A Study on Image Analysis of Graphene Oxide Using Optical Microscopy (광학 현미경을 이용한 산화 그래핀 이미지 분석 조건에 관한 연구)

  • Experimental considerations have been performed to obtain the clear optical microscopic images of graphene oxide which are useful to probe its quality and morphological information such as a shape, a size, and a thickness. In this study, we investigated the contrast enhancement of the optical images of graphene oxide after hydrazine vapor reduction on a Si substrate coated with a 300 nm-thick $SiO_2$ dielectric layer. Also, a green-filtered light source gave higher contrast images comparing to optical images under standard white light. Furthermore, it was found that a image channel separation technique can be an alternative to simply identify the morphological information of graphene oxide, where red, green, and blue color values are separated at each pixels of the optical image. The approaches performed in this study can be helpful to set up a simple and easy protocol for the morphological identification of graphene oxide using a conventional optical microscope instead of a scanning electron microscopy or an atomic force microscopy.

  • Recently, import supply of glass eels from Philippines has been increased in South Korea, and the species distinction was very important to import traders and Anguilla marmorata and A. bicolor pacifica are major species at Philippines. In this study, the species distinction and the rate were researched targeting imported glass eels from the estuaries of North Luzon and the South Mindanao in May and July, 2014. Dorsal fin position and pigmentation pattern at caudal fin are the trait standard for species distinction. The results about morphological identification were coincide with the result of the genetic identification, and the regional distribution of A. marmorata rate was over 90% at North Ruzon in May. However, the rates of A. bicolor pacifica and A. mamorata were 63.3% and 36.7% in May, and were 53.3% and 46.7% in July respectively at South Mindanao.

Effects of Turbid Water on Fish Ecology in Streams and Dam Reservoirs

  • Turbid water or suspended sediment is associated with negative effects on aquatic organisms; fish, aquatic invertebrate, and periphyton. Effects of turbid water on fish differ depending on their developmental stage and a level of turbidity. Low turbid water may cause feeding and predation rates, reaction distance, and avoidance in fish, and it could make fish to die under high turbidity and long period. Therefore, it is very important to find out how turbid water or suspended sediment can affect fish in domestic watersheds. The objectives of this study were 1) to introduce international case studies and their standards to deal with suspended sediment, 2) to determine acute toxicity in 4 major freshwater fishes, and 3) to determine in relation to adverse effect of macroinvertebrates and fish. Impacts of turbid water on fish can be categorized into direct and indirect effects, and some factors such as duration and frequency of exposure, toxicity, temperature, life stage of fish, size of particle, time of occurrence, availability of and access to refugia, etc, play important role to decide magnitude of effect. A review of turbidity standard in USA, Canada, and Europe indicated that each standard varied with natural condition, and Alaska allowed liberal increase of turbidity over natural conditions in streams. Even though acute toxicity with four different species did not show any fatal effect, it should be considered to conduct a chronic test (long-term) for more detailed assessment. Compared to the control, dominance index of macroinvertebrates was greater in the turbid site, whereas biotic index, species diversity index, species richness index, and ecological score were smaller in the turbid site. According to histopathological analysis with gills of macroinvertebrate and fishes, morphological and physiological modification of gills due to suspended sediments can cause disturbance of respiration, excretion and secretion. In conclusion, in order to maintain good and healthy aquatic ecosystem, it is the best to minimize or prevent impact by occurrence of turbid water in stream and reservoir. We must make every effort to maintain and manage healthy aquatic ecosystem with additional investigation using various assessment tools and periodic biomonitoring of fish.

Pilot Development of Supporting Tools for Automatic Detection of Safety Standards (안전기준 자동검색을 위한 지원도구 시범개발)

  • With the development of society, the scale of the statute is not only increasing, but also the content is getting complicated. The scale of safety standards existing in the law is also increasing and specialized, making it difficult to integrate and manage to minimize conflicts or overlaps among safety standards. For the integrated management of safety standards, a technology that searches for and extracts safety standards in laws and regulations must first be secured. In this study, considering the limitations of time and manpower, a tool for automatic detection of safety standards is developed based on several specific cases. The safety standards classified in the previous studies and the safety standards announced by the Ministry of Interior and Safety were analyzed, and also statute information which includes safety standards extracted by the National Disaster Management Institute in 2018 was collected. After the collected laws were refined and morphological analysis was performed, a safety standard thesaurus was constructed and indexed to develop a safety standard search tool. When automatic search tools are routinely applied to find safety standards in the future, it is expected that these tools will help to solve overlapping or conflicting problems of complex safety standards.

A Highly Pathogenic Strain of Bacillus thuringiensis serovar kurstaki in Lepidopteran Pests

  • In order to detect and identify the most toxic Bacillus thuringiensis strains against pests, we isolated a B. thuringiensis strain (Bn1) from Balaninus nucum (Coleoptera: Curculionidae), the most damaging hazelnut pest. Bn1 was characterized via morphological, biochemical, and molecular techniques. The isolate was serotyped, and the results showed that Bn1 was the B. thuringiensis serovar, kurstaki (H3abc). The scanning electron microscopy indicated that Bn1 has crystals with cubic and bipyramidal shapes. The Polymerase Chain Reactions (PCRs) revealed the presence of the cry1 and cry2 genes. The presence of Cry1 and Cry2 proteins in the Bn1 isolate was confirmed via SDS-PAGE, at approximately 130 kDa and 65 kDa, respectively. The bioassays conducted to determine the insecticidal activity of the Bn1 isolate were conducted with four distinct insects, using spore-crystal mixtures. We noted that Bn1 has higher toxicity as compared with the standard B. thuringiensis subsp. kurstaki (HD-1). The highest observed mortality was 90% against Malacosoma neustria and Lymantria dispar larvae. Our results show that the B. thuringiensis isolate (Bn1) may prove valuable as a significant microbial control agent against lepidopteran pests.

Antagonistic Potential of Native Trichoderma viride Strain against Potent Tea Fungal Pathogens in North East India

  • Indigenous strains of Trichoderma species isolated from rhizosphere soils of Tea gardens of Assam, north eastern state of India were assessed for in vitro antagonism against two important tea fungal pathogens namely Pestalotia theae and Fusarium solani. A potent antagonist against both tea pathogenic fungi, designated as SDRLIN1, was selected and identified as Trichoderma viride. The strain also showed substantial antifungal activity against five standard phytopathogenic fungi. Culture filtrate collected from stationary growth phase of the antagonist demonstrated a significantly higher degree of inhibitory activity against all the test fungi, demonstrating the presence of an optimal blend of extracellular antifungal metabolites. Moreover, quantitative enzyme assay of exponential and stationary culture filtrates revealed that the activity of cellulase, ${\beta}$-1,3-glucanase, pectinase, and amylase was highest in the exponential phase, whereas the activity of proteases and chitinase was noted highest in the stationary phase. Morphological changes such as hyphal swelling and distortion were also observed in the fungal pathogen grown on potato dextrose agar containing stationary phase culture filtrate. Moreover, the antifungal activity of the filtrate was significantly reduced but not entirely after heat or proteinase K treatment, demonstrating substantial role of certain unknown thermostable antifungal compound(s) in the inhibitory activity.

  • Concentrated localized torrential rains due to global warming and climate change have resulted in much water damage each year. GIS is used as a tool for predicting the peak-outflows caused by these regional torrential rains in mountainous rivers. However, the research of the resolution of the data is limited, and most of approaches are about hydrological geographic. This paper estimates the flood discharge needed for decision of standard rainfall of automatic rainfall warning system by using GIS with GcIUH model, and establishes the criteria of flash flood warning. It also has analyzed the terrain in river basin, extracted the morphological characteristics parameters of water shed such as stream width, channel slope, channel length, shape factor, and GcIUH parameters, and analyzed the relationship between them.

Description of the Post Larva of Star Pipefish, Halicampus punctatus (Syngnathidae, Gasterosteiformes) First Found in the Southwestern East Sea, Korea

  • Larval specimens of Halicampus punctatus were collected off Ulsan and Uljin in December 2002 (three specimens) and off Ulsan in December 2003 (one specimen). These specimens are characterized by the following morphological characteristics: rings, 14 + 35 = 49; subdorsal rings, 1 + 3 = 4; dorsal fin rays, 19 - 20; pectoral fin rays, 14 - 15; anal fm rays, 9; head length (HL), 5.8 - 6.7 in the standard length; snout length, 1.9 in HL; snout depth, 3.7 - 5.2 in snout length. The number of caudal fin rays 9 is less than those of the other species in the same genus 10. Wide stripe bands composed of small pigments are shown in the trunk and the tail. Melanophores are not found in the dorsal fin, the pectoral fin, and the anal fin except the caudal fin. The supraoccipital crest is on the head. The frontal ridge is on the dorsal side of front trunk. The blanched ridges on the opercular are fused with a main ridge like a tree branch. A few branched ridges that are small and narrow on the trunk and the tail are fused with the main ridges. We report these specimens as the first record in Korea and name them 'Byeol-sil-go-ki' in Korean.

  • A highly strained nanostructure comprising crystallographically aligned HgTe nanoinclusions and a surrounding PbTe matrix has been synthesized using a precipitation process of supersaturated HgTe-PbTe alloys. From the early precipitation stage, HgTe nanoinclusions take disk shape, which is transformed from initial HgTe nuclei, although there is no lattice constant difference of the two end components at standard state. As a primary reason for the morphological transformation of the initial spherical HgTe nuclei to HgTe nanodisks, the induced lattice mismatch is suggested. On the condition that the HgTe nanodisks maintain perfect coherent nature with PbTe matrix, the stress-free lattice constant of constrained HgTe nanodisks has been calculated based on the defined concept of the strain-induced tetragonality, the linear elasticity and the actual measurement in HRTEM images.

  • This study covers centrality of villas after Renaissance and architectural forms corresponding to the nature in the aspect of Karl Popper and Gilles Deleuze. In addition, the researcher tries to understand the changes in architectural forms according to the changes in places. To do that, Pascal and Hans Meyer's theory was introduced as a major theory. The researcher tries to set up the standard by analyzing the architectural form related with the nature and the form of center in the housing through Villa Rotonda and Villa Emo. And with key analysis of the centrality and architectural form corresponding to the nature and the four houses that are historically and morphologically important. It is found that due to the development in modern technology and materials, a variety of architectural approach is developed and they are implemented as various architectural forms. However, due to various theories of modern architectural approach, it is found that the morphological differences of suburban housing exists by forming a universal relationship constructed by four architects. The researcher expects to be understood and realized in real life construction.
