Size Characterization of Sodium Hyaluronate by Field Programming Frit Inlet Asymmetrical Flow Field-Flow Fractionation/Multiangle Light Scattering

  • Sodium hyaluronate (NaHA), water soluble polymer having ultra-high molecular weight, is characterized by using on-line frit inlet asymmetrical flow field-flow fractionation (FI-AFlFFF) and multiangle light scattering (MALS). This study demonstrates the capability of power programming FI-AFlFFF for the separation of NaHA and the applicability of FI-AFlFFF with MALS for the characterization of molecular weight distribution and their structural information. Since sample injection and relaxation in FI-AFlFFF are achieved by using hydrodynamic relaxation, separation of high molecular weight polymers can be achieved smoothly without halting the separation flow. Experiments are carried out with the two different NaHA products (a raw NaHA sample and a thermally degraded NaHA product) and molecular weight distribution and conformations in solution are determined. Influence of sample filtration on the change of molecular weight distribution is also discussed.

Purification and Characterization of Recombinant Tadpole H-Chain Ferritin in Escherichia coli

  • The tadpole H-ferritin produced in E. coli was purified and its molecular properties were investigated to obtain information about the contribution of the H-subunit in the reaction of iron core formation. All the expressed subunits were assembled into complete holoprotein in vitro, presumably 24-mer, and the protein was heat-stable. Electron microscopy revealed that the recombinant ferritin forms spherically and contains iron core. No difference was observed in the absorption spectrum of the expressed protein compared to that of the natural ferritin. The Ouchterlony double diffusion of the expressed protein showed that the H-chain ferritin shares an antigenic determinant with natural tadpole ferritin. Rabbit anti-horse spleen ferritin discriminated the H-ferritin from natural ferritin. The rate of ferritin formation by the recombinant H-chain apoferritin was determined to be higher than that shown by natural tadpole ferritin, which consists of H, M and L-subunits. This phenomenon may be caused by the absence of M and L-subunits in the recombinant H-chain apoferritin.

  • Club root caused by Plasmodiophora brassicae is found in crucifers. Among the over hundreds of Streptomyces isolated from soil in Korea. One strain showed prominent activity against P. brassicae. The strain was identified based on 16S rDNA sequencing and the morphology by a method of scanning electron microscopy. An active compound in the fermented broth obtained from the strain was separated. Even though the complete assignments of the compound remain for future work, the results regarding the isolation and characterization of the strain with a certain activity against P. brassicae are shown in this paper.

Aspergillus caninus (Syn: Phialosimplex caninus): a New Isolate from Field Soils in Korea

  • During the study of indigenous fungal communities in soil samples collected from various field soils in Sancheong, Gyeongsangnam-do, Korea in 2017, several species of Aspergillus were discovered. Aspergillus caninus (KNU17-7) was isolated, identified, and described based on the results from macro and micro morphological characteristics and molecular characterization. Morphologically, it was identified using five different growth media: potato dextrose agar, oatmeal agar, yeast extract sucrose agar, czapek yeast extract agar, and malt extract agar. For the molecular identification, sequencing of internal transcribed spacer, ${\beta}-tubulin$, and calmodulin genes was performed. Based on this characterization, our study isolate was identified as Aspergillus caninus. This fungal species has not been officially reported in Korea before, and we report here with its morphological and molecular phylogenetic characterization.

Recent Advances in Electrochemical Studies of π-Conjugated Polymers

  • We review the evolution of electrochemical studies of conducting polymers into the current state-of-the-art based primarily on our work. While conventional electrochemical experiments sufficed for the needs in the phase of studies of both electrochemical synthesis and characterization of conducting polymers, developments of various new experimental techniques have led to their introduction to this field for more refined information. As a result, the conventional electrochemical, spectroelectrochemical, electrochemical quartz crystal microbalance, impedance, and morphological as well as electrical characterization studies all made important contributions to a better understanding of the polymerization mechanisms and the conductive properties of these classes of polymers. From this review, we hereby expect that the electrochemical techniques will continue to play important roles in bringing this field to the practical applications such as nanoscale electronic devices.

Flanking Sequence and Copy-Number Analysis of Transformation Events by Integrating Next-Generation Sequencing Technology with Southern Blot Hybridization

  • With the continual development of genetically modified (GM) crops, it has become necessary to develop detailed and effective molecular characterization methods to select candidate events from a large pool of transformation events. Relative to traditional molecular analysis methods such as the polymerase chain reaction (PCR) and Southern blot hybridization, next generation sequencing (NGS) technology for whole-genome sequencing of complex crop genomes had proven comparatively useful for in-depth molecular characterization. In this study, four transformation events, including one in Bacillus thuringiensis (Bt)-resistant rice, one in resveratrol-producing rice, and two in beta-carotene-enhanced soybeans, were selected for molecular characterization. To merge NGS analysis and Southern blot-hybridization results, we confirmed the transgene insertion sites, insertion construction, and insertion numbers of these four transformation events. In addition, the read-coverage depth assessed by NGS analysis for inserted genes might provide consistent results in terms of inserted T-DNA numbers in case of complex insertion structures and highly duplicated donor genomes; however, PCR-based methods can produce incorrect conclusions. Our combined method provides an effective and complete analytical approach for whole-genome visual inspection of transformation events that require biosafety assessment.
