Identification of Productive Mulberry Silkworm Hybrids Resistant to Densonucleosis Virus Type 1 (BmDNV1)

  • The use of commercial silkworm hybrids resistant to important silkworm diseases is economical and better option particularly in tropical areas. This necessitated the evolution of productive bivoltine silkworm breeds non-susceptible to $BmDNV_1$. Non-susceptibility to $BmDNV_1$, infection was found to be controlled by a single recessive gene, nsd-l or a dominant gene, Nid-l. A major dominant/recessive gene confers resistance to $BmDNV_1$, from potent donor parents have been transferred to 10 productive but susceptible bivoltine silkworm strains through conventional breeding methods. By utilizing these breeds prepared 25 hybrids $(5{\times}5)$ and hybrid evaluation was carried out to identify most promising hybrids resistant to $BmDNV_1$. All these hybrids are inoculated with $BmDNV_1$ inoculum along with productive control hybrid $CSR2{\times}CSR4$ and reared under standard rearing procedure. Based on inoculated rearing and test reeling results, two most promising hybrids $(CSR18DR{\times}CSR29DR\;and\;CSR21DR{\times}CSR50DR)$ were selected for commercial exploitation. The selected hybrids have shown a survival rate of >85% with productive traits, where as control hybrid have shown 11.1% survival with inferior cocoon traits. The methodologies adopted were discussed.

Quantitative Analysis of 1-deoxynojirimycin in Mulberry Leaves (뽕나무 유전자원의 1-deoxynojirimycin 함량 변이)

  • Countries of the world are competing for the collection and utilization of genetic resources, which is a resource of value-added industrialization. We are building database of silkworm and mulberry resources with morphological, genetic characteristics data etc. Moreover, recent bioactive compound has been receiving increasing attention. So we analyzed 1-deoxynojirimycin(1-DNJ) content as a this item in addition to the basic information using 363 strains(varieties) of mulberry genetic resources. They were grown under the same environment and conditions. Mulberry leaves were collected and then freeze-dried and powdered for 1-DNJ test. As a result, 1-DNJ mean content of 363 mulberry strains was $0.176{\pm}0.077%$, and the coefficient of variation (CV) was 43.5%. The variation between the strains was greatly severe. Among the tested strains, 'C1D89/29' was showed the highest content of 0.47%, whereas 'Pumbo 24' and 'Turkey E' were showed the lowest content of 0.05% respectively. The content of 1-DNJ of 16 mulberry varieties for silkworm rearing were compared. Average concentration was $0.17{\pm}0.04%$, and the coefficient of variation (CV) was 22.8%. Variation among cultivars was not significant. Finally we selected 1-DNJ high-containing 11 strains. They are as follows. 'C1D89/29', 'Hiihak', 'Jeokasibmunja', 'Gweonchil', 'Botongsibmunja', 'Jeokchuk', 'bulguksang', 'Geunsookgojo', 'Busanggeum', 'Guksang 20' and 'Taekwang' which are more than twice of the overall average content.

  • Surveying, laboratory and field trial were performed to elucidate the causes of non-spinning syndrome of silkworm, Bombyx mori, abnormal symptoms of silkworm instars observed in certain sericultural fanning region. Mortality and growth of silkworm was still influenced by feeding the mulberry leaves sprayed with 10,000 fold diluted solution of standard spray of diflubenzuron WP (25%). Mortality and growth of silkworm fed with diflubenzuron-sprayed mulberry leaves with recommended spray solution were entirely affected even six weeks after spraying irrespective to the instars. Larval period of 5th instar and cocooning duration of silkworm was not affected in case 50m of buffer zone has maintained in near-by orchard field sprayed with diflubenzuron WP (25%). However, survival rate of pupae was not influenced in case 100 m of buffer zone has kept. Insect growth regulating insecticides were required to keep a certain distance of buffer zone when sprayed with water-diluted solution, 100 m from the application site at least in order to prevent contamination of the spraying drift by the chemicals.

On the Measurement of Biomass and the Productivity of the Cultivated Mulberry Plants (뽕나무의 현존량추정법과 생산력에 대한 연구)

  • With the cultivated mulberry plant which feeds silkworm on its leaves in sericulture, the measuring methods of biomass of terrestrial organs were compared and the annual net productivity was estimated. The measurement of the standing crops by means of allometric method on basis of relation between the parameter D230H, square of the diameter of the branch on 30cm high above ground($D{\frac}{2}{30}$) multiplied by its height(H), and the amount of leaves(WL) or of branch (Ws) was more accurate than other methods on basis of correlation between a character of the branch such as H, D30 or D230 and WL or Ws. The estimate of value of the net productivity of terrestrial parts of mulberry was 9.06-12.54 ton/ha.year, which was similar to that obtained from secondary forests in cool temperate zone.

  • On the purpose of exploiting cellulose resourse which is needed for artificial diet of silkworms, mulberry branch, eastern cotton wood branch, pitch pine pulp, poplar pulp, chaff, sawdust and rice straw were examined. Among the matters above mentioned. mulberry branch and pitch pine pulp were found to be replaceable as cellulose component in the artificial diet. By means of some chemical treatments, it is found that one of the extracted cellulose powder from mulberry branches is more useful in the larval growth.

  • Eight new three-way cross heterozygotic hybrid genotypes of the mulberry silkworm Bombyx mori, resulted from hybridization of multivoltine as female and bivoltine Fl hybrid as male component, have been evaluated for the cumulative effect of nine traits of commercial economic importance with the objective of selecting out the most prospective ones for their exploitation as an intermediate technology during unfavorable season. Five of them namely, G(P5${\times}$NBl8) (E.I. =57.23); S(NB18${\times}$P5) (E.I. : 55.48); S(P5${\times}$NBl8) (E.I. = 52.44); S(KBN${\times}$B7) (E.I. = 50.42) and S(NB7${\times}$KB) (E.I. : 50.16) have scored higher values of the Evaluation Index (E.I.) and could be selected in the same order for exploitation during unfavorable seasons in tropical climates.

Studies on the Accelerative Function for the Silkworm Maturation with Ecdysis Hormone (탈피 Hormone의 누에 숙화 촉진에 관한 연구)

  • It is very important to make mounting numerous silkworms by their normal growth when considering the sericultural management. There were several studies on the utilization of repellent to the matured silkworms to mounting, studies concerning forcing maturity of silkworms were developed considerably after studies on the insect ecdysis hormone. Commercial named MSH(II) (Inoksterone) similar substance of ecdysterone and DAT repellent (Dodesil alcohl) were used for this study, and the results can be summarized as follows; 1. MSH (II) mixed with the mulberry leaves shown the matured effect on the growth of silkworms and increased amount of MSH (II) shortened the last stage in their growth. 2. MSH (II) mixed with the mulberry leaves when 5% of early matured silkworms appeared were most effective. 3. DAT was hastened their mounting. 4. Utilization of MSH (II) and DAT together were hastened silkworms mounting and those chemicals can be used practically in the silkworm rearing.

  • In the light of the present situation of fertilizing presently apllied in farmer's mulberry fields, it was considered to be value enough to carry out the experiment on whether its degree of fertilizing have no affect on silkworm growth and their cocoon harvest as well, so that the results thus obtained from the experiment are as following; The amount of fertilizing, N 44.4kg, P$_2$O$\sub$5/ 18.4kg, and K$_2$O 34.4kg, respectively were minimum for the . east affect on the silkworm growth and cocoon yields.
