Effect of Poly(ethylene glycol) dimethyl ether Plasticizer on Ionic Conductivity of Cross-Linked Poly[siloxane-g-oligo(ethylene oxide)] Solid Polymer Electrolytes

  • Cross-linked network solid polymer electrolytes were prepared by means of in situ hydrosilylation between poly[hydromethylslioxane-g-oligo(ethylene oxide)] and diallyl or triallyl group-containing poly(ethylene glycols). The conductivities of the resulting polymer electrolytes were greatly enhanced upon the addition of poly(ethylene glycol) dimethyl ether (PEGDME) as an ion-conducting plasticizer. Conductivities of the cross-linked polymer electrolytes were more dependent on the molecular weight of PEGDME than on the cross-linkers. The maximum conductivity was found to be 5.6${\times}$10$\^$-4/ S/cm at 30$^{\circ}C$ for the sample containing 75 wt% of PEGDME (M$\_$n/ =400). These electrolytes exhibited electrochemical stability up to 4.5 V against the lithium reference electrode. We observed reversible electrochemical plating/stripping of lithium on the nickel electrode.

Mono and Multilayer Assembly of Zeolite Microcrystals on Substrates

  • We have shown that zeolite microcrystals can be readily organized in the form of uniformly oriented monoand multilayers on various substrates by well-defined chemical linkages based on covalent, ionic, and hydrogen bondings between the microcrystals and the substrates. This finding establishes the fact that micrometer-scale building blocks can be readily organized into organized entities through interconnection of the surface-tethered large number of functional groups. Since zeolite crystals have highly regular and uniform nanochannels and nanopores within them, the resulting mono and multilayers of zeolite microcrystals bear great potential to be utilized in various novel applications.

  • QDs-LEDs(quantum dot light emitting device) should contain well-organized arrays of QDs on an electron transport layer. Thin films of CdSe/ZnS core-shell QDs were successfully fabricated on $TiO_2$ substrates by using PDMS stamp and micro contact printing method. 2-Carboxyethylphosphonic acid(CAPO) and 1,6-hexanedithiol(HDT) were employed as molecular linkers in assembling CdSe/ZnS core-shell QDs with high-density and uniform array. The CAPO increased the binding strength between the QDs and the substrates, and the HDT induced the strong inter-particle attractions of assembled QDs. The assembling properties of QDs thin films were characterized by SEM, AFM, optical microscope and photoluminescence spectroscope(PL).

Identification of Novel Target Proteins of Cyclic GMP Signaling Pathways Using Chemical Proteomics

  • For deciphering the cyclic guanosine monophosphate (cGMP) signaling pathway, we employed chemical proteomics to identify the novel target molecules of cGMP. We used cGMP that was immobilized onto agarose beads with linkers directed at three different positions of cGMP. We performed a pull-down assay using the beads as baits on tissue lysates and identified 9 proteins by MALDI-TOF (Matrix-Assisted Laser Desorption/Ionization Time-of-Flight) mass spectrometry. Some of the identified proteins were previously known cGMP targets, including cGMP-dependent protein kinase and cGMP-stimulated phosphodiesterase. Surprisingly, some of the co-precipitated proteins were never formerly reported to associate with the cGMP signaling pathway. The competition binding assays showed that the interactions are not by nonspecific binding to either the linker or bead itself, but by specific binding to cGMP. Furthermore, we observed that the interactions are highly specific to cGMP against other nucleotides, such as cyclic adenosine monophosphate (cAMP) and 5'-GMP, which are structurally similar to cGMP. As one of the identified targets, MAPK1 was confirmed by immunoblotting with an anti-MAPK1 antibody. For further proof, we observed that the membrane-permeable cGMP (8-bromo cyclic GMP) stimulated mitogen-activated protein kinase 1 signaling in the treated cells. Our present study suggests that chemical proteomics can be a very useful and powerful technique for identifying the target proteins of small bioactive molecules.

DNA Structural Perturbation Induced by the CPI-Derived DNA Interstrand Cross-linker : Molecular Mechanisms for the Sequence Specific Recognition

  • The highly potent cytotoxic DNA-DNA cross-linker consists of two cyclopropa[c]pyrrolo[3,4-3]indol-4(5H)-ones insoles [(+)-CPI-I] joined by a bisamido pyrrole (abbreviated to "Pyrrole"). The Pyrrole is a synthetic analog of Bizelesin, which is currently in phase II clinical trials due to its excellent in vivo antitumor activity. The Pyrrole has 10 times more potent cytotoxicity than Bizelesin and mostly form DNA-DNA interstrand cross-links through the N3 of adenines spaced 7 bp apart. The Pyrrole requires a centrally positioned GC base pair for high cross-linking reactivity (i.e., $5^1$-T$AT_2$A*-$3^1$), while Bizelesin prefers purely AT-rich sequences (i.e., $5^1$-T$AT_4$A*-$3^1$, where /(equation omitted) represents the cross-strand adenine alkylation and A* represents an adenine alkylation) (Park et al., 1996). In this study, the high-field $^1$H-NMR and rMD studies are conducted on the 1 1-mer DNA duplex adduct of the Pyrrole where the 5′(equation omitted)TAGTTA*-3′sequence is cross-linked by the drug. A severe structural perturbation is observed in the intervening sequences of cross-linking site, while a normal B-DNA structure is maintained in the region next to the drug-modified adenines. Based upon these observations, we propose that the interplay between the bisamido pyrrole unit of the drug and central C/C base pair (hydrogen-bonding interactions) is involved in the process of cross-linking reaction, and sequence specificity is the outcome of those interactions. This study suggests a mechanism for the sequence specific cross-linking reaction of the Pyrrole, and provides a further insight to develop new DNA sequence selective and distortive cross-linking agents.

  • To find out rational design and synthetic strategies toward efficient hydrogen storage materials, molecular modeling and quantum mechanical studies have been carried out on the MOFs(Metal-Organic Frameworks) having various organic linkers and nanocube frameworks. The calculation results about the free volume ratio, surface area, and electron density variation of the frameworks indicated that the capacity of the hydrogen storage of MOFs was largely dependent on the specific surface area and electron localization around benzene ring rather than the free volume of MOFs. The prediction of the modeling study could be supported by the hydrogen adsorption experiments using IRMOF-1 and -3, which showed more enhanced hydrogen storage capacities of IRMOF-3 compared with the IRMOF-1's at both experimental conditions, 77K, ∠ $H_2$ 1 atm and ambient temperature, ∠ $H_2$ 35 atm.

Preparation of Immunotoxin Herceptin-Botulinum and Killing Effects on Two Breast Cancer Cell Lines

  • Background: Worldwide, breast cancer is the most common cancer diagnosed among women and a leading cause of cancer deaths. The age of onset in Iran has become reduced by a decade for unknown reasons. Herceptin, a humanized monoclonal antibody, is a target therapy for breast cancer cells with over expression of HER2-neu receptors, but it is an expensive drug with only 20% beneficial rate of survival. This study introduces a novel approach to enhance the efficacy of this drug through immunoconjugation of the antibody to botulinum toxin. Decreasing the cost and adverse effects of the antibody were secondary goals of this study. Materials and Methods: Botulinum toxin was conjugated with Herceptin using heterobifunctional cross linkers, succinimidyl acetylthiopropionate (SATP) and sulfo-succinimidyl-4-(N-maleimidomethyl) cyclohexane-1-carboxylate (SMCC) according to the supplier's guidelines and tested on two breast cancer cell lines: SK-BR-3 and BT-474. Toxin and Herceptin were also used separately as controls. The cytotoxicity assay was also performed using the new bioconjugate on cultured cells with Alamar blue and a fluorescence plate reader. Results: Herceptin-Toxin bioconjugation significantly improved Herceptin efficacy on both breast cancer cell lines when compared to the control group. Conclusions: Toxin-Herceptin bioconjugation can be a potential candidate with increased efficiency for treating breast cancer patients with over expression of the HER2 receptor.

Ultrasound-Aided Monolayer Assembly of Spherical Silica Nanobeads (초음파 방법을 이용한 실리카 나노비드의 단층 정렬에 관한 연구)

  • Uniform silica nanobeads were synthesized by St$\ddot{o}$ber method and assembled in the form of monolayer on glass substrate using sonication method. Before the assembly of silica nanobeads, glass substrates were treated with molecular linkers, such as CP-TMS and PEI, and nanobeads were dispersed in toluene. In attachment test, SO (sonication without stacking) method and SS (sonnication with stacking) method was used and sonication time was controled. After the experiment, microbalance was use to measure deviation between before and after the attachment experiment then calculate percent of coverage. Minutely observe with SEM (Scanning Electron Microscopy) then select the most close-packed and monolayer assembled cover glass and calculate DOC (Degree of Coverage). In SO method, DOC increased very sharply and reach over 140 percent point, also got lots of multi-layer region. On the other hand, in SS method DOC increased slower than SO method but more close-packed and monolayer assembled.

Recent Progress on Metal-Organic Framework Membranes for Gas Separations: Conventional Synthesis vs. Microwave-Assisted Synthesis (기체분리용 금속유기구조체 분리막의 최근 연구 동향 및 성과)

  • Metal-organic frameworks (MOFs) are nanoporous materials that consist of organic and inorganic moieties, with well-defined crystalline lattices and pore structures. With a judicious choice of organic linkers present in the MOFs with different sizes and chemical groups, MOFs exhibit a wide variety of pore sizes and chemical/physical properties. This makes MOFs extremely attractive as novel membrane materials for gas separation applications. However, the synthesis of high-quality MOF thin films and membranes is quite challenging due to difficulties in controlling the heterogeneous nucleation/growth and achieving strong attachment of films on porous supports. Microwave-based synthesis technology has made tremendous progress in the last two decades and has been utilized to overcome some of these challenges associated with MOF membrane fabrication. The advantages of microwaves as opposed to conventional synthesis techniques for MOFs include shorter synthesis times, ability to achieve unique and complex structures and crystal size reductions. Here, we review the recent progress on the synthesis of MOF thin films and membranes with an emphasis on how microwaves have been utilized in the synthesis, improved properties achieved and gas separation performance of these films and membranes.

Effects of Silicone Polymer Blends on Physical Properties of Dental Polyvinylsiloxane Impression Materials (실리콘폴리머의 혼합사용이 폴리비닐실록산 치과용 인상재의 물성에 미치는 영향)

  • The purpose of this study was to investigate the effects of combined use of several types of silicone polymers on the physical properties of the dental polyvinylsiloxane impression materials (PVS). Four types of silicone prepolymers having different molecular weight and vinyl group position, and two types of cross-linkers having differently located silyl hydride functional groups were used in various combinations for the formulation. The samples containing bimodal or trimodal prepolymers showed higher tensile strength, elongation at break, and elastic deformation than those containing only one type of prepolymer. The samples using CR210 cross-linker which has side- and terminal-silyl hydride groups showed higher elastic deformation and elongation at break than those using CR101 cross-liker which has side-only silyl hydride group. High vinyl content prepolymer having side vinyl group delayed setting even though it enhanced tensile strength. Further studies are needed to clarify the specific role of this component on setting time and to find appropriate controlling methods for making improved PVS with optimum workability.
