Discrimination of Pinellia tuber through Morphological characteristics and Genetic analysis (형태적 특징과 유전자분석을 통한 반하(半夏) 감별 연구)

  • The following is a list of morphologic and genetic characteristics of Pinellia tuber. 1. The original plant of Pinellia tuber is Pinellia ternata$(T_{HUNB})$$B_{REIT}$. With regards to its external morphology, it is smaller than other Araceae species and its spadix is longer than its leaves, which trifurcate. 2. As regards its internal morphology, its mucous cell is elliptical and the vessel is helical or annular-shaped. Granules exist in abundance and in various shapes. 3. Distribution and size of laticifers are the key criteria on which to differentiate between domestic and imported Pinellia tuber. Laticifers are mainly distributed in the epidermis in domestic Pinellia tuber and in the cortical parenchyma in imported Pinellia tuber. The size of laticifers is somewhere between 1,3 and $8{\mu}m$ in diameter in imported Pinellia tuber bigger than its domestic counterpart. 4. RAPD markers display a great similarity in bands between domestic and Chinese Pinellia tuber. However, RAPD primers 352, 358, 365, 368 and 374 are distinctive markers for domestic Pinellia tuber. In the meantime, North Korean Pinellia tuber, morphologically similar to domestic Pinellia tuber, is genertically distinctive from its domestic counterpart in primers 354, 358, 365, 368, 374 and 379, a finding that supports the postulation that North Korean Pinellia tuber is tuber of another Araceae species.

Therapeutic effect of the traditional herbal formula, Hwanglyeonhaedok-tang, on rhinitis: A Review of the experimental study (황련해독탕이 비염 개선에 미치는 효과: 기초실험 연구 문헌 분석)

  • Objectives : This study aimed to assess the scientific evidence for the use of Hwanglyeonhaedok-tang, a traditional herbal formula, in the treatment of rhinitis, and prepare the basis for the investigational new drug application by analyzing the experimental studies. Methods : Ten electronic databases were searched up to December, 2016 without language limitation. Experimental studies on the anti-inflammatory and anti-allergic effects of Hwanglyeonhaedok-tang against rhinitis were included. We extracted data about study design, characteristics of intervention, outcomes, and pharmacological effects from the included studies and summarized them. Results : Eight hundred and thirty-three potentially relevant studies were identified, of which 18 experimental studies met our inclusion criteria. Of 18 included studies, 5 had conducted cell viability test, and all studies had reported that Hwanglyeonhaedok-tang was non-cytotoxic. Hwanglyeonhaedok-tang exhibits anti-inflammatory effect by regulating the inflammation-related cytokines including nitric oxide(NO), prostaglandin $E_2(PGE_2)$, interleukin-6(IL-6), and tumor necrosis $factor-{\alpha}(TNF-{\alpha})$ in vitro and in vivo. Hwanglyeonhaedok-tang exhibits anti-allergic effect by suppressing eosinophil, and histamine levels. Hwanglyeonhaedok-tang helps in the recovery of nasal mucous membrane by supressing goblet cells, heat shock protein 70, and substance P. Conclusions : This study suggests that Hwanglyeonhaedok-tang has the potential to be developed as therapeutic agent for rhinitis. Further experimental and clinical studies needed to be performed to prove the safety and efficacy.

Systemic Pemphigus Vulgaris: A Case Report (전신성 심상성 천포창의 치험례)

  • Purpose: Pemphigus vulgaris, a rare autoimmune blistering disease of the skin and mucous membranes remains a challenging disease to treat. Management is focused on immunotherapy against autoimmune antibodies that target keratinocyte cell adhesion molecules, and antibiotics preventing secondary infections. There is no established dressing protocol and skin is usually manipulated the least amount possible in order to minimize irritation. The authors suggest that early initiation of aggressive bathing and debridement of skin lesions, with nutritional support, is essential in accelerating resolution. Methods: A 40 year-old male previously diagnosed with pemphigus vulgaris was admitted due to exacerbation of mucocutaneous lesions involving the epidermis and mucosa of the whole body. Steroids, immunosuppressants, intravenous immunoglobulin and antibiotics were administrated, but infection and de-epithelialization progressed, while his general condition deteriorated with a weight loss of over 20 kilograms. The plastic surgery department intervened with daily bathing, debridement of unhealthy debris and non-traumatizing coverage of growing epithelium. Total parenteral nutrition and mobilization with rehabilitation therapy was initiated as early as possible. Results: After bathing, healthy epithelium gradually covered the patient's entire body, while his general condition improved with a corresponding weight gain of 14 kgs. Conclusion: Treatment of pemphigus vulgaris focuses on immunotherapy and infection control. However, an equal amount of attention should be laid on early intervention with daily dressings including bathing and irrigation, nutritional support, and exercise as this accelerates resolution of existing infections, promotes healthy epithelialization and leads to faster recovery.

  • Chitosan is applied as a dressing for oral mucous wound and a tampon following radical treatment of maxillary sinus. Furthermore, it is being investigated as an absorbing membrane for endodontic and periodontic surgeries. A few studies have reported osteoconduction and osteogenesia at the site of chitosan implant in vivo. However, compared with soft tissue healing processes, the mechanisms concerning effects of chitosan for biological mineralization have not yet been resoil In the present study, we studied the gene expression pattern using cDNA microarray and RT-PCR analyses in hard tissue forming osteoblasts cultured with water-soluble and low molecular weight chitooligosaccharide. cDNA microarray analysis revealed that 16 genes were expressed at 〉1.5-fold higher signal ratio levels in the experimental group compared with the control group after 3 days. RT-PCR analysis showed that chitosan oligomer induced an increase in the expression of two genes, CD56 antigen and tissue-type plasminogen activator. Furthermore, the expression of mRNAs for BMP-2 was almost identical in the experimental and control groups after 3 days of culture, but slightly increased after 7 days of culture with chitosan oligomer. These results suggest that a super-low concentration of chitooligosaccharide could modulate the activity of osteoblastic cells through mRNA levels and that the genes concerning cell proliferation and differentiation can be controlled by water-soluble chitosan.

Acute Toxicity Study of the 2-butoxyethanol by Intratracheal Instillation in Male Sprague-Dawley Rats (수컷 랫드(Sprague-Dawley)에서 2-부톡시에탄올(2-butoxyethanol)의 단회 기도내 투여에 따른 급성 독성시험)

  • Objectives: The present study aimed to evaluate the potential toxicity of 2-butoxyethanol after intratracheal instillation in male rats. Methods: In order to calculate median lethal dose (LD50) of 2-butoxyethanol using Probit analysis with SAS program, the 2-butoxyethanol was administered with dose levels of 0, 101.64, 203.28 and 406.56 mg/kg by once intratracheal instillation to male rats. During the test period, clinical signs, mortality, body weights, organ weights, hematology, and serum biochemistry were examined. At the end of 14 days observation period, all animals were sacrificed and gross finding and histopathological examination were performed. Results: All animals of 406.56 mg/kg group died within 2 weeks after the administration of 2-butoxyethanol. Treatment-related clinical signs, gross observation and histopathological changes (mucous cell hyperplasia, alveolar macrophage aggregation, and hemorrhage) of lung exhibited an increased in 2-butoxyethanol treated groups in a dose dependent manner. However, there were no changes in the organ weights, hematology and serum biochemistry, and histopathology of any other organ except lung. Conclusions: On the basis of the results, it was concluded that a single intratracheal instillation of 2-butoxyethanol in male Sprague-Dawley rats resulted in some adverse effects on mortality, clinical sign, and histopathology in the lung. In the experimental conditions, the LD50 of 2-butoxyethanol was considered to be 287.2 mg/kg and lung was founded to be the target organ of 2-butoxyethanol.

Influence of Panax Ginseng on DNA Synthesis of Submandibular Gland in Mice Receiving ACTH(II) (고려인삼이 ACTH를 받은 마우스의 악하선 조직 DNA 합성능에 미치는 영향( II ))

  • It was planned to investigate, in mice that received ACTH, the influence of Panax Ginseng upon DNA synthesis of submandibular gland by observing incorporation of $[^3H]$ thymidine into the tissue cells. Thirty male mice $(body\;weight:\;18{\sim}20\;g)$ were divided equally into the ginseng-ACTIH and the saline-ACTH groups. Each animal of the ginseng-ACTH and the saline-ACTH groups received every day (subcutaneously) 0.05 m1/10 g body weight of ginseng extract(4 mg of ginseng alcohol extract in 1 ml of saline) and the same amount of saline, respectively, for 5 days. On the 5th experimental day, all animals received 0.01 unit of ACTH intraperitoneally one hour after the last medication, and $1\;{\mu}Ci/g$ body weight of $[^3H]$ thymidine after one more hour. Five animals, at a time, of each group were sacrificed 1, 10, and 24 hr after $[^3H]$ thymidine administration, and the radioactivity of cells in their mandibular gland was measured autoradiographically in terms of the % number of radioactive cells in 1,000 cell counts (Radioactive Index, R.1.). Results obtained were as follows: 1. The radioactive indices obtained from submandibular gland of the saline-ACTH group 1, 10 and 24 hr after $[^3H]$ thymidine administration were $15.2{\pm}0.32,\;20.1{\pm}0.30,\;and\;24.5{\pm}0.52(mean{\pm}S.D.)$ in the mucous cells, $13.0{\pm}0.22,\;10.2{\pm}0.05,\;and\;7.5{\pm}0.42$ in the serous cells. and $10.5{\pm}0.40,\;13.6{\pm}0.32,\;and\;15.9{\pm}0.42$ in the duct cells, while the $mean{\pm}S.D.$ of the values obtained from the 3 cell types 1, 10 and 24 hr after $[^3H]$ thymidine were $10.9{\pm}0.28,\;12.4{\pm}0.31,\;and\;10.0{\pm}0.39.$ Thus the radioactive indices obtained from the ginseng-ACTH group were generally lower than those obtained from the saline-ACTH group. It is inferred from the above results that the ginseng tends to promote the suppressive action of ACTH upon DNA synthesis of cells in the mandibular gland.

Effect of Chamaecyparis obtusa tree Phytoncide on Candida albicans (편백 피톤치드가 Candida albicans에 미치는 영향에 대한 연구)

  • Phytoncide, essential oil of trees, has microbicidal, insecticidal, acaricidal, and deodorizing effect. The present study was performed to examine the effect of phytoncide on Candida albicans, which is a commensal colonizer of the mucous membranes but has become an opportunistic pathogen. C. albicans was incubated with or without phytoncide extracted from Hinoki (Chamaecyparis obtusa Sieb. et Zucc.; Japanese cypress) and then changes were observed in its optical density, cell viability and morphology. As concentrations of phytoncide added to the culture medium increased, optical density and cell viability of C. albicans decreased. Minimum inhibitory concentration of phytoncide for C. albicans was observed to be 0.25%, and minimum fungicidal concentration was 0.5%. Numbers of morphologically atypical cells with electron-dense cytoplasm and granules and increased with increasing concentration of the phytoncide. At higher concentrations of phytoncide, compartments and organelles in the cytoplasm became indistinguishable. The overall results indicate that the phytoncide used for this study has a strong antimicrobial activity against C. albicans. Therefore, the phytoncide may be used as a candidate for prevention and therapeutic agent against oral candidiasis.

Ultrastructure of the Mantle Epidermis in the Ark Shell, Scapharca broughtonii (Bivalvia: Acridae) (피조개, Scapharca broughtonii 외투막 상피층의 미세구조)

  • Histology and ultrastructure of the mantle epidermis in the ark shell, Scapharca broughtonii are described using light and electron microscopy. The mantle of the ark shell is composed of outer epidermis, connective tissue and inner epidermis. Both epidermis are simple and consists of supporting cells, ciliated cells and secretory cells. Connective tissue is composed of mainly collagen and muscle fibers. The supporting cells in the inner epidermis are usually columnar and covered with microvilli. The ciliated cell have cilia and microvilli on the free surface, and numerous tubular mitochondria are observed in the apical cytoplasm. Secretory cells are mainly observed in the outer epidermis, and it can be divided into four types of A, B, C and D with morphological features of the secretory granules. Type A cells of mucous cell are found in the marginal and central mantle. And these cells contains numerous secretory granules of non-bounded and low electron density. Type B cells contains numerous rough endoplasmic reticula, well-developed Golgi complex and secretory granules of membrane-bounded and high electron density. Secretory granules of type C cells are divided into fibrous core layer and homogeneous peripheral layer. Type D cells are found in the outer epidermis of the central and umbonal mantle. And secretory granules of these cells are divided into homogeneous core layer and granular peripheral layer. This results suggest that the outer and inner epidermis of the mantle are related with shell formation and cleaning of the mantle cavity, respectively.

Mantle Ultrastructure of the Spiny Top Shell, Batillus cornutus (Gastropoda: Turbinidae) (소라(Batillus cornutus) 외투막의 미세구조)

  • The histochemical characteristics and ultrastructure of the mantle in the spiny top shell, Batillus cornutus were described using light and electron microscopy. The simple epidermal layer wrapped on the top and bottom of the centrally located connective tissue. And then the epidermal layer were divided into the outer epidermal layer near a shell and the inner epidermal layer closed to the visceral mass. The connective tissue layer was composed of the collagen fiber muscularfiber bundle and hemolymph sinus. Mucous cells in the apical mantle contained acid and neutral mucopolysaccaride, and acidic carboxylated mucopolysaccaride in the mid and marginal mantle. The mantle thickness, epidermal layer thickness and hemolymph sinus area displayed a trend of reduction from the marginal zone to the apical zone. From TEM observation, it was possible to distinguish epithelium, ciliated cell, absorptive cell and secretory cell in the epidermal layer. The epithelia were columnar and the nucleus was elliptical. The free surface were covered with microvilli. The lateral membranes of epithelium was con nected with neighboring cells by the zonular occludens, zonular adherens and membrane interdigitation. Ciliated cell on free surface had cilia and microvilli, and numerous mitochondria in the apical cytoplasm. In the epidermal layer, it observed 2 type cells having absorptive function. The absorptive cells were columnar in shape, and contained microvilli, pinocytotic vesicles, mitochondria and lysosomes of various electron density. Secretory cells can be divided into four types (A, B, C, D) depending on the cell shape and characteristics of secretory granules. These cells were unicellular glands and had similar characteristics to previously reported on the mantle of the gastropod and bivalves.

Effects of Dansambohyultang on the Gastrointestinal Mucosa of Rats Stressed by Heating, Immobilization and Starvation (단삼보혈탕(丹蔘補血湯)이 Stress로 유발(誘發)된 백단삼보혈탕(白丹蔘補血湯)이 Stress로 유발(誘發)된 백서(白鼠) 위장관(胃腸管)의 조직손상(組織損傷)에 미치는 영향(影響))

  • Objectives : This is the experimental paper to study the curative and preventive effects of Dansambohyultang, Methods : Oriental herb medicine for gastro-intestinal disease caused by stress, on the rats put under the stress through starvation, heating and immoblization, the author used four different group; the normal group, which was not put under stress and well fed, and the control group, to which the drug was not administered and put under the stress through starvation, heating and immoblization, the Sample I, to which the drug was administered before they were put under stress, the Sample lI, to which the drug was administered after they were put under stress. Results : When a Masson's trichrome stain method was applied to the control group, a small size of ulcer was found in the fundus and the pylorus, and atrophy was observed in the neck region of mucous membrane. When the drug was administered to the Sample I and the Sample II, the former recovered from the ulcer and atrophy to almost normal and showed better results than the latter. When a Masson's trichrome stain method was applied to the control group, atrophy of mucous membrane was found all over the intestines. When the drug was administered to the Sample I and the Sample ll, the curative and recovered effects was seen in both, but the former showed better results than the latter. After application of Mayer's hematoxylin stain method, the observation of numerical changes of goblet cells in the small intestines showed the most significant decrease in the control group. In the Sample I, the number of the cells in the duodenum, the jejunum and the ileum returned to an almost normal level. In the Sample II, a slight increase was observed only in the ileum. After application of Mayer's hematoxylin stain method, the observation of numerical changes of goblet cell in the large intestines showed a significant decreased in the control group. Both the Sample I and the Sample II showed an increase in the number of goblet cells in comparison with the control group. When a serotonin-immunoreaction test was applied, the control group showed a general decrease in the number of gastro-entero-endocrine cells, The Sample I showed the effect in all over gastro.intestinal tract as compared with control group and the Sample II showed the effect in all over gastro-intestinal tract except the duodenum and the former showed better results than the latter. Conclusion : According to the above results, the regions which are most sensitive to stress were observed to the ileum and the colon. It was concluded that Dansambohyultang has a remedical value particularly on the abnormalities caused by stress in all over gastro-intestinal tract and it was found to be more effective in the Sample I than in the Sample ll. Judging from the fact that the Sample I showed better results than the Sample ll, Dansambohyultang can be said to have a preventive effect more than a curative effect on gastro-intestinal tract disease.
