Morphological Characteristics and Classification of Zizyphus Cultivars in Korea by Multivariative Analysis (다변량 분석에 의한 국내산 대추나무 품종의 형태적 특성과 유연관계)

  • The objectives of this study, an analysis of fruit and leaf morphological characteristics among the five Zizyphus cultivars could be used for the investigation of cultivars classification and could provide information to make out the UPOV TG(Test Guidelines). ANOVA tests showed that there were statistically significant differences in all fruit and leaf morphological characteristics among the five Zizyphus cultivars at 1% level. But, for kernel characteristics, differences were statistically non-significant among the cultivars. Approximately, the Wolchul and Boeun cultivars showed larger and smaller values in overall characteristics and cultivars, respectively. The results of principal component analysis(PCA) for the fruit and leaf morphological characteristics showed that the first for principal components(PC's) explained about 65.3% of the total variation. The first PC was correlated with those characteristics that were mainly related to the terminal leaf length(TLL), leaf length(LL), fruit length(FL), terminal leaf width(TLW), and leaf petiole length(LPL). The second and third PC was mainly correlated with the terminal leaf morphological index(TLMI). Therefore, these characteristics were important to analysis of the fruit and leaf morphological characteristics and classification among the five Zizyphus cultivars. Cluster analysis using UPGMA method based on principal components showed that five Zizyphus cultivars could be clustered into two groups. Group I comprises Mudung, Wolchul, and Bokjo and Geumsung cultivars, Group II is Boeun cultivar. These results well similar to that of principal component analysis.

  • Mahalanobis Taguchi System (MTS) is a pattern information technology, which has been used in different diagnostic applications to make quantitative decisions by constructing a multivariative measurement scale using data analytic methods. In MTS approach, Mahalanobis distance (MD) is used to measure the degree of abnormality of patterns and principles of Taguchi methods are used to evaluate accuracy of predictions based on the scale constructed. The advantage of MD is that it takes into consideration the correlations between the variables and this consideration is very important in pattern analysis. The purpose of this study is constructing admission diagnosis system and define the effect of admission requirements for student's academic accomplishment.

Study of Metabolic Profiling Changes in Colorectal Cancer Tissues Using 1D 1H HR-MAS NMR Spectroscopy

  • Metabolomics is a field that studies systematic dynamics and secretion of metabolites from cells to understand biological pathways based on metabolite changes. The metabolic profiling of intact human colorectal tissues was performed using high-resolution magic angle spinning (HR-MAS) NMR spectroscopy, which was unnecessary to extract metabolites from tissues. We used two different groups of samples, which were defined as normal and cancer, from 9 patients with colorectal cancer and investigated the samples in NMR experiments with a water suppression pulse sequence. We applied target profiling and multivariative statistical analysis to the analyzed 1D NMR spectra to identify the metabolites and discriminate between normal and cancer tissues. Cancer tissue showed higher levels of arginine, betaine, glutamate, lysine, taurine and lower levels of glutamine, hypoxanthine, isoleucine, lactate, methionine, pyruvate, tyrosine relative to normal tissue. In the OPLS-DA (orthogonal partial least square discriminant analysis), the score plot showed good separation between the normal and cancer groups. These results suggest that metabolic profiling of colorectal cancer could provide new biomarkers.

The Osteoporotic Condition as a Predictive Factor for Hemorrhagic Transformation in Acute Cardioembolic Stroke

  • Objective : Hemorrhagic transformation (HT) can be occurred after acute cerebral infarction. HT can worse symptoms in severe cases and adversely affect long-term prognosis. As bone and vascular smooth muscle are composed of type 1 collagen, we aimed to identify a potential relationship between bone mineral density (BMD) and HT after acute cardioembolic stroke. Methods : As an indicator of BMD, we used mean frontal skull Hounsfield unit (HU) values on brain computed tomography (CT). Multivariative hazard ratios were calculated using Cox regression analysis to identify whether the osteoporotic condition was an independent predictor of HT after acute cardioembolic stroke. Results : This 11-year analysis enrolled 506 patients who diagnosed as acute cardioembolic infarction. The first tertile of skull HU value was an independent predictor of HT development compared to the third tertile (hazard ratio, 2.12; 95% confidence interval, 1.13-3.98; p=0.020). We observed no interactions between age and skull HU with respect to HT statistically. Conclusion : The results of this study revealed an association between osteoporotic conditions and HT development after acute cardioembolic stroke. A convenient method to measure the cancellous bone HU value of the frontal skull using brain CT images may be useful for predicting HT in patients with acute cerebral infarction.

Characterization of the Effects of Silver Nanoparticles on Liver Cell Using HR-MAS NMR Spectroscopy

  • AgNPs (silver nanoparticles) has been widely used for the commercial products, which have antimicrobial agent, medical devices, food industry and cosmetics. Despite, AgNPs have been reported as toxic to the mammalian cell, lung, liver, brain and other organs and many researchers have investigated the toxicity of AgNPs. In this study, we investigated toxicity of the AgNPs to the liver cell using metabolomics based on HRMAS NMR (High Resolution Magic Angle Spinning Nuclear Magnetic Resonance) technics, which could apply to the intact tissues or cells, to avoid the sample destruction. Target profiling and multivariative statistical analysis were performed to analyze the 1D $^1H$ spectrum. The results show that the concentrations of many metabolites were affected by the AgNPs in the liver cell. The concentrations of glutathione (GSH), lactate, taurine, and glycine were decreased and most of amino acids, choline analogues, and pyruvate were increased by the AgNPs. Moreover, the levels of the metabolites were recovered upto similar level of metabolites in the normal cell by the pre-treatment of NAC, external antioxidant. The results suggest that the depletion of the GSH by the AgNPs might induce the conversion of lactate and taurine to the pyruvate.

Changes of the Clinicopathological Characteristics and Survival Rates of Gastric Cancer with Gastrectomy - 1990s vs early 2000s (위절제 위암 환자의 임상병리학적 특성과 생존율의 변화: 1990년대와 2000년대 초기의 비교)

  • Purpose: The incidence of upper gastric cancer and especially the diffuse type have increased in western countries. The aim this study was to investigate the chronologic changes of the clinicopathological features and survival rates of Korean upper gastric cancer patients. Materials and Methods: 1,638 gastric cancer patients who underwent gastrectomy were included in this study and they were divided into two groups; the 1990's (1991~1999, n=987) and the early 2000's (2000~2003, n=651). We evaluated the differences of the clinicopathologic features and the factors that affected the survival rates by univariative and multivariative analysis. Results: The older age (>60) patients increased from 42.7% to 50.7% respectively. Being overweight (body mass index$\geq$23) also increased from 31.5% to 43.2%. For the pathology, the incidence of stage Ia gastric cancer increased (29.8% to 44.5%) and the incidence of stage IV gastric cancer decreased (23.5% to 11.8%). Yet there was no difference according to the WHO classification, Lauren's classification and the location of tumor between the groups. The 5 year survival rates increased 67.7% to 83.7%, according to the group. Multivariative analysis showed that the odd ratios of the early 2000s was 0.715 (95% CI; 0.555~0.921) as compared to that of the 1990s. Conclusion: There were no changes of the clinicopathologic features, like the pattern in western countries, although the incidence early gastric cancer, old age patients and overweight patients increased. The survival rate of early 2000s was better that that of the 1990s.

Classification of Garlic Germplasms Based on Agronomic Characteristics and Multivariative Analysis (마늘 유전자원의 작물학적 특성과 다변량 분석에 의한 품종군 분류)

  • This research was conducted to investigate the genetic diversity and select useful accession with agronomic characteristics of garlic (Allium sativum L.). germplasms at Garlic Research Institute in Chungbuk Agricultural Research and Extension Service. Morphological diversity and relationships among 160 germplasms collected from 26 countries were assessed by methods of clustering and principal component analysis. Among 11 types of leaves and bulbs characteristics, emergence days of leaf showed the highest variation with coefficient of variation of 84.8%, and the bulb weight and the number of scales showed higher variability with 24.3%. Correlation analysis based on 11 quantitative traits showed that bulb weight and bulb length have very high positive correlation with bulb quantity. Plant height, leaf length, and number of leaves showed positive correlation with bulb weight as collections with better performance in growth produced large bulb with higher quality. The cluster analysis based on 5 principal components generated 6 clusters with an average distance of 1.6 among clusters. Domestic genetic resources were the largest with 36 species (22.5%) in group II.

Treatment Result and Prognostic Factors in Pateints with Esophageal Cancer (식도암의 근치적 치료성적 및 예후인자)

  • Purpose : To analyse clinical outcome and prognostic factors according to treatment modality, this paper report our experience of retrospective study of patients with esophageal cancer Materials and Methods : One hundred and ten patients with primary esophageal cancer who were treated in Presbyterian Medical Center from May 1985 to December 1992. We analysed these patients retrospectively with median follow up time of 28 months, one hundred and four patients($95{\%}$) were followed up from 15 to 69 months. In methods, twenty-eight patients were treated with median radiation dose irradiated 54.3Gy only. Fifty-six patients were treated with combined chemoradiotherapy. Sixteen cases of these patients were treated with concurrent chemoradiation and the other patients(forty cases) were treated sequential chemoradiotherapy. In concurrent chemoradiotherapy group, patients received 5-FU continuous IV infusion for 4 days. Cisplatin IV bolus. and concurrent esophageal irradiation to 30 Gy. After that patients received 5-FU continuous IV, Cisplatin bolus injection and Mitomycin-C bolus IV, Bleomycin continuous IV, and irradiation to 20 Gy. In sequential chemoradiotherapy group, the chemotherapy consisted of 5-FU 1,000mg/$m^2$ administered as a continuous 24 hour intravenous infusion during five days and Cisplatin 80-100mg/$m^2$ bolus injected, or Bleomycin, Vinblastine, Cisplatin, Methotrexate were used of 1 or 2 cycles. After preoperative concurrentm chemoradiation twenty-six patients underwent radical esophagectomy. Results : Ninety-three patients could be examined for response assessment, By treatment modality, response rates were $85.1{\%}$ for radiation alone group and $86.3{\%}$ for combined chemoradiation group. But in operation group, after one cycle of concurrent chemoradiation treatment, response rate was $61.9{\%}$. The pathologic complete response were $15.4{\%}$ in operation group. Overall median survival was II months and actuarial 5-year survival rate was $8{\%}$. The median survival interval was 6 months for radiation alone group, 11 months for combined chemoradiation group and 19 months for operation group. And also median survival was 19 months for complete responder group that 8 months for noncomplete responder group. In univariative analysis, statistically significant prognostic factors were tumor size, clinical stage, tumor response, and operation. In multivariative analysis, significantly better survival was associated with clinical stage, tumor response, radiation dose, and operation. Conclusion : Compared with radiotherapy alone, combined multimodality may improve the median survival in patients with localized carcinoma of the esophagus and toxicity is acceptable.

Evaluation of factors affecting sensory neural hearing loss (감각신경성 청력손실에 영향을 미치는 제 요인에 관한 연구)

  • Reports on a potential relationship between sensory neural hearing loss(SNHL) and cardiovascular or hematologic factor show that the results are controversial. A detailed analysis of risk factors in the development of SNHL was carried out in 3,050 non-noise exposed healthy worker. The mean hearing threshold of both ears at 500, 1000, 2000, 4000, 8000Hz was measured and the effect of age, possible cardiovascular risk factor and hematologic factor (blood viscosity and hemostatic factors) on SNHL were evaluated. first, each of these were associated with loss of hearing sensitivity when univariately and multivariatively analysed. In a multiple regression model, age, sex, body mass index, WBC and total cholesterol level were independently associated with the mean of hearing sensitivity decrease at 4000 and 8000Hz. Second, study subjects were divided into two group (normal vs SNHL) and we compare the possible risk between both groups, and analysed univariate and multivariative logistic model. In a multiple logistic regression model, age, sex, body mass index, WBC and total cholesterol level, total protein, platelet were independently associated with SNHL. Our results show that we have some reliable indices of susceptibility to SNHL using cardiovascular measures or biochemical factor, but future, more extensive studies are required.
