Characterizing Fluorescence Properties of Dissolved Organic Matter for Water Quality Management of Rivers and Lakes (하천 및 호소 수질관리를 위한 용존 자연유기물질 형광특성 분석)

  • Fluorescence measurements of dissolved organic matter(DOM) have the superior advantages over other analysis tools for applying to water quality management. They are simple and fast and require minimal pretreatment of samples. Fluorescence index($F_{450}/F_{500}$), synchronous spectra, and fluorescence excitation-emission matrices(EEM) of various DOM samples were investigated to discriminate autochthonous/allochthonous composition, protein-like fluorescence, fulvic-like fluorescence, humic-like fluorescence, terestrial humic-like fluorescence by comparing among the real DOM samples of different origins with the help of literature. The samples used included standard purified DOM, lake, river and wastewater treatment effluent. The relative distribution of various DOM composition was derived from the ratios of each fluorescence region. The results were very consistent with those expected from the sample properties. Allochthonous and terrestrial humic-like fluorescence were more prominent in the samples with abundant soil-derived DOM components. In addition, the protein-like fluorescence property was more pronounced in the samples where strong algal or microbial activities were expected. It was also shown that the ratio of protein-like/terrestrial humic-like fluorescence obtained from synchronous spectrum and fluorescence EEM could be used as an indicator for the evaluation of wastewater treatment on the downstream water quality of rivers and for the prediction of the degree of algal/microbial activities in lakes. It is expected that the results of this study will provide the basic information to develop the future water quality management techniques using DOM fluorescence measurements.

Reliability Evaluation of ACP Component under a Radiation Environment (방사선환경에서 ACP 주요부품의 신뢰도 평가)

  • This study deals with the irradiation effects on some selected components which are being used in an Advanced Spent Fuel Conditioning Process (ACP). Irradiation test components have a higher priority from the aspect of their reliability because their degradation or failure is able to critically affect the performance of an ACP equipment. Components that we chose for the irradiation tests were the AC servo motor, potentiometer, thermocouples, accelerometer and CCD camera. ACP facility has a number of AC servo motors to move the joints of a manipulator and to operate process equipment. Potentiometers are used for a measurement of several joint angles in a manipulator. Thermocouples are used for a temperature measurement in an electrolytic reduction reactor, a vol-oxidation reactor and a molten salt transfer line. An accelerometer is installed in a slitting machine to forecast an incipient failure during a slitting process. A small CCD camera is used for an in-situ vision monitoring between ACP campaigns. We made use of a gamma-irradiation facility with cobalt-60 source for an irradiation test on the above components because gamma rays from among various radioactive rays are the most significant for electric, electronic and robotic components. Irradiation tests were carried out for enough long time for total doses to be over expected threshold values. Other components except the CCD camera showed a very high radiation hardening characteristic. Characteristic changes at different total doses were investigated and threshold values to warrant at least their performance without a deterioration were evaluated as a result of the irradiation tests.

Development and performance evaluation of the porous tube dilutor for real-time measurements of fine particles from high humidity environments (고수분 환경에서 미세먼지 실시간 측정을 위한 다공 튜브형 희석장치의 개발 및 성능 평가)

  • Real-time measurements of fine particles from stack emission gases are necessary due to the needs of continuous environmental monitoring of PM10 and PM2.5. The porous tube dilutor using hot and cold dilutions was developed to measure fine particles without condensable particles from highly humid emission gases and compared to the commercialized ejector-type dilutor. Particle size distributions were measured at the emission gases from a diesel engine and a coal-fired boiler. The porous tube dilutor could successfully measure the accumulation mode particles including relatively large particles more than $3{\mu}m$ without nuclei particles, while the ejector dilutor detected some condensable particles and could not detect large particles. The porous tube dilutor could successfully remove the already condensed water droplet particles generated by a humidifier in a $30m^3$ chamber.

Evaluation of Influent Water Quality Using Indicator Microorganisms in Lake Shiwha (지표미생물을 이용한 시화호 유입수의 수질평가)

  • Lake Shiwha, an artificial lake located near metropolitan Seoul, offers a unique water environment and has been suspected to have high levels of chemical and microbiological contaminations. Lake Shiwha was originally connected to the sea but currently has four major surface water inputs from agricultural, municipal, industrial areas and in addition an occasional inflow from the sea. The objectives of this study are to investigate the relative contribution of microbial contaminants from each of the inflowing surface waters and to identify appropriate microbial indicator organisms in this unique water environment. We measured the levels of microbial contaminations in the four inflowing surface waters. A number of microbial indicator organisms including total coliform (TC), fecal coliform (FC), E. coli, Enterococci, somatic and male-specific coliphages were analyzed. Bacterial indicator microorganisms were detected and quantified by the $Colilert^{(R)},\;Enterolert^{(R)}$ kit. Surface water (50 l) was sampled by $ViroCap^{TM}\;5"$ cartridge filters and analyzed by the single agar layer method for detecting coliphages. The concentrations of TC, FC, E. coli, and Enterococci were 1543 CFU/100 ml${\sim}1.99{\times}10^6$ CFU/100 ml, 0 CFU/100 ml${\sim}202$ CFU/100ml, 0 CFU/100 ml${\sim}1.80{\sim}10^5$ CFU/100ml, 74 CFU/100 ml${\sim}3408$ CFU/100 ml, respectively. The male-specific and somatic coliphages were detected in three different inflowing surface waters. Isolated E. coli and Enterococci strains were further analyzed by 16s rDNA amplification and subsequent phylogenetic analysis from Jungwang-chun, Ansan-chun, Banwol-chun and penstock of inflowing surface water. Our results indicated that the concentrations of different fecal indicator microorganisms might not be highly correlated with each other. Multiple microbial indicator organisms should be used for monitoring microbial contamination and microbial source tracking methods.

A Survey on the Safety of the Imported Foods in Gwangju (광주지역 수입식품의 안전성에 대한 조사연구)

  • In order to provide fundamental data of food circulation policy, we performed monitoring the safety of the imported food which was circulating through Gwangju from March to October, 2005. Acid and peroxide value which are barometers for evaluation of the quality of lipid were investigated in 130 samples of imported oil treatment food. Not-permitted tar pigment and artificial sweetner were investigated in 139 candies by TLC and HPLC. The content of sulfur dioxide in 129 samples of dried fishery products and dried fruits was investigated by Monier-Williams method. In 130 samples of imported oil treatment food, 9 samples (6.9%) were incongruent with acid value, 6 samples (4.6%) with peroxide value and 4 samples (3.1%) with acid value simultaneously with peroxide value. In 139 imported candies, not-permitted artificial sweetner were found in 2 samples (1.4%). In 129 samples in which sulfur oxide was analyzed,4 samples (3.1%) were incongruent. Finally, in total 398 samples in which this study was analyzed, 25 samples (6.3%) were incongruent.

Critical Evaluation of and Suggestions for the VOCs Measurement Method Established as the Korean Indoor Air Quality Standard Method (실내공기질 공정시험법 중 VOCs 측정방법의 문제점 고찰 및 개선방안에 관한 연구)

  • During the last two decades, indoor air quality and volatile organic compounds (VOCs) have been of concern in Korean society due to their nature of potential health impacts. In order to investigate the pollution levels of VOCss in indoor environments, establishment of a solid test method for monitoring the airborne VOCss is essential. In Korea, a method based on adsorbent sampling and GC analysis coupled with thermal desorption was proclaimed as the Korea Standard Method for Indoor Air Quality Test. This study was carried out to examine some inherent problems of the VOCs measurement method. The VOCs method does not describe in detail preparing the standard samples. The standard samples may be prepared by impregnation of either liquid standard solutions or a mixture of standard gases. In this study, we investigated the optimal temperature condition for transferring the liquid standards onto a standard adsorbent tube. As a result, keeping the impregnation temperature at $250^{\circ}C$ will be recommended in regard of the boiling points of multiple target analytes and the thermal stability of the adsorbent. We also demonstrated some problems associated with handling of a syringe used for transferring the standard solutions onto the adsorbent tubes, and a best way to get rid of the syringe problems was suggested. Finally, a number of field works were conducted to evaluate the performance of adsorbent sampling methods. Comparison of different adsorbent tubes, i.e. tube packed with single sorbent (Tenax) and double sorbents (Tenax with Carbotrap), revealed that 30 to 40% differences between the two groups, implying that sampling efficiency is depending on the volatility and the strength of adsorbents. However, duplicate precisions for VOCs sampling with a same type of adsorbent and at same flow rates appeared to be satisfactory to be all within 20%, which is a quality control guideline. Distributed volume precisions were also found to be within a guideline value, 25%, although the precision was in general inferior to the duplicate precision. The Korea indoor VOCs test method should be more refined and improved in many aspects, particularly procedure and instrumentation for preparing the standard samples and specification of quality control assessment.

The Usefulness of Transcutaneous Tissue Oxygen Pressure ($TcpO_2$) for Determination of the Point of Time at Venous Revascularization (적절한 정맥 재혈류 시점의 판단을 위한 조직산소분압의 유효성)

  • Purpose: The purpose of this study is to delineate the optimal time of venous revascularization for preventing the flap necrosis due to venous occlusion, and to clarify the usefulness of tissue oxygen pressure ($TcpO_2$) in the determination of the point of time for venous revascularization. Methods: Thirty-six, $3{\times}3\;cm$ sized epigastric island flap was elevated in left abdomen of male Sprague-Dawley rat weighing 250 gram. Flaps were randomly assigned to six groups of six flaps according to the duration of venous occlusion with microvascular clamp; 10 minutes in the group I as the control, 60 minutes in the group II, 2 hours in the group III, 3 hours in the group IV, 4 hours in the group V, and 6 hours in the group VI, respectively. Just before removal of clamp after flap was reposed in situ, the ratio of $TcpO_2$ (tissue oxygen pressure) of the island flap to that of right abdomen was calculated in each group, and tissue specimen was harvested from the distal area of the flap for histological evaluation of vascular change. Five days later, survival area of the flap was estimated, and evaluated the correlation between the tissue oxygen pressure and the rate of flap survival. Results: The $TcpO_2$ and the survival rate of flap were decreased proportionally with the duration of venous occlusion. The ratio of the $TcpO_2$ of the flap is decreased abruptly to below sixty percentile compared to the $TcpO_2$ of normal tissue, and the survived area of the flap is decreased to nine-tenth of the designed size after three hours of total venous occlusion. Histologically, the number of congested vessels was increased according to venous occluded time, and proportionally increased after 3-hours of occlusion significantly. Conclusion: There is a close correlation between the $TcpO_2$ and the survival rate of flaps according to the duration of venous occlusion. Therefore, the $TcpO_2$ represents the hemodynamic changes within the flap, and thought to be an alternative effective tool in the flap monitoring for venous revascularization.

Construction of In Vivo Fluorescent Imaging of Echinococcus granulosus in a Mouse Model

  • Human hydatid disease (cystic echinococcosis, CE) is a chronic parasitic infection caused by the larval stage of the cestode Echinococcus granulosus. As the disease mainly affects the liver, approximately 70% of all identified CE cases are detected in this organ. Optical molecular imaging (OMI), a noninvasive imaging technique, has never been used in vivo with the specific molecular markers of CE. Thus, we aimed to construct an in vivo fluorescent imaging mouse model of CE to locate and quantify the presence of the parasites within the liver noninvasively. Drug-treated protoscolices were monitored after marking by JC-1 dye in in vitro and in vivo studies. This work describes for the first time the successful construction of an in vivo model of E. granulosus in a small living experimental animal to achieve dynamic monitoring and observation of multiple time points of the infection course. Using this model, we quantified and analyzed labeled protoscolices based on the intensities of their red and green fluorescence. Interestingly, the ratio of red to green fluorescence intensity not only revealed the location of protoscolices but also determined the viability of the parasites in vivo and in vivo tests. The noninvasive imaging model proposed in this work will be further studied for long-term detection and observation and may potentially be widely utilized in susceptibility testing and therapeutic effect evaluation.

Test-bed evaluation of developed small constructed wetland for using in urban areas (도시지역에 적용하기 위한 소규모 인공습지 Test-bed 시설 평가)

  • Conventional construction technologies have been continually applied without consideration of its impact to the environment. This resulted to various problems including the negative responses of local citizens that regarded some constructed facilities as aversive facilities causing environmental and hydraulic problems in the urban area, etc. To prevent these problems, therefore, alternative methods should be undertaken. A new approach termed "Low Impact Development (LID)" technology is currently adapted in developed countries around the world. This study aims to investigate the efficiency of the developed small constructed wetland (SCW) with horizontal subsurface flow as a LID technique applicable in urban areas. Two test-bed facilities were constructed and monitoring had been conducted between July 2010 and June 2011. Based on the findings, the removal efficiencies achieved for TSS, $COD_{Cr}$, TN, TP, Total Fe, Total Pb for the SCW-1 were 66, 53, 46, 55, 67 and 50%, respectively. On the other hand, the SCW-2 attained 82, 62, 51, 48, 74 and 42% efficiency for TSS, $COD_{Cr}$, TN, TP, Total Fe, Total Pb, respectively. The results indicated that the removal of particulate matter and heavy metals which are considered as main pollutants from stormwater runoff in urban areas was satisfactory in the system. Therefore, the test-beds proved to be appropriate for the treatment of pollutants in urban landuses such as road, parking lot, etc. The results of this study can contribute to the conservation of aquatic ecosystems and restoration of natural water cycle in the urban areas.

The application of Fourier transform near infrared (FT-NIR) spectroscopy in the wine industry of South Africa

  • Fourier transform near infrared (FT-NIR) spectroscopy was used as a rapid method to measure the $^{o}Brix$ content and to discriminate between different must samples in terms of their fee amino nitrogen (FAN) values. FT-NIR spectroscopy was also used as a rapid method to discriminate between Chardonnay wine samples in terms of the status of the male-lactic fermentation (MLF). This was done by monitoring the conversion of malic to lactic acid and thereby determining whether MLF has started, is underway or has been completed followed by classification of the samples. Furthermore, FT-NIR spectroscopy was applied as a rapid method to discriminate between table wine samples in terms of the ethyl carbamate (EC) content. EC in wine can pose a health threat and need to be monitored by determining the EC content in relation to the regulatory limits set by the authorities. For each of the above mentioned parameters, $QUANT+^{TM}$ methods were built and calibrations derived and it was found that a very strong correlation existed in the sample set for the FT-NIR spectroscopic predictions of $^{o}Brix$ (r = 0.99, SECV = 0.306), but the correlations for the FAN (r = 0.61, SECV = 272.1), malic acid (r = 0.58, SECV = 1.06), lactic acid (r = 0.51, SECV = 1.14) and EC predictions (r = 0.47, SECV = 3.67) were not as good. Soft Independent Modeling by Class Analogy (SIMCA) diagnostics and validation was applied as a sophisticated discrimination method. The must samples could be classified in terms of their FAN values when SIMCA was applied, obtaining results with recognition rates exceeding 80%. When SIMCA diagnostics and validation were applied to determine the progress of conversion of malic to lactic acid and the EC content, again results with recognition rates exceeding 80% were obtained. The evaluation of the applicability of FT-NIR spectroscopy measurement of FAN, $^{o}Brix$ values, malic acid, lactic acid and EC content in must and wine shows considerable promise. FT-NIR spectroscopy has the potential to reduce the analytical times considerably in a range of measurements commonly used during the wine making process. Where conventional FT-NIR calibrations are not effective, SIMCA methods can be used as a discriminative method for rapid classification of samples. SIMCA can replace expensive, time-consuming, quantitative analytical methods, if not completely, at least to some extent, because in many processes it is only needed to know whether a specific cut off point has been reach or not or whether a sample belongs to a certain class or not.
