Five strains of Pleurotus sajor-caju were collected from some countries including India and Papua New Guinea. Four strains produced brown fruitbody but the other strain from Papua New Guinea produced white fruitbody. Monokaryons obtained from both strains producing brown fruitbody and white fruitbody were mated each other. They showed different mating types such as brown strain of $A_1A_2B_1B_2$ and white strain of $A_3A_4B_3B_4$. DNAs were isolated from mycelia of five strains of P. sajor-caju. Mitochondrial DNA was seperated from nuclear DNA by bisbenzimide-CsCl ultracentrifugation. Digestion of the fungal mitochondrial DNA with Eco RI restriction endonuclease yielded from ten to fourteen fragments. Two strains of the five strains showed different restriction pattern of mitochondrial DNA from the other three strains. Summation of the fragment sizes gave a mitochondrial DNA size of about 60 to 65kb.
This study was carried out to optimize the conditions for a chemotaxonomic classification of Ganoderma species. The mycelia of Ganoderma species were extracted with 100% MeOH, and the concentrated extracts were further purified and partitioned with column chromatography (HP20) and n-BuOH, respectively. From the result of high-performance liquid chromatography (HPLC), the constituents of the samples were efficiently separated with a Chemco Pak $C_{18}$ column ($250mm{\times}4.6mm$) by linear gradient elution using water and acetonitrile as mobile phase components at a flow rate of 0.8 ml/min and detector wavelength at 210 nm. However, the samples without purification or partition were not detected the characteristic peaks. This profile could be used to classify and identify the various Ganoderma species.
The effects of environmental and nutritional requirement such as temperature, pH, different media, carbon, nitrogen, and carbon-to-nitrogen ratio on the mycelial growth of Cordyceps militaris strain 'Yedang 3' were studied. The optimum temperature and pH for the growth of mycelium were $20-25^{\circ}C$ and pH 6-7, respectively. Out of ten media tested, mushroom complete media (MCM) was the best medium for fast mycelial growth, and Sabouraud's dextrose agar yeast extract (SDAY), malt extract yeast extract agar (YMA) also were favored. The color and shape of colonies varied in different media. The best carbon sources for mycelial growth were fructose, mannitol, and sucrose, whereas the best nitrogen sources were tryptone and peptone. However, mycelia grew slowly in inorganic nitrogen compounds such as $NH_4Cl$, $(NH_4)_2SO_4$, $NH_4NO_3$, and $NaNO_3$. The optimum C:N ratio observed on the culture media was 30-40 range. These results provided basic information on cultural characteristics of vegetative growth and might be useful for spawn production in Cordyceps militaris.
Artomyces microspora is a genus of coral fungi from the family Auriscalpiaceae that have sporophores which are clavarioid, profusely and pyxidately branched, and devoid of a conspicuous stipe. These fungi can be found in summer and fall. This study aimed to decipher fundamental information regarding optimal growth conditions of Artomyces microsporus mycelia, including pH, temperature, carbon sources, and nitrogen sources. Based on the assessment of colony diameter and mycelial density, the optimal culture medium, temperature, and pH for mycelial growth were found to be PDA, 25 ℃, and pH 5.0, respectively. Furthermore, the study revealed that the optimal carbon and nitrogen sources for mycelial growth were 1% soluble starch and 2% malt extract, respectively. The other suitable inorganic nitrogen sources were deemed to be 0.1% NH4H4PO4 and 0.1% aspartic acid.

Newly, nutrients recovery by bioconversion in the swine waste which caused serious problems due to its high organic fraction and content of nutrients such as phosphorus and nitrogen is viewed as a considerable approach since it produces valuable product as well as recycling of resources. Consequently, it is necessary to find new methods to treat swine waste. One possible solution to this problem is to use this potential pollutant as a growth substrate for economically valuable products. The study for the fundamental improvement of bioconversion efficiency by finding optimum growth conditions using statistical models and biotechnology was performed. A novel approach to utilize swine waste by cultivating mycelia of the mushroom Phellinus linteus are described. A central composite face-centered design (CCF) for the experiments was used to develop empirical model providing a quantitative interpretation of the relationships among the three variables, which were substrate concentration, pH, and temperature. The maximal radial extension rate (2.78mm/d) of P.linteus was determined under the condition of 5.0 g COD/L, pH 5.0, and temperature $29.7^{\circ}C$. The results of this study suggest that swine waste could be utilized as a growth substrate for the cultivation of mushroom mycelia enhancing an efficiency of utilizing this by-product of the livestock industry.
The promoting effect of Pseudomonas sp. P7014 on the mycelia growth of Pleurotus eryngii was investigated. An ethyl acetate fraction (F5) from the culture supernatant of the bacteria was confirmed to contain the growth promoting compound (GPC). The GPC was identified to be indole acetic acid (IAA) by TLC, HPLC, MS/MS, and NMR analyses. P. eryngii mycelia grew rapidly both on PDA and in PDB after the treatment of GPC. The promoting concentration of GPC was as low as 1.0 nM. Tryptophan, the aminated form of IAA, was confirmed to be the precursor of IAA. These results suggested that bacterial secreted compound was IAA and plays an important role in promoting growth of mushroom mycelia.

This study was carried out to select cell wall degrading enzymes for maximizing protoplast yield from Basidiomycetes. The protoplasts were released from spore suspension, mycelia cultured on cellophane membrane, and homogenized mycelia of Flammulina velutipes using commercial cell wall degrading enzymes. The highest yield of protoplasts was obtained from the homogenized mycelia treated with the enzyme combination of $Glucanex^R$ 200G and cellulase onozuka R-10. The protocol was also available for Pleurotus ostreatus, P. eryngii, and Hypsizygus marmoreus.

Pleurotus species are the most consumed and cultivated mushrooms in Korea. Although oyster mushrooms (P. ostreatus) can be cultivated automatically, their storability is slightly lower than that of king oyster mushrooms (P. eryngii) and winter mushrooms (Flammulina velutipes); therefore, the export proportion of oyster mushrooms is very low. Since Korean mushrooms are highly preferred across Southeast Asian, the export of oyster mushrooms in the form of complete substrates is expected to be more promising than that of fresh mushroom. Here, 1 and 2.5 kg complete substrates of P. ostreatus 'Soltari' and P. sajor-caju 'Sambok' were prepared and stored at different temperature from 0 to 15℃ for 10 days. Thereafter, the formation of fruiting bodies was induced. Since the 2.5 kg complete substrates required 70 days of incubation, their mycelia were at an advanced age and their fruiting bodies did not grown normally. When 70%-incubated complete substrates were stored at 5-10℃, the growth was faster and more uniform and stable fruiting bodies were formed. Export test of complete substrates to Vietnam using distribution containers set at 0℃ and 15℃ revealed that the growth period was shortened by 1-2 days when the distribution containers were set at 15℃ and the yield of 'Soltari' increased by approximately 10%. In addition, even though the yield of 'Sambok' was similar between treatments at 0℃ and 15℃, the quality of fruiting bodies from 15℃-distributed complete substrates was much better than that of those from 0℃-distributed substrates.

To obtain basic data for the better use of Cordyceps pruinosa we reassessed the effect of different medium, culture method, pH, and carbon and nitrogen sources on the mycelial growth properties of four C. pruinosa isolates. The growth of mycelia differed among the four isolates depending on medium type and cultivation days. Among the tested 8 kinds of solid media, the four isolates grew well on PDA and MMMA(mushroom minimal medium agar). While, among the tested 8 kinds of liquid media, all the isolates grew well in SDYM(Sabourand's dextrose yeast extract medium). The isolates also grew well in the SDYM with pH from 4.0 to 9.5 without any inhibition. One isolate could best grow at pH 8 to 9.5. Regarding the ability of utilizing carbon source, the difference of mycelia growth among the isolates was the most with xylose. Regarding nitrogen source, three isolates could utilize urea which is new fact in this species. These results provide new points on the growth properties of the fungal mycelium which has not been explored before. Overall, this reassessed study concluded that it is necessary to check in advance the growth properties of mycelium when a new isolate of C. pruinosa is expected to be used for application.

This study was conducted to investigate the effect of mycelia of Lentinus edoes mushroom-cultured Lonicera japonica Thunberg (LLJ) on proliferation of the cancer cell lines (Hep3B, MCF-7 and HeLa), sarcoma 180 (S-180) and antiallergy. In an anti-cancer test using Hep3B (hepatic cancer cell), MCF-7 (breast cancer cell) and HeLa (uterine cancer cell), LLJ extract showed higher antiproliferating effect than that of LJ (Lonicera japonica Thunberg) extract. In an anti-cancer testing using Hep3B cells, LLJ extract showed growth-inhibitory effect of $85.60{\pm}4.66%$ at 3mg/rnL. In an anti-cancer testing using MCF-7 cells, LLJ and LJ extracts showed high antiproliferating effect. LLJ showed the tumor suppressive effect in mice injected with S-180 cells. The growth-inhibitory rates against tumor cells were 61% for LLJ, 37% for LJ. LLJ inhibited histamine release from rat peritoneal mast cells activated by compound 48/80. These results suggest that Lentinus edodes mushroom-cultured herb has an antiproliferating effect against cancer cell lines (Hep3B, MCF-7 and HeLa) and S-180 tumor, and will be beneficial in the treatment of allergic reaction.
