Microvascular Decompression for Familial Hemifacial Spasm : Single Institute Experience

  • Objective : The purpose of this study was to evaluate the characteristics and surgical outcomes of familial hemifacial spasm (HFS) and to discuss the role of genetic susceptibility. Methods : Between 2001 and 2011, 20 familial HFS patients with ten different pedigrees visited our hospital. The data from comprehensive evaluation of these patients, including clinical, radiological and electrophysiological data and surgical outcomes were reviewed to characterize familial HFS and to compare the characteristics between familial HFS and sporadic HFS. Results : According to the family tree, the inheritance pattern was difficult to define clearly using these data. Radiologic findings suggested that the vertebral artery (VA) was a more frequent offender in familial HFS than in sporadic cases (35.0% vs. 10.0%, p<0.001). Chi-square test showed that there were no correlation between VA tortuosity and underlying morbidity such as diabetes or hypertension (p=0.391). Eighteen out of 19 patients who underwent microvascular decompression showed no residual spasm. Other features of familial HFS overlap with sporadic cases. These findings suggest that certain genetic susceptibilities rather than hypertension or diabetes may influence vascular tortuosity and HFS development. Conclusion : In this study, familial HFS seems not so different from sporadic cases. Authors thought familial HFS could have heterogeneous etiology. Further study of familial HFS including clinical, anatomic, genetic, and molecular information may help identify a gene or trait that can provide insight into the mechanisms of sporadic and familial HFS.

Population genetic analysis of Salurnis marginella (Hemiptera: Flatidae)

  • Salurnis marginella Guérin-Méneville, 1829 (Hemiptera: Flatidae) is an invasive species first reported in 2003 in Iksan, which is located in the mid-western region of South Korea, and subsequently found in the nearby regions in 2005. However, molecular-perspective reports on their invasive characteristics are not yet available. In this study, population genetic characteristics of Korean S. marginella were evaluated using the mitochondrial COI region and sequencing 124 individual samples collected in 11 Korean localities. A total of 12 haplotypes were identified with a maximum sequence divergence of 1.368% (9 bp). Haplotype diversity was relatively higher than that of other insect species invaded into Korea, providing 2-6 haplotypes per populations, indicating that introduction to Korea may have happened rather extensively and consistently. Nucleotide diversity (π) was the highest in Iksan but owing to the limited sample size (three individuals) from this locality, additional studies are required for drawing conclusive inference regarding the place of entry to Korea. Ulsan, the easternmost population in the present study, revealed nearly the lowest diversity estimates, such as the lowest H and the second-lowest π; a unique haplogroup with a higher frequency; and an independent genetic cluster, suggesting that the introduction of S. marginella to Ulsan was an independent event. Further collection in Korea and neighboring countries, including the original distributional range is necessary to elucidate the invasive dynamics of S. marginella

Prevalence and Genetic Characteristics of Japanese Encephalitis Virus among Mosquitoes and Pigs in Hunan Province, China from 2019 to 2021

  • Japanese encephalitis virus (JEV), the causative agent of Japanese encephalitis (JE), is an importantly zoonotic, vector-borne virus widely prevalent in Asia. Although JE has been well controlled in China, its prevalence remains a huge threat to the pig industry as well as human health. Herein, we report on our molecular and serological investigations of JEV among pigs from different regions in Hunan Province of China from 2019 to 2021. Collectively, 19.27% (583/3026, 95% Confidential Interval (CI) 17.86-20.68) of sampled pigs were positive for JEV IgG antibody as revealed by indirect enzyme-linked immunosorbent assay, and the seroprevalence of JEV among pigs was significantly associated with the development stage and breeding scale (p < 0.01). Meanwhile, 10.99% (42/382, 95% CI 7.86-14.13) of tissue samples of pigs with suspected clinical symptoms of JE and 23.44% (15/64, 95% CI 13.06-33.82) of mosquito batches were JEV-positive via reverse polymerase chain reaction. In addition, the complete E gene sequences of 14 JEV strains identified in this study were amplified and sequenced. Phylogenetic analysis showed that all 14 JEV strains belonged to genotype I-b and displayed a distinct genetic relationship to the present JEV vaccine strain (SA14-14-2). In conclusion, our results revealed not only the severe prevalence of JEV in Hunan Province, but also that JEV I-b might be the predominant genotype in Hunan Province, suggesting therefore that effective measures for JE control are urgently needed.

Characterization of a Brown Rot Fungus Isolated from Dwarf Flowering Almond in Korea

  • The fruits showing brown rot symptom on dwarf flowering almond were found in Gongju, Chungchungnam-Do in Korea in July 2005. Small water-soaked lesions on the fruits were initiated, and gradually developed to soft rot covered with gray conidia. Then the diseased fruits were shrunk and became grayish-black mummies. A fungus was isolated from the diseased fruit and its morphological, cultural and molecular genetic characteristics were investigated. Typical blastospores of Monilinia spp. were observed under a light microscope both from tissues of the diseased fruits and from PDA-grown cultures. The fungus grew well at $25^{\circ}C$ and on PDA. The ITS ribosomal DNA region (650 bp) of the fungus was amplified by PCR and analyzed. Comparative data on ITS sequence homology among Monilinia spp., ITS sequence-based phylogram and morphological characteristics showed that the fungus is Monilinia fructicola. This is the first report on Monilinia fructicola causing brown rot on fruits of dwarf flowering almond in Korea.

AFLP를 이용한 단감나무 탄저병 병원균 Colletotrichum spp.의 유연관계 분석 (Analysis of genetic relationships of Colletotrichum spp. isolated from sweet persimon with AFLP)

  • Colletotrichum spp.는 광범위한 기주 범위를 갖는 다범성 균으로 각종 작물에 피해를 야기시키는 중요한 식물병원 진균이다. 최근 국내에서 널리 재배되고 있는 단감, 사과, 복숭아, 포도 등에 탄저병이 발생하여 많은 경제적 손실을 초래하고 있다. 탄저병 병원균의 경우 기존에는 주로 형태적 특성이나 배지상에서의 특성, 기주에 대한 병원성의 차이에 의존하여 분류를 해 왔다. 그러나 최근에는 병원균의 분류에 있어 문제점을 해결하기 위하여 분자생물학적 방법을 이용하고 있다. 최근에는 RAPD(ramdomly amplified polymorphic DNAs)와 RFLP(restriction fragment length polymorphism)의 장점만을 살린 AFLP(amplified fragment length polymorphism)기법이 유연관계 분석에 있어서 각광을 받고 있고 rDNA부위를 증폭하여 제한효소를 이용하여 다양성을 보는 방법이 많이 이용되고 있다. 이에 본 실험에서는 AFLP 기법 이용하여 단감나무에 탄저병을 일으키는 균들간에 유연관계를 밝혔다. AFLP결과에서 variation이 심하여 각각의 특징적인 band를 검출할 수 있었다. 특히 단감나무에 병해를 일으키는 탄저병 병원균 2개의 종이 복합적으로 관여하는 사실을 알 수 있었다.

Differences and Variations among Anguilla japonica, Muraenesox cinereus and Conger myriaster from the Yellow Sea

  • Genomic DNAs were extracted from the muscle of twenty-one specimens of three eel species collected in Anguilla japonica (AJ), Muraenesox cinereus (MC) and Conger myriaster (CM) from the Yellow Sea, respectively. In the present study, 7 oligonucleotides primers generated 191 specific loci in the AJ species, 226 in the (MC) species and 181 in the CM species, respectively. The primer BION-02 generated the most loci (a total of 83), with an average of 11.86 in the AJ species. The specific loci generated by oligonucleotides primers exhibited inter-individual-specific characteristics, thus revealing DNA polymorphisms. With regard to average bandsharing value (BS) results, individuals from Conger myriaster species (0.808) exhibited higher bandsharing values than did individuals from Muraenesox cinereus species (0.729) (P<0.05). The longest genetic distance (0.430) displaying significant molecular difference was also between individual no. 01 within Anguilla japonica eel species and individual no. 04 within Anguilla japonica species. In this study, the dendrogram resulted from reliable seven oligonucleotides primers, indicating three genetic clusters composed of group I (ANGUILLA 01~ANGUILLA 07), group II (MURAENESOX 08~MURAENESOX 14) and group III (CONGER 15~CONGER 21). The existence of species differentiation and DNA polymorphisms among three eel species were detected by PCR analysis. As mentioned above, a dendrogram revealed close relationships between individual identities within three eel species. High levels of a significant genetic distance among three eel species showed this PCR approach is one of the most suitable tools for individuals and/or species biological DNA studies.

ITS2 부위의 염기서열 및 RAPC-PCR에 의한 Pseudo-nitzschia 4종의 유연관계 (Phylogenetic Relationships Using ITS2 Sequence and RAPD-PCR Data from Four Species of Korean Pseudo-nitzschia (Bacillariophyceae))

  • ITS2 부위를 시퀀싱하여 Pseudo-nitzschia delcatissima, P. multiseries, P. pungens, P. subfraudulenta 상호간의 유전자 다양도를 조사함과 아울러 RAPD-PCR pattern을 이용하여 유사도를 구하였다. 유전자 거리를 근거로 했을 때 P. delicatissima 종은 P. multiseries와 P. pungens와는 유전적 거리가 상당히 요원하였고, 심지어 P. subfraudenlta와도 거리를 보였다. 유사도의 경 P. multiseries와 P. pungens는 0.31로 보인 반면에, P delicatissima는 다른 세종과 0.81를 나타내었다. 따라서 P. delicatissima 종은 P. multiseries, P. pungens, P. subfraudulenta와는 유전적으로 밀접하지 않는 관계로 보였다. ITS2부위는 Pseudo-nitzschia 동정에 사용될 수 있는 유용한 도구로 보이며 형태적으로 구분할 수 없는 P. multiseries와 P. pungens을 구분할 수 있다. 또한 RAPD-PCR 방법도 단시간에 Pseudo-nitzchia을 분리시키는데 사용될 것으로 보인다.

Conservation Biology of Endangered Plant Species in the National Parks of Korea with Special Reference to Iris dichotoma Pall. (Iridaceae)

  • The aim of this study was to provide basic guidelines for conservation and management of endangered plants in the national parks of Korea. Iris dichotoma Pall. (Iridaceae), which is a popular garden plant, is considered a second-class endangered species by Korean government and it is listed as a EN (Endangered) species in Red Data Book of Korea. We analyzed ecological conditions of I. dichotoma habitats based on vegetation properties and soil characteristics. This species which is known to inhabit in grassland adjacent to the ocean of lowlands slope and its population was located at an elevation of 8 m to 11 m. In the study sites, the mean of soil organic matter, total nitrogen and soil pH were 6.16%, 0.234% and 5.39 respectively. Additionally, the genetic variation and structure of three populations were assessed using ISSR (Inter Simple Sequence Repeat) markers. The genetic diversity of I. dichotoma (P = 59.46%, H = 0.206, S = 0.310) at the species level was relatively high. Analysis of molecular variance (AMOVA) showed 82.1% of the total genetic diversity was occurred in within populations and 17.9% variation among populations. Lastly, we developed predicted distribution model based on climate and topographic factors by applying SDMs (Species Distribution Models). Consequently, current status of I. dichotoma habitats is limited with natural factors such as the increase of the coverage rate of the herbs due to ecological succession. Therefore, it is essential to establish in situ and ex situ conservation strategies for protecting natural habitats and to require exploring potential and alternative habitats for reintroduction.

Monitoring of genetically close Tsaiya duck populations using novel microsatellite markers with high polymorphism

  • Objective: A set of microsatellite markers with high polymorphism from Tsaiya duck were used for the genetic monitoring and genetic structure analysis of Brown and White Tsaiya duck populations in Taiwan. Methods: The synthetic short tandem repeated probes were used to isolate new microsatellite markers from the genomic DNA of Tsaiya ducks. Eight populations, a total of 566 samples, sourced from Ilan Branch, Livestock Research Institute were genotyped through novel and known markers. The population genetic variables were calculated using optional programs in order to describe and monitor the genetic variability and the genetic structures of these Tsaiya duck populations. Results: In total 24 primer pairs, including 17 novel microsatellite loci from this study and seven previously known loci, were constructed for the detection of genetic variations in duck populations. The average values for the allele number, the effective number of alleles, the observed heterozygosity, the expected heterozygosity, and the polymorphism information content were 11.29, 5.370, 0.591, 0.746, and 0.708, respectively. The results of analysis of molecular variance and principal component analysis indicated a contracting Brown Tsaiya duck cluster and a spreading White Tsaiya duck cluster. The Brown Tsaiya ducks and the White Tsaiya ducks with Pekin ducks were just split to six clusters and three clusters when K was set equal to 6 and 3 in the Bayesian cluster analysis. The individual phylogenetic tree revealed eight taxa, and each individual was assigned to its own population. Conclusion: According to our study, the 24 novel microsatellite markers exhibited a high capacity to analyze relationships of inter- and intra-population in those populations with a relatively limited degree of genetic diversity. We suggest that duck farms in Taiwan could use the new (novel) microsatellite set to monitor the genetic characteristics and structures of their Tsaiya duck populations at various intervals in order to ensure quality breeding and conservation strategies.

Survey of genetic structure of geese using novel microsatellite markers

  • Objective: The aim of this study was to create a set of microsatellite markers with high polymorphism for the genetic monitoring and genetic structure analysis of local goose populations. Methods: Novel microsatellite markers were isolated from the genomic DNA of white Roman geese using short tandem repeated probes. The DNA segments, including short tandem repeats, were tested for their variability among four populations of geese from the Changhua Animal Propagation Station (CAPS). The selected microsatellite markers could then be used to monitor genetic variability and study the genetic structures of geese from local geese farms. Results: 14 novel microsatellite loci were isolated. In addition to seven known loci, two multiplex sets were constructed for the detection of genetic variations in geese populations. The average of allele number, the effective number of alleles, the observed heterozygosity, the expected heterozygosity, and the polymorphism information content were 11.09, 5.145, 0.499, 0.745, and 0.705, respectively. The results of analysis of molecular variance and principal component analysis indicated a contracting white Roman cluster and a spreading Chinese cluster. In white Roman populations, the CAPS populations were depleted to roughly two clusters when K was set equal to 6 in the Bayesian cluster analysis. The founders of private farm populations had a similar genetic structure. Among the Chinese geese populations, the CAPS populations and private populations represented different clads of the phylogenetic tree and individuals from the private populations had uneven genetic characteristics according to various analyses. Conclusion: Based on this study's analyses, we suggest that the CAPS should institute a proper breeding strategy for white Roman geese to avoid further clustering. In addition, for preservation and stable quality, the Chinese geese in the CAPS and the aforementioned proper breeding scheme should be introduced to geese breeders.
