산삼의 배양 및 그 응용에 관한 연구

  • Korea mountain ginseng known as oriental miracle drug is an important medicinal plant. The effect of mountain ginseng adventitious roots extract has been described. The valuable root of mountain ginseng contained several kinds of ginsenosides that have been confirmed to have many active functions for the human body. However, the study of mountain ginseng has a limit because the price of wild ginseng is very expensive and rare. The mountain ginseng adventitious roots were derived from mountain ginseng callus that were induced from mountain ginseng roots. Adventitious roots were separated from callus and grown in solid media(Murachige and stoog media). It was cultured in a 20L bioreactor. After culturing for 40days, adventitious roots were harvested. Afterwards the harvested mountain ginseng adventitious roots were dryed and extracted. We examined the effect on melanogenesis of mountain ginseng adventitious roots extrac. Here, we report the inhibitory effect of melanin biosynthesis on the adventitious roots extract of In vitro test. Also, we assessed the safety of adventitious roots extract. In vitro, cytotoxicity of adventitious roots extract was assessed in mouse fibroblast using two method: The neutral red uptake assay and the MTT assay. In vivo, the allergic and irritant were patch tested in 30 patients. Consequently, extract of mountain ginseng adventitious roots have inhibitory effect on melanin biosynnthesis in B-16 melanoma cell test, tyrosinase inhibitory test and DOPA auto-oxidation test. There were decreased 86%(0.5% concentration), 45%(1% concentration) and 61%(1% concentration), respectively

  • Korean ginseng(Panax Ginseng C.A. Meyer) known as a oriental miracle drug is an important medicinal plant. Ginseng has been used for geriatric, tonic, stomachic, and aphrodisiac treatments for thousands years. Also, it is an antibiotic and has therapeutic properties against stress and cancer. Ginseng is widely distributed all over the world. Among them, Korean mountain ginseng has the most valuable effect on pharmaceuticals. The roots of mountain ginseng contained several kinds of ginsenosides that have many active functions for the human body. However, the study of mountain ginseng has a limit because the mountain ginseng is very expensive and rare. So, we artificially cultured mountain ginseng adventitious roots using the bioreactor culture system. We induced callus from original mountain ginseng, directly dug up in mountain and aged about one hundred ten years. Separated adventitious roots were precultured in 500ml conical flasks and then, transferred in 20L bioreactors. The adventitious roots of mountain ginseng were harvested after culturing for 40days, dried and then, extracted with several solvents. In this study, we investigated the whitening effect, anti-wrinkle effect and the safety of tissue cultured adventitious roots extract of mountain ginseng in order to identify the merit as a cosmetic ingredient. Particularly, extract of mountain ginseng adventitious roots showed whitening and anti-wrinkle effects. The inhibitory effect of this extract on the melanogenesis was examined using B-16 melanoma cell. When B-16 melanoma cells were cultured with adventitious root extract, there was a dramatically decrease in melanin contents of 8-16 melanoma cell. And we identified this extract inhibited Dopa auto-oxidation significantly. Also, when transformed mouse fibroblast L929 cells were treated with this extract, there was a significant increase in collagen synthesis. The results show significant inhibited melanization and wrinkle without inhibiting cell viability.

Flow Regimes of Continuously Stratified Flow over a Double Mountain (두 개의 산악 위에서의 연속적으로 성층화된 흐름의 흐름 체계)

  • The flow regimes of continuously stratified flow over a double mountain and the effects of a double mountain on wave breaking, upstream blocking, and severe downslope windstorms are investigated using a mesoscale numerical model (ARPS). According to the occurrence or non-occurrence of wave breaking and upstream blocking, three different flow regimes are identified over a double mountain. Higher critical Froude numbers are required for wave breaking and upstream blocking initiation for a double mountain than for an isolated mountain. This means that the nonlinearity and blocking effect for a double mountain is larger than that for an isolated mountain. As the separation distance between two mountains decreases, the degree of flow nonlinearity increases, while the blocking effect decreases. A rapid increase of the surface horizontal velocity downwind of each mountain near the critical mountain height for wave breaking initiation indicates that severe downslope windstorms are enhanced by wave breaking. For the flow with wave breaking, the numerically calculated surface drag is much larger than theoretically calculated one because the region with the maximum negative perturbation pressure moves from the top to the downwind slope of each mountain as the internal jump propagating downwind occurs.

A Numerical Case Study Examining the Orographic Effect of the Northern Mountain Complex on Snowfall Distribution over the Yeongdong Region (북한 지역의 산맥군이 영동 지역의 겨울철 강설 분포에 미치는 영향에 관한 수치 연구)

  • Numerical experiments using the Weather Research and Forecasting (WRF) model were done to identify the role of the mountain ranges in the northern part of the Peninsula (referred as "the northern mountain complex"), in the occurrence of two heavy snowfall events over the Yeongdong region on 7-8 December 2002 and 20-21 January 2008. To this end, control simulations with the topography of the northern mountain complex and other simulations without the topography of the mountain complex were performed. It was revealed that the amount of snowfall over the Yeongdong region from the control simulation much more exceeded that of the simulation without the topography of the mountain complex. This increase of the snowfall amount over the Yeongdong region can be explained as follows: As the upstream flow approached the northern mountain complex, it deflected around the northern mountain complex due to the blocking effect of the mountains with a low Froude number less than ~0.16. This lead to the strengthening of northeasterly over the East Sea and over the Yeongdong region. The strong northeasterly is accompanied with much more snowfall over the Yeongdong region by intensifying air-mass modification over the sea and the orographic effect of the Taeback mountains. Thus, it was concluded that the topography of the northern mountain complex is one of the main factors in determining the distribution and amount of precipitation in the Yeongdong region when there is an expansion of the Siberian High toward the East Sea.

The Effects of Mountain Climbing Motivation and Mountain Climbing Wear Benefits Sought on Clothing Usage Behavior (등산동기가 등산복 추구혜택 및 등산복 착용행동에 미치는 영향)

  • This study investigated 1) the effects of mountain climbing motivation on the desired mountain climbing wear benefits, and 2) the desired effects of mountain climbing wear benefits on clothing usage behavior. Data were collected via a questionnaire administered to 498 consumers who make at least one mountain climbing wear purchase for personal use. All items were evaluated on a 5-point Likert scale, and SPSS 20.0 and Amos 7.0 were used for data analysis. Confirmatory factor analysis for the research variables were performed and three factors of mountain climbing motivation (social interaction, health improvement, and mental rejuvenation), four factors of mountain climbing wear benefits sought (ostentation, ideal healthy figure, conformity, and functions), and two factors regarding clothing usage behavior (sportswear usage and daily wear usage) were identified. The results from the model test were as follows: 1) Mountain climbing motivation was found to vary depending on the mountain climbing wear benefits sought. Social interaction had a positive effect on all 4 factors of mountain climbing wear benefits sought. Health improvement was the only motivating factor for consumers who sought the benefit of functions; however, the functions benefit was not found to be a consideration for consumers who climbed for mental rejuvenation. 2) Mountain climbing wear usage was affected by mountain climbing wear benefits sought. The benefits for conformity and functions had a positive effect on sportswear usage and daily wear usage; however, the ideal healthy figure only affected daily wear usage. The twofold usage of mountain climbing wear reflects a cultural trend toward the usage of mountain climbing wear as daily wear; however, consumers show this twofold usage to conform successfully to mainstream society and not to pursue the ideal healthy figure.

A Study on the Effect of Mountain Ginseng Adventitious Roots Extract (산삼부정근 추출물의 효능${\cdot}$효과에 관한 연구)

  • This study reviewed the application of an extract from mountain ginseng adventitious roots which had been grown through tissue culture as a cosmetic ingredient. The mountain ginseng adventitious roots were derived from mountain ginseng callus that was induced from mountain ginseng root whose origin is estimated to date back about one hundred years ago. The adventitious roots were separated from callus and grown in a 20 L bioreactor. In order to proliferate the adventitious roots, they were cultured for 5 weeks in bioreactor. Then the harvested mountain ginseng adventitious roots were dried and extracted. For verifying skin whitening effect of an extract from the tissue-cultured mountain ginseng adventitious roots in vivo, we performed the clinical test of it. The research showed the significant skin whitening effect of a mountain ginseng adventitious roots extract and the statistical analysis showed a significant difference (p<0.0001) between sample ($2\%$ mountain ginseng adventitious roots extract) and placebo. But, some saponins showed below $10\%$ inhibitory effect of tyrosinase and melanin synthesis in B-16 melanoma. The extracts of red ginseng and ginseng which were the same concentration as the tissue-cultured mountain ginseng adventitious roots extract's showed little inhibitory effect of tyrosinase and melanin synthesis in B-16 melanoma. In DPPH test, Anti-hydroxyl radical activity of $0.5\%$ the tissue-cultured mountain ginseng adventitious roots extract was $86\%.$.

Effect of pulse-wave factors in Middle Aged Women by Mountain Cultivated Ginseng Pharmacopuncture Original Articles (산삼 약침이 정상 중년여성의 맥파요인에 미치는 영향)

  • Objectives: The aim of this experiment is to know about Effect of pulse-wave factors in Middle Aged Women by Mountain Cultivated Ginseng Pharmacopuncture. Methods: First 20 Middle Aged women are diagnosed by pulse diagnosis, and then Mountain Cultivated Ginseng Pharmacopuncture(1 injection 20cc) were injected. 30 minutes later, pulse diagnosis again performed. As a result, method of one-group pretest-posttes design were used for evaluation. Results: T(Total pulse cycle) time statistically significant increased on both left and right chon, kwan, cheok. T4 time statistically significant increased on both left and right chon, kwan, cheok. T4-T1/T indexs except left cheokmaek, Right cheokmaek observation area decreased significantly in four sites. Wm(indicating high pressure retention time) indexs increased significantly in the five sites were observed except right chon maek. Conclusions: Effect of pulse-wave factors in Middle Aged Women by Mountain Cultivated Ginseng Pharmacopuncture increased T, T4, Wm and decreased T4-T1/T indexs. The results of this experiment, Mountain Cultivated Ginseng Pharmacopuncture induced to increase the Pulse-wave's stability and strength.

  • This is a research with two purposes, in order to understand the situation with which mountain villages region gets faced present time. First, to examine the conception and the formative process of mountain villages region. Second, the measure the possibility of development in a project on mountain villages region to increase the evenings as a continuous project by bringing ecology tourism. The future plan for mountain villages region project will focus on building up natural resting places for citizens by offering the actual living area it self mountain villages region should be made a place where people can release fatigue or stress, rest, think, have fun and get relationship each other, villagers and citizens. For this, it, is needed to find the way to increase economic effect in mountain villages region after enough study of development peculiarity each mountain villages region has, propensity to consume, appropritate method and example, negative and positive influence.

Effect of forest road establishment based on forest management on occurrence of suspended sediment (산림경영기반의 임도개설이 부유사 발생에 미치는 영향)

  • Forest management starts from forest road facility, which is designated as generation source of muddy water in mountain stream during initial stage of establishment. Therefore, this study reviewed the effect of suspended sediment generated in forest road surface on the muddy water in mountain stream with respect to marsh area of forest. As a result, characteristics of outflow of suspended sediment was understood, and it was judged that generation of suspended sediment due to establishment of forest road is diluted by mountain stream this charged from drainage area so as to have small effect on muddy water in total mountain stream.
