The efficient synthesis of a tetrasaccharide, the suitably protected form of the repeat unit, →2)-α-D-Manp-(1→2)-β-D-Manp-(1→3)-α-D-GlcpNAc-(1→6)-α-D-Manp-(1→, of the O-antigen polysaccharide of the lipopolysaccharide from E. coli O77 has been accomplished. Glycosylation reactions for the coupling of four monosaccharide building blocks of the tetrasaccharide were carried out employing the CB glycoside method, the mannosyl 4-pentenoate/PhSeOTf method, and the glycosyl trichloroacetimidate method with complete stereoselectivities in excellent yields.
Extract of water-soluble polysaccharide (CFWx), showing inhibiting activity on ${\alpha}-glucosidase$, was prepared from the fruiting bodies of Cordyceps militaris by hot-water extraction, and ethanol precipitation. Chemical characteristics of CFWx were as follows: carbohydrate content 30% including 16% of uronic acid; 51% protein content; monosaccharide composition, Man:Glu:Gal (30:43:27); molecular weight $3-5{\times}10^4$. CFWx was further purified by ion-exchange, gel-permeation, and affinity chromatography and $CFWx-AH-{\alpha}$ fraction was isolated. Fundamental structure of $CFWx-AH-{\alpha}$ was deduced as ${\alpha}-(1{\to}4$)-D-glucan with ${\alpha}-(1{\to}3$)- and/or ${\alpha}-(1{\to}6$)-D-glycosidic side chains based on methylation analysis.

In order to investigate the cultural characteristics of Lentinus lepideus, media, pH, carbon sources, nitrogen sources, vitamin and organic acids were tested in submerged culture. It grew well in GPB(broth), and the optimum temperature and pH was $25^{\circ}C$ and 4.2, respectively. The carbon sources such as galactose belong to monosaccharide and maltose belong to disaccharide were effective for mycelial growth. Peptone which is compound nitrogen source was good for mycelial yield but the other nitrogen sources were not effective. Among the various vitamins, pyridoxine is suitable for mycelial yield. Among the various organic acids, citric acid promoted the mycelial yield but acetic acid interrupted the mycelial growth of L. lepideus.
This study was carried out to investigate potential applications of the extract of Castanopsis cuspidata var. sieboldii $N_{AKAI}$ nut as a functional food ingredient. The pH and $^{\circ}Brix$ of nut were 6.43 and 3.17, respectively. L, a and b values as Hunter's color were 83.07, 1.49 and 10.48, respectively. Total content of monosaccharide was 54.26 mg% and organic acids were composed of oxalic acid 495.37 mg%, formic acid 200.03 mg%, malic acid 93.65 mg%, citric acid 27.80 mg%, and succinic acid 16.61 mg%. Total phenolic contents in various solvent extracts were as follows: water 27.69 mg%, 75% ethanol 16.50 mg%, ethyl acetate 16.50 mg%, and methanol 10.30 mg%. The antioxidant activity ($SC_{50},\;{\mu}g/mL$) of the nut extracts by various solvents was in the order of ethyl acetate 74.88 > methanol 155.00 > n-hexane 213.33 > ethanol 249.33 > butanol 274.78 > chloroform 314.67 > 75% ethanol 848.33 > water extracts 869.67. The results indicated that the extract of C. cuspidata nut contained a potential food ingredient.

Citrus canker is very important disease in international trade of citrus. The disease was usually take place from late of June, and severe middle of July to middle of August, though disease occurrence was affected by environmental conditions. In pathogenicity test, three varieties, orange, lemon and kiyomi among 7 varieties, were succeptible, two varieties, satsuma mandarin and iwasachi, intermediate resistant. On the other hand, shiranuhi and yuzu were resistant relatively. The pathogen, Xanthomonas axonopodis pv. citri, grew well in PD broth adjusted to pH 7.0 at 26$^{\circ}C$. It's growth was best in medium containing group of monosaccharide as a carbon source and group of ammonium as a nitrogen source. Tow isolates were resistant to streptomycin among 11 isolates isolated from diseased leaves in field in Jeju-Do. The streptomycin sensitives isolate was controlled by in greenhouse test. On the other hand, the resistant and sensitive isolates were controlled by treatment with copper sulfate, the control value is 88.7% and 90.6%, respectively.
Lactan gum produced by Rahnella aquatilis is a high viscous, anionic polysaccharide and has shear thinning behaviour. Lactan gum yield and cencentration was greater on disaccharide such as lactose and sucrose than on monosaccharides such as glucose and galactose. When initial carbon source concentration was 45g/l of sucrose of lactose, the microorgnisms produced 28 g/l and 27 g/l of lactan, respectively with a yield more than 60%. $\beta$-Galactosidase, hydrolyzing lactose into galactose and glucose, was induced by lactose or galactose. When initial corbon source was 45 g/l of mixed carbon I (glucose:galactose=1:1), lactan gum concentaration was higher than that from 45 g/l of monosaccharide (glucose pf galactose) but was similar to the result from 45 g/l of lactose. Therefore, lactose hydrolysis reaction by $\beta$-galactosidase does not seem to be a rate determining step in lactan gum biosynthesis. When initial carbon source was 45 g/l of mixed carbon II (glucose:fructose=1:1). total carbon source consumption rate was slower than that from sucrose, but glucose consumption rate was faster than that from fructose.
This study was conducted to identify the soluble carbohydrates in soybean seeds using on-line HPLC-RID-ES/MS and HPLC behavior, and to determine their contents for high quality soybean breeding. The monosaccharide (glucose) and three oligosaccharides (sucrose, raffinose, and stachyose) were identified in Korean soybeans by their chromatographic behavior and results of on-line HPLC-RID-MS with Electrospray Ionization mode. On the basis of HPLC with a RID detector, the 32 Korean major soybeans contain $0.37{\pm}0.26\%$ glucose, $4.55{\pm}0.91\%$ sucrose, $1.19{\pm}0.19\%$ raffinose, and $2.72{\pm}0.37\%$ stachyose on a dry basis. In 468 soybean germplasms, the ranges of glucose, sucrose, raffinose, and stachyose were $0.03 - 0.98\%$, $2.33 - 6.96\%$, $0.08 -1.87\%$ and $0.75 - 3.18\%$, respectively. Among 500 soybean samples, oligosaccharide contents of 32 Korean major cultivated soybeans and 468 soybean germplasms were varied $5.83 - 10.06\%$ and $3.66 - 10.32\%$, respectively. The composition of glucose, sucrose, raffinose, and stachyose in soluble carbohydrates of 500 soybean samples were $2.07 {\pm} 1.75\%$, $58.01{\pm}5.82\%$, $10.13{\pm}2.28\%$ and $29.80{\pm}4.54\%$, respectively. Sucrose appeared to be most prevalent in soybean soluble carbohydrates.
Fucoidan has been shown to exhibit a host of biological activities, including anti-coagulant, anti-thrombotic, anti-tumourigenic, anti-inflammatory, anti-viral, anti-complementary and neuroprotective effects. In the present study, we attempted to determine the effects of Fucoidan on both apoptosis and cell proliferation in the hippocampal CA1 region and the dentate gyrus of gerbils after the induction of transient global ischemia. This experiment involved the use of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay as well as immunohistochemisty for caspase-3 and 5-bromo-2'-deoxyuridine (BrdU). The monosaccharide composition of the purified Fucoidan which had been extracted from Laminaria religiosa was utilized in this study. The present study clearly induces that apoptotic cell death and cell proliferation in the gerbil's hippocampal regions increased significantly following the induction of transient global ischemia and the results of this study also indicate that Fucoidan exerted a suppressive effect on this observed ischemia-induced increase in apoptosis within the CA1 and dentate gyrus, and also suppressed cell proliferation in the dentate gyrus.

The bacterial strain WC-17 able to produce chitinase was isolated from soil using an enrichment technique. The isolated strain was identified as Acinetobacter sp. judging by their morphological and physiological characterisitics. The optimal culture conditions for the production of chitinase of Acinetobacter sp. WC -17 are 1.5% colloidal chitin and 1 % tryptone at $30^{\circ}C$ with pH 6.5. Since the enzyme was rapidly produced in a culture supplied with chitin, glucose, or N-acetylglucosamine but not with other polymers and monosaccharide, the enzyme was considered to be an inducible enzyme. Notably N- acetylglucosamine and glucose were found to be effective inducers at low concentrations but repressors at excessive concentrations. The cultural supernatant of Acinetobacter sp. WC-17 inhibited the growth of phytopathogenic fungi such as P.oryzae, R.solani, and F.solani. Among the phytopathogenic fungi tested, P.oryzae was the most sensitive. The conventional agar plate (PDA containing 1 % colloidal chitin) method also produced the same result.
There is a significant amount of experimental evidence suggesting that polysaccharides from mushrooms enhance the host immune system by activating various mechanisms in immune cells, including macrophages. In this study, polysaccharides from Lentinus edodes were found to stimulate the functional activation of macrophages to secrete inflammatory mediators and cytokines and increase the phagocytotic uptake. The chemical properties of the stimulatory polysaccharides, CPFN-G-I, CPBN-G, and CPBA-G, were determined based on their monosaccharide composition, which mainly consisted of glucose and mannose. According to FT-IR and GC/MS, the structure of CPFN-G-I, purified from the fruiting body of L. edodes, was found to consist of a $\beta$-1,6-branched-$\beta$-1,4-glucan, whereas CPBN-G and CPBA-G, purified from the liquid culture broth, were found to be composed of a heteromannan. The configuration of the p-linkage and triple helical conformation of each polysaccharide were confirmed using a Fungi-Fluor kit and Congo red, respectively.
