Anti-inflammatory Effects of Enzymatic Extract from Ecklonia cava on TPA-induced Ear Skin Edema

  • Anti-inflammatory potential of the enzymatic extract prepared by Kojizyme (ECK), a component of brown seaweeds Ecklonia cava (Alariaceae, Phaeophyta) in vivo was investigated. For the application of mouse ear edema model, 12-O-tetradecanoylphorbol acetate (TPA) was used, a topical inducer of a long-lasting inflammatory response. Our results demonstrated that ECK inhibited ear edema when topically applied to mouse ear skin. In histological evaluation, the inhibition activity of ECK on TPA-induced inflammation is similar to that of dexamethasone, although less strong. In addition, the mRNA expression levels of IL-$1{\beta}$, IFN-$\gamma$, TNF-$\alpha$, and cyclooxygenase-2 (COX2) and the immunoreactivity to inducible nitric oxide synthase (iNOS) and COX2 expressed mainly in inflammatory cells were down-regulated by ECK. These results indicate that ECK has anti-inflammatory effects through the inhibition of Th1 cytokines and 2 inducers of inflammation in TPA-induced ear skin edema.

Inhibitory Effects of Opuntia humifusa on 7, 12-Dimethyl-benz[a]anthracene and 12-O-tetradecanoylphorbol-13-acetate Induced Two-stage Skin Carcinogenesis

  • Opuntia humifusa, member of the Cactaceae family, was previously demonstrated to have radical scavenging, anti-inflammatory and anti-proliferative effects in in vitro models. It was suggested that O. humifusa could function in the prevention of carcinogenesis. To investigate the in vivo chemopreventive effect of O. humifusa, mice were fed a diet containing either 1% or 3% following 7, 12-dimethylbenz[a] anthracene (DMBA) and 12-O-tetradecanoylphorbol-13-acetate (TPA) induction of skin carcinogenesis. Significant decrease in the numbers of papilloma and epidermal hyperplasia were observed in mice fed with O. humifusa, compared to the control group. O. humifusa also upregulated high total antioxidant capacity and level of phase II detoxifying enzyme such as superoxide dismutase and glutathione S-transferase activity in the skin. Lipid peroxidation activity level was measured in skin cytosol and significantly inhibited in 3% OH fed group compared to the control group. These results suggest that O. humifusa exerts chemopreventive effects on chemical carcinogenesis in mouse skin and that prevention effects are associated with reduction of oxidative stress via the modulation of cutaneous lipid peroxidation, enhancing of total antioxidant capacity especially in phase II detoxifying enzyme system and partial apoptotic influence.

DEVELOPMENT OF PORTABLE NEAR INFRARED SYSTEM FOR HUMAN SKIN MOISTURE

  • In this study, portable near infrared (NIR) system was newly integrated with a photodiode array detector, which has no moving parts and this system has been successfully applied for evaluation of human skin moisture. The good correlation between NIR absorbance and absolute water content of separated hairless mouse skin was, in vitro, showed depending on the water content (7.42-84.94%) using this portable NIR system. Partial least squares (PLS) regression was used for the calibration with the 1100-1650 nm wavelength range. For the practical use for the evaluation of human skin based on moisture, PLS model for human skin moisture was, in vivo, developed using the portable NIR system based on the relative water content values of stratum corneum from the conventional capacitance method. The PLS model showed a good correlation. This study indicated that the portable NIR system could be a powerful tool for human skin moisture, which may be much more stable to environmental conditions such as temperature and humidity, compared to conventional methods. Furthermore, in order to confirm the performance of newly integrated portable NIR system, scanning type conventional NIR spectrometer was used in the same experiments and the results were compared.

The Effect of Oleic Acid and Propylene Glycol on the Electrical Properties of Skin (올레인산 및 프로필렌글리콜이 피부의 전기적 성질에 미치는 영향)

  • The effects of oleic acid, propylene glycol and 5% (w/w) oleic acid in propylene glycol on the electrical properties of hairless mouse skin were studied and the results were compared. The complex electrical impedance was measured as a function of frequency, and resistance and capacitance were determined from the Nyquist plot. Immediately after the treatment with oleic acid, resistance was 145% of the pretreatment value. However it decreased with time and, after 20 hours, it was about 25% of its pretreatment value. Capacitance increased; immediately after the treatment, it was about 125% of pretreatment value and it seemed to increase slowly with time. When the skin was treated with propylene glycol, resistance decreased about 5O% and capacitance increased about 65%. Similar results were observed when the skin was treated with 5% (w/w) oleic acid in propylene glycol, except that the magnitude of resistance drop was much larger. Oleic acid acted synergistically with propylene glycol. Together with the flux data in the literature, the results obtained in this work indicate that electrical resistance is closely related to the permeability of drug molecules through the skin. The results are discussed in terms of the mechanism of action of these penetration enhancers. Overall, this work provided further mechanistic insight into the role of SC lipids in skin resistance and capacitance.

Inhibition of Chronic Skin Inflammation by Topical Anti-inflammatory Flavonoid Preparation, Ato $Formula^{\circledR}$

  • Flavonoids are known as natural anti-inflammatory agents. In this investigation, an anti-inflammatory potential of new topical preparation (SK Ato $Formula^{\circledR}$) containing flavonoid mixtures from Scutellaria baicalensis Georgi roots and Ginkgo biloba L. leaves with an extract of Gentiana scabra Bunge roots was evaluated in an animal model of chronic skin inflammation. Multiple 12-O-tetradecanoylphorbol-13-acetate treatments for 7 consecutive days on ICR mouse ear provoked a chronic type of skin inflammation: dermal edema, epidermal hyperplasia and infiltration of inflammatory cells. When topically applied in this model, this row formulation $(5-20\;{\mu}L/ear/treatment)$ reduced these responses. Furthermore, it inhibited prostaglandin $E_2$ generation (17.1-33.3%) and suppressed the expression of proinflammatory genes, cyclooxygenase-2 and $interleulin-1{\beta}$ in the skin lesion. Although the potency of inhibition was lower than that of prednisolone, all these results suggest that Ato $Formula^{\circledR}$ may be beneficial for treating chronic skin inflammatory disorders such as atopic dermatitis.

  • A novel retinol derivative, polyethoxylated retinamide (Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of [$^3H$]-proline incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells (effective diffusion area: $1, 766{\;}\textrm{cm}^2$) and the excised skin of female hairless mouse aged 8 weeks were used The stabilities of retlnoids were evaluated at two different temperature ($25{\;}^{\circ}C$ and $40{\;}^{\circ}C$) and under UV in solubilized state and in OW emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.

An Eye Location based Head Posture Recognition Method and Its Application in Mouse Operation

  • An eye location based head posture recognition method is proposed in this paper. First, face is detected using skin color method, and eyebrow and eye areas are located based on gray gradient in face. Next, pupil circles are determined using edge detection circle method. Finally, head postures are recognized based on eye location information. The proposed method has high recognition precision and is robust for facial expressions and different head postures, and can be used in mouse operation. The experimental results reveal the validity of proposed method.

  • To clarifiy the activating effects of Buthus martensi Karsch(BMK) on tumor promotion in two-stage carcinogenesis in mice was investigated. In vivo system, BMK was seen to gave an inhibitory activity on TPA-induced mouse ear edema. In addition, the BMK was proved to have antitumor-promoting activity in two-stage mouse skin carcinogenesis induced by DMBA and two-stage mouse lung carcinogenesis induced by 4-NQO as a initiator plus TPA and glycerol as a promoter. Moreover, BMK significantly exhibited an cytolytic effect in HepG2 cells and showed significant antitumor activity against Sarcoma-180 bearing mice by oral administration. These results suggest that BMK could be effective in adjuvant chemotherapy for human cancer.

  • When the inhibitory effect of ginsenoside (G) Re isolated from ginseng and its metabolites G-Rg1, G-F1, G-Rh1 and protopanaxatriol in mouse ear skin psoriasis stimulated by oxazolone was investigated, G-Re and its metabolites suppressed mouse ear swelling stimulated by oxazolone. Among these agents tested, G-Rh1 most potently suppressed ear swelling as well as mRNA expression of COX-2 and proinflammatory cytokines $IL-1{\beta},\;TNF-{\alpha}$ and $interferon-{\gamma}$. These findings suggest that G-Rh1 may improve chronic dermatitis and psoriasis.

  • We have synthesized acetylsalicylic acid derivative of eugenol. And also, physico-chemical properties and analysis on this compound were examined. The correlation coefficient of the calibration curve in methanol solution was 0.9998. The hair growth stimulation of eugenol derivative on the hair of black mouse (C57BL/6), was also carried out using Implant method. When its ethanol solution was administered to the black mouse by route of skin, eugenol derivative promoted the growth of hair.
