This study was conducted to investigate the effects of anti-inflammatory plant extracts on the in vitro rumen fermentation characteristics and methane emission. Anti-inflammatory plant extracts from Morus bombycis Koidz, Mallotus japonicus L., Morus alba L., Paulownia coreana Uyeki, Isodon japonicus Hara and Ginkgo biloba L. were used in the study. The ruminal fluid(5 mL), McDougall buffer(10 mL), timothy as a substrate(0.3 g) and each anti-inflammatory plant extract(5% of substrate) were dispensed anaerobically into 50mL serum bottle. The mixtures were incubated for 3, 9, 12, 24, 48 and 72h at $39^{\circ}C$ without shaking. Supplementation of the anti-inflammatory plant extracts did not effects characteristics(pH, digestibility of dry matter, glucose concentration, ammonia concentration, protein concentration, VFA) on rumen fermentation. Total gas was showed a different pattern depending on treatments. Carbon dioxide was significantly(p<0.05) higher in Morus alba and Isodon japonicus than in control at 48h. Methane was significantly(p<0.05) lower in treatment than in control at initial fermentation. However the more incubation time was increased, the more methane emission was higher in treatment than in control. The concentrations of polyphenol and flavonoid were higher in Ginkgo biloba. In conclusion, supplementation of the anti-inflammatory plant extracts did not effect on rumen fermentation and methane emission was decreased in initial fermentation.
Objectives : This study was performed to investigate the efficacy and safety of the Mulberry Extract Complex in a placebo-controlled randomized clinical trial and animal study on degenerative arthritis. Methods : Animal study: Mulberry Extract Complex is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To evaluate the serum level of interleukin-2, interferon-$\gamma$, and prostaglandin E2, an animal model of degenerative arthritis induced by monosodium iodoacetate was employed. Clinical study: The efficacy index (VAS, K-WOMAC) was compared among patients with symptoms of degenerative arthritis before and after Mulberry Extract Complex ingestion as well as the one in groups. Evaluations of the improvement by the subjects and by doctor assessment were also performed. Results : Animal study: Mulberry Extract Complex reduced the serum level of interferon-$\gamma$ and prostaglandin E2 in an animal model with degenerative arthritis. Clinical study: The VAS change showed statistical significance in the experimental groups after 4 weeks (PP set) and 8 weeks (ITT set) of ingestion. When the K-WOMAC was analyzed using a modified ITT set to determine the effectiveness, statistically significant results were obtained in the fields of pain & symptom within the Mulberry Extract Complex group as well as between the Mulberry Extract Complex and placebo groups after 8 weeks of ingestion. Results from the improvement evaluation by subjects and the assessment of improvement by doctors showed statistical significance in the experimental groups (PP set) after 8 weeks. Conclusions : Mulberry Extract Complex could be useful for the improvement of various symptoms of degenerative arthritis based on its anti-inflammatory activity and its reduction of VAS and K-WOMAC pain scores.

A few clues about correlation between endoplasmic reticulum (ER) stress and mulberry (Morus alba) leaves were investigated in only the experimental autoimmune myocarditis and streptozotocin-induced diabetes. To investigate whether a novel extract of mulberry leaves fermented with Cordyceps militaris (EMfC) could suppress ER in fatty liver, alterations in the key parameters for ER stress response were measured in high fat diet (HFD)-induced obese C57L/6 mice treated with EMfC for 12 weeks. The area of adipocytes in the liver section were significantly decreased in the HFD+EMfC treated group as compared to the HFD+Vehicle treated group, while their level was higher in HFD+Vehicle treated group than No treated group. The level of the eukaryotic initiation factor 2 alpha ($eIF2{\alpha}$) and inositol-requiring enzyme 1 beta ($IRE1{\alpha}$) phosphorylation and CCAAT-enhancer-binding protein homologous protein (CHOP) expression were remarkably enhanced in the HFD+Vehicle treated group. However, their levels were restored in the HFD+EMfC treated group, although some differences were detected in the decrease rate. Similar recovery was observed on the ER stress-induced apoptosis. The level of Caspase-3, Bcl-2 and Bax were decreased in the HFD+EMfC and HFD+orlistat (OT) treated group compared to the HFD+Vehicle treated group. The results of the present study therefore provide first evidence that EMfC with the anti-obesity effects can be suppressed ER stress and ER stress-induced apoptosis in the hepatic steatosis of HFD-induced obesity model.

A silkworm (Bombyx mori L.) extract known to contain naturally occurring iminosugars, including 1-deoxynojirimycin (1-DNJ) derived from the mulberry tree (Morus alba L.), was evaluated in surrogate HCV and HBV in vitro assays. Antiviral activity of the silkworm extract and one of its purified constituents, 1-DNJ, was demonstrated against bovine viral diarrhea virus (BVDV) and GB virus-B (GBV-B), both members of the Flaviviridae family, and against woodchuck hepatitis virus (WHV) and hepatitis B virus (HBV), both members of the Hepadnaviridae family of viruses. The silkworm extract exhibited a 1,300 fold greater antiviral effect against BVDV in comparison to purified 1-DNJ. Glycoprotein processing of BVDV envelope proteins was disrupted upon treatment with the naturally derived components. The glycosylation of the WHV envelope proteins was affected largely by treatment with the silkworm extract than with purified 1-DNJ as well. The mechanism of action for this therapy may lie in the generation of defective particles that are unable to initiate the next cycle of infection as demonstrated by inhibition of GBV-B in vitro. We postulate that the five constituent iminosugars present in the silkworm extract contribute, in a synergistic manner, toward the antiviral effects observed for the inhibition of intact maturation of hepatitis viral particles and may complement conventional therapies. These results indicate that pre-clinical testing of the natural silkworm extract with regards to the efficacy of treatment against viral hepatitis infections can be evaluated in the respective animal models, in preparation for clinical trials in humans.
The antioxidant and antimicrobial activities of pork patties supplemented with medicinal herb extract mix were investigated. The medicinal herb extract powder was mixed at a ratio of 48.5:48.5:3.0 for Morus alba L, Lonicera flos, and Coptis chinensis, respectively. The pork was prepared and supplemented with medicinal herb extract mix (0, 0.5, 1, or 2%) and the antioxidant and antimicrobial activities were tested during storage at $4^{\circ}C$. The pH value decreased during cold storage at $4^{\circ}C$ for allsamples, however the pH of samples supplemented with herb extract mix rapidly decreased by days 5 and 10 ($p{\leq}0.05$). The total phenol content in the pork patties with herb extract mix was higher than in the control patties. The ABTS+ radical scavenging activities increased with increasing concentrations of herb extract mix from 0.5% to 2%. In addition, pork patties supplemented with herb extract mix showed an approximately 1 decimal reduction in total aerobic counts. Therefore, the addition of herb extract mix into pork patties increased the antioxidant activity and slightly improved the antimicrobial activity of pork patties during cold storage. However, the levels of added medicinal herb extract mix should be considered prior to its use in order to maintain proper sensory acceptance.

Anti-fungal potential of 5 plant extracts viz., Eucalyptus citriodora, Allium sativum, Cassia sophera, Chromolaena odorata and Datura metel on the growth of mulberry brown leaf spot pathogen Myrothecium roridum were examined. Except fur the aqueous extract of Allium bulb, ethanolic leaf extract of all other plants more efficiently reduced the colony growth of the fungus on potato-dextrose-agar, Of which, Allium and Eucalyptus extracts were more effective. Initiation of radial growth of M. roridum on solid media was deferred maximum 6 days by ethanolic Eucalyptus extract and 4 days by aqueous Allium extract at $0.4 mg.ml^{-1}$. In the liquid media amended with Eucalyptus extract ($0.4 mg.ml^{-1}$) complete inhibition of sporulation was noticed upto 8 days, and initial inhibition of mycelial bio-mass generation was considerably diminished with time and reduction was 1.3 fold 14 days after application. While, complete inhibition of mycelial growth for 6-14 days was recorded with $\geq$0.1 mg.ml$^{-1}$ commercial eucalyptus oil. However, rejuvenation of growth appeared when fungus was re-inoculated in fresh media. Post-inoculate application of different doses Of Eucalyptus and Allium extracts significantly (p < 0.05) reduced the disease severity in pot-ted mulberry. However, persistence of the effect up to 28 days was apparent at $\geq$ 1.0 mg.ml$^{-1}$ and effectively was on par with carbendazim (1 mg.ml$^{-1}$ ). Almost equal control ability of 1.0 mg.ml$^{-1}$ Eucalyptus extracts can be achieved by ca. 10 times lowered dose of commercial eucalyptus oil. It seems, the toxic principle of E. citrodora to M. roridum is fungistatic in nature and may have essential oil based origin.
Objectives : This study was performed to evaluate the efficacy of ampule containing herbal extract(Ulmus davidiana, Ginkgo biloba, Perilla ocymoides, Morus alba, Glycyrrhiza uralensis (licorice), and Angelica gigas) on skin wrinkles in adult women with facial wrinkles. Methods : A total of 12 women, 30 to 46 years of age, with wrinkles in the corner of eyes, applied ampule containing herbal extract twice daily for up to 12 weeks. Silicon replicas of right crow's feet area and antecubital fossa were taken before use of test product and at 4, 8 and 12 weeks. The replicas were analyzed by optical profilometry with Skin Visiometer SV600. The wrinkle and roughness parameters, R1, R2, R3, R4 and R5 were calculated and statistically analyzed. In addition, a subjective evaluation of product efficacy was conducted by patient's assessment. Results : The mean values of all the skin roughness parameters were decreased at 4-week, and decreased significantly at 8 and 12-week except R4 at 12-week after test product use. The subjective evaluation of wrinkle by patient's assessment was also improved. However, one patient noted stiffness of face after use of test product. Conclusions : These study suggested that the application of ampule containing herbal extract twice daily for 12 weeks may effectively improve the facial wrinkles without severe side effect.
The water extract from the root bark of Cortex Mori (CM, Morus alba L.: Sangbaikpi), a mulberry tree, has been known in Chinese traditional medicine to have antiphlogistic, diuretic, and expectorant properties. In this study, the cytotoxicity of CM against tumor cells and its mechanism was examined . CM exhibited cytotoxic activity on K-562, B38O human leukemia cells and B16 mouse melanoma cells at concentrations of > 1 mg/ml. A DNA fragmentation, PARP cleavage, and nuclear condensation assay showed that those cells exposed to CM underwent apoptosis. The water extract of Scutellarie Radix (SR) was used as a negative control and showed no cytotoxicity in those cells. The flow cytometric profiles of the CM-treated cells were also indicative of apoptosis. However, they did not appear to exert the G1 arrest, which is observed in other tubulin inhibitor agents such as vincristine, taxol. The protein-binding test using Biacore and a microtubule assembly-disassembly assay provided evidence showing that CM bound to the tubulins resulting in 3 markets inhibition of the assembly, but not the disassembly of microtubules. The possible nonspecific effect of the CM extract could be excluded due to the results using SR, which did not affect the assembly process. Overall, the water extract of CM induces apoptosis of tumor cells by inhibiting microtubule assembly.

This study evaluated several in vitro activities including the preliminary assessment of the anti-cancer, anti-inflammatory, and anti-diabetic effects of tree sprout extracts. Chlorogenic, caffeic, and p-coumaric acid contents in tree sprouts were analyzed using high-performance liquid chromatography and an ultraviolet detector. Among the studied tree sprout extracts, the ethanol (EtOH) extract of Rhus verniciflua exhibited the most potent anti-cancer effect by suppressing the cell viability of a human gastric adenocarcinoma cell line, with an IC50 of 7.06 ㎍/mL. The EtOH extract of Morus alba (MAB) inhibited the secretion of nitric oxide (NO) at a concentration of 100 ㎍/mL, with an IC50 of 83.44 ㎍/mL. Moreover, the EtOH extract of Securinega suffruticosa inhibited NO secretion with the lowest IC50 of 54.42 ㎍/mL. The EtOH extract of Fraxinus mandschurica was the only extract with effective α-glucosidase inhibitory activity. The total content of chlorogenic, caffeic, and p-coumaric acids was the highest in MAB (14.63 mg/g ext.). In conclusion, the beneficial activities of the tree sprout extracts with high phenolic acid content were generally high. Our results provide a theoretical basis for the development of health-promoting supplements and functional foods.

Over the past decade there has been a resurgence of interest in botanical products for their medicinal uses. This present study was designed to screen medicinal plants for the treatment of brain diseases such as Alzheimer's disease or aging. We tested the effects of the water extracts from 41 species medicinal plants on acetylcholinesterase (AChE) inhibition and antioxidant activity in vitro. Among them, Euodia rutaecarpa, Rubus coreanus, Achyranthes japonica, Morus alba, Salvia miltiorrhiza, Liriope platyphylla, and Rheum palmatum showed relatively AChE inhibition potency over 80%. Among the 7 medicinal plants investigated, E. rutaecarpa showed an 25%, 65%, and 93% inhibitions on AChE activity at 2.5, 12.5, and 25 ㎍/㎖ of the extract, respectively and was found to be one of most potent AChE inhibitor. The water extracts from 4 species (E. rutaecarpa, R. coreanus, A. iaponica, and M. alba) were tested on their antioxidant capacities using radical scavenging effect against ABTS/sup ㆍ+/. Among the 4 medicinal plants investigated, both R. coreanus and E. rutaecarpa showed significant antioxidant capacity. Therefore, E. rutaecarpa and R. coreanus are expected to ameliorate the clinical symptoms in Alzheimer's disease due to significant AChE inhibition and radical scavenging effect.
