Studies on the Antineoplastic Components of Korean Basidiomycetes (한국산(韓國産) 담자균류(擔子菌類) 항암(抗癌) 성분(成分)에 관한 연구(硏究))

  • To investigate antitumor components of Korean higher fungi, the carpophores of Ganoderma lucidum (Fr.)Karst. were collected at Gal-mae-ri in Gyeong-gi Province and extracted with 0.1N sodium hydroxide solution. The extract was purified by dialyzing through Visking tube and a dark brownish power was obtained. The antitumor activity of the fraction was tested against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction against the tumor was 87.6% in doses of 50 mg/kg/day for the period of ten days. The tumor in five of 15 mice was completely regressed. The antitumor fraction was chemically analyzed and found to be a complex of polysaccharide and protein, from which four monosaccharides and eighteen amino acids were identified.

  • A thermostable xylanase from Thermotoga maritima (Xyn B) cleaves several pNP-glycosides of monosaccharides. We found that the initial product of the cleavage of pNP-xyloside (pNP-Xy1) was a disaccharide, not xylose, indicating that xylosyl unit of pNP-Xyl was transglycosylated to another pNP-Xyl. We determined that the disaccharide was xylobiose which has the linkage of the ${\beta}$ 1-4, and described the reaction mechanism of the Xyn B. Also, we produced the several pNP-glycosides and propyl-disaccharides from the transglycosylation of Xyn B with varial glycosides and/or 1-propanol. All reaction products were purified by column chromatography (Toyo-pearl HW-40C, 45 cm${\times}$2.5 cm or 45 cm ${\times}$ 2.5 cm${\times}$ 2). The isolated products were analyzed by means of 1D and 2D NMR.

Plant Cell Wall Degradation with a Powerful Fusarium graminearum Enzymatic Arsenal

  • The complex enzyme pool secreted by the phytopathogenic fungus Fusarium graminearum in response to glucose or hop cell wall material as sole carbon sources was analyzed. The biochemical characterization of the enzymes present in the supernatant of fungal cultures in the glucose medium revealed only 5 different glycosyl hydrolase activities; by contrast, when analyzing cultures in the cell wall medium, 17 different activities were detected. This dramatic increase reflects the adaptation of the fungus by the synthesis of enzymes targeting all layers of the cell wall. When the enzymes secreted in the presence of plant cell wall were used to hydrolyze pretreated crude plant material, high levels of monosaccharides were measured with yields approaching 50% of total sugars released by an acid hydrolysis process. This report is the first biochemical characterization of numerous cellulases, hemicellulases, and pectinases secreted by F. graminearum and demonstrates the usefulness of the described protein cocktail for efficient enzymatic degradation of plant cell wall.

A New Synthetic Approach to 3-Amino-2-phenylthietanes as Potential Monoamine Oxidase Inhibitors

  • 3-Amino-2-phenylthietane derivatives were considered as a useful tool to elusidate the mechanism of inhibiton of MAO by tranylcypromine-type inhitors. The synthesis of 3-benzoylamino-2-phenylthieetane, 3-amino-2-phenylthietane, and 3-N, N-dimentylamino-2-p-nitrophenythietane was attempted using the reaction between 1, 3 dihalogeno alkanes with alkali sulfide. When 1-pheny1-1, 3-dihalo-2-benzolaminopropane was treated with sodium sulfide, 2-pheny 1-4 benzylidene-2-oxazoline was isolated, indicating the case of elimination reaction compared to ring formation. The reaction of 1-p-nitropheny1-1, 3-dichloro-2-N, N-dimethylaminopropane with sodium sulfide gave bis (1-p-nitropheny1-2-N, N-dimethylamino-3-chloropropane)sulfide. The mechanism of reaction was discussed.

  • The carpophores of a mushroom, Naematoloma fasciculare (Fr.) Karst, were extracted with 0.2 N NaOH and the extract was dialyzed through visking tube. It was found to contain an antitumor activity against sarcoma 180 implanted in mice. The components of this aqueous extract were found to be a polysaccharide and a protein by color reactions including Anthrone and Lowery-Folin tests. The hydrolysis of the polysaccharide with 3% HCL-Me-OH and trimethylsily lation of the hydrolysate yielded five monosaccharides, i. e. glucose, frutose, mannose, galactose and xylose, which were detected and analyzed by GLC. After hydrolysis of the protein with 6N HCl, 15 amino acids, including aspartic acid and glutamic acid, were detected and analyzed by an auto amino acid analyzer.

  • A protein-polysaccharide fraction was prepared from the carpophores of Laetiporus sulphureus. This fraction suppressed growth of sarcoma 180 in A-strain mice when administered i. p. To investigate the mechanism of antitumor action of this fraction, plaque assay was conducted by administrating i. p. to the mise at a dose level of 50mg/kg for five days. Ten days later, the mice were immunized with 1 * 10$^{7}$ sheep red blooc cells. The number of hemolytic plaque forming cells was significantly greater than that of the control mice. Three monosaccharides and fifteen amino acids were identified in the protein-polysaccharide fraction.

Development of an improved selective media for differentiation of emetic and diarrheal type Bacillus cereus

  • The objective of this study was to develop a differential medium with improved selectivity for the isolation of Bacillus cereus. Mannitol egg yolk polymyxin medium supplemented with D-galactose allowed the differentiation of diarrheal- and emetic-type B. cereus through pH monitoring. The pH of the medium decreased significantly when incubating the emetic-type B. cereus, whereas the pH change was not significant when incubating the diarrheal-type. The addition of pH indicators, such as methyl red and phenol red, to the medium allowed visual differentiation between diarrheal- and emetic-type B. cereus. A solid agar medium was also developed by optimizing the concentrations of medium components such as monosaccharides, agar, egg yolk enrichment, pH indicators, and antibiotics. This study indicates the possibility of applying selective media for the differentiation of diarrheal- and emetic-type B. cereus.

  • This guidance is an updated version of the irritable bowel syndrome (IBS) guidelines based on evidence-based medicine. IBS is a common chronic gastrointestinal syndrome that occurs in approximately 10% of the population and causes chronic abdominal pain as well as bowel habit changes, such as stool frequency or consistency. The final diagnosis of IBS is based on the exclusion of organic diseases that would explain the symptoms and the absence of endoscopic abnormalities. IBS can reduce the quality of life and cause a major disease burden, such as repeated examinations and continuous drug use, by mistaking organic diseases including malignancy. The major changes are as follows: 1) when to perform a colonoscopy under the impression of IBS; 2) effect of a low-fermentable oligosaccharides, disaccharides, monosaccharides, and polyol diet; 3) impact of probiotics in IBS; and 4) role of antibiotics in IBS. The Korean Society of Neurogastroenterology and Motility recently updated these guidelines to support physicians for qualified medical services and reduce the socioeconomic burden of IBS.

Factors Affecting the Survival Rates of Frozen-Thawed Spermatozoa in Jindo Dog by Monosaccharides and Freezing Rate (진도개 정액 동결 시 단당류 및 동결 속도가 생존율에 미치는 영향)

  • This studies were conducted to investigate the survival rate of frozen-thawed spermatozoa of Jindo Dog by monosaccharide and freezing rates. Experimental animals were prepared 12 males within 1~8 year's old and collected once in a couple of weeks by digital manuplation methods. Collected semen was diluted 1:1 with Tris-egg yolk extender and added 4, 6 or 8% of glycerol and none, 4 mM glucose or 4 mM fructose as cryoprotectant and was equilibrated for 2 hrs in $4^{\circ}C$. In monosaccharide groups, the freezing rate was 5 cm-5 min. above $LN_2$. The survival rates without monosaccharide were $50.7{\pm}19.0%$, $58.6{\pm}18.0%$, $40.0{\pm}10.0%$ in 4, 6 or 8% glycerol, respectively. In addition of glucose, the survival rates were $43.1{\pm}14.7%$, $38.1{\pm}16.5%$, $33.3{\pm}4.0%$ in 4, 6 or 8% glycerol, respectively and in fructose, were $47.9{\pm}21.1%$, $61.3{\pm}6.2%$, $34.3{\pm}12.6%$ in 4, 6 or 8% glycerol, respectively. There showed significantly different between glycerol groups and monosaccharides groups (p<0.05). The survival rates of freezing rate in 5 cm-5 min. group was $64.5{\pm}15.8%$, $51.9{\pm}27.6%$, $29.7{\pm}24.8%$ and in 10 cm-10 min. group was $62.5{\pm}20.3%$, $64.9{\pm}23.6%$, $34.5{\pm}27.4%$ in 4, 6 or 8% glycerol, respectively. There were significantly different between freezing rates (p<0.05). These results suggest that the addition of fructose with 6%-glycerol and slow freezing improve the survival of frozen-thawed sperm in Jindo Dog.

Variation of Chemical Components and Their Interaction with Isoflavones in Maturing Soybean Seeds

  • This study was focuses on the variation of isoflavone contents during seed development and their interaction with major chemical components such as protein, amino acids, saccaharides, lipid and fatty acids. During maturing, lipid, protein, and amino acid contents in soybean seeds showed the highest values at R7 stages, but isoflavone contents were increased until R8 stage. It was noted that malonyl glucosides $(64.2\%)$ are predominant forms among conjugated isoflavones followed by glucosides $(30.7\%)$, acetyl glucosides $(4.1\%)$ and aglycones $(0.9\%)$. Sucrose and stachyose were presented as a major saccharide in soybean seeds. As maturing days progressed, they were constantly increased and the highest contents were observed at R8 stage. While small quantities of raffinose, fructose, glucose, maltose, DP3 (DP: degree of polymerization), DP6, and DP7 were detected. These results showed that saccharide composition at the beginning of seed development is primarily monosaccharides with little sucrose and oligosaccharides, but as maturing days proceeds, sucrose and starch increase with concomitant decrease in monosaccharides. Sucrose and stachyose were positively correlated with isoflavone (r=0.780, 0.764 at p<0.01, respectively), while fructose, glucose, maltose, and DP7 were negatively correlated (r=-0.651, -0.653, -0.602, and -0.586 at p<0.05, respectively). Soybeans at R8 stage were high in protein and amino acid, but low in free amino acid contents. Protein and amino acid contents showed positively significant correlations with isoflavone (r=0.571 and 0.599 at p<0.05, respectively), but free amino acid content were negatively correlation with isoflavone (r=-0.673, p<0.01). The lipid content reaches its final content relatively early stage of seed development and remains constant as compared with other chemical components. Among the fatty acids, although varietal difference was presented, stearic acid and linolenic acid were gradually decreased, while oleic and linoleic acid were increased as seed maturing progressed. Lipid was significantly correlated (r=0.754, p<0.01) with isoflavones. However, neither saturated fatty acid nor unsaturated fatty acids significantly affected the isoflavone contents of maturing soybean seeds.
