연쇄상구균(Streptococcus mutans GS-5)의 항원단백질 AgI/II의 N-terminus절편에 대한 항체형성 (Generation of antibodies against N-terminus fragment of AgI/II protein from Streptococcus mutans GS-5)

  • 치아 우식은 구강 내 미생물에 의해 치아 석회 조직의 일부가 용해되고 파괴되는 감염성 질환이다. 치아 우식의 원인균은 Streptococcus mutans와 같은 Mutans streptococci로 알려져 있고, 이 미생물이 치면에 접착하여 군집을 형성하는 능력이 균의 독성에 중요한 역할을 한다. S. mutans가 치면의 타액성 피막에 부착하는 데에는 AgI/II와 같은 세포표면의 섬유성 단백질을 매개로 한다. 그러므로, AgI/II는 S. mutans GS-5에 대한 백신 개발에 적절한 목표가 된다. 본 실험은 S. mutans GS-5로부터 AgI/II 유전자를 복제하고 염기서열분석을 하였다. 복제된 AgI/II와 앞서 보고된 S. mutans GS-5의 해당 부위의 280개의 핵산은 완벽하게 일치하였다. 복제된 유전자를 두 부위로 절단하여 형질전환을 통해 재조합 단백질인 AgI/IImr을 얻었고, 정제된 재조합 단백질을 쥐에게 주입하여 다클론 항체를 얻었다. 추출된 다클론 항체는 AgI/IImr항원단백질에 반응하였고, 대조군으로 쓰인 단백질에는 반응하지 않았다.

전문가구강관리가 요양병원 장기입원환자들의 구강 내 Streptococcus mutans 수에 미치는 영향 (Effects of Professional Oral Care for Long-term Patients in Nursing Facilities on the Streptococcus mutans Population in the Intraoral Region)

  • 본 연구는 전문가구강관리가 요양병원 장기입원환자들의 구강내 Streptococcus mutans 수에 미치는 영향을 파악하기 위하여 D광역시에 소재한 일개 노인 요양병원 장기입원환자를 대상으로 2011년 7월 24일 부터 10월 25일 까지 3개월간 연구대상자 43명을 대상으로 주 2일, 1일 3회 치과위생사가 전문가구강관리를 제공하고 1개월 간격으로 3회에 걸쳐 타액분비량과 타액 내 mutans streptococci 집락 수를 세어 구강건강상태를 비교하였다. 연구결과 타액분비량은 관리 전 5.8 ml에서 1개월 후 6.4 ml, 2개월 후 7.5 ml로 증가하였고, 세균 수는 약 $6{\times}10^8CFU/ml$에서 2개월 후 약 $3{\times}105CFU/ml$로 20배 이상 감소하였다. 전신질환에 따른 구강관리 전후의 세균 수의 변화는 고혈압과 당뇨병이 있는 환자의 세균 수가 유의하게 감소하였다. 충전치아의 수와 고정성보철물의 수가 많을수록 세균 수는 유의하게 감소하였다. 전문가구강관리는 요양병원 장기입원 환자들의 구강내 Streptococcus mutans 수와 밀접한 관련이 있는 것으로 나타났다. 따라서 향후 요양병원 장기입원환자의 특성을 고려한 실질적이고 지속적인 구강관리가 이루어지도록 구강보건프로그램을 개발하는데 필요한 다양한 연구가 실시되어야 한다.

Inhibitory Effects of Resina Pini on the Growth and Glucosyltransferase activity of Streptococcus mutans

  • The purpose of this study is to evaluate the inhibitory effects of Resina Pini against Streptococcus mutans (S. mutans) that is one of the major causes of dental caries and oral diseases. Topically applied Resina Pini (RP) would be incorporated in saliva and thus the factor associated with water solubility should be considered. In this paper, therefore, effects of various treatment for RP and activities of water extracts from unprocessed and processed RP were compared. The crude RP (RP1) and the recrystallized RP (RP2) in ethanol solution showed strong antimicrobial activities (d.>15mm) against S. mutans. All RP samples exhibited considerable inhibitory effect against glucosyltransferase produced by S. mutans $(IC_{50}=91.2\;to\;276.2\;{\mu}g/ml)$. The very considerable increase in cellular permeability of S. mutans was observed with RP1, RP2 and their water extracts. These results suggest that RP1 and RP2 may be a potential source for pharmaceutical products used for prevention and/or treatment of dental caries and periodontal disease.

Constituents Released from Streptococcus mutans Attenuate Arecoline-mediated Cytotoxicity in HGF Cells by Altering Intracellular $Ca^{2+}$ Signaling

  • Streptococcus mutans (S. mutans) is a facultative anaerobic bacterium mainly found in the oral cavity and is known to contribute to tooth decay and gingivitis. Recent studies on intestinal microbiota have revealed that microorganisms forming a biofilm play important roles in maintaining tissue homeostasis through their own metabolism. However, the physiological roles of oral microorganisms such as S. mutans are still unclear. In our current study, we identified that constituents released from S. mutans (CR) reduce arecoline-mediated cytotoxicity without producing toxic effects themselves. Arecoline, as a major alkaloid of areca nut, is known to mediate cytotoxicity on oral epithelial cells and induces a sustained intracellular $Ca^{2+}$ ($[Ca^{2+}]_i$) increase that is cytotoxic. The exposure of human gingival fibroblast (HGF) cells to CR not only inhibited the sustained $[Ca^{2+}]_i$ increase but also the initial $[Ca^{2+}]_i$ elevation. In contrast, CR had no effects on the gene regulation mediated by arecoline. These results demonstrate that S. mutans has physiological role in reducing cytotoxicity in HGF cells and may be considered a novel pharmaceutical candidate.

Antimicrobial Effect of Polyphenon 60 against Streptococcus mutans and Streptococcus sobrinus

  • Polyphenon 60 refers to the mixture of catechins present in green tea. The aim of this study was to investigate the antimicrobial activities of polyphenon 60 against 4 strains of Streptococcus mutans and 2 strains of Streptococcus sorbrinus, which are the major causative bacteria of dental caries. The minimum bactericidal concentration (MBC) values of polyphenon 60 for S. mutans and S. sobrinus were determined and the effect of biofilm formation inhibition of that was evaluated. The MBC value of polyphenon 60 against the bacterial strains was 2.5 mg/ml except for one particular strain, S. mutans KCOM 1128 for which the value was 1.25 mg/ml. The results of biofilm formation inhibition assay revealed that polyphenon 60 inhibited biofilm formation more than 90% at a concentration of 2.5 mg/ml. It was apparent that polyphenon exhibited biofilm formation inhibition activity along with bactericidal effect against S. mutans and S. sobrinus. Therefore, it is proposed that polyphenon 60 as one of the components of bactericidal agents could be useful in developing oral hygiene products, toothpaste or gargling solution.

Acrylic resin 표면의 거칠기에 따른 세균부착 비교 (EFFECT OF SURFACE ROUGHNESS OF ACRYLIC RESIN ON THE ADHESION OF BACTERIA)

  • Statement of problem: The microbial adhesion on the surface of materials used in prosthodontics and restorative dentistry significantly influences microbial infection. Purpose: The purpose of this study was to evaluate the effect of how the degree of surface roughness of acrlyic resin affect the adhesion of bacteria. Material and methods: Resins were finished with $50{\mu}m$ and $250{\mu}m$ aluminium oxide particles by using sandblaster, by using stone point, and high polished with $Opa^{(R)}$ and Lace $motor^{(R)}$. The surface of acrylic resin attached by bacteria was directly touched on the surface of BHI agar, which was incubated. Bacteria colonies formed on BHI agar were counted in accordance with the degree of the surface roughness. Results: 1. The viable cell number of Streptococcus mutans increased on the acrylic resins incubated in BHI broth than in PBS. 2. The viable cell number of Streptococcus mutans increased on the acrylic resins incubated without agitation than with agitation, washed three times than six times, and incubated in broth added with 5% sucrose than without sucrose. 3. When Streptococcus mutans incubated in BHI broth, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins finished with $250{\mu}m$ aluminium oxide particle using sandblaster. But when incubated in BHI broth containing sucrose, the number of colonies formed on that was the largest on the acrylic resins high polished using $Opal^{(R)}$ and Lace $motor^{(R)}$. 4. When Streptococcus sanguis was incubated in BHI broth with or without sucrose, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins finished with $250{\mu}m$ aluminium oxide particle using sandblaster. 5. When Actinomyces viscosus was incubated in BHI broth with or without sucrose, the number of Streptococcus mutans colonies formed on BHI agar was the largest on the acrylic resins high polished using $Opal^{(R)}$ and Lace $motor^{(R)}$. Conclusion: These results indicated that when acrylic resins attached by bacteria were touched on the surface of BHI agar, the number of bacterial colonies formed on the agar was dependent on the bacterial species. Also, the result of this study was showed that increase in the surface roughness and the addition of sucrose increased retention of microbial cells.

고삼의 Streptococcus mutans에 대한 항세균 효과 (Antibacterial Effects of Sophora flavescens on Streptococcus mutans)

  • 고삼에서 치아우식원인세균인 S. mutans에 대한 항세균효과와 GTase의 활성을 저해하는 물질을 탐색, 개발하여 식품 또는 치아우식예방제로 응용하기 위하여 연구를 수행하였다. 고삼 에틸 아세테이트추출물의 3차 분획의 소분획에서 S. mutans에 대한 최소억제농도가 3.13 $\mu$g/ml으로 나타나, 높은 항세균효과를 관찰할 수 있었다. GTase 활성은 고삼의 에틸 아세테이트 추출물에 의하여 glucan의 형성을 저해하였으며, 100 $\mu$g/ml의 농도에서 77%의 저해율이 나타났고, 통계학적으로 유의한 효과가 나타났다 (p<0.05), 이상의 결과로 고삼은 치아우식예방제로 개발 가능성이 있다고 기대된다.

  • 최근 대체 감미료로서 당알코올과 합성감미제에 관한 연구가 활발히 진행되고 있다. 에리스리톨은 포도당을 원료로 효모에 의해 생산되는 포도당 발효감미료로서 과실류, 버섯, 포도주, 청주, 간장등의 발효식품에 함유되어 있는 천연당질로 $C_4H_{10}O_4$의 분자구조를 가지는 4탄당의 당알코올이다. 구강내에서 산생성균인 Streptococcus mutans의 증식과 산생성, 치면세균막 형성 및 치아우식증 등은 여러 종류의 영양물질이 혼합되어 있는 구강내에서 일어나는 과정이므로 본 연구는 에리스리톨과 다른 감미제에서 S. mutans의 성장력과 산생성력을 분석하여 에리스리톨이 치아우식증의 예방에 관여하는 중요한 기전을 밝혀내어 치아우식예방을 위한 제품을 생산하거나 활용시 올바른 정보를 제공하고 개선방안을 마련하기 위한 기초자료를 제공하고자 시행하였다. 연구결과 에리스리톨은 우식활성균의 성장을 억제시키고 산생성능을 감소시켜 치아우식을 유발하지 않으며 우식예방효과가 있는 자당 대체 감미제로 우수한 것으로 확인이 되었다.

타액, 모유, 분유에서의 Streptococcus mutans 활성도 차이 (Effect of Streptococcus mutans colinization activing in saliva, breast milk, formula milk)

  • Objectives : This study was performed to provide the basic data for oral health of infants, through the comparison of Streptococcus mutans activities(the representative bacteria of initial dental caries), in main nutritional foods for infants: breast milk, milk powder, and soya milk. Methods : The study targeted 30 infants and the saliva was sampled after the oral examination. The pH of sampled saliva was measured; and the sample of 'the saliva', 'the saliva and breast milk', and 'the saliva and formula milk' by researcher were produced; and the measurement was repeated 7 times. Results : As a result of examining the change of colony number of Streptococcus mutans by hour, the bacterial activity took in the source of nutrition took place in the first place; and in the group with dental caries, breast-feeding at night occurreds most frequently. Due to the combined activities, such as the increase in the contact time, the existence rate of dental caries appeared high. However, as the improvement of breast-feeding is closely related to the nutrition supply to babies, rather than the improvement of breast-feeding, the 12 hours after the culture in which Streptococcus mutans activities started was considered. Conclusions : In case of breast-feeding at night, it is necessary to remove the dental plaque in the mouth, before 12 hours has passed. Especially, using toothbrush and toothpaste to decrease the occurrence of dental caries, after the eruption of teeth, is recommended for oral administration.

세륨옥사이드나노입자(Cerium oxide nano particles: CNPs)를 함유한 치면열구전색재의 Streptococcus mutans 부착량 변화 (Changes in the amount of adhesion of Streptococcus mutans to pit and fissure sealant incorporating cerium oxide nano particles(CNPs))

  • Objectives: The aim of this study was to investigated the surface roughness and change in the amount of adhesion of Streptococcus mutans to the commercial pit and fissure sealant containing cerium oxide nano particles(CNPs). Methods: The CNPs was incorporated into a commercial pit and fissure sealant at 0-4.0 wt%. Disk Specimens (ϕ 10 mm × 2 mm) were prepared by light polymerization the front and back for 40s. Average surface roughness was measured and Streptococcus mutans adhesion was observed under confocal laser scanning microscopy (CLSM) after 24 hour. Data were statistically analyzed by one-way ANOVA and Tukey HSDa post-hoc test. Results: Difference of the surface roughness(Ra) between groups was not statistically significant in both non CNPs group and CNPs group(p>0.05). In CNPs group, the amount of S. mutans adhesion was significantly different between control group and decreased in order of CNPs 4.0, CNPs 0.5, CNPs 1.0 and CNPs 2.0(p<0.05). Conclusions: Within the limitation of this study, these aspects of oral bacteria performances suggest potential usefulness of the CNPs incorporation, especially CNPs 1% and 2%, in pit and fissure sealant for inducing antibacterial effect.
