Immunohistochemical Localization of Heat Shock Protein 70 in the Central Nervous System of Nicotine-treated Rat Embryo (태서 중추신경계의 Heat Shock Protein 70 분포에 대한 Nicotine 영향)

  • This study was investigated to determine whether nicotine causes the morphological changes and expression of heat shock protein(HSP) 70 in the central nervous system of rat embryo. The pregnant rats were injected s.c. twice daily with 3 mg nicotine per 100g body weight from day 0 to 14 of gestation and embryos were removed on gestation day 15. As morphological changes, retardation of cell proliferaton was observed in the telencephalon of nicotine-treated groups and no changes in the other region were found. Minimal HSP 70 was expressed over chole central nervous system was similar between control and nicotine-treated group, the expression of blood cells in the meinges and chroid plexus was significantly greater in nicotine-treated group than in control.

  • Periodic mesoporous organosilicas with rope-based morphology from a reaction gel composition of 1 BTME : 0.57 ODTMABr : 2.36 NaOH : 353 H2O were synthesized. While long rope-shaped product dominated in case of static synthesis condition , gyroid type products instead of rope shaped product appeared and rope shaped product disappeared with agitation. PMO with such a long rope shaped morphology is firstly reported. Additionally, various rope-based morphologies depending on the degree of bending, twisting, folding and winding of rope such as spirals, discoids, toroids, and worm-like aggregates were observed. White powdered products were characterized by X-ray diffraction, N2 sorption measurement, SEM and TEM. From XRD pattern and TEM image, ODTMA-PMO with hexagonal symmetry was identified. The pore diameter and BET surface area of ODTMA-PMO are $32.9{\AA}$ and 799 m2g-1 , respectively. Hexagonally arrayed channels run with long axis of rope and rope-based shapes with various degree of curvature, which was elucidated by using TEM images.

An Analysis and Control of Pb Pollutions of Soils by Pollens (화분에 의한 납의 오염도 분석과 그 방제에 관한 연구)

  • In this paper, pollen types of plants and Pb, N, P, K, Ca and Na in pallens of trees were investigated to control the air and soil pollutions. Morphological types of about 500 species of plants were identified to make the best use of paleontology, medicine and pharmacy. There were the positive correlations between Pb, N, P, K,Ca and Na contents in pollens and soils, respectively. A heavy metal, Pb, contained in pollens was not absorbed by rats. This fact should be utilized to purify air and soils polluted by Pb. Key words: Pb, Pollen, Pollution, puripication, Control.

Effects of Er:YAG Laser Irradiation on Removal of Subgingival Calculus. (Er:YAG laser 조사에 의한 치은연하치석 제거효과)

  • The purpose of this in vitro study was to evaluate the efficiency of Er:YAG laser on calculus removal and the morphologic changes and hardness of the irradiated surface at different power settings. This experiment used human teeth which were extracted due to periodontal disease and had a band of calculus. Forty root slabs ($5{\times}5mm$) were made and divided into control group and irradiated groups. Experimental groups were as follows; Control group (root planing), Group1 (irradiated with laser at 30mJ), Group2 (irradiated with laser at 60mJ), Group3 (irradiated with laser at 100mJ). Twelve root slab embedded in resin block were used in each group. Er:YAG laser was applied under water irrigation with the tip held perpendicular to the root surface in contact mode. The treatment time was measured until the calculus was removed completely under naked eyes. The efficiency of calculus removal was evaluated by the time for removal. Morphological changes of laser irradiated site were observed under SEM and the surface hardness was measured using a VH tester. The results were as follows; 1. The efficiency of laser scaling was increased with increasing the energy level of irradiation(p<0.05). 2. The morphological changes such as carbonization, crater and scale-like defects in the irradiated root surface were frequently observed with increasing the energy level. 3. The surface hardness tended to increase at 60mJ and 100mJ irradiated groups than that of control group. From the results evaluting on the efficiency, morphological change and surface hardness, lower energy level was suggested for the clinical application of the Er:YAG laser in scaling.

Cultural and Morphological Characteristics of a New White Button Mushroom Cultivar 'Saedo'

  • Since development of the first Korean button mushroom cultivar 'Sae-Ah' in 2010, mushroom farmers have demanded an improved cultivar. Although a new cultivar, 'Saejeong', was developed in 2011, this cultivar has thus far been difficult to cultivate in typical mushroom farms. To solve this problem, another new cultivar, 'Saedo', was developed in 2012. This new cultivar was selected by genetic analysis from crossing the heterokaryon line A175 and the homokaryon line ASI1346-15. The heterokaryon A175 was a crossed line between strains ASI1038-211 and ASI1346-20. The mycelium of 'Saedo' showed good growth on compost dextrose agar at $25^{\circ}C$. The morphological traits such as the pileus (cap) and stipe of the 'Saedo' cultivar were thicker than those of the reference control 'Saejeong', and the production yield was increased by 1.89-fold compared to that of the reference control. A field experiment of the 'Saedo' cultivar has been ongoing since 2013. The yield of the 'Saedo' cultivar was found to be substantially higher than that of the reference control during the $2^{nd}$ flush.

Morphological defects of sperm and their association with motility, fertility, and hatchability in four Korean native chicken breeds

  • Objective: This study was conducted to compare morphological defects, viability, motility (MOT), fertility (F), and hatchability (H) in four Korean native chicken breeds (KNCBs), and to evaluate whether defective segments of spermatozoa are associated with MOT, F, and H. Methods: Four KNCBs, including Korean Ogye (KO), Hwangbong (HB), Hyunin Black (HH), and Hoengseong Yakdak (HY) were used. White Leghorn (WL) was used as a control. Nine cocks from each breed were randomly assigned into three groups. Semen was collected by abdominal massage method. Eosin-nigrosin staining method was used to identify live-dead spermatozoa. Different segments and specific morphological defects of spermatozoa were identified using 4', 6-diamidino-2-phenylidole and MitoTracker Red CMXRos. F and H rates were evaluated following artificial insemination (AI). Results: KO had the highest MOT rate compared to HY. Viable normal sperm rates of KO and HH were high and comparable with WL. HY spermatozoa had the highest viable abnormal sperm (VAS) or morphological defect rate followed by HB. Likewise, HB spermatozoa had the highest dead sperm (dead) rate compared to KO, HY, and WL. Bent, coiled, detached, broken, and knotted were common identified specific morphological defects for all breeds. Most morphological defects were at the head and tail in all breeds. VAS showed strong negative correlation with MOT (r = -0.697) and F (r = -0.609). Similarly, defective tail was negatively correlated with MOT (r = -0.587), F (r = -0.797), and H (r = -0.448). The F and H rates of KO and WL were comparable. Conclusion: These data indicate that most identified specific morphological defects are at the head and tail. VAS and defective tail were associated with poor motility, F, and H. KNCBs showed more morphological defects than WL. Finally, these results will facilitate successful AI and semen cryopreservation.

Effect of Anti-juvenile Hormone Analogue (AJH) on the Larval Ecdysis of the Silkworm, Bombyx mori L. (항유약호르몬 활성물질이 누에탈피에 미치는 영향)

  • To clarify the effect of anti-juvenile hormone analogue (AJH) on the larval ecdysis by feeding at early stage of the 4th instar, the total amount of protein and activity of chitinolytic enzymes in the integument of Bombyx mori were analyzed, PAGE pattern of the protein was observed and the morphological changes of integument during molting period were also observed and the morphological changes of integument during molting period were also observed by means of TEM. The total amount of protein was greatly increased in premolting, then reached maximum level just before ecdysis, and rapidly decreased after the larval ecdysis in the control, while in the AJH treatment, increased 12 hr later than the control and its maximum was only 82.6% of the control. Two specific proteins, which were presumed as the protein originated from endocuticle, also appeared 12 hr later than the control and were maintained to 132 hr after AJH treatment from the aspects of the Native- and SDS-PAGE patterns, although those of the control disappeared instantly after ecdysis. Chitinase and $\beta$-N-acetylglucosaminidase activities were also suppressed and delayed by AJH treatment. Furthermore, it was observed that the apolysis took place 12 hr later than the control but new epicuticle was not formed at least until 132 hr after AJH treatment. From these results, it is suggested that the larval molting process of silkworm develops 12 hr later than the control but new epicuticle was not formed at least until 132 hr after AJH treatment. From these results, it is suggested that the larval molting process of silkworm develops 12 hr later than the control by AJH treatment but no further processing takes place just after apolysis.

The effects of aqua-exercise on the muscle atrophy of hind limb in rats

  • This study was peformed to investigate the effects on skeletal muscle recovery with aqua-exercise; swimming to take the muscle endurance for 20 days on two group of white rats which were the low extremity atrophy group(control groups) by fixed for two weeks and aqua-exercise group(experimental groups) after it. The effects was observed with light and electron microcope to measure the morphological changes of muscle fibers. The results obtained were as follow. 1. Light microscope: In the case of control groups, quadriceps fibers had been irregular alignment, decreased muscle width and the irregular alignment nuclear appeared, as it is degenerative muscle fibers. In the case of experimental groups, the fibers had been regular alignment cells and fibers. The nucleus of muscle had been normal characterized by oval shape and fiber sarcomere clearly classified. 2. Electron microscope: In the case of control groups, there were the quadriceps which was Z-line streaming phenomenon induced at the sarcomere and cells nuclear separated from basal membrane. It was not only observed the sarcomere alignment irregularly and mitochondria damaged, but also vacuoles found. In the case of experimental groups, A band, I band, H band had been clearly appeared, classified at the myofibrils of quadriceps, and electronic dense M-line found in sarcomere. There were observed satellite cells and basal laminas that usually to be appeared at the time of mitochondrial development, skeletal muscle fiber regeneration or development. This results suggest that the aqua-exercise assisted to inhibit the degenerative morphological changes of skeletal muscle cells and help to recover from abnormal states. Especially, it is considered to effect on a normal structural formation.

Morphological and Growth Characteristics of Poa pratensis L. 'Pureundul' ('푸른들' 왕포아잔디(Poa pratensis L. 'Pureundul')의 형태 및 생육특성)

  • The research was carried out to define the morphological and growth characteristics of Poa pratensis L. (Kentucky bluegrass) 'Pureundul' native to Korea. Poa pratensis 'Pureundul' was collected from the belt highway around Mt. Uam located in Naedeok-dong, Cheongju City, Chungcheongbuk-do Province on May 5, 1995. The morphological characteristics of 'Pureundul' were compared with those of 205 individuals of native Poa pratensis collected throughout the nation from June 11, 2000 to June 21, 2003. The findings were as follows : The height of plant was 28.2cm but increased to 40.0cm at the flowering time. The height of first node from the base was 6.8cm and the length and width of leaf were 17.3cm and 2.9mm, respectively. The length of a spike was 7.9cm. Based on these morphological characteristics, 'Pureundul' was defined medium type Kentucky bluegrass. Visual quality(1~9), visual color and visual density were measured. The visual quality, color and density of 'Pureundul' were not superior to those of introduced species but above the average other native species. The damages from the diseases during summer resulted in poorer visual quality of color of 'Pureundul'. But the spread of the diseases was slower in 'Pureundul' than other native species, and accordingly 'Pureundul' didn't suffer from more quality deterioration from July to September during which the diseases broke out. Entering fall, 'Pureundul' showed high quality comparable to that of the introduced species. If 'Pureundul' is put to breeding by implementing pest control in summer and introducing disease tolerant genes, it will be used to make greens of greater quality than now.
