In this paper, we fabricated and evaluated polydimethylsiloxane(PDMS)-based flexible and implantable micro electrodes. The electrode patterning was carried out with the photolithography and chemical etching process after e-beam evaporation of 100 ATi and 1000 A Au. The PDMS substrate was treated by oxygen plasma using reactive ion etching(RIE) system to improve the adhesiveness of PDMS and metal layers. The minimum line width of fabricated micro electrode was 20 $\mu$m. After finished patterning, we did packaging with PDMS and then brought up the electrode's part about 40 $\mu$m with gold electroplating. The Hank's balanced salt solution(HBSS) test was carried out for 6 month for endurance of fabricated micro electrode. We carried out in-vivo test for the evaluation of biocompatibility by implanting electrodes under the ICR mouse skin for 42 days.
Anti-allergic action of each water extracts of some crude drugs was investigated in mice and rats. The activity of hyaluronidase which was used in the screening test of anti-allergic action was inhibited significantly by Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Cinnamomi Ramulus, Glycyrrhizae Radix and Scutellariae Radix. The 48-hour homologous passive cutaneous anaphylaxis(48-hr PCA) in mouse ear was inhibited significantly by intraperitoneal(i.p.) injection of Amomi Semen, Cimicifugae Rhizoma, and ketotifen, a comparative drug of an anti-allergic action. The increase of vascular permeability induced by histamine or serotonin was inhibited significantly by i.p. injection of Amomi Semen, Cimicifugae Rhizoma, Cinnamomi Ramulus and ketotifen. In rat dorsal skin, the increase of vascular permeability which was induced by histamine, serotonin or compound 48/80 was inhibited significantly by i.p. injection of Amomi Semen, Asiasari Radix, Cimicifugae Rhizoma, Scutellariae Radix and ketotifen. Armeniacae Semen and Liriopis Tuber which had not inhibited hyaluronidase activity did not inhibit 48-hr PCA and the increase of histamine, serotonin or compound 48/80-induced vascular permeability in mice and rats. These results suggest that each water extract of Amomi Semen and Cimicifugae Rhizoma has anti-allergic action.

Atopic dermatitis is an allergic inflammatory skin disease caused by aberrant and overreactive immune responses including overactivation of $T_H2$ immune responses, high levels of IgE as well as proinflammatory cytokines and chemokines. We examined whether BHOSB, a traditional herbal medicine, has modulatory effects on various immunological factors related to pathogenesis of atopic dermatitis in ONCB treated NC/Nga mice. Oral administration of BHOSB at the concentration of 10.8 mg/mouse/day significantly inhibited the production of IgE compared with control, and the levels of IgG2a and IgG2b, but not IgG1, were also significantly reduced. Production of IL-6 and TNF-a was greatly decreased. The results from flowcytometry of peripheral blood mononuclear cells indicated that the percentages of C03+C069+ cells and C04+ were significantly decreased by BHOSB. Theses results suggested that BHOSB has suppressive effects on aberrant and overreactive immunological activities in ONCB-induced dermatitis mice of NC/Nga.

Lidocaine(2-Diethylaminoaceto-2', 6'-xylidide) was transdennally delivered by iontophoresis and the effect of enhancer on the delivery of lidocaine was studied. We delivered lidocaine through the skin of hairless mouse using diffusion cell and investigated the effect of the amount of cation salts such as sodium chloride, calcium acetate, zinc acetate and aluminum acetate on the drug delivery. The amounts of transported drugs and adsorbed metal ions were measured by HPLC(High Perfonnance Liquid Chromatography) and AAS(Atomic Absorption Spectrophotometry), respectively. The addition of zinc acetate and aluminum acetate greatly enhanced the delivery of lidocaine. The detection of two metal ions by AAS seemed to support the idea that the astringency effect of these ions were the main reason for the enhancement of transdermal delivery.
Atopic dermatitis (AD) is a common chronic inflammatory disease of preceding the development of allergic disorders. The incidence of AD is increasing and it causes problems with administrative costs. Therefore, no side effects, easyto- use development of AD treatment is needed. The aim of this study was to evaluate whether PBMCAT, a functional extract from herbal complex was effective to treat the AD mice. AD was induced by patching ovoalbumin on the backside in BALB/ c mouse and then mice were topically treated with PBMCAT. Elidel $cream^{(R)}$ (pimecrolimus, PL) was used as a control. Scratching counts (SC) and clinical skin severity (CSS) were measured, and total serum IgE level was also measured. After inducing AD, SC and CSS were increased. The total serum IgE level was also increased in AD-induced mice. Treatment with PBMCAT significantly decreased SC, CSS, and serum IgE concentration in mice. Especially, treatment of PBMCAT 0.1% in BALB/c mice more effected than PL. These results suggest that the ointment of PBMCAT may enhance the process of AD healing by influencing phase of allergic reacting.
This study was carried out to investigate the effect of Origanum vulgare extracts on cell proliferation of human hair dermal papilla cell (HHDPC) using sulforhodamine B (SRB) assay, antioxidant activity by 1,1-diphenyl-2-picryl hydrazyl (DPPH) method, expression of insulin-like growth factor-1 (IGF-1) by analyzing reverse transcriptase polymerase chain reaction (RT-PCR) and hair growth in a shaving animal model of C57BL/6 mice topically applying with an amount of 0.1 mL once a day for 3 weeks. The mice were divided into 4 groups including normal group (saline, N), negative control group (dimethyl sulfoxide, NC), positive control group (5 mg/mL minoxidil, PC), and experimental group (Origanum vulgare extracts, OV). Treatment of OV didn't show cytotoxicity in HHDPC up to 10 ${\mu}g/mL$ and exhibited antioxidant activity with $IC_{50}$ of 31.0 ${\mu}g/mL$. IGF-1 expression in the skin was significantly (p<0.05) increased in the PC and OV compared to the N or NC. PC and OV also showed a prominently promoted hair regrowth compared to the N or NC in hair growth observation. The hair regrowth of OV was significantly higher than that of PC (p<0.05). Therefore, these results indicate that O. vulgare extracts effectively stimulated hair growth in an animal model.

This study was performed to investigate the effect of ultrasound irradiation on epidermal growth factor(EGF) expression in rat wound tissue. Skin wounds were created below 5mm to both sides of scapular inferior angle. The right wound was used experimental side and left was used control side. Ultrasound was irradiated pulse rate $20\%$, frequency 1MHz, intensity $0.5W/cm^2$ for 5 minutes during 3days. After sonication during 3 days, rats were sacrified. The expression of epidermal growth factor evaluated immunohistochemistry on mouse anti-EGF. In the control side, a little expression of EGF was observed at epidermis and dermis. In the experimental side, A strong immunostaining was seen at epidermis and dermis. This study suggests that ultrasound irradiation is effective on EGF expression in wound tissue.

Forsythiae fructus has traditionally been used for the treatment of erysipelas, skin rash and acute or chronic inflammatory disorders. The effect of Forsythiae fructus against lipopolysaccharide-induced inflammation was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcription-polymerase chain reaction (RT-PCR), on mouse RAW 264.7 macrophages. Forsythiae fructus extract suppressed the expression of iNOS, COX-2 and NF-$_K$B mRNAs on the lipopolysaccharide-stimulated enhancement in RAW 264.7 macrophages. We examined the expression of iNOS and COX-2 in both mRNA and protein levels to investigate the mechanism by which Forsythiae fructus extract inhibits NO production. Forsythiae fructus extract significantly reduced iNOS, NF-$_K$B and PGE$_2$, but didn't inhibit COX-2 expression which was induced by LPS treatment in Raw 264.7 cells. These results suggest that Forsythiae fructus exerts anti-inflammatory effects probably by suppression of the iNOS and NF-$_K$B expressions.
The effect of DHU001, a mixed herbal formula consisted of 7 types aqueous extracts for various respiratory disorders were evaluated on 2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis, type I allergic model. Contact dermatitis was induced by sensitization with dinitrophenyl-derivatized ovalbumin (DNP-OVA) and DNFB challenge as antigen. Two different dosages of DHU001 (300 and 150 mg/kg) were orally administered to DNP-OVA sensitization mice once a day for 7 days with reference material, dexamethasone (15 mg/kg, intraperitoneal treatment). End of 7 days oral administration of DHU001 extracts or intraperitoneal treatment of dexamethasone, the changes on the edematous changes and scratching behavior were measured. Immediate after DNFB challenge on ear or paw of DNP-OVA sensitized mice, increases of ear and paw thicknesses and weights were detected with anterior ear skin (dermis to epidermis) thickness and paw scratching behavior increases. However, these contact dermatitis signs induced by DNFB treatment were reduced by treatment of the both different dosages of DHU001 and dexamethasone, respectively. The results obtained in this study suggest that oral treatment of DHU001 extracts also has relatively favorable effects on contact dermatitis.

Vaccinium uliginosum L. (VU) is a flowering plant in the genus Vaccinium has berry fruit. This study was performed to investigate the effect of water extract of Vaccinium uliginosum L. and fractions on inhibition of melanogenesis and wrinkle formation. One hundred grams of the Vaccinium uliginosum L. was extracted with 2,000 mL of water ($90^{\circ}C$, 16h, 2 times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Vaccinium uliginosum L. extracts showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical, dose dependently. And VU extract and fractions reduced melanin contents on B16F10 melanoma and inhibited the expression of melanogenesis-related proteins, tyrosinase, tyrosinase-related protein (TRP-1) and dapachrometa utomerase (Dct, TRP-2). Moreover VU extract and fractions stimulated procollagen production and inhibited MMP-1 production in human fibroblast. And it decreased degree of wrinkle formation in hairless mouse skin that induced by DVB irradiation for 9 weeks. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be the health functional food and functional cosmetics that have anti-melanogenesis and anti-wrinkle effect.
