Effect of Ethanol on Mouse Liver Monoamine Oxidase

  • The effects of ethanol and acetaldehyde on monoamine oxidase activity in mouse liver mitochondrial fraction were studied. In vivo, a single dose of ethanol increased the hepatic monoamine oxidase activity compared to control group, and chronic ethanol consumption also increased the enzyme activity using tyramine, benzylamine or serotonin as substrate. Acetaldehyde, the metabolite of ethanol, significantly increased monoamine oxidase activity more than ethanol did. In contrast to the in vivo results, it was found that the monoamine oxidase activity was inhibited in vitro by ethanol or acetaldehyde in the dose-dependent manner.

정신분렬병의 혈소판 Monoamine Oxidase 활성도에 관한 연구 (Studies on Platelet Monoamine Oxidase Activity in Schizophrenics)

  • 만성 정신분렬병 환자에서 성(性)에 따른 혈소판 MAO 활성도의 변화양상을 분석하고 이에 관련하여 혈장 Testosterone 및 Estradiol농도의 변화를 검토하고자, 135명의 정상대조군과 78명의 만성 정신분렬병 환자를 대상으로 하여 혈소판 MAO활성도와 혈장 Testosterone 및 Estradiol농도를 측정한 결과는 다음과 같다. 1) 만성 정신분렬병 환자에서 혈소판 MAO활성도는 남녀 모두 그 정상대조군에 비하여 낮은 사람이 더 많았다. 2) 혈소판 MAO활성도의 남녀간의 차이는 정상대조군에서는 없었지만 만성 정신분렬병 환자가 여자 환자에 비해 혈소판 MAO활성도가 낮은 사람이 더 많았다. 3) 남자 정신분렬병 환자에서는 혈장 Estradiol농도만 정상대조군에 비하여 저하되어 있었고, 여자 정신분렬병 환자에서는 혈장 Testosterone농도만 정상대조군에 비하여 저하되어 있었다.

  • Eight natural or semistynthesized monoterpenes were examined for their effects in rat brain monoamine oxidase(MAO) using benzylamine as substrate. Thujone and 3-carene were found to have the inhibition effects on rat brain MAQ activity, 38% and 95% inhibition at 103M respectively. The kinetic study on 3-carene, the most potent inhibitive type. But (+) pulegon and (-) isopulegon was found to activate MAO slightly.

Ethanol이 유도한 간장중 MAO 활성변동에 미치는 인삼의 영향 (Effect of Panax Ginseng in Mouse Liver with Ethnol-Induced Monoamine Oxidase Activity)

  • 본 실험에서는 ethanol 및 acetaldehyde가 유도하는 MAO 활성변동에 인삼 butanol 분획이 미치는 영향을 검토하였다. Ethanol(1 g/kg)을 mouse 복강내로 투여하였을 때 간장 중 MAO 활성이 현저히 증가하였으며 이러한 작용은 인삼butanol 분획 전처리로 개선되었다. 또한 pyrazole(200mg/kg)을 전처리하였을 때 ethanol에 의해 증가된 간장중 MAO활성이 정상수준으로 감소되었다. 만성적으로 ethanol을 투여한 경우에 있어서도 MAO활성이 증가되었으며 인삼 butaonl분획 전처리로 MAO 활성이 감소됨이 관찰되었다. Ethanol의 대사산물인 acetaldehyde(100mg/kg)을 복강내로 투여하고 관찰한 실험에서도 간장중 MAO활성이 현저하게 증가되었으며 인삼 butanol분획을 전처리함으로써 정상수준 가깝게 MAO활성이 감소되었다. 이 연구결과로 ethanol에 의한 간장중 MAO활성증가는 ethanol의 대사산물인 acetaldehyde에 기인된 것으로 생각되어지며 인삼 butanol분획이 acetaldehyde에 의해 유도되는 간장중 MAO활성 변동을 조절하고 있을 것으로 사료된다.

  • Monoamine oxidases catalyze the oxidative deamination of dietary amines and amine neurotransmitters, and assist in maintaining the homeostasis of the amine neurotransmitters in the brain. Dysfunctions of these enzymes can cause neurological and behavioral disorders including Parkinson's and Alzheimer's diseases. To understand their physiological roles, efficient assay methods for monoamine oxidases are essential. Reviewed in this Perspective are the recent progress in the development of fluorescent probes for monoamine oxidases and their applications to enzyme assays in cells and tissues. It is evident that still there is strong need for a fluorescent probe with desirable substrate selectivity and photophysical properties to challenge the much unsolved issues associated with the enzymes and the diseases.

  • Fluorides were supposed to exert a stimulatory action on the catecholamine release. In this study, the authors attempted to investigate the action of sodium fluoride on the catecholamine release from the isolated perfused cow adrenal gland and rat heart. And also the inhibitory effect of sodium fluoride on the monoamine oxidase activity in rat heart and liver mitochondria was investigated. The monoamine oxidase activity was measured by the conversion of benzylamine to benzaldehyde. The results obtained were follows; 1. Sodium fluoride stimulated the release of catecholamine from the isolated perfused cow adrenal gland and rat heart. 2. Sodium fluoride inhibited the rat heart and liver mitochondrial monoamine oxidase activity.

  • The age-related changes of monoamine contents and monoamine oxidase (MAO) activities in the whole brain and blood pressures were measured and compared every week during the ages of 5 weeks to 12 weeks in two types of animals; (1) spontaneously hypertensive rats (SHR) and (2) normotensive Sprague-Dawley rats (NR). Blood pressures in SHR began to rise at the ages of 7 or 8 weeks and were significantly higher thereafter than in NR. Abnormally low MAO activities in the brain of SHR were detected from the ages of 8 weeks. Comparisons were also made between brain monoamine-norepinephrine, dopamine and 5-hydroxytrytamine contents of SHR and those of NR at the same ages throughout the test period. In SHR, none of those monoamine values was consistently higher or lower than in NR during the test period. Protein contents of the brain between SHR and NR were not significantly different at any ages. These observations suggest that the abnormally low MAO activities in SHR brain may be one of the underlying neurological factors for the susceptibilities to hypertension and the deficit of MAO activities may be due to the changes in properties rather than in amount of this enzyme.

Evodiamine의 Monoamine Oxidase 활성 저해작용 (Inhibition of Monoamine Oxidase by Evodiamine)

  • The effects of evodiamine on monoamine oxidase (MAO) activity were investigated. MAO was purified from mouse brain and the $K_m\;and\;V_{max}$ values of MAO were $78.5{\pm}5.28{\mu}M$ and $0.68{\pm}0.07$ nmol/min/mg protein, respectively (n=4). Evodiamine at $30-120{\mu}M$ showed an inhibitory effect on MAO activity using a substrate kynuramine with an $IC_{50}$ value of $104.2{\mu}M$ (n=4). Evodiamine also exhibited a non-competitive inhibition on MAO. The $K_i$ value for evodiamine was $72.5{\pm}10.8{\mu}M$ (n=4). These results suggest that evodiamine partially contributes to the regulation of monoamine content.

  • 1. 전신가토(全身家兎) 및 척수가토(脊髓家兎)에서 MAO 억제하(抑制下)에서의 brethylium의 tyramine 승압효과(昇壓效果)에 미치는 영향(影響)을 관찰(觀察)하였다. 2. 전신가토(全身家兎) 및 척수가토(脊髓家兎)에서 bretylium 정주(靜注) 후(後)에는 tyramine의 승압효과(昇壓效果)는 강화(强化)되었으나 MAO 억제제(抑制劑)인 catron을 정주(靜注)하고 bretylium을 추가(追加) 주입(注入)한 후(後)에는 tyramine의 승압효과(昇壓效果)는 catron 투여전(投與前)의 그것보다 감약(減弱)되었다. 3. Bretylium에 의한 tyramine 승압효과(昇壓效果)의 강화(强化)는 MAO 억제작용(抑制作用)으로 설명(說明)되고, tyramine 승압효과(昇壓效果)의 감약(減弱)은 교감신경말단(交感神經末端)의 차단작용(遮斷作用)으로 설명(說明)된다.

구속(拘束)스트레스로 유발된 생쥐 시상하부내(視床下部內) Monoamine의 변화에 대한 시호소간산(柴胡疎肝散)과 시호억간탕(柴胡抑肝湯)의 효능 (Effect of Chaihushugansan and Chaihuyigantang on the change of Monoamine Contents in the Hypothalamus of Mice induced by the Immobilization Stress)

  • This study aimed to investigate the effects of Chaihushugansan and Chaihuyigantang on the change of monoamine-contents in the hypothalamus of mice induced by the immobilization stress. Each formula(1.0g/kg) was injected intragastrically to mice 1 hour prior to the immobilzation stress(15 minutes) before measuring the change of monoamine-contents level in mice hypothalamus. The results were as follows: 1. DA content at control group showed significant change. CHSGS group and CHYGT group showed significant decrease.(P<0.05) 2. NE content of the control group was increased than normal group. NE content of the CHYGT group was shown a significant decrease than control group.(P<0.05) 3. DOPAC contents didn't show significant changes at any groups. 4. 5-HT contents didn't show significant changes at any groups. 5. 5-HIAA contents at CHSGS group and CHYGT group showed signifiant changes.(P<0.05, 0.01) These data revealed that chaihushugansan and chaihuyigantang might have the anti-stress effect through the control of DA and NE contents in the hypothalamus.
