A case of Moniliformis moniliformis (Acanthocephala) infection in Iran

  • Only a few cases of Acanthocephala infections have been reported in humans, and Moniliformis moniliformis is the most common species around the world. We report here a case of infection with M. moniliformis, which passed in the stool of a 2-year-old girl in Iran. The patient had abdominal pain, diarrhea, vomiting, and facial edema. According to her mother, the patient had habit of eating dirt and once a cockroach was discovered in her mouth. In stool examination, eggs of M. moniliformis were not found. She was treated with levamisole and the clinical symptoms reduced within 2 weeks. The specimen contained 2 pieces of a female worm with a total length of 148 mm lacking the posterior end. The spiral musculature of the proboscis receptacle and the shape of the trunk allowed its generic determination. Previously 2 cases of M. moniliformis infection were reported in Iran. This is the 3rd case of M. moniliformis infection in Iran.

  • 두식물 군락에서 Complexipes moniliformis가 발견되었다. 발견된 균은 형태적으로 많은 차이점이 관찰되었으며, 이는 다른 보고와 동일한 결과를 얻었다. 그러나, 형태적인 차이점은 두식물군락에서 동일하였으나, 포자의 직경의 크기는 두 식물군락에서 유이성이 관찰되었다.

Chemical Investigation on an Endophytic fungus Gibberella moniliformis JS1055 Derived from a Halophyte Vitex rotundifolia

  • Chemical investigation of the ethyl acetate extract of Gibberella moniliformis JS1055 endophytic fungus derived from a halophyte, Vitex rotundifolia, led to the isolation of nine compounds including 7-butyl-6,8-dihydroxy-3(R)-pent-11-enylisochroman-1-one (1), 7-butyl-6,8-dihydroxy-3(R)-pentylisochroman-1-one (2), 7-butyl6,8-dihydroxy-3(R)-pentylisochroman-1-one (3), $5{\alpha},8{\alpha}$-epidioxyergosta-6,9(11),22-trien-3-ol (4), ergosterol peroxide (5), tetradecanoic acid (6), 8-O-methylfusarubin (7), nicotinic acid (8) and adenosine (9). They were identified by extensive spectroscopic data analysis including 1D, 2D ($^1H-^1H$ COSY, HSQC, HMBC) NMR, and ESIMS. All the isolates (1-9) are reported for the first time from this endophytic fungus.

Proteomic Comparison of Gibberella moniliformis in Limited-Nitrogen (Fumonisin-Inducing) and Excess-Nitrogen (Fumonisin-Repressing) Conditions

  • The maize pathogen Gibberella moniliformis produces fumonisins, a group of mycotoxins associated with several disorders in animals and humans, including cancer. The current focus of our research is to understand the regulatory mechanisms involved in fumonisin biosynthesis. In this study, we employed a proteomics approach to identify novel genes involved in the fumonisin biosynthesis under nitrogen stress. The combination of genome sequence, mutant strains, EST database, microarrays, and proteomics offers an opportunity to advance our understanding of this process. We investigated the response of the G. moniliformis proteome in limited nitrogen (N0, fumonisin-inducing) and excess nitrogen (N+, fumonisin-repressing) conditions by one- and two-dimensional electrophoresis. We selected 11 differentially expressed proteins, six from limited nitrogen conditions and five from excess nitrogen conditions, and determined the sequences by peptide mass fingerprinting and MS/MS spectrophotometry. Subsequently, we identified the EST sequences corresponding to the proteins and studied their expression profiles in different culture conditions. Through the comparative analysis of gene and protein expression data, we identified three candidate genes for functional analysis and our results provided valuable clues regarding the regulatory mechanisms of fumonisin biosynthesis.

Diversity of Endophytic Fungi Isolated from Korean Ginseng Leaves

  • We investigated the diversity of the foliar endophytes of Korean ginseng. Endophytic fungi were isolated from healthy leaves of mountain-cultivated ginseng (MCG) and field-cultivated ginseng (FCG) at 4 sites in Chungbuk Province. A total of 24 species of fungal endophytes were identified using molecular approaches. Additionally, the diversity of these endophytic fungi was compared between MCG and FCG. The major isolated endophytes were Edenia gomezpompae and Gibberella moniliformis in the MCG and FCG samples, respectively. The results suggest that ginseng endophytes have different community structures in different environments, and this understanding may prove useful in ginseng cultivation.

Endoparasites of Small Mammals in Edo State, Nigeria: Public Health Implications

  • Some small mammals occur as household pests and harbour a number of parasites that could be of public health importance. This study profiled the helminth and protozoan parasites in trapped small mammals within and around human dwelling places (houses) located across 4 major towns (Auchi, Benin, Ekpoma, and Uromi) and environs in Edo state, Nigeria. Six genera (Apodemus sp., Crocidura sp., Mastomys natalensis, Mus musculus, Rattus sp., and Sorex sp.) were identified from 502 trapped small mammals. Overall, M. musculus (71.9%) and Rattus rattus (20.1%) were the most frequently trapped. In total, on examination of blood, gastrointestinal contents, and brain tissues, 12 helminth taxa (Angiostrongylus sp., Aspicularis sp., Capillaria sp., Gongylonema sp., Heterakis spumosa, Hymenolepis diminuta, Hymenolepis nana, Mastophorus muris, Moniliformis moniliformis, Nippostrongylus brasiliensis, Strongyloides sp., Trichosomoides sp., and Trichuris sp.), and 6 protozoan parasites (Babesia sp., Trypanosoma lewisi, Plasmodium sp., Eimeria sp., Isospora sp., and Toxoplasma gondii) were isolated. Most prevalent helminths with relatively heavy mean intensity were Strongyloides sp. and Heterakis spumosa, while Plasmodium, Eimeria, and Isospora were the most prevalent protozoan parasites. Generally, intrinsic factors like sex and age had marginal influence on the rate and burden of infection in M. musculus and R. rattus. Although the infection rate and prevalence of zoonotic parasites were low, they were largely recovered in rodents from Ekpoma. This study elucidates the public health implication of the presence of zoonotic parasites in these small mammals.

종 특이 primer를 이용한 옥수수 오염 Fusarium verticillioides의 PCR 검출 (Detection of Fusarium verticillioides Contaminated in Corn Using a New Species-specific Primer)

  • Fusarium verticillioides(완전세대: Gibberella moniliformis)는 Gibberellea fujikuroi 종 복합체에 속하는 식물병원균으로서 옥수수의 줄기와 이삭에 썩음병을 일으킬 뿐 아니라 인축에 중독증을 일으키는 fumonisin 곰팡이 독소를 생산한다. 본 연구의 목적은 옥수수에 주로 발생하는 fumonisin 생성가능 G. fujikuori 종 복합체 소속 Fusarium 곰팡이 중 F. verticillioides와 그 외 F. proliferatum, F. fujikuori 등을 서로 구별할 수 있는 종 특이적 PCR primer 조합을 개발하는 것이다. RNA polymerase II beta subunit 유전자(RPB2)의 염기서열로부터 제작된 특이 primer 조합(RVERT1와 RVERT2)은 우리나라 옥수수에서 분리한 잠재적인 fumonisin 생성 G. fujikuori 종 복합체 균주 중 오직 F. verticillioides로부터 208 bp 크기의 단일 DNA 절편을 증폭하였다. 한편 F. verticillioides를 포함한 모든 조사균주는 fumonisin 생합성에 필수적인 FUM1 유전자를 포함하고 있었다. 개발된 특이 primer 조합의 검출한계는 분석 곰팡이 DNA 0.125 pg/${\mu}l$ 수준이었다. 한편, 같은 primer 조합으로 Fusarium spp.에 오염된 옥수수 시료의 게놈 DNA로부터 F. verticillioides 특이 DNA 절편이 증폭되었다. 이와 같은 결과를 종합할 때, 본 연구에서 개발된 primer 조합은 여러 곡물 시료에 오염되어 있는 F. verticillioides 균주의 검출과 종 동정에 유용하게 사용될 것이다.

Enhanced Homologous Recombination in Fusarium verticillioides by Disruption of FvKU70, a Gene Required for a Non-homologous End Joining Mechanism

  • Fusarium verticillioides (teleomorph Gibberella moniliformis) is associated with maize worldwide causing ear rot and stalk rot, and produces fumonisins, a group of mycotoxins detrimental to humans and animals. While research tools are available, our understanding of the molecular mechanisms associated with fungal virulence and fumonisin biosynthesis in F. verticillioides is still limited. One of the restraints that hampers F. verticillioides gene characterization is the fact that homologous recombination (HR) frequency is very low (<2%). Screening for a true gene knock-out mutant is a laborious process due to a high number of ectopic integrations. In this study, we generated a F. verticillioides mutant (SF41) deleted for FvKU70, a gene directly responsible for non-homologous end-joining mechanism, with the aim of improving HR frequency. Here, we demonstrate that FvKU70 deletion does not impact key Fverticillioides phenotypes, e.g., development, secondary metabolism, and virulence, while dramatically improving HR frequency. Significantly, we also confirmed that a high percentage (>85%) of the HR mutant strains harbor a desired mutation with no additional copy of the mutant allele inserted in the genome. We conclude that SF41 is suitable for use as a type strain when performing high-throughput gene function studies in F. verticillioides.

  • 포장(圃場)에서 키다리병(病)에 걸린 줄기와 종자(種子)로 부터 얻은 자낭각이 어느 종류(種類)의 Gibberella에 존재(存在)하며, 수확후(收穫後)에 얻은 자낭각의 생존(生存)여부를 조사(調査)하기 위하여 처리한 결과(結果)는 다음과 같다. 1. 종자소독(種子消毒)을 하지 않은 포장(圃場)에서 키다리병(病)의 이병율(罹病率)은 평균(平均) 12%이었다. 2. 수확전(收穫前)에 얻은 자낭각은 키다리병의 자낭각이 적미병의 자낭각보다 많았으나 종자(種子)나 봄에 채집한 자낭각은 거의 적미병에 있었다. 3. 키다리병에 이병된 벼에서 채종한 종자(種子)에서 키다리병, 적미병, Ophiobolus sp.는 10, 25, 8%로 나타났으며 이병(罹病)된 줄기로부터 얻은 포자(胞子)로 접종(接種)한 종자(種子)에서는 3, 10, 2%로 나타났다. 4. 적미병자낭각을 야외 땅속 실내 집눌속에 놓고 5개월이 지난다음 생존(生存)여부를 조사(調査)한 결과(結果) 야외에 놓은 것만 발아하지 않았다.

벼 종자에서 분리한 Fusarium속 균주들의 prochloraz에 대한 저항성 검정 및 교차 저항성 조사 (Detection for the Resistance of Fusarium spp. Isolated from Rice Seeds to Prochloraz and Cross-resistance to Other Fungicides Inhibiting Sterol Biosynthesis)

  • Fusarium spp. 균주들에 대한 Prochloraz의 살균효과를 비교하기 위하여 한천희석법을 사용하여 각 균주에 대한 prochloraz의 $EC_{50}$값을 구하였다. 전체 균주에 대한 평균 $EC_{50}$값은 $0.25{\mu}g\;mL^{-1}$이었으며, 최저값과 최고값은 0.02과 $1.78{\mu}g\;mL^{-1}$이었다. Fusarium의 종별로 조사한 평균 $EC_{50}$값을 보면 F. fujikuroi의 경우가 $0.31{\mu}g\;mL^{-1}$, F. moniliformis와 F. proliferatum의 경우는 0.09과 $0.11{\mu}g\;mL^{-1}$이었다. 실험에 사용한 36개 Fusarium 속 균주의 Prochloraz에 대한 감수성과 저항성을 판단할 수 있는 기준 $EC_{50}$ 값은 $0.5{\mu}g\;mL^{-1}$가 타당하며, 이 기준에 의한 저항성 균주의 분리 비율은 13.8%이었다. Prochloraz에 대한 저항성과 Fusarium 균주의 병원성간에는 전혀 상관관계가 없었다. 또한 prochloraz에 대해서 저항성인 균주들은 triflumizole, hexaconazole, difenoconazole, tebuconazole과 교차 저항성을 보이지 않았다.
