Polyurethane (PU) was prepared with the compositions of polytetramethylene glycol (PTMG) having two different molecular weight (250, 1000 g/mol). The optimum composition of PTMG 250/1000 was 60/40 based on the mechanical properties. PU/organoclay nanocomposites were prepared with several kinds of organoclay. The mechanical properties of nanocomposite prepared with 93A were considerable. The improvement in tensile strength and modulus for PU/organoclay nanocomposite with the application of ultrasound compared to the PU/organoclay nanocomposite without the application of ultrasound was factors of 1.2, and hardness (shore A type) increased from 90 to 95. The difference in thermal degradation was not observed. The results of transmission electron micrographs and X-ray measurements suggest that the intercalated organoclay in PU matrix was observed.
This study was undertaken to find out the effect of phytate on the protein digestibility of various soybean foods, including soy milk, bean curd, curd residue, cheongkukjang, soy sauce, and soy paste. The phytate content of soybean was 2.4%, which decreased to 0.2%, 0.7%, and 0.4% in soy milk, bean curd, and curd residue, respectively, and to 0.2% and 1.0% in soy sauce and soy paste, respectively. The phytate/protein ratio was not correlated with protein digestibility by pepsin whereas the ratio was highly correlated with pancreatin digestibility (p<0.01, r= -0.73). According to SDS-PAGE for the soluble protein fractions, soaked bean showed an alteration in soluble components and bean curd residue exihibited newer low molecular weight bands. Fermented soy products showed no protein band, likely due to degradation.
Psychrotrophic bacterial strains utilizing crude oil as their sole carbon and energy sources were isolated from Antarctic soil and sea sediments. One of the strains named AI-I showed the hightest activity for emulsification of crude oil and the best growth. This strain was identified as Acinetobacter calcoaceticus. A. calcoaceticus AI-I strain contains a plasmid (OCT plasmid) which was related to the utilization of alkane compounds. The molecular weight of this plasmid was estimated to be about 110 Md by agarose gel electrophoresis. The cured strain of A. calcoaceticus AI-I strain (OCT ) was not able to utilize normal hydrocarbon compounds ($C_6C_{17}$) as carbon and energy sources. A. ca/coaceticus AI-1 was resistant to ampicillin and sensitive to streptomycin, kanamycin, chloramphenicol, tetracycline. The results suggested that this strain carries a plasmid (OCT) responsible for oil utilization which is quite stable and might be concerned with antibiotics resistancy.

We investigated the recycling method to re-plasticize cross-linked polyethylene by using supercritical methanol. The cross-linked polyethylene is successfully fragmented to thermoplasticized polyethylene with little degradation reactions in supercritical fluids. The thermo-plasticization reaction was accelerated with increase in temperature in the range from $360^{\circ}C$ to $400^{\circ}C$, resulting in decrease in crosslinking density, molecular weight and mechanical properties. However, the thermoplasticized polyethylene at $360^{\circ}C$ showed comparable tensile strength and impact strength with a raw resin of crosslinked polyethylene. Chemical structure of main chain of polyethylene was not affected by reaction condition.
The collagenase gene from Vibrio parahaemolyticus 04 was subcloned into an expression vector pET-29b. The recombinant collagenase was expressed in Escherichia coli BL2l(DE3) and partially purified by Hi-Trap affinity and Sephacryl S-100 size exclusion chromatographies. The recombinant enzyme was purified by 43.7-fold and the yield was 73%. SDS-PAGE revealed that the molecular weight of the enzyme was approximately 35 kDa. Substrate specificity study of the enzyme displayed that the enzyme showed the highest activity with the type I collagen and the synthetic peptide, Z-GPGGPA, indicating that the enzyme was indeed a collagenase. The enzyme showed broad pH optimum around pH 6-12 and was stable between pH 5.5 and 11.5. The optimum temperature for the type I collagen degradation was $35^{\circ}C$. The thermostability measurement of the enzyme indicated that the enzyme was stable up to $55^{\circ}C$, but the activity was diminished quickly above $60^{\circ}C.$
Pseudomonas sp. K82 has been reported to be an aniline-assimilating soil bacterium. However, this strain can use not only aniline as a sole carbon and energy source, but can also utilize benzoate, p-hydroxybenzoate, and aniline analogues. The strain accomplishes this metabolic diversity by using dif-ferent aerobic pathways. Pseudomonas sp. K82, when cultured in p-hydroxybenzoate, showed extradiol cleavage activity of protocatechuate. In accordance with those findings, our study attempted the puri-fication of protocatechuate 4,5-dioxygenase (PCD 4,5). However the purified PCD 4,5 was found to be very unstable during purification. After Q-sepharose chromatography was performed, the crude enzyme activity was augmented by a factor of approximately 4.7. From the Q-sepharose fraction which exhibited PCD 4,5 activity, two subunits of PCD4,5 (${\alpha}$ subunit and ${\beta}$ subunit) were identified using the N-terminal amino acid sequences of 15 amino acid residues. These subunits were found to have more than 90% sequence homology with PmdA and PmdB of Comamonas testosteroni. The molecular weight of the native enzyme was estimated to be approximately 54 kDa, suggesting that PCD4,5 exists as a het-erodimer (${\alpha}$$_1$${\beta}$$_1$). PCD 4,5 exhibits stringent substrate specificity for protocatechuate and its optimal activity occurs at pH 9 and 15 $^{\circ}C$. PCR amplification of these two subunits of PCD4,5 revealed that the ${\alpha}$ subunit and ${\beta}$ subunit occurred in tandem. Our results suggest that Pseudomonas sp. K82 induced PCD 4,5 for the purpose of p-hydroxybenzoate degradation.

Thermal degradation characteristics of polypropylene and polystyrene have been studied in the thermogravimetric(TG) reactor and batch-type microreactor. The dynamic thermogravimetric curve of TG provided a valuable information about pyrolysis temperature. It was found that PS was thermally degraded at lower temperature of $30{\sim}50^{\circ}C$ than PP. It was found that the yield and molecular weight of liquid product in the microreactor were decreased with the increase of reaction temperature and time in the case of PP. The production of styrene monomer was significantly increased by the promotion of depolymerization with the increase of temperature and time. The chain-end scission rate parameters were determined to be 50.0 kcal/mole of PP, 45.2 kcal/mole of PS by the Arrhenius plot.
1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU, carmustine) is one of the effective chemotherapeutic agents which has been used clinically for treating malignant glioma. Poly(D,L-lactide-co-glycolide) (PLGA, molecular weight: 20000 g/mole. mole ratio of lactide to glycolide 75 : 15) is a well known biodegradable polymer used as a drug carrier for drug delivery system. In this study, we investigated the BCNU release behaviour of BCNU-loaded PLGA wafers containing poly (N-vinylpyrrolidone) (PVP) or polyethyleneoxide (PEO) and the effect of hydrophilic polymers incoporated in the wafers. BCNU-loaded PLGA microparticles with or without hydrophilic polymers were prepared by a spray drying method and fabricated into wafers by direct compression. Encapsulation efficiency of BCNU-loaded PLGA microparticles containing PVP and PEO was 85 ∼ 97% and crystallinity of BCNU encapsulated in PLGA decreased significantly initial release amount and release rate of BCNU increased with the increasing PVP or PEO amount. Morphological change and mass loss of wafers during the release test were confirmed that hydration and degradation of PLGA would be facilitated with an increase of hydrophilic polymers.
In order to obtain more detailed information concerning the degradation of lignin in the oxygen alkali pulping single stage isothermal delignification of pine wood meal (Pinus koraiensis S. et Z.) was studied in the oxygen alkali system at five temperature level ($110^{\circ}C$, $120^{\circ}C$, $130^{\circ}C$, $140^{\circ}C$, $150^{\circ}C$) for 60 min.. The rate constant, activation energy, oxygen and alkali consumption during the oxygen alkali delignification were determined by the kinetic method. The 2/5 of total lignin was eliminated at the start of the reaction. The delignification rate constant was about 3 times that of caustic soda pulping. The activation energy was about 1/3 lower than in caustic soda pulping. Like oxygen consumption, alkali consumption was also rapid early at the reaction and almost ceased after about 10 min.. The degradation reaction of lignin was strongly dependent upon the pH decrease of the cooking liquor by organic acid generated in pulping. The lignin in the oxygen alkali pulping degraded into lover molecular weight and had more hydrophillic properties. The methoxyl group decreased considerably at the first of oxygen alkali delignification, while the carbonyl, carboxyl and phenolic hydroxyl group increased rapidly.
Antheraea yamamai silk fibroin powder was prepared by treatment with HCl. The prepared A. yamamai fibroin hydrolysate was characterized by gel filtration chromatography, amino acid analysis, X-ray diffractometry, circular dichroism analysis, differential thermal analysis, and thermogravimetry. The average molecular weight of A. yamamai powder was about 430 and the major amino acids composed of the powder were Ala and Ser. According to XRD analysis, A. yamamai silk powder showed sharp diffraction peaks at $2{\theta}=20.34^{\circ}\;and\;31.5^{\circ}$. CD spectrum showed a peak around 220 nm and a should 215 nm, assigned to ${\alpha}-helix\;and\;{\beta}-sheet$ structure, respectively. DSC and TGA showed that the maximum degradation temperature of powder was around $250{\sim}270^{\circ}C$. Moreover, no harmful heavy metal was contained in the A. yamamai silk fibroin hydrolysate.
