Estimating free-living human energy expenditure: Practical aspects of the doubly labeled water method and its applications

  • The accuracy and noninvasive nature of the doubly labeled water (DLW) method makes it ideal for the study of human energy metabolism in free-living conditions. However, the DLW method is not always practical in many developing and Asian countries because of the high costs of isotopes and equipment for isotope analysis as well as the expertise required for analysis. This review provides information about the theoretical background and practical aspects of the DLW method, including optimal dose, basic protocols of two-and multiple-point approaches, experimental procedures, and isotopic analysis. We also introduce applications of DLW data, such as determining the equations of estimated energy requirement and validation studies of energy intake.

창출(蒼朮) Alkaloid 가 Schedule Controlled Behavior 에 미치는 영향(影響) (The Effect of Atractylis Alkaloid on Schedule-Controlled Behavior)

  • The effect on schedule controlled behavior and toxicity of Atractylis alkaloid was investigated in the pigeons which were trained on a multiple fixed-ratio fixed-interval schedule of food presentation. Atractylis alkaloid decreased the rate of responding during both the fixed-interval and fixed ratio component of the schedule at 10 mg/kg. Further depression occurred at 30 mg/kg. This 'flat dose-response curve for depression of conditioned behavior was typical of tranquilizers. Conclusively it was suspected that Atractylis alkaloid had major tranquilizing activity.

Combination Therapy of Prednisolone and Toceranib Phosphate in a Dog with Malignant Metastatic Insulinoma

  • A 14-year-old intact female Yorkshire terrier was presented with a 2-month history of shivering, intermittent pelvic limb weakness and collapse. Biochemical abnormalities revealed inappropriately increased serum insulin concentration with persistent hypoglycemia. Abdominal ultrasound revealed multiple various sized nodules in liver and fine-needle aspirates of the nodule showed typical neuroendocrine cells with high cellularity. Computed tomography (CT) revealed well-defined hyperattenuating mass in the right pancreatic lobe with homogenous enhancement. CT findings were consistent with a pancreatic tumor with malignant metastasis. Treatment was initiated with low-dose prednisolone and toceranib phosphate. The dog was maintained stable with no more progression of clinical signs and it is worth to try toceranib phosphate in a dog with metastatic insulinoma for improving the quality of life.

Deformable image registration in radiation therapy

  • The number of imaging data sets has significantly increased during radiation treatment after introducing a diverse range of advanced techniques into the field of radiation oncology. As a consequence, there have been many studies proposing meaningful applications of imaging data set use. These applications commonly require a method to align the data sets at a reference. Deformable image registration (DIR) is a process which satisfies this requirement by locally registering image data sets into a reference image set. DIR identifies the spatial correspondence in order to minimize the differences between two or among multiple sets of images. This article describes clinical applications, validation, and algorithms of DIR techniques. Applications of DIR in radiation treatment include dose accumulation, mathematical modeling, automatic segmentation, and functional imaging. Validation methods discussed are based on anatomical landmarks, physical phantoms, digital phantoms, and per application purpose. DIR algorithms are also briefly reviewed with respect to two algorithmic components: similarity index and deformation models.

Evaluation of anti-inflammatory activity of Solanum trilobatum roots

  • This study evaluated the anti-inflammatory potential of the crude alkaloidal fraction (CAF) of methanol extract of Solanum trilobatum Linn. (Solanacea) root in animal models of inflammation. Crude alkaloidal fraction at doses of 25, 50 and 100 mg/kg significantly (p < 0.01) reduced carrageenan induced rat paw volume at 3 h after carrageenan challenge as compared to control group of animals. CAF (25, 50 and 100 mg/kg) significantly (p < 0.01) and dose dependently suppressed cotton pellet induced granuloma formation. Topical application of CAF (1, 5 and 10 mg/ear) markedly inhibited multiple application of TPA in mice. CAF elicited pronounced inhibitory effects on formaldehyde and adjuvant induced arthritis in rats. These results indicate that CAF of methanol extract of the Solanum trilobatum has anti-inflammatory activity in acute and chronic inflammation.

복어 균질액의 Caco-2 인간 결장직장암세포 성장 억제 효과에 대한 연구 (Inhibitory Effect of Globefish Homogenate on the Growth of Caco-2 Human Colorectal Cancer Cells)

  • Colorectal cancer is a leading cause of cancer mortality worldwide. Many studies show that most cases of human colorectal cancer arise from adenomatous polyps, which are usually dysplastic, nonmalignant precursor lesions; however, accumulation of multiple somatic mutations leads some to develop into advanced adenoma, which ultimately progresses to an invasive colorectal cancer. Notwithstanding the efforts made to improve chemotherapy, most colorectal cancers are unresponsive to this form of treatment, and malignant colorectal cancers remain incurable. To reduce the incidence of colorectal cancer mortality, further studies to improve colorectal cancer treatment are needed. Here, we show that Globefish homogenate suppresses the growth of Caco-2 human colorectal cancer cells, and that the homogenate inhibits Caco-2 cell proliferation in a dose-dependent manner. These data suggest that Globefish homogenate may suppress colorectal cancer development.

Transcriptional Response According to Strength of Calorie Restriction in Saccharomyces cerevisiae

  • To characterize gene expression that is dependent on the strength of calorie restriction (CR), we obtained transcriptome at different levels of glucose, which is a major energy and carbon source for budding yeast. To faithfully mimic mammalian CR in yeast culture, we reconstituted and grew seeding yeast cells in fresh 2% YPD media before inoculating into 2%, 1%, 0.5% and 0.25% YPD media to reflect different CR strengths. We collected and characterized 160 genes that responded to CR strength based on the rigorous statistical analyses of multiple test corrected ANOVA (adjusted p value < 0.1 or raw p value < 0.0031) and Pearson correlation (|r| > 0.7). Based on the individual gene studies and the GO Term Finder analysis of 160 genes, we found that CR dose-dependently and gradually increased mitochondrial function at the transcriptional level. Therefore, we suggest these 160 genes are markers that respond to CR strength and that might be useful in elucidating CR mechanisms, especially how stronger CR extends life span more.

The Heterochromatin Protein 1 (HP1) Family: Put Away a Bias toward HP1

  • Heterochromatin protein 1 (HP1) was first described in Drosophila melanogaster as a heterochromatin associated protein with dose-dependent effect on gene silencing. The HP1 family is evolutionarily highly conserved and there are multiple members within the same species. The multi-functionality of HP1 reflects its ability to interact with diverse nuclear proteins, ranging from histones and transcriptional co-repressors to cohesion and DNA replication factors. As its name suggests, HP1 is well-known as a silencing protein found at pericentromeres and telomeres. In contrast to previous views that heterochromatin is transcriptionally inactive; noncoding RNAs transcribed from heterochromatic DNA repeats regulates the assembly and function of heterochromatin ranging from fission yeast to animals. Moreover, more recent progress has shed light on the paradoxical properties of HP1 in the nucleus and has revealed, unexpectedly, its existence in the euchromatin. Therefore, HP1 proteins might participate in both transcription repression in heterochromatin and euchromatin.

푸들개에서 발생한 특발성 결절성 피부지방염의 발생 예 (Idiopathic Nodular Panniculitis in a Poodle Dog)

  • A seven-year-old male poodle was presented with multiple non-pruritic draining skin lesions. On physical examination the dog was depressed and had three draining nodules 3.0 to 4.0 cm in diameter on the neck, flank and back. The multifocal sparing of fat lobules, macrophages, and numerous neutrophils were observed in aspiration cytologic examination of nodule. Haematological examination demonstrated a moderate leukocytosis and neutrophilia. An aerobic bacterial culture was undertaken from draining lesions on the flank. But there was no bacterial growth in aerobic culture. Therapy was initiated with oral cephalexin at a dose of 20 mg/kg, body weight three times daily and prednisolone at 1.0 mg/kg twice daily. The (lug returned to normal within 24 hours of treatment and its general attitute and appetite were improved. However, within 1 month new nodules had appeared on the dorsum over the rump, coalescing into a mass of draining nodules and tracts.

Carbon Ion Therapy: A Review of an Advanced Technology

  • This paper provides a brief review of the advanced technologies for carbon ion radiotherapy (CIRT), with a focus on current developments. Compared to photon beam therapy, treatment using heavy ions, especially a carbon beam, has potential advantages due to its physical and biological properties. Carbon ion beams with high linear energy transfer demonstrate high relative biological effectiveness in cell killing, particularly at the Bragg peak. With these unique properties, CIRT allows for accurate targeting and dose escalation for tumors with better sparing of adjacent normal tissues. Recently, the available CIRT technologies included fast pencil beam scanning, superconducting rotating gantry, respiratory motion management, and accurate beam modeling for the treatment planning system. These techniques provide precise treatment, operational efficiency, and patient comfort. Currently, there are 12 CIRT facilities worldwide; with technological improvements, they continue to grow in number. Ongoing technological developments include the use of multiple ion beams, effective beam delivery, accurate biological modeling, and downsizing the facility.
