Specially shaped concrete blocks are used for the armor layer of rubble structure for breakers, seawalls, or other shore protection work. In this study, the hydraulic characteristics of the Modified-Tribar(MTB), which addresses the shortcomings of the Arch-Tribar, and the most widely used Tetrapod(TTP) in Korea are examined through hydraulic model tests. The MTB are much more stable than the TTP, as shown through the stability model tests under non-breaking and non-overtopping condition. The value of the stability coefficient(KD) was obtained at around 30. The model tests show that the TTP random two layers and MTB uniform 1.5 layers have similar effects, but the MTB one layer shows slightly low effects in dissipating wave energy. The TTP random two layer model is the most effective in reducing wave overtopping rate, under overtopping condition, while the MTB uniform one layer and the MTB uniform 1.5 layer models follow respectively.
Spectrophotometric Determination of Rare Earths by Ternary CompleMTB와 희토류 원소들 간에 이루어진 착물에 cetylpyridinium chloride(CPC), dodecyltrimethylammonium bromide(DTMAB), cetyltrimethylammonium bromide (CTMAB), Triton X-100 등의 계면활성제를 가하였을 때의 흡광도 변화로부터 이들 원소를 정량하는 방법과 조성비를 연구하였다. 양이온 계면활성제들인 CPC, DTMAB, CTMAB 등이 있을 때 MTB와 희토류 원소들간의 흡광도가 계면활성제들이 없을 때보다 더 컸다. 가장 큰 흡광도 증가는 CPC에서 얻어졌으므로 희토류 정량에는 이를 선택하였다. REE-MTB-CPC 삼성분 착물은 650nm에서 최대흡광도를 갖고 0~100 ng/ml 범위에서 Beer 법칙에 따르고 몰흡광계수는 $6.6{\sim}9.4{\times}10^4\;mol^{-1}l\;cm^{-1}$이다.
Tuberculosis (TB) remains an unsolved community health problem since identification of its causing microorganism called Mycobacterium tuberculosis (MTB) by Robert Koch in 1882. Annually, eight million TB cases are newly reported and 2~3 million patients die from TB. Pulmonary TB is highly infectious and untreated pulmonary TB patients are believed to infect >10 people in a year. The conventional methods for diagnosis of TB are chest X-ray and isolation of the causing microorganisms from patient specimens. Screening of TB is conducted with smeared sputum in slides, and TB is confirmed by identification of MTB in cultured specimens. One of the fatal pitfalls of screening detection for smeared sputum is that it is impossible to distinguish MTB and other acid-fast bacilli (AFB) because they are stained equally with Ziehl-Neelsen (ZN) stain. Culture of MTB is the most reliable method for diagnosis of TB but it takes 4~8 weeks. In this report, we suggest a fast and highly-reliable MTB detection method that distinguishes AFB in sputum samples. Purified DNA from the AFB stained slide samples offered by The Korean Institute of Tuberculosis were used to detect infected MTB in patients. PCR, real-time PCR and reverse blot hybridization assay (REBA) methods were applied to purified DNA. Conclusively, the real-time PCR method was confirmed to produce high sensitivity and we were able to further detect drug-resistant MTB with REBA.

Background:N-acetyl transferase (NAT) inactivates the pro-drug isoniazid (INH) to N-acetyl INH through a process of acetylation, and confers low-level resistance to INH in Mycobacterium tuberculosis (MTB). Similar to NAT of MTB, NAT2 in humans performs the same function of acetylation. Rapid acetylators, may not respond to INH treatment efficiently, and could be a potential risk factor, for the development of INH resistance in humans. Methods: To understand the contribution of NAT of MTB and NAT2 of humans in developing INH resistance using in silico approaches, in this study, the wild type (WT) and mutant (MT)-NATs of MTB, and humans, were modeled and docked, with substrates and product (acetyl CoA, INH, and acetyl INH). The MT models were built, using templates 4BGF of MTB, and 2PFR of humans. Results: On the basis of docking results of MTB-NAT, it can be suggested that in comparison to the WT, binding affinity of MT-G207R, was found to be lower with acetyl CoA, and higher with acetyl-INH and INH. In case of MT-NAT2 from humans, the pattern of score with respect to acetyl CoA and acetyl-INH, was similar to MT-NAT of MTB, but revealed a decrease in INH score. Conclusion: In MTB, MT-NAT revealed high affinity towards acetyl-INH, which can be interpreted as increased formation of acetyl-INH, and therefore, may lead to INH resistance through inactivation of INH. Similarly, in MT-NAT2 (rapid acetylators), acetylation occurs rapidly, serving as a possible risk factor for developing INH resistance in humans.

We aimed to investigate the proportions of MTB- and NTM-positive tests and the distribution patterns of species isolated by contracted testing agencies in South Korea. Respiratory specimens submitted to contracted testing agencies in South Korea for AFB culture from January 2014 to December 2021 were included (533,713 specimens in total). Trends based on MTB and NTM detection, patient sex and age, culture medium type, and testing year were analyzed. MTB and NTM positive detection increased in the patients. The average ages of MTB- and NTM-positive patients increased in those aged ≥61 years. For solid culture, the MTB detection rate decreased from 5.9% in 2014 to 3.3% in 2018 and increased to 4.7% in 2021; the NTM detection rate increased from 2.1% in 2014 to 3.4% in 2018 and 3.7% in 2021. For liquid culture, the MTB detection rate decreased from 8.3% in 2014 to 5.5% in 2018 and increased to 6.0% in 2021; the NTM detection rate increased from 3.5% in 2014 to 5.5% in 2018 and decreased to 5.3% in 2021. An isolation ratio reversal between MTB and NTM was observed in 2018. In this study, we provide information on mycobacterial isolation rates and species distributions using AFB culture test results from Korea's referral laboratories. Increased MTB- and NTM-isolation rates were observed in individuals aged ≥60 years, indicating the need for regular testing and focused management for them. Expanding liquid culture applications, which show higher positivity rates than solid culture methods, is necessary.
연구배경 : IL-8은 강력한 화학주성인자로서 결핵감염 부위로 염증세포들을 동원함으로서 결핵균에 대한 숙주의 방어기전에 있어서 중요한 역할을 한다. IL-8의 유전자의 발현에 있어서 NF-${\kappa}B$가 중요한 역할을 한다. 저자들은 결핵 감염시 폐상피세포가 NF-${\kappa}B$ 의존성으로 IL-8을 분비하는지 알아보고자 하였다. 방 법 : 말초혈액단핵구에 결핵균을 감염시키고 24시간 배양 후 배양상층액(CoMTB)을 얻었다. 결핵균, CoMTB로 자극한 A549 세포주의 IL-8 분비 정도를 ELISA 방법으로 측정하였다. CoMTB로 자극한 A549 세포주의 IL-8 mRNA 의 발현 정도를 RT-PCR로, $I{\kappa}B{\alpha}$의 분해를 western blot 분석으로, NF-${\kappa}B$의 핵이동과 DNA 결합은 electrophoretic mobility shift assay(EMSA)를 이용하여, 그리고 NF-${\kappa}B$ 의존성 IL-8 유전자의 전사활성은 luciferase reporter gene assay를 이용하여 측정하였다. 결 과 : A549 세포주를 CoMTB로 24시간 자극하여 얻은 배양액의 IL-8 농도는 $46.8{\pm}4.8\;ng/ml$로 분비하여 결핵균으로 직접 자극하였을 때의 $6.8{\pm}2.9\;ng/ml$보다 높았다. CoMTB로 A549 세포주를 자극하였을 때 IL-8 mRNA의 발현이 증가하였고, $I{\kappa}B{\alpha}$의 분해가 일어났으며, NF-${\kappa}B$의 핵이동과 DNA 결합이 일어났으며, NF-${\kappa}B$ 의존성 IL-8 유전자의 전사활성이 증가하였다. 결 론 : 결핵병변에서 폐상피세포는 결핵균을 탐식한 단핵식 세포와의 상호작용에 의해 NF-${\kappa}B$ 의존성으로 IL-8을 분비한다.
Tuberculosis (TB), mainly disseminated by infection of the respiratory tract, remains an unsolved community health problem by Mycobacterium tuberculosis (MTB). However, because of the different susceptibility to MTB, people infected with MTB do not all develop TB. These differences of disease arise from individual genetic susceptibility as well as the property of the microorganisms itself. CD226, one of the genetic factors that influences TB, interact with its ligand PVR and ITGB2. It is induced various cellular responses that contribute multiple innate and adaptive responses. In a previous study, CD226 enhanced immune efficacy induced by Ag85A DNA vaccination that is secreted protein by MTB. The aim of this study was to investigate the association between six genetic polymorphisms of CD226 gene and TB status with Korean population. Our results show that two SNPs of CD226 were identified to associate with tuberculosis. The highest significant SNP was rs17081766 (OR=0.70, CI: 0.54~0.90, $P=5.4{\times}10^{-3}$). According to this study, polymorphisms of CD226 gene affect the outbreak of TB in MTB-infected patients. It is suggested that polymorphism of other genes also associated with immune responses results in susceptibility to TB. The results from this study suggest that not only the characteristics of the microorganism itself but also the genetic background of the individual may affect progression of TB in MTB-infected patients.
Purpose: This study sought to explore the effects of sport enjoyment on leisure commitment. Research design, data, and methodology: Subjects of this study are members of MTB participation in Seoul and Gyeonggi area and were selected by purposive sampling out of non-probability sampling. 295 questionnaires were collected and 288 questionnaires were used for the study. To analyze data, frequency analysis, factorial analysis, t-test, ANOVA, correlation and regression analysis were employed. Results: First, as for leisure satisfaction and leisure commitment pursuant, there was significant difference in gender, age, the number of participation in weekly leisure, and hours of participation in weekly leisure. Second, regarding MTB participants' amusement and leisure satisfaction competence had significant influence on all the sub-factors of leisure satisfaction and that both achievement and competence had impacts on social leisure satisfaction. Third, regarding effects on leisure commitment, competence of amusement had impacts on cognitive and behavioral leisure commitment. Fourth, regarding relationships between leisure satisfaction and leisure commitment, satisfaction of mental leisure, social leisure, relaxation leisure, physiological leisure and leisure satisfaction had significant influence on cognitive commitment. Conclusions: Summary of above results showed that MTB participants' amusement factor had significant influence on leisure satisfaction and leisure commitment. Implications were discussed. The tuberculin skin test (TST) and interferon gamma (IFN-${\gamma}$) release assay (IGRA) have been widely used for diagnosis of latent tuberculosis infection (LTBI). In order to overcome limitations of current LTBI diagnostic methods, the development of a novel molecular assay which is able to measure the IFN-${\gamma}$ messenger RNA (mRNA) expression level after stimulation with Mycobacterium tuberculosis (MTB) specific antigen was recently developed. The ability of a molecular assay to detect MTB infection was similar to commercial IGRA however, the optimal incubation time for stimulating IFN-${\gamma}$ was not yet established. Therefore, in this study the direct comparisons of MTB Ag stimulation times (4 and 24 hrs) were performed for diagnosis of MTB infection. Data showed that the coincident rate between QFT-GIT IFN-${\gamma}$ ELISA and IFN-${\gamma}$ RT-PCR (4 hrs) was 88.35% and that of QFT-GIT and IFN-${\gamma}$ RT-PCR (24 hrs) was 70.85%. Based on a receiver operating characteristic (ROC) curve, the 4 hrs-MTB specific Ag stimulation time for IFN-${\gamma}$ RT-PCR had the significant P value, 95% CI value, and AUC (P < 0.0001, 95% CI=0.82 to 1.02, and AUC=0.9214) in comparison with 24 hrs-MTB specific Ag stimulation time (P = 0.009, 95% CI=0.06 to 0.94, and AUC=0.7711). These results show that 4-hr was the most optimal MTB Ag stimulation time for performing IFN-${\gamma}$ RT-PCR. Although semi-quantitative RT-PCR had a few analytical limitations, it might be useful as an alternative molecular diagnostic method for detecting MTB infection.
Background: The purpose of this study was to evaluate recently developed real-time polymerase chain reaction (PCR) assay kit to detect Mycobacterium tuberculosis (MTB) and nontuberculous mycobacteria (NTM) in respiratory specimens. Methods: We assessed the positive rate of the real-time PCR assay to detect MTB and NTM in 87 culture-positive specimens (37 sputum, 50 bronchial washing), which were performed real-time PCR by using $Real-Q_{TM}$ MTB&NTM Kit from January 2009 to June 2009, at Gyeongsang University Hospital. To compare the efficacy with the TB-PCR assay, we evaluated 63 culture-positive specimens (19 sputum, 44 bronchial washing) for MTB or NTM, which were performed TB-PCR by using ABSOLUTETM MTB II PCR Kit from March 2008 to August 2008. Results: Among 87 specimens tested using real-time PCR, MTB and NTM were cultured in 58 and 29, respectively. The positive rate of real-time PCR assay to detect MTB was 71% (22/31) and 92.6% (25/27) in AFB stain-negative and stain-positive specimens. For NTM, the positive rate of real-time PCR was 11.1% (2/18) and 72.7% (8/11) in AFB stain-negative and stain-positive specimens. Among 63 specimens performed using TB-PCR, MTB and NTM were cultured in 46 and 17, respectively. The positive rate of TB-PCR was 61.7% (21/34) and 100% (12/12) in AFB stain-negative and stain-positive specimens. TB-PCR was negative in all NTM-cultured 17 specimens. Conclusion: TB/NTM real-time PCR assay is useful to differentiate MTB and NTM in AFB stain-positive respiratory specimens and it is as effective in detecting MTB with TB-PCR.
