아위버섯균의 생리적 특성 (The physiological characteristics of Pleurotus ferulae Lanzi)

  • 본 연구의 결과는 다음과 같다. 국내외에서 수집된 아위버섯 4균주(F-1, F-2, F-3, F-4)를 공시하여 우량 균주를 선발한 결과 균사생장이 빠르고 균사밀도가 좋은 F-2 균주를 선발하였다. 선발된 아위버섯균은 MYPA배지에서 균사생장이 가장 좋았으며, 균사생장 최적 온도는 $25{\sim}30^{\circ}C$이며 $30^{\circ}C$에서 가장 빨리 자랐다. 그리고 균사배양 최적 산도는 pH 5.0~6.0 이었다. 아위버섯균의 균사생장을 위한 최적배지조성은 탄소원에서는 다당류인 soluble starch이고 그 다음으로는 maltose였으며, 최적 탄소원인 soluble starch의 적정농도는 1%(w/w)였다. 질소원에서는 yeast extract 0.25%(w/w), malt extract 0.25%(w/w)를 혼합하였을 때 가장 빠른 생장속도를 보여주었으며, malt extract 만을 첨가하였을 때는 균사생장이 극히 부진하였다. 아위버섯균의 현미경 관찰 결과 일부 세포벽의 박벽화와 침식 현상이 관찰되었지만, 이 같은 현상이 광범위하게 관찰되지 않았다는 점에서 아위버섯균에 의한 리기다소나무재의 분해는 매우 미미한 것으로 사료된다. 아위버섯균에 의해 분해된 리기다소나무재의 편광현미경 관찰 결과 일부 세포의 S3에서 복굴절성이 감소하는 것으로 나타나 아위버섯균이 부분적으로 셀룰로스를 분해하고 있음이 관찰되었다. 그러나 이 같은 복굴절성이 소멸되는 가도관은 많지 않아 아위버섯균에 의한 리기다소나무재의 셀룰로스의 분해 역시 크지 않았음을 보여주고 있다. 아위버섯균의 졸참나무재의 분해결과 리기다소나무재에 비하여 방사조직과 축방향유세포에서 분해가 상당히 이루어졌고, 목섬유에서도 부분적으로 침식과 박벽화가 상당히 진행되었음을 알 수 있었다. Bavendam Test결과 아위버섯균은 탄닌산을 분해하여 갈색의 대사물질을 형성하여 전형적인 백색부후균의 부후형태를 나타내었다.

Characteristics and suitability of various cereal grains in spawn production of button mushroom

  • Spawn is the vegetative growth of the mycelium and serves as the inoculum or seed for cultivating mushrooms. Various cereal grains, such as wheat, millet, barley, sorghum, brown rice, rye, and oat were assessed to compare their characteristics and suitability for spawn production of button mushroom (Agaricus bisporus). The mycelial growth rates, density, and the number of completely colonized grains were measured from the twentieth day of inoculation. Wheat grains showed fastest mycelial growth with 8.4 cm followed by rye, oat, barley with 8.2, 7.5 and 7.3 cm, respectively. In the mycelial density, foxtail millet, barley, and sorghum were best compared with that of wheat grains. Especially, the number of grains which were completely colonized by mycelia were greatest in foxtail millet with 5,123 grains followed by proso millet, and wheat with 3,052 and 914, respectively. Based on the results obtained, barley, foxtail millet, and sorghum grains would be appropriate substituting for wheat grain in spawn production of button mushroom.

Purification, Characterization, and Cloning of Fibrinolytic Metalloprotease from Pleurotus ostreatus Mycelia

  • A fibrinolytic protease (PoFE) was purified from the cultured mycelia of the edible oyster mushroom Pleurotus ostreatus, using a combination of various chromatographies. The purification protocol resulted in an 876-fold purification of the enzyme, with a final yield of 6.5%. The apparent molecular mass of the purified enzyme was estimated to be 32 kDa by SDS-PAGE, fibrin-zymography, and size exclusion using FPLC. The optimal reaction pH value and temperature were pH 6.5 and $35^{\circ}C$, respectively. PoFE effectively hydrolyzed fibrinogen, preferentially digesting the $A{\alpha}$-chain and the $B{\beta}$-chain over the ${\gamma}$-chain. Enzyme activity was enhanced by the addition of $Ca^{2+},\;Zn^{2+},\;and\;Mg^{2+}$ ions. Furthermore, PoFE activity was potently inhibited by EDTA, and it was found to exhibit a higher specificity for the chromogenic substrate S-2586 for chymotrypsin, indicating that the enzyme is a chymotrypsin-like metalloprotease. The first 19 amino acid residues of the N-terminal sequence were ALRKGGAAALNIYSVGFTS, which is extremely similar to the metalloprotease purified from the fruiting body of P. ostreatus. In addition, we cloned the PoFE protein, encoding gene, and its nucleotide sequence was determined. The cDNA of cloned PoFE is 867 nucleotides long and consists of an open reading frame encoding 288 amino acid residues. Its cDNA showed a high degree of homology with PoMEP from P. ostreatus fruiting body. The mycelia of P. ostreatus may thus represent a potential source of new therapeutic agents to treat thrombosis.

송이 감염묘 육성을 위해 야외 조건에서도 송이균 생존이 가능한 소나무의 최소 크기 결정 (Determination of the Minimum Size of Seedlings with Matsutake Mycelia That Can Survive in the Field for Matsutake-infected Pine Tree Production)

  • 송이는 세계적으로 상용 가치가 높은 버섯이나, 아직까지 자연 채취에 의존하고 있다. 현재까지 송이 감염묘를 통해 송이를 한 개씩 발생시키는 것까지는 한국과 일본에서 성공하였다. 본 연구에서는 송이 감염묘법에서 송이균이 생존할 수 있는 소나무의 최소 크기를 결정하고자 하였다. 송이 감염묘에서 송이균이 생존한 것은 이식 2년째에 14본(50%)과 이식 1년째에 10본(71%)이었다. 이식 후 송이균이 생존한 송이 감염묘는 이식 당시 평균 수고가 25 cm (최소 12 cm ~ 최대 40 cm) 였다. 결론적으로 송이 감염묘의 수고가 최소 12 cm 이상이 되면, 야외 조건에서 송이균이 생존할 수 있을 것으로 판단되었다. 이 결과는 기존의 송이 감염묘에서 사용하는 기주식물의 크기를 획기적으로 줄여 송이 감염묘 방법을 개선하였고, 송이 인공재배를 위해 어느 정도 크기의 소나무를 활용하는 것이 좋을 지의 기준을 제시하였다.

버섯균사체 배양물로부터 면역증진 기능성 소재 개발 (Immuno enhancing and chemopreventing agent from mushroom mycelial culture)

  • This study relates to low and medium molecular weight isoflavone-${\beta}$-D-glucan produced by submerged liquid culture of Agaricus blazei, a method of producing the isoflavone-B-D-glucan using autolysis enzyme of Agaricus blazei mycelia, and use of the isoflavone-B-D-glucan for anti-cancer and immunoenhancing effect. In acordance with one aspect of the present study, it deals with a method of producing isoflavone-${\beta}$-D-glucan, which comprises the followings; 1) culturing and separating mushroom mycelia, 2) producing low-medium molecular weight isoflavone-${\beta}$-D-glucan from high molecular weight one. The cytotoxicity on human cnacer cell line (Caco-2, MCF-7), the expression of Cyclin D, Bcl-2, Bax protein, p21 protein, p53 protein in MCF-7 cells assessed by SDS-PAGE and immunoblotting, and other immuno related factors such as TNF-${\alpha}$ and IL-1B activities were examined. Structural identification of isoflavone-${\beta}$-D-glucan which showed cytotoxicity against cancer cell and immunoenhancing effects was carried by separation with DEAE-cellulose column chromatography, TLC, HPLC, IR, NMR. Clinical test for the cancer patients (n=119) for 6 month was carried out, and immunoenhancing factors (NK. cell number, ratio of T4/T8) were checked. We concluded the identified isoflavone-${\beta}$-D-glucan has immuno enhancing effects and could be useful for cancer chemoprevention.

Occurrence of dsRNA Mycovirus (LeV-FMRI0339) in the Edible Mushroom Lentinula edodes and Meiotic Stability of LeV-FMRI0339 among Monokaryotic Progeny

  • dsRNA was found in malformed cultures of Lentinula edodes strain FMRI0339, one of the three most popular sawdust cultivated commercial strains of shiitake, and was also found in healthy-looking fruiting bodies and actively growing mycelia. Cloning of the partial genome of the dsRNA revealed the presence of the RdRp sequence of a novel L. edodes mycovirus (LeV), and sequence comparison of the cloned amplicon showed identical sequences sequence to known RNA-dependent RNA polymerase genes of LeV found in strain HKA. The meiotic stability of dsRNA was examined by measuring the ratio of the presence of dsRNA among sexual monokaryotic progeny. More than 40% of the monokaryotic progeny still contained the dsRNA, indicating the persistence of dsRNA during sexual reproduction. Comparing the mycelia growth of monokaryotic progeny suggested that there appeared to be a tendency toward a lower frequency of virus incidence in actively growing progeny.

Mycelial Yield of Pleurotus ostreatus Using Thinned Apple, Pear, and Peach on Submerged Culture

  • The effect of thinned fruits, apple, pear and peach, on the mycelial growth of mushrooms was investigated. The growth of mycelia with the addition of thinned fruit was clearly better than that in the control for all the tested mushrooms. The growth rate of Pleurotus ostreatus was faster than any other mushroom. The optimal concentrations of thinned apple, pear, and peach in a solid culture were 1.0%, 1.0%, and 3%, respectively, while in a liquid culture the optimal concentrations were 5,0%, 3.0%, and 5.0%, respectively. When Pleurotus ostreatus was incubated in a 20-L pilot scale fermenter with 10 L of a liquid medium containing 3% thinned fruit at 25$^{\circ}C$ and 6 vvm for 10 days, the mass-production of mycelia was 74.2 g/10 L (apple), 96.2 g/10 L (pear), and 86.3 g/10 L (peach). The mycelial yield of Pleurotus ostreatus in a medium containing thinned fruit was 2 ∼ 3 times higher than that in the control.

Antimelanogenesis Effects of Fungal Exopolysaccharides Prepared from Submerged Culture of Fomitopsis castanea Mycelia

  • Fungal exopolysaccharides are important natural products having diverse biological functions. In this study, exopolysaccharides from Fomitopsis castanea mycelia (FEPS) were prepared, and the highest mushroom tyrosinase inhibitory activity was found. FEPS were prepared from cultivation broth by ethanol precipitation method. The extraction yield and protein concentration of FEPS were 213.1 mg/l and 0.03%, respectively. FEPS inhibited mushroom tyrosinase with the half maximal inhibitory concentration ($IC_{50}$) of 16.5 mg/ml and dose-dependently inhibited cellular tyrosinase activity (63.9% at $50{\mu}g/ml$, and 83.3% at $100{\mu}g/ml$) in the cell-free extract of SK-MEL-5 human melanoma cell and ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$)-stimulated melanin formation in intact SK-MEL-5 human melanoma cell. The $IC_{50}$ of FEPS against NO production from RAW264.7 macrophage cells was $42.8{\pm}0.64{\mu}g/ml$. By in vivo study using a zebrafish model, exposure of FEPS at $400{\mu}g/ml$ to dechorionated zebrafish embryos for 18 h decreased the pigment density, compared to that without FEPS-treated control.

A Comprehensive Review of Tropical Milky White Mushroom (Calocybe indica P&C)

  • A compressive description of tropical milky white mushroom (Calocybe indica P&C var. APK2) is provided in this review. This mushroom variety was first identified in the eastern Indian state of West Bengal and can be cultivated on a wide variety of substrates, at a high temperature range ($30{\sim}38^{\circ}C$). However, no commercial cultivation was made until 1998. Krishnamoorthy 1997 rediscovered the fungus from Tamil Nadu, India and standardized the commercial production techniques for the first time in the world. This edible mushroom has a long shelf life (5~7 days) compared to other commercially available counterparts. A comprehensive and critical review on physiological and nutritional requirements viz., pH, temperature, carbon to nitrogen ratio, best carbon source, best nitrogen source, growth period, growth promoters for mycelia biomass production; substrate preparation; spawn inoculation; different supplementation and casing requirements to increase the yield of mushrooms has been outlined. Innovative and inexpensive methods developed to commercially cultivate milky white mushrooms on different lignocellulosic biomass is also described in this review. The composition profiles of milky white mushroom, its mineral contents and non-enzymatic antioxidants are provided in comparison with button mushroom (Agaricus bisporus) and oyster mushroom (Pleurotus ostreatus). Antioxidant assay results using methanol extract of milky white mushroom has been provided along with the information about the compounds that are responsible for flavor profile both in fresh and dry mushrooms. Milky white mushroom extracts are known to have anti-hyperglycemic effect and anti-lipid peroxidation effect. The advantage of growing at elevated temperature creates newer avenues to explore milky white mushroom cultivation economically around the world, especially, in humid tropical and sub-tropical zones. Because of its incomparable productivity and shelf life to any other cultivated mushrooms in the world, milky white mushroom could play an important role in satisfying the growing market demands for edible mushrooms in the near future.

표고 수집균주의 재배적 특성 및 갈변과의 상관관계 (Cultural characteristics on collected strains of Lentinula edodes and correlation with mycelial browning)

  • 표고 수집균주의 특성을 파악하고 재배적 특성과 갈변과의 상관관계를 알아봄으로써 표고 톱밥재배를 안정적으로 실시하고 재배기간을 단축시킬 수 있는 방법을 모색하고자 하였다. 한천배지에서의 균사생장은 균주와 배지에 따라 다르게 나타났으며, MEA와 PDA에서는 큰 차이가 없었다. 균사생장 최적온도는 $20{\sim}25^{\circ}C$ 이었다. 200lux의 광조건에서 갈변패턴에 따라 균주를 분류할 수 있었다. 갈변기간은 한천배지에서는 30-40일, 톱밥봉지에서는 70~100일이 소요되었다. 자실체 특성을 관찰한 결과 300g 이상 수량을 보이는 균주가 12균주였으며, 자실체 수량 및 재배특성을 고려할 때 ASI 3046이 가장 우수한 것으로 판단되었다. 균사생장과 갈변 간의 유의성은 인정되지 않았으며, PDA배지와 톱밥 균사생장과는 유의성이 인정되었으며, 한천배지에서 갈변되는 기간과 톱밥배지에서 갈변되는 기간과의 관계는 고도 유의 상관을 나타내었다. 따라서 균이 갈변되는 성질은 영양분의 차이보다는 균의 고유한 성질이라고 생각되며 갈변이 빠른 균주를 선발하기 위해서는 한천배지를 이용하여 균을 선발할 수 있을 것으로 생각된다. 앞으로 톱밥재배에 필수적인 갈변에 대한 이해를 실제 재배에 접목시켜 톱밥재배 기간을 단축시키거나 생산성을 비약적으로 증대시키는 등 톱밥재배를 활성화시킨다면 품질이 높은 생표고를 낮은 가격으로 생산해 낼 수 있으리라 생각된다.
