial Protein Synthesis in the Rumen for Sheep

  • The efficiency of the luminal fermentation process influences overall efficiency of luminal production, animal health and reproduction. Ruminant production systems have a significant impact on the global environment, as well. Animal wastes contribute to pollution of the environment as ammonia volatilized to the air and nitrate leached to ground water. Microbial protein synthesis in the rumen satisfies a large proportion of the protein requirements of animals. Quantifying the microbial synthesis is possible by using markers for lumen bacteria and protozoa such as nucleic acids, purine bases, some specific amino acids, or by isotopic $^{15}N,^{32}P,\;and\;^{35}S$ labelled feeds. All those methods require cannulated animals, they are time-consuming and some methods are very expensive as well. Many attempts have been made to find an alternative method for indirect measurement of microbial synthesis in intact animals. The present investigations aimed to assess possibilities of NIRS for prediction of purine nitrogen excretion and ruminal microbial nitrogen synthesis by NIR spectra of urine. Urine samples were collected from 12 growing sheep,6 of them male, and 6- female. The sheep were included in feeding experiment. The ration consisted of sorghum silage and protein supplements -70:30 on dry matter basis. The protein supplements were chosen to differ in protein degradability. The urine samples were collected daily in a vessel containing $60m{\ell}$ 10% sulphuric acid to reduce pH below 3 and diluted with tap water to 4 liters. Samples were stored in plastic bottles and frozen at $-20^{\circ}C$ until chemical and NIRS analysis. The urine samples were analyzed for purine derivates - allantoin, uric acid, xantine and hypoxantine content. Microbial nitrogen synthesis in the lumen was calculated according to Chen and Gomes, 1995. Transmittance urine spectra with sample thickness 1mm were obtained by NIR System 6500 spectrophotometer in the spectral range 1100-2500nm. The calibration was performed using ISI software and PLS regression, respectively. The following statistical results of NIRS calibration for prediction of purine derivatives and microbial protein synthesis were obtained.(Table Omitted). The result of estimation of purine nitrogen excretion and microbial protein synthesis by NIR spectra of urine showed accuracy, adequate for rapid evaluation of microbial protein synthesis for a large number of animals and different diets. The results indicate that the advantages of the NIRS technology can be extended into animal physiological studies. The fast and low cost NIRS analyses could be used with no significant loss of accuracy when microbial protein synthesis in the lumen and the microbial protein flow in the duodenum are to be assessed by NIRS.

Dietary Effect of Liriopis Tuber Water Extracts on the Level of Blood Glucose and Serum Cholesterol in Streptozotocin-Induced Diabetic Rat (맥문동 물 추출물의 식이가 Streptozotocin으로 유도한 당뇨 흰쥐의 혈당과 혈청 콜레스테롤 함량에 미치는 영향)

  • The dietary effect of water extracts of Liriopis tuber(WELT) in the diabetic SD-rats on the level of blood sugar and serum cholesterol was investigated. The experimental plots divided into normal group(N), diabetic control(DC), 5% WELT-group(WELT-I) and 10% WELT-group (WELT-II). Each group was fed for 6 weeks, then continuously fed for 1 more week after streptozotocin injection. The loss of the body weight fed for one week after induction of the diabetes was 2.2~6.3% in the WELT-I and -II groups, but it was 18.6% in the DC-group. There was no significant difference in the feed intakes after diabetes induction between N-group and WELT-group, while it was significantly increased in DC-group. The feed efficiency ratios before diabetes induction were 1.70 in WELT-I group, 1.53~1.59 in the N, DC and WELT-II group, while the ratios after diabetes induction were 0.92 in DC-group, 1.51~1.83 in the N, WELT-I and -II group. While the amounts of water intakes for one week after diabetes induction was 625.4 mL in the DC-group, and it were 364.3~371.1 mL in the WELT-groups showing no significant difference with N-group. The excretion amounts of urine were 431.96 mL in DC-group for one week after diabetes induction, and it was 182~192.84 mL in WELT-groups. The ratios of liver weight against body weight were 2.74% in N-group, 2.93~2.96% in WELT-groups, but it was 4.01% in DC-group. The level of blood glucose in WELT-groups fed for one week after diabetes induction were 136.8~138.6 mg/dL showing no significant difference with N-group, but it was 357.8 mg/dL in DC-group. The level of serum triacylglycerol and serum total cholesterol were 93.8 and 68.7 mg/dL in N-group, 120.1 and 101.6 mg/dL in DC-group, 97.4~100.6 and 60.8~67.7 mg/dL in WELT-groups, respectively, showing no significant difference between N-group and WELT-groups. HDL-cholesterol/total cholesterol ratio were 0.63 in N-group, 0.57~0.67 in WELT -groups, which was significantly higher than that of DC-group(p<0.05). Atherogenic index were 0.58 in N-group, 0.49~0.74 in WELT-groups, but it was 1.32 in DC-group. The above results suggest that the WELT diets may have both preventive and curing effects against the diabetes.

Effects of by-product feed-based silage on feeding, rumination, and excretion in growing Hanwoo heifers

  • This study investigated the effects of feeding by-product feed (BF)-based silage on the behavior of growing Hanwoo heifers. Twelve Hanwoo heifers (13.2 months-old, 315 kg body weight; four heifers per pen) were assigned to three diets: a rice straw (RS) diet (concentrate mix and free access to RS), a RS and BF-based silage (RSBFS) diet (concentrate mix and free access to RS and BF-based silage), and a BF-based silage (BFS) diet (concentrate mix and free access to BF-based silage). Behavior was recorded for 5 days using camcorders. Compared to the RS group, the BFS group showed 21.7% higher dry matter intake, shorter feeding, rumination, and chewing times, as well as longer resting time (p < 0.05). Although all groups exhibited similar drinking, urination, and defecation frequencies, the BFS group exhibited higher feeding rates, rumination efficiency, and chewing efficiency than the RS group (p < 0.05). Compared to the BFS group, the RSBFS group showed higher $peNDF_{8.0}$ intake (15.2% vs. 25.0% dry matter intake), longer feeding and sitting times, lower defecation frequency (p < 0.05), and similar rumination efficiency. In conclusion, complete replacement of conventional RS with BF-based silage reduced rumination and chewing activity in growing Hanwoo heifers, and BF-based silage feeding with large-particle straw is an effective approach in improving heifer behavior.

Analysis of Urine Iodine Excretion Decrease by Two-Week Stringent Low Iodine Diet for Remnant Thyroid Ablation with Radioactive Iodine in Korean Patients with Thyroid Cancer; Prospective Study (한국 갑상선암 환자들에서 잔여갑상선 제거를 위한 방사성요오드 치료 전 2주간의 엄격한 저요오드식이에 의한 소변 내 요오드량 감소 분석; 전향적 연구)

  • A low iodine diet (LID) is the recommended preparation for radioactive iodine treatment. However, the recommended duration and stringency of LID are different among each recommendation. More stringent LID is expected in Korea because Korea is a iodine-rich region. We investigated the decrement of urine iodine excretion by two-week stringent LID for remnant thyroid ablation with radioactive iodine in Korean patients with thyroid cancer, prospectively. Material and Method: From November 2006, patients who referred to our hospital for remnant ablation after total thyroidectomy were included in this study. To decrease total body iodine, our protocol included three strategies. First, we checked medication which could inhibit the radioactive iodine uptake. Second, the date of 1-131 treatment was scheduled at least 3 months later if contrast agent had been used. The last strategy was two-week stringent LID education by specialized nutritionist. Before and after two-week stringent LID, 24hr-urine iodine was analyzed respectively. 24hr-urine creatinine was also analyzed for determining more valid 24hr urine sampling subgroup. Results: Total 51 patients were finally enrolled. Average of 24hr-urine iodine excretion was significantly lowered ($787{\pm}2242\;{\rightarrow}\;85{\pm}85\;{\mu}g/d$, p=0.03) after LID and 74.4% of patients reached below the recommended urine iodine excretion level ($<100\;{\mu}g/d$). In subgroup (n=14), similar results was showed ($505{\pm}666\;{\rightarrow}\;99{\pm}116\;{\mu}g/d$, p=0.05) and 78.6% of patients met the criteria. Conclusion: Most patients could reach below the recommended urine iodine level after two-week stringent LID. Therefore, in our opinion, at least two-week stringent LID should be recommended in Korea.

Mineral Metabolism in Singleton and Twin-pregnant Dairy Goats

  • During pregnancy, the maternal body undergoes significant physiological changes. The present study assessed the changes on calcium (Ca), phosphorus (P), magnesium (Mg), sodium (Na) and potassium (K) metabolism in singleton and twin-pregnant dairy goats. The 42 goats used ($49.5kg{\pm}7.6$ body weight [BW]) were assigned at random to treatments that were factorially arranged to account for 2 breeds (Oberhasli and Saanen), 2 pregnancy types (singleton and twin) and 3 gestation periods (80, 110, and 140 days). Digestibility trials were performed at 80, 110, and 140 days of gestation. Mineral retention during pregnancy was determined in the maternal body, femur, uterus, mammary gland, fetus and fetal fluid. Blood samples were taken during pregnancy before and after a meal, and Ca, P, Mg, Na, K ions and alkaline phosphatase activity determined in serum. Bone mineral density was determined in the right femur. Statistical analyses were performed using the SAS MIXED procedure. Dry matter intake decreased linearly up to 140 days of gestation. Maternal BW gain, and Ca, P, and Mg retention (g/kg) decreased linearly with the advance of gestation days. Macromineral retention in maternal body (g/kg) was greater in Oberhasli than Saanen goats, and their fetuses had higher Ca, P, and Mg deposition (mg/g). Mineral retention (mg/g) increased in fetuses according to pregnancy development, with no differences between singleton and twin pregnancy. In the mammary gland, the retention of all minerals (g) increased with the days of pregnancy. In conclusion, related to Ca, P, and Mg metabolism can be divided into two stages. Up to 80 days of gestation, was characterized by the preparation of the maternal body reserves for future mineral demands. From 80 days of gestation onward, was characterized by the transfer of maternal body reserves for fetal development and colostrum production. Na and K supply was provided by adjustments in endogenous excretion and an increase in intestinal absorption. Finally, mineral metabolism was specific to each genotype and, except for Na, was not affected by the number of fetuses.

Study on Composition and Fertilization of Feces from Laying Hens Fed Dietary Phytase (인분해효소를 급여한 산란계에서 배출되는 계분의 성상과 비료화에 관한 연구)

  • This study was conducted to produce a nutrient-balanced complex fertilizer using compost from laying hens fed with phytase levels. A total of 30 laying hens were randomly assigned in individual cages. The dietary treatments were fed from 51 to 60 wks of age, and included a phytase; 0, 1,000, and 2,000 FTU/kg in basal diets. Fresh feed (110 g) and drinking water were supplied to the laying hens every day. Feces from hens were collected daily and analyzed for N, P2O5, and K2O after compost maturity. This result showed there was no difference on the fecal excretion per feed intake of laying hens supplemented with phytase levels in the diets. On the other hand, the excretions of dried feces, N, P2O5, and K2O were significantly (P<0.05) reduced in 2,000 FTU/kg treatment than 0, 1,000 FTU/kg treatments. There was no difference on the compost composition and nutrient contents in compound fertilizer using compost from hens fed dietary phytase levels. However, compost content in the fertilizer was 69.6~71.6% when compost of 25~30% moisture content was used, and 13.0~47.1% at compost of 40~60% moisture content. As a result of this study, it was confirmed that hen's compost controlled moisture content could be produced as a nutrient-balanced compound fertilizer.

사료중 어유가 급성기 반응중인 육계 병아리에서 에너지 대사와 항산화효소 활성에 미치는 영향

  • Three experiment were designed to determine the effects of dietary(n-3) fatty acids on the growth-suppressive effects of the inflammatory response and anti-oxidant enzyme activity and balances of nitrogen and energy. Day-old chicks were fed diets containing 1.0 % soybean oil in experiment 1, or diets containing 1.0 % of fish oil in experiment 2, and then on 7-d of age chicks were fed diets containing 1.0 % of either soybean oil or fish oil. In experiment 3, day-old chicks were fed on diets containing 1.0 % of either soybean oil or fish oil. In chicks of half pens within each dietary treatments of each experiments, the acute phase response were induced by injecting with Salmonella typhymurium lipopolysaccaride(LPS) i.p. 3 times at 8, 10 and 12-d of age. During acute phase response, chicks fed dietary fish oil from 1-d of age enhanced energy balances and did not give negative effect on nitrogen balance and uric acid excretion, but elevated MnSOD and Cu/ZnSOD activity In liver and erythrocyte cytosols compared with those fed dietary soybean oil. The results indicated that enhanced daily gain and metabolizable energy value of diet in birds fed fish oil from one-d of age seems due to change on the antioxidants systems during acute phase response.

Emission Rate of Greenhouse Gases from Bedding Materials of Cowshed Floor: Lab-scale simulation study (우사깔짚에서 발생되는 온실가스 배출량 산정: 모의 실험결과)

  • To know the emission amount of greenhouse gases from bedding materials of cowshed floor, the emission rates of methane ($CH_4$) and nitrous oxide ($N_2O$) gases from a simulated cowshed floor (SCF) with sawdust that manure loading rate into the bedding material could be accurately controlled were assessed in this study. The manure loading rates of Korean beef and Holstein dairy cattle into the SCF of $0.258m^2$ surface area with 10 to 15 cm height sawdust were $1.586kg/m^2/d$ and $3.588kg/m^2/d$, respectively, and those were calculated on the basis of "Standard model for sustainable livestock" and "Data for excretion amount of manure from livestock". All experiments were done in triplicates in three different seasons (May to July, Sep. to Nov., and Feb. to Apr.) using 12 SCFs. The effects of bedding material thickness on $CH_4$ and $N_2O$ emission from SCFs for both Korean beef cattle and Holstein dairy cattle were not statistically significant (p<0.05). Emission amount of $CH_4$ and $N_2O$ per square meter of SCF for Holstein dairy cattle was 7.5 and 1.2 times higher than that of Korean beef cattle, respectively. The yearly $CH_4$ amount per head was 17.7 times higher in Holstein dairy cattle, obtaining 130.4 g/head/year from SCF for Holstein dairy cattle and 7.4 g/head/year from SCF for Korean beef cattle, and $N_2O$ was also 3.8 times higher in Holstein dairy cattle (3,267 g/head/year in Korean beef cattle and 14,719 g/head/year in Holstein dairy cattle). However, the $N_2O$-N per loaded nitrogen into SCF was higher in Korean beef cattle, having 0.2148 and 0.1632 kg $N_2O$-N/kg N in Korean beef cattle and Holstein dairy cattle, respectively, and those values were 3.07 and 2.33 times higher than that of Intergovernmental Panel on Climate Change (IPCC) 2006 guideline (GL) (0.07 kg $N_2O$-N/kg N).
