Screening of Alcohol Dehydrogenase Inhibitors from Natural Products (천연물로부터 알코올 탈수소효소 저해제 검색)

  • Excessive or long term ingestion of alcohol may cause hepatitis, cirrhosis, hepatic tumor and so on. Aldehyde and active form of free oxygen that are metabolites of alcohol in liver are the cause of liver cell damage. The main system of alcohol metabolism is composed of alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ALDH) and cytochrome P450. In connection with in vivo alcohol metabolism, more than one hundred natural products were screened for inhibition or activation of alcohol dehydrogenase. As a results, we found significant inhibition ($IC_50$) of ADH by methanolic extracts of Puerariae Radix ($61.2{\;}\mu\textrm{g}/ml$), Glycyrrhizae Radix ($105.0{\;}\mu\textrm{g}/ml$), Cinnamomi Ramulus ($7.0{\;}\mu\textrm{g}/ml$), Rhei Rhizoma ($36.7{\;}\mu\textrm{g}/ml$), Mori Cortex Radicis ($106.2{\;}\mu\textrm{g}/ml$), Chrysanthemi Flos ($112.2{\;}\mu\textrm{g}/ml$), Erycibes Caulis ($36.7{\;}\mu\textrm{g}/ml$), and Scutellariae Radix ($122.5{\;}\mu\textrm{g}/ml$)

Inhibitory Effects of Crude Drugs on $\alpha$-Glucosidase

  • The inhibitory activity of several crude drugs on $\alpha$-glucosidases, which are the key enzymes for carbohydrate digestion and the prevention of diabetic complications, was investigated. Several crude drugs including Terminaliae Fructus, Mori Cortex Radicis, Caesalpiniae Lignum and Cyrophora esculenta potently inhibited maltase and sucrase isolated from rat intestine, while Arecae Semen and Corni Fructus remarkably inhibited $\alpha$-amylase. Caesalpiniae Lignum and Gyrophora esculenta exhibited significant reductions of blood glucose elevation in mice loaded with maltose and sucrose.

  • The immunomodulating activity of a polysaccharide isolated from Morus alba (PMA) root bark was examined in murine splenic lymphocytes. PMA enhanced proliferation of splenic lymphocytes in a synergistic manner in the presence of mitogens. However, PMA suppressed pri-may IgM antibody production from B cells, which was activated with lipopolysaccharide, a polyclonal activator, or immunized with a T-cell dependent antigen sheep red blood cells. Our observations showed that the immunomodulating activity of PMA increased lymphocyte proliferation and that PMA decreased antibody production from B cells, which was distinct from those of other plant-originated polysaccharides.

  • This study was carried out to investigate sulfur dioxide in herb drugs.(Puerariae Radix, Lycii Fructus, Platycodi Radix, Dioscoreae Rhizoma, Mori Radicis Cortex, Nelumbo Seed, Paeoniae Radix, Remotiflori Radix, Astragali Radix, Polygonatum Rhizome) Experimental subjects included 105 cases of 10 species of herb drugs collected in Kyung-dong market from December in 2001 to February in 2002. Sulfur dioxide was determined by Optimized Monier-Wiliams method. In 65 cases(61.9%) of the 105 cases, we detected $SO_2$ over 10mg/kg. (omitted)

Inhibition of Biofilm Formation in Yersinia enterocolitica by Edible Plant Extracts Including Polygoni Multiflori Radix

  • Yersinia enterocolitica, which causes yersiniosis, is a bacterium that produces biofilms effectively. The inhibition of biofilm formation provides a method for preventing infections with Y. enterocolitica. In this study, the inhibitory activity of Y. enterocolitica biofilm formation was investigated in a library of 140 edible plant methanol extracts including forest products. It was identified that the biofilm formation could be inhibited by 12 extracts of plants, Agastachis Herba, Agrimoniae Herba, Diospyros kaki leaves, Elsholtziae Herba, Ginkgonis Semen, Lycopi Herba, Melonis Pedicellus, Menthae Herba, Mori Radicis Cortex, Polygoni Multiflori Radix, Prunellae Spica, and Schizonepetae Spica. After changing the solvent to ethanol and water, the greatest inhibition of biofilm formation was produced by a 50% ethanol extract of Polygoni Multiflori Radix. A method to effectively prevent yersiniosis can be developed using the edible plant extracts identified in this study.

The Growth Promotion Effect of Useful Enterobacteria Bifidobacterium aolescentis KCTC 3216 by Combination of Natural Products Bearing Antioxioative Capacity (장내 유용세근 Bifidobacterium adolescentis KCTC 3216의 성장을 촉진시키는 항산화 천연산물의 조합구성)

  • The growth of enterobacteria, Bifidobacteriurn adolescentis KCTC 3216 was promoted by natural products bearing antioxidative capacity and combined two, three and four kinds of them. B. adolescentis was showed a good growth by Sophorae flos, Phellodendri cofex, Mori cortex radicis, Aurantii nobilis pericarpium, Angelicae gigantis radix, alone, and two mixed combinations were composed of Paeonia japonica and Theae foiium, Epimedii herba and Angelicae gigantis radix, Paeonia japonica and epimedii herba, Atractylodis rhizoma alba and angelicae gigantis radix, and three mixed combinations were oraganized with Theae folium, Paeonia japonica and epimedii herba, Theae folium, Beiamcanda chinensis and Paeonia japonica, Theae foiium, Astragaii radix and Mori cortex radicis, and four mixed combinations were formed with Seiamcanda chinensis, Angelicae gigantis radix, Epimedii herba, Theae folium, and Angeiicae gigantis radix, Epimedii herba, Paeonia japonica, Theae folium, and Epimedii herba, Paeonia japonica, Sophorae flos, Theae folium. The best four mixed combination for the growth of B. adolescentis was mixture of Epimedii herba, Paeonia iaponica, Sophorae flow and Theae foiium, which promoted 2.6 times than that of control, and its antioxidative capacity was also 5.6 times higher, and the ratio of elimination of hydroxyl radical was more than 80% in each dilution rate. As these combinations of natural products will activate some parts of body, they may be applied to pharmaceuitcal applications, functional foods, antiaging tea, also expected to promote useful entero bacterial growth for fermentative beverage bearing multifunction.

The Growth Inhibiting Effect of Vibrio parahaemolyticus KCTC 2471 by the Natural Products Bearing Antioxidative Capacity (Vibrio Parahaemolyticus KCTC 2471에 대한 항산화 천연산물의 성장 억제 효과)

  • The growth of V. parahaemolyticus was inhibited by the combination of natural products having antioxidative capacity. The growth of V. parahaemolyticus was controlled effectively by each or the combination of Paeonia japonica, Acanthodanacis cortex, Phellodendri cortex, Mori cortex radicis, Theae folium, Galla rhois, Coptidis rhizomn and Astragali radix. The growth inhibition ratio of the combination of Astragali radix, Phellodendri cortex, Coptidis rhizotna and Paeonia japonica was 95%, and its antioxidative capacity was also 5.4 times higher, and the elimination ratio of hydroxyl radical was more than 80% compared with control. As natural products might activate some parts of human body, they could be utilized as pharmaceuitcal applications, functional foods, antiaging tea, and multifunctional fermentative beverages.

  • The growth of C. pefringens was inhibited by the combination of natural products which have antioxidative capacity C. perfringens was controlled very effectively by each or the combination of Paeonia japonica, Atractylodis rhizome alba, Epimedii herba, Acori graminei rhizoma, Schizandraefructus, Lyciifructus, Cinnamomi cortex Sophorae flos Phellodendri cortex, Nelumbo nuclfera gaertner, Asparagi tuber, Mori cortex radicis, Astrafali radix and Coptidis rhizome The combination of Phellodendri cortex, Epimedii herba, Astragali radtx and Glycyrrhizae radix inhibited the growth of C. pefringen by 75% and increased the antioxidative activity by 4.4 times. This combination also eliminated more than 80% of hydroxyl radical. These natural products can be utilized as pharmaceuticals, functional foods, antiaging tea, and multifunctional fermentative beverages.

Study on Pathology of Taeeumin Medicinal Herb in Sasang Constitutional Medicine (태음인 약재의 사상의학적 병리 연구)

  • Objective This study was designed to define the efficacy of medicinal herbs of Taeeumin in Sasang constitutional medicine. Method We searched and analyzed original text such as Donguisusebowon Sinchuk edition written in 1901, Donguisusebowon Gabo edition written in 1894, Donguisusebowon Sansang Chobongwon, and posthumous manuscripts left by Je-ma Lee published by the Ministry of Health of North Korea. Results Seven herbs such as Ephedrae Herba, Coicis Semen, Castaneae Semen, Fel Ursi, Mori Cortex Radicis, Ginkgonis Semen, and Farfarae Flos regulate the mechanism of Esophagus-Cold (Wiwanhan). Three herbs such as Melonis Pedicellus, Ailanthi Radicis Cortex, and Quisqualis Fructus regulate the mechanism of Cold Lung-Dry (HanpaeJo). Sixteen herbs such as Rhei Rhizoma, Angelicae Tenuissimae Radix, Puerariae Radix, Cimicifugae Rhizoma, Angelicae Dahuricae Radix, Moschus, Gleditsiae Spina, Mume Fructus, Aurum, Glycine Semen Germinatum, Ampelopsis Radix, Cornu rhinocerotis, Antelopis Cornu, Bomeolum, Bezoar Bovis, and Typhae Pollen regulate the mechanism of Liver-Heat (Ganyeol). Three herbs such as Chrysanthemi Indici Flos, Nelumbinis Semen, and Spirodelae Herba regulate the mechanism of Heat Lung-Dry (YeolpaeJo). Conclusion Forty four herbs of Taeeumin regulate the mechanisms of Esophagus-Cold (Wiwanhan), Cold Lung-Dry (HanpaeJo), Liver-Heat (Ganyeol), Heat Lung-Dry (YeolpaeJo) and correct the energy-fluid pathology of Taeeumin.

Screening of Leukotriene $B_4$ Receptor Antagonist Activity from the Herbal Drugs (생약의 류코트리엔 $B_4$ 수용체결합 저해작용 검색)

  • Leukotriene $B_4\;(LTB_4)$ is a pro-inflammatory mediator synthesized in myeloid cells from arachidonic acid. Elevated levels of $LTB_4$ have been found in a number of inflammatory diseases and levels are related to disease activity in some of these. Because $LTB_4$ interacts with cells through specific cell surface receptors, $LTB_4$ receptor blockade is the most specific approach to reduce the pathogenic role of $LTB_4$. In order to find $LTB_4$ receptor antagonist from plants, we screened the $LTB_4$ receptor antagonistic activity of the methanol extract and solvent fractions of herbal drugs. The ability of samples to inhibit specific binding of $[^3H]-LTB_4$ to human peripheral neutrophils was used as assay to evaluate the antagonistic activity of plant materials. Among the tested methanol extracts of herbal drugs, Mori Radicis Cortex, Perillae Semen, Armeniacae Semen and Sophorae subprostratae Radix showed potent inhibitory activity above 70% at the concentration of $100\;{mu}g/ml$. The inhibitory activities of $LTB_4$ binding to human neutrophils were evaluated for several solvent fractions at three different concentrations. Especially, hexane soluble fractions of Anemarrhenae Rhizoma and Embeliae Radix, and ethyl acetate soluble fractions of Aristolochiae Fructus, Magnoliae Cortex and Zingiberis Rhizoma crudus showed moderate activity at $25\;{mu}g/ml$. These fractions were promising candidates for the study of the activity-guided chromatographic purification of active compounds. Silica gel column chromatography of hexane soluble fractions of Anemarrhenae Rhizoma and Embeliae Radix gave very active sub-fractions, AA-4 and ES-4, and their inhibition activities of $LTB_4$ binding to human neutrophil at $30\;{mu}g/ml$ were 78% and 62%, respectively. From these results we could anticipate new $LTB_4$ receptor antagonist from herbal drugs, and the block of $LTB_4$ effects may provide beneficial in neutrophil mediated diseases such as inflammation and bronchial asthma.
