Genetic Diversity Estimation of the Rice Mutant Lines Induced by Sodium Azide

  • To investigate dose-effect of a chemical mutagen, sodium azide on a rice elite line, Suweon472, seed aliquots were treated with five different concentrations of sodium azide. The degree of mutation levels of each aizde concentration were estimated by using DNA fingerprinting techniques such as RAPD and AFLP. Some selected mutant lines ($M_4$) were also subjected for DNA fingerprinting to estimate their mutation levels by comparing the banding patterns of the wild type, Suweon 472. RAPD and AFLP fingerprinting patterns indicated that dose-effect of different azide concentrations was not clear. With allele description of detected AFLPs among favorable mutant lines, it was possible to discriminate each mutant line from others which have similar phenotypes and reactions against pathogens. AFLP fingerprinting patterns of waxy mutant lines, otherwise, were highly homogeneous as well as their phenotypic and agronomic characters.

  • Always, emotion is mutant. This is principle of literary treatment. In the literature, sadness is not sadness, and 'loving emotion' is not 'loving emotion.' Despite loving of our, loving is sadness. Also loving is to cry. This crying becomes love. This study is to show the mutant emotion which is to be able to code Deep Learning AI. We explored the Sijo "Streams that cried last night", because this Sijo was useful to study mutant emotion. The result was that this Sijo was coding the mutant emotion. Almost continuously, the sadness codes were spawning and concentrating. So this Sijo was making the emotion of love with the sadness. If this study is continued, It is believed that our lives will be much happier. And the method of literary therapy will be able to more upgrade.

Development of Pyropia yezoensis Mutant with Improved Amino Acid Content Using Gamma Rays (방사선 돌연변이 육종기술을 통한 고 아미노산 함유 김(Pyropia yezoensis) 돌연변이 개발)

  • Gamma irradiation is one of the simple methods used to induce mutagenesis. Therefore, it is widely used for the development of breeding lineages of plants and algae. In this study, it was developed a new variety of Pyropia yezoensis using gamma irradiation. It was applied a dose of 1 kGy and named the developed mutant Py1k. The blade with width of the mutant was narrower and the blade length was longer than those of the wild type. To further investigate the mutant, it was analyzed the nutrient composition and antioxidant activity. In comparison to those in the wild type, it was found a higher amino acid composition and marginally increased antioxidant activity in Py1k. Based on these results, it was suggests that our protocol can be utilized to develop Pyropia species with improved nutritional quality through gamma irradiation.

Selection and Characterization of Pseudomonas aeruginosa EMS1 Mutant strain Showing Enhanced Biosurfactant Production

  • A new bacterial strain, was isolated from activated sludge, identified and named P. aeruginosa EMS1. The new strain produced surface-active rhamnolipids by batch cultivation in mineral salts medium with waste flying oils. The mutant strain KH7, designated P. aeruginosa EMS1, derived by random mutagenesis with N-methyl-N-nitro-N-nitrosogoanidine treatment producing high levels of the biosurfactants was selected by an ion-pair plate assay. The mutant strain KH7 showed 4-5 times more hydrocarbon emulsification as compared to the parent when grown on waste frying oils and various hydrocarbons. Furthermore, P. aeruginosa EMS1 and mutant strain KH7 was also able to use whey as a co-substrate for growth and biosurfactant production. As results of this study, mutant strain KH7 is a very efficient biosurfactant producer, and its culture conditions are relatively inexpensive and economical. Rhamnolipid is synthesized by the rhlAB-encoded rhamnosyltransferase. To be convinced of these genes, we performed PCR based on P. aeruginosa PAO1 whole-genome database. rhl gene cluster nucleotide and amino acid sequences were compared for both parent and mutant. Comparison of nucleotide sequence of rhlAB, there were usually terminal's codons exchange.

  • The isolate SPO 3, an oligosporogenous and crystalliferous mutant, which was isolated from Bacillus thuringiensig subsp, israelensis by heat treatment at 42$^{\circ}C$ for 24 hit. The $\delta$-endotoxin of the mutant had a relatively low hemolytic activity on human red blood cells; the $\delta$-endotoxin of the mutant had 25 times less hemolytic activity compared to that of wild strain. The loss of 28 KDa hemolytic protein subunit in $\delta$-endotoxin of the mutant was confirmed by means of double immunodiffusiori, immunoelectrophoresis, and SDS-PAGE.

  • Pleurotus ostreatus 'Miso' is a mutant strain showing white color in pileus from the known parent strain 'Wonhyeong 1'. Shape and several other characters also vary with culture conditions. Mating experiments were performed to understand interstrain mating relationship using monokaryons of the parent and the mutant strains. All monokaryons were grown from single spores isolated from freshly collected fruit bodies. Pairings were performed in 90 mm petri dishes on PDA. They were allowed to grow at 25 until two fronts of the advancing mycelia met and developed a conspicuous contact zone. The contact zone and the outer edges of paired colonies on each plate were examined for clamp connections. The parent and the mutant resulted in tetrapolar incompatibility in intrastrain crosses. In interstrain crosses, each monokaryotic tester strain of the parent strain was out-crossed to monokaryotic tester strains of the mutant. As a result of these crosses it was found that both strains share the same A and B incompatibility factors yielding 25% compatibility.

Generation of an Arginine Auxotrophic Mutant of Colletotrichum acutatum as a Recipient Host for Insertional Mutagenesis

  • Colletotrichum acutatum was the main cause of the recent outbreaks of anthracnose on pepper fruit in Korea. To facilitate molecular analysis of C. acutatum, we generated an arginine auxotrophic mutant of the C acutatum strain JC24 using a targeted gene replacement strategy. A 3.3-kb genomic region carrying an ortholog (designated CaARG2) of the fungal gene encoding N-acetylglutamate synthase, the first enzyme of arginine biosynthesis in fungi, was deleted from the fungal genome. The mutant exhibited normal growth only when arginine was exogenously supplied into the culture medium. Transformation of the arginine auxotrophic mutant with a plasmid DNA carrying an intact copy of CaARG2, which was smaller than the deleted region in the mutant, not only caused random vector insertions in the fungal genome, but also recovered both hyphal growth and pathogenicity of the mutant to the wild-type level. Using this new selection system, we have successfully developed a restriction enzyme-mediated integration procedure, which would provide an economically efficient random mutagenesis method in C. acutatum.

  • Tridecanedioic acid (DC-13), starting material of the valuable musk ethylene brassylate, was obtained from n-tridecane by the Candida tropicalis mutant. The mutants were first obtained from primary screening step using the selective medium and then solid phase extraction sampling method was used for the selective isolation of organic acids from the cultured media of mutants. The resulting acids were directly converted to volatile tert-butyldimethyl silyl delivatives, which were then analyzed by gas chromatography. The efficient GC profiling method was used for the rapid identification of the mutant producing DC-13 in large quantity, and for the optimization of the culture conditions of mutant. The optimal culture conditions were found as follows: pH 8.0, 30$^{\circ}C$, 250rpm, 48hour of culture and $(NH_4)_2HPO_4$ as nitrogen source.

유사 생합성 경로를 가진 Streptomyces sp.의 혼합배양을 이용한 Doxorubicin 생합성

  • We selected two mutants namely strain D5 and Nu23 by mutagenesis of anthracycline producing Streptomyces: the former is an $\varepsilon$-rhodomycinone overproducing mutant selected from Streptomyces sp. C5, a baumycin producer and the latter, a blocked mutant of early pathway for doxorubicin biosynthesis obtained from Streptomyces peucetius ATCC 27952, a doxorubicin producer. The mutant strain Nu23 does not produce anthracycline metabolites but retains the most of enzyme activities converting aklavinone to doxorubicin and the mutant strain D5 produced $\varepsilon$-rhodomycinone at a level of 150 $\mu$g/ml. These strains were grown separately in NDYE medium and each was mixed at day 3 by equal volume of culture broth but the quantity of doxorubicin produced was far below an estimation based on the level of $\varepsilon$-rhodomycinone normaly produced by the strain D5. On the other hand doxorubicin was reached at maximum level after 4 days in the mixed culture condition which was composed of culture broth of strain D5 grown for 6 day and that of strain Nu23 grown for 3 day. It was turned out that the growth of mutant strain D5 was inhibited by the accumulation of daunorubicin and doxorubicin in mixed culture broth, which cause the limitation of $\varepsilon$-rhodomycinone.

A Genome-wide Approach for Functional Analysis Using Rice Mutant

  • Rapid extension of genomic database leads to the remarkable advance of functional genomics. This study proposes a novel methodology of functional analysis using 5-methyltrytophan (5 MT) mutant together with their 2-DE analysis and public microarray database. A total of 24 proteins was changed in 5 MT mutant and four remarkably different expressed proteins were identified. Among them, three spots were converted to Affymetrix probe. A total of 155 microarray samples from Gene Expression Omnibus (GEO) in NCBI was retrieved and followed by constructing gene co-expression networks over a broad range of biological issues through Self-Organising Tree Algorithm. Three co-expressing gene clusters were retrieved and each functional categorization with differential expression pattern was exhibited from 5 MT resistance mutant rice. It was indicated new co-expression networks in the mutant. This study suggests that on investigating possibility which correspond 2-DE to microarray database with their full potential.
