선천성 정신지체가 있는 der(8)t(8;13)(p23.3;q32.1) 핵형의 성인여성 (Unbalanced translocation der(8)t(8:13)(p23.3;q32.1)dn identified by array CGH and subtelomeric FISH in a patient with mental retardation)

  • 염색체 말단부위의 결실, 혹은 중복 등의 재배열은 정신지체나 기형의 중요한 원인이 되며, 정신지체 환자 2,500명에서 5%미만으로 보고되고 있다. 그러나 일반적으로 행하여지는 핵형 분석으로는 미세한 염색체의 재배열을 정확히 설명하기 어렵다. 본 증례는 불균형 전좌를 가진 성인여성의 정확한 핵형 분석을 위해 말초혈액의 분석 시 기존의 세포유전학적인 방법에 분자유전학적인 방법을 함께 병행한 보고이다. 환자는 31세 여성으로 심각한 정신지체, 행동발달과 언어 발달 장애를 보였으며, 그 원인분석을 위해 Trypsin과 Giemsa를 이용한 GTG 염색법으로 핵형분석을 시행하였다. 그 결과, 46,XX,add(8)(p23.3)으로 확인되었으며, 기원을 확인하기 위하여 부모 염색체 검사를 통해 유전력의 여부를 확인하고, array CGH와 FISH를 시행하여 기원을 알 수 없는 염색체 조각의 기원을 확인한 결과 46,XX,der(8)t(8;13)(p23.3;q32.1)dn의 최종 핵형을 확인하였다. 따라서, FISH 또는 array-CGH 등의 분자유전학적 방법의 적절하고 적극적인 적용은 기존의 세포유전학적 방법을 보완하여, 환자의 정보를 빠르고 정확하게 보고하는데 매우 유용하고, 효과적인 방법이라 하겠다.

Clinical, Cytogenetic and CYP1A1 exon-1 Gene Mutation Analysis of Beedi Workers in Vellore Region, Tamil Nadu

  • Background: Beedi rollers are exposed to unburnt tobacco dust through cutaneous and pharyngeal route and it is extremely harmful to the body since it is carcinogenic in nature and can cause cancer during long exposure. This indicates that occupational exposure to tobacco imposes considerable genotoxicity among beedi workers. Materials and Methods: In the present study, 27 beedi workers and age and sex matched controls were enrolled for clinical, cytogenetics and molecular analysis. Clinical features were recorded. The workers were in the age group of 28-67 years and were workers exposure from 8-60 years. Blood samples were collected from workers and control subjects and lymphocyte cultures were carried out by using standard technique, slides were prepared and 50 metaphases were scored for each sample to find the chromosomal abnormalities. For molecular analysis the genomic DNA was extracted from peripheral blood, to screen the variations in gene, the exon 1 of CYP1A1 gene was amplified by polymerase chain reaction (PCR) and then screened with Single Strand Conformation Polymorphism (SSCP) analysis. Results: A statistically significant increase was observed in the frequencies of chromosomal aberrations in exposed groups when compared to the respective controls and variations observed in Exon 1 of CYP1A1(Cytochrome P450, family 1, subfamily A, polypeptide 1) gene. Conclusions: This study shows that, the toxicants present in the beedi that enter into human body causes disturbance to normal state and behavior of the chromosomes which results in reshuffling of hereditary material causing chromosomal aberrations and genomic variations.

  • DNA methyltransferase 1 (DNMT1) is a relatively large protein family responsible for maintenance of normal methylation, cell growth and survival in mammals. Toxic industrial chemical exposure associated methylation misregulation has been shown to have epigenetic influence. Such misregulation could effectively contribute to cancer development and progression. Methyl isocyanate (MIC) is a noxious industrial chemical used extensively in the production of carbamate pesticides. We here applied an in silico molecular docking approach to study the interaction of MIC with diverse domains of DNMT1, to predict cancer risk in the Bhopal population exposed to MIC during 1984. For the first time, we investigated the interaction of MIC and its hydrolytic product (1,3-dimethylurea) with DNMT1 interacting (such as DMAP1, RFTS, and CXXC) and catalytic (SAM, SAH, and Sinefungin) domains using computer simulations. The results of the present study showed a potential interaction of MIC and 1,3-dimethylurea with these domains. Obviously, strong binding of MIC with DNMT1 interrupting normal methylation will lead to epigenetic alterations in the exposed humans. We suggest therefore that the MIC-exposed individuals surviving after 1984 disaster have excess risk of cancer, which can be attributed to alterations in their epigenome. Our findings will help in better understanding the underlying epigenetic mechanisms in humans exposed to MIC.

SRY 유전자를 가진 46,XX 남성 1례 (A Case of a 46,XX Male with SRY Gene)

  • 46,XX 남성은 여성의 핵형을 가지나, 남성의 표현형을 나타내는 경우를 말한다. SRY 유전자는 Y 염색체(Yp 11.31)의 단완에 위치하며 인간에서 성을 결정하는 중요한 요인이다. 본 증례는 무정자증의 임상증상이 보이는 46,XX 남성의 정확한 원인분석을 위해 말초혈액분석에서 세포유전학적인 방법과 분자유전학적인 방법을 함께 병행한 보고이다. 남성 표현형과 비교하여 성에 불일치 소견이 보여, 그 원인을 찾기 위해 QF-PCR, FISH와 Multiplex PCR 분석 등의 분자유전학적 방법을 적용하였다. 그 결과, 여성의 성 염색체 XX를 가지되 SRY 유전자가 존재하여 남성 표현형을 보이면서 무정자증이 된 경우이다. 따라서, 일반적인 세포유전학 방법을 기초로 QF-PCR, FISH와 Multiplex PCR 분석 등과 같은 분자유전학적 방법을 병행하면 환자의 정보를 빠르고 정확하게 제공하는데 효과적이고 유용하다.

Analysis of Research Papers Published in the Korean Journal of Clinical Laboratory Science from 1985 to 2012

  • The author surveyed and analyzed the research papers of korean journal of clinical laboratory science (KJCLS) that have been posted for 28 years from 17 volumes in 1985 to 44 volumes in 2012 in the time of 50th anniversary of foundation of the korean association of medical technologists (KAMT) of in 2012. This study is aimed to provide members with basic materials helpful to research development and suggest development measures of journal. The author analyzed the number of papers, research field, type of papers, and number of authors based on the title of paper. The total number of papers is 916 and average number of paper is 33. The research field was biochemistry 167 (18.2%), microbiology 160 (17.4%), histology & cytology 99 (10.8%), molecular genetics 77 (8.5%), hematology 69 (7.5%), physiologic function 64 (7.0%), immunology 60 (6.5%), blood bank 33 (3.6%), radioimmunoassay 33 (3.6%), parasitology 27 (2.9%), quality control 18 (2.0%), urinalysis & body fluid 13 (1.4%), cytogenetics 12 (1.3%), flow cytometry analysis 6 (0.7%), and other articles were 78 (8.5%). Regarding the type of papers, original article was 777 (84.8%), case report 52 (5.7%), review 23 (2.5%), others 64 (7.0%). Regarding the number of paper authors, single author was 208 (22.7%), 2-man joint authors 178 (19.4%), 3-man joint authors 181 (19.8%), 4-man joint authors 151 (16.5%), over-5-man joint authors 198 (21.6%). The average number of papers was 33 for 28 years from 1985 to 2012, it is fewer than number of technologists and professors working currently regardless of the level of quantity and quality. The KAMT needs paper promoting measures and strategic investment on the scholarly journals that can aggressively promote to members and inspire research desire for korea citation index (KCI) registered article selection of KJCLS in the future.

A Study on DNA Sequences and Mutation of Integrase Region of Korean-type Bovine Leukemia Virus (BLV) pol Gene

  • Bovine leukemia virus (BLV) is a causative agent for lymphoma disease in cattle including cows worldwide. BLV shares similar virion structure and characteristics with other retroviruses. The pol gene of the BLV genome produced reverse transcriptase (RT) and integrase (IN) for important roles for BLV genome integration into host cell chromosomes that is known to be coded in the 3' side of the BLV pol gene (one third portion). In this study, we have sequenced 978 bp in the 3' side of the BLV pol gene from BLV 10C3 in order to determine the BLV IN region of it. And we compared it to the nucleotide sequences of an Australian BLV isolate. As a result, nucleotide sequences of the IN region of the Korean-type BLV pol gene were mutated at a rate of 3.7%. We can confirm that the typical mutations are such as Arg (AGG) $\rightarrow$ Lys (AAG), Thr (ACG) $\rightarrow$ Met (ATG), Ile (ATT) $\rightarrow$ Val (GTT), Asn (ACC) $\rightarrow$ His (CAC), Phe (TTT) $\rightarrow$ Leu (TTG) and Asn (ACC) $\rightarrow$ Asp (GAC). From the analysis of the sequencing data, we were able to determine the zinc-finger-like "HHCC" motif in the amino terminus of BLV IN, that was H-$X_3$-H-$X_{25}-C-X_2$-C. It was also found the DD35E motif in the IN catalytic domain as D-$X_{56}$-D-$X_{35}$-E. It fits very well to the consensus sequences of retroviral IN as well as HHCC motif.

Generation of FISH Probes Using Laser Microbeam Microdissection and Application to Clinical Molecular Cytogenetics

  • Chromosome microdissection and the reverse FISH technique is one of the most useful methods for the identification of structurally abnormal chromosomes. In particular, the laser microbeam microdissection (LMM) method allows rapid isolation of a target chromosome or a specific region of chromosomes without damage of genetic materials and contamination. Isolated chromosomes were directly amplified by the degenerate oligonucleotide-primed polymerase chain reaction (DOP-PCR), and then the FISH probes labeled with spectrum green- or spectrum red-dUTP were generated by nick-translation. Whole chromosome painting (WCP) probes were successfully generated from only 5 copies of the chromosome. With this method, we produced 24 WCP probes for each human chromosome. We also tried to characterize a marker chromosome, which seemed to be originated from chromosome 11 on conventional banding technique. The marker chromosomes were isolated by the LMM method and analyzed by reverse FISH. We elucidated that the marker chromosome was originated from the short arm of chromosome 5 ($5p11{\to}pter$). A fully automated and computer-controlled LMM method is a very simple laboratory procedure, and enables rapid and precise characterization of various chromosome abnormalities.

The Biocontrol Activity of Chromobacterium sp. Strain C-61 against Rhizoctonia solani Depends on the Productive Ability of Chitinase

  • A chitinolytic bacterium, Chromobacterium sp. strain C-61, was found strongly antagonistic to Rhizoctonia solani, a causal agent of damping-off of eggplant. In this study, the biocontrol activity and enzymatic characteristics of strain C-61 were compared with its four Tn5 insertion mutants (C61-A, -B, -C, and -D) that had lower chitinolytic ability. The chitinase activity of a 2-day old culture was about $76\%,\;49\%\;and\;6\%$ level in C61-A, C61-B and in C61-C, respectively, compared with that of strain C-61. The $\beta-N-acetylhexosaminidase$(Nahase) activity was little detected in strain C-61 but increased largely in C-61A, C61-B and C61-C. Activities of chitinase and Nahase appeared to be negatively correlated in these strains. Another mutant, C-61D, produced no detectable extracellular chitinase and Nahase. The in vitro and in vivo biocontrol activities of strain C-61 and its mutants were closely related to their ability to produce chitinase but not Nahase. No significant differences in population densities between strain C-61 and its mutants were observed in soil around eggplant roots. The results of SDS-PAGE and isoelectrofocusing showed that a major chitinase of strain C-61 is 54-kDa with pI of approximately 8.5. This study provides evidence that the biocontrol activity of Chromobacterium sp. strain C-61 against Rhizoctonia solani depends on the ability to produce chitinase with molecular weight of 54-kDa and pI of 8.5.

  • Tremendous progress has been made over the past quarter-century studying the genetics of gametogenesis and the resulting gametes and embryos. Studies merging molecular techniques and conventional cytogenetics are now beginning to bridge the gap between what we have learned about the meiotic process in males and females and what we know of the mitotic chromosomes of zygotes. Numerical abnormalities in sperm, oocytes and embryo can now diagnosed by fluorescence in situ hybridization (FISH). "At risk" couples can, therefore, have only unaffected embryos replaced in the sterus and avoid the possibility of terminating a pregnancy that might only be diagnosed as affected later gestation. Single-cell genetic analysis has also provided powerful tools for studying genetic defects arising during early human development. Recent studies of sperms, oocytes and cleavage-stage human embryos have revealed an unexpectedly high incidence. These genetic abnormalities are likely to contribute to early pregnancy loss and have important implications for improving pregnancy rates in infertile couples by assisted reproduction. The widespread use of preimplantation genetic diagnosis (PGD) awaits further documentatio of safety and accuracy. Other issues also must be addressed. First, the ethical issues regarding germ cell and embryo screening must be addressed including what diseases are serious enough to warrant the procedure. Another concern is the use of this technology for non-genetic disorders such as gender selection. Finally, the experimental nature of these procedure must continually be discussed with patients, and long-term follow-up studies must be undertaken. Development of more accurate and less expensive assays coupled with improved assisted reproductive technology success rates may make PGD a more widely use clinical tool. The future awaits these development.velopment.
