Penicillium purpurogenum 유래의 Mannanase에 의한 Mannooligosaccharide의 생산 (Production of Mannooligosaccharides by the Penicillium purpurogenum Mannanase)

  • Penicillium purpurogenum , which produces a copra galactomannan degrading enzyme extracellularyl, was isolated from soil , and its properties and formation condition of mannooligosaccharides were investigated. The optimum ph and temperature for the activity of the mannanase were 5.5 and 55$^{\circ}C$, respectively. The mannanase was stable in between pH 3.5 and 7.0 after 2 hr incubation at 3$0^{\circ}C$ lost 90% of the original activity after incubation at 55$\AA$ and pH 5.5 for 2 hr. With two different substrate concentration, hydrolysis of white coprameal proceeded rapidly at the early stage of the reaction, but gradually solwed thereafter especially at a higher concentration of copra meal (20 %). The enzyme hydrolyzed white copra meal to monosaccharides, mannobiose and mannotriose at the final stage of the reaction.

상엽의 혈당강하 활성 (Blood Glucose-Lowering Effects of Mori Folium)

  • Mori Folium(MF) methanol extract and its water soluble fraction showed significant blood glucose lowering effects alloxan-induced hyperglycemic mice. Their hypoglycemic activities seemed to nothing to do with the stimulation of insulin release or insulin-like action, according to our experiments. On the other hand, MF prevents the hyperglycemic responses from an oral load of starch and glucose in vivo. Since complex carbohydrates present in a diet must be degraded to monosaccharides by $\alpha$-glucohydrolase before being absorbed in the gastrointestinal tract, it is thought that blood glucose lowering effects of MF may be related to the inhibition of $\alpha$-glucohydrolase catalyzed enzymatic reaction. In addition, experiments that examined an effect of MF water soluble fraction on gastrointestinal movement showed no significant GI movement inhibitory effect. In conclusion, MF water soluble fraction may possess active component which is a potential candidate as an orally active agent for the treatment of diabetes mellitus.

Enzymatic transesterification for the synthesis of amino acid-sugar conjugates

  • Among the tested ten enzymes, Optimase M-440 showed the highest activity in transesterification of N-t-Boc-L-Phe-OTFE with D-glucose. Monosaccharides and their derivatives acted as good acyl acceptors in the Optimase M -440 catalyzed transesterification of N-t-Boc-L-Phe-OTFE. Optimase M-440 showed a preferable catalytic activity on the primary hydroxyl group of saccharides and a good regioselectivity. Optimase M-440 showed the highest activity in pyricline among the tested solvents. As acyl donors, trifluoroethyl esters of amino acids showed a high reactivity in transesterification. Optimase M-440 showed a broad substrate specificity towards amin 。 acid esters and saccharides.

  • Production of inulo-oligosaccharides from inulin was carried out with the maximum yield of 94% using a purified endoinulinase from Aspergillus ficcum. The optimum reaction temperature and pH were 55-60$^{\circ}C$ and pH 5.5-6.0, respectively. The Michaelis constant and maximum reaction velocity of the endoinuinase for inulin were 13.27 g/L and 0.13 g/L$.$min at 55$^{\circ}C$, pH 5.5, respectively. Inulin source had no significant effect on oligosaccharide yield and product composition, although initial production rate differed according to inulin origins. The reaction pH was a critical factor in inulo-oligosaccharide production because considerable free monosaccharides were released, decreasing oligosaccharide yield at low pH conditions. An empirical relationship describing the reaction performance was developed from kinetic data: the time to reach maximum oligosaccharide yield (tw) as a function of initial concentration of inulin (lo) and enzyme (Eo); i.e., log tM = -1.025 log Eo - 0.011 l0 + 2.655.

오탄당(五炭糖) Triazine 유도체(誘導體)의 Acetylation 과 생리활성(生理活性)에 관(關)하여 (Acetylation of s-Triazine Derivatives from Uronic Acids and their Biological Activity)

  • 오탄당(五炭糖) triazine유도체(誘導體)를 합성(合成)하였으며 이들을 acetylation하여 원소분석(元素分析), UV, IR, NMR 등(等)의 기기분석(機器分析)에 의하여 Methyl $[T]-\beta-L-acetyl-arabo-psyranoside$ 그리고 $[T]-\alpha-D-acetyl\;lyxopyranoside$로 각각 동정(同定)되었다. 오탄당(五炭塘) triazine유도체(誘導體)와 이들의 acetylation한 화합물(化合物)의 무우, 벼, 종자발아(種子發芽)에 대(對)한 생리활성(生理活性)은 없었다. 그리고 곰팡이, 세균에 대하여도 별다른 생리활성(生理活性)은 나타나지 않았다.

  • 상황버섯 자실체의 열수 및 알카리 추출 다당류에 대한 성분 분석을 조사하여 다음과 같은 결과를 얻었다. 1. 상황버섯 자실체를 열수 및 알칼리 추출시 수율은 각각 1.1%와 4.4% 였으며, 당은 각각 81.1%와 37.4%를 함유하고 있었다. 2. 구성당은 glucose와 galactose가 주를 이루고, 아미노산은 열수 추출분획은 Tre와 Ser을 다량 함유하고 있었으며, 알칼리 추출분획은 Asp와 Ala가 주를 이루는 것으로 조사되었다.

당알콜이 밀전분이 호화 및 노화에 미치는 영향 (Effect of Sugar Alcohol on Wheat Starch Gelatinization and Retrogradation)

  • The sugar alcohols are useful alternatives to sucrose in confections because they provide desirable taste but lessen the potential risks for dental caries. The effects of sugar alcohol and their interaction on starch pasting properties were determined by the viscoamylograph with the concentration of 30%, 20%, 10% sugar alcohol in 1% CMC. The disaccharides, lactitol, maltitol, and isomalt, delayed pasting more than did monosaccharides, sorbitol and xylitol. With regard to the solubility in water, sorbitol and xylitol are more soluble than sucrose, maltitol and lactitol are almost equal and isomalt is less soluble than sucrose. Sorbitol and xylitol were highest on gelatinization. But their retrogradation occurred faster than other sugar alcohols. Lactitol and maltitol decreased gel strength more than did sucrose and other sugar alcohols.

Saccharina japonica를 이용한 전처리 및 분리당화발효와 동시당화발효로부터 에탄올 생산 (Ethanol Production by Separate Hydrolysis and Fermentation and Simultaneous Saccharification and Fermentation Using Saccharina japonica)

  • Ethanol fermentations were carried out using simultaneous saccharification and fermentation (SSF) and separated hydrolysis and fermentation (SHF) processes with monosaccharides from seaweed, Saccharina japonica (sea tangle, Dasima) as the biomass. The pretreatment was carried out by thermal acid hydrolysis with $H_2SO_4$ or HCl. Optimal pretreatment condition was determined at 10% (w/v) seaweed slurry with 37.5 mM $H_2SO_4$ at $121^{\circ}C$ for 60 min. To increase the yield of saccharfication, isolated marine bacteria Bacillus sp. JS-1 was used and 48 g/L of reducing sugar were produced. Ethanol fermentation was performed using SSF and SHF process with Pachysolen tannophilus KCTC 7937. The ethanol concentration was 6.5 g/L by SSF and 6.0 g/L by SHF.

Optimal Conditions for the Production of Sphimin, a Sphingomyelinase Inhibitor from Steptomyces sp. F50970

  • We isolated a sphingonyelinase (SMase) inhibitor, which would be a potential reagent to regulate cell proliferation, oncogenesis, and inflammation, from a strain of Streptomyces sp.. In this paper, we report the optimal conditions for the production of SMase inhibitor, designed as sphinin, from Streptomyces sp. F50970. The optimal carbon and nitrogen source were 1% soluble starch and 0.05%-0.15% trypton. Most of monosaccharides and high concentration of soluble starch above 1.0% caused falling of pH and sphinin production. Zn2+, Cu2+, Fe2+, Mn2+, and Co2+inhibited cell growth and the production of sphinin. Inorganic phosphate promoted the sphinin production. Optimal initial pH for the production of sphinin was 7.5-8.0. Addition of CaCO3 to the medium resulted in an increase of inhibitor production. Based on these results, we designed a fermentation medium for the production of a SMase inhibitor, sphinin, from Streptomyces sp. F50970.

  • To investigate constituents of Russula pseudodelica Lange and Microporus affinis(Blume et Nees) Kuntze, quantitative analyses of free and total amino acids were carried out with G. L.C. and an amino acid autoanalyzer. Polysaccharides of the two mushrooms were extracted with hot water and the filtrate was concentrated. The addition of three volumes of ethanol to the concentrate formed precipitation of crude polysaccharides which were analyzed by G.L.C. and H.P.L.C. Sixteen amino acids and four monosaccharides were identified in the protein-bound poly­saccharides of the two mushrooms. The polysaccharides of M. affinis showed antineoplastic activity against sarcoma 180 implanted in mice.
