Nanopharmaceutical Approach for Enhanced Anti-cancer Activity of Betulinic Acid in Lung-cancer Treatment via Activation of PARP: Interaction with DNA as a Target -Anti-cancer Potential of Nano-betulinic Acid in Lung Cancer-

  • Objectives: This study examined the relative efficacies of a derivative of betulinic acid (dBA) and its poly (lactide-co-glycolide) (PLGA) nano-encapsulated form in A549 lung cancer cells in vivo and in co-mutagen [sodium arsenite (SA) + benzo[a]pyrene (BaP)]-induced lung cancer in mice in vivo. Methods: dBA was loaded with PLGA nanoparticles by using the standard solvent displacement method. The sizes and morphologies of nano-dBA (NdBA) were determined by using transmission electron microscopy (TEM), and their intracellular localization was verified by using confocal microscopy. The binding and interaction of NdBA with calf thymus deoxyribonucleic acid (CT-DNA) as a target were analyzed by using conventional circular dichroism (CD) and melting temperature (Tm) profile data. Apoptotic signalling cascades in vitro and in vivo were studied by using an enzyme-linked immunosorbent assay (ELISA); the ability of NdBA to cross the blood-brain barrier (BBB) was also examined. The stage of cell cycle arrest was confirmed by using a fluorescence-activated cell-sorting (FACS) data analysis. Results: The average size of the nanoparticles was ~ 110 nm. Confocal microscopy images confirmed the presence of NdBA in the cellular cytoplasm. The bio-physical properties of dBA and NdBA ascertained from the CD and the Tm profiles revealed that NdBA had greater interaction with the target DNA than dBA did. Both dBA and NdBA arrested cell proliferation at G0/G1, NdBA showing the greater effect. NdBA also induced a greater degree of cytotoxicity in A549 cells, but it had an insignificant cytotoxic effect in normal L6 cells. The results of flow cytometric, cytogenetial and histopathological studies in mice revealed that NdBA caused less nuclear condensation and DNA damage than dBA did. TEM images showed the presence of NdBA in brain samples of NdBA fed mice, indicating its ability to cross the BBB. Conclusion: Thus, compared to dBA, NdBA appears to have greater chemoprotective potential against lung cancer.

Impact of IL-2 and IL-2R SNPs on Proliferation and Tumor-killing Activity of Lymphokine-Activated Killer Cells from Healthy Chinese Blood Donors

  • One of the goals of tumor immunotherapy is to generate immune cells with potent anti-tumor activity through in vitro techniques using peripheral blood collected from patients. However, cancer patients generally have poor immunological function. Thus using patient T cells, which have reduced in vitro proliferative capabilities and less tumor cell killing activity to generate lymphokine-activated killer (LAK) cells, fails to achieve optimal clinical efficacy. Interleukin-2 (IL-2) is a potent activating cytokine for both T cells and natural killer cells. Thus, this study aimed to identify optimal donors for allogeneic LAK cell immunotherapy based on single nucleotide polymorphisms (SNP) in the IL-2 and IL-2R genes. IL-2 and IL-2R SNPs were analyzed using HRM-PCR. LAK cells were derived from peripheral blood mononuclear cells by culturing with IL-2. The frequency and tumor-killing activity of LAK cells in each group were analyzed by flow cytometry and tumor cell killing assays, respectively. Regarding polymorphisms at IL-2-330 (rs2069762) T/G, LAK cells from GG donors had significantly greater proliferation, tumor-killing activity, and IFN-${\gamma}$ production than LAK cells from TT donors (P<0.05). Regarding polymorphisms at IL-2R rs2104286 A/G, LAK cell proliferation and tumor cell killing were significantly greater in LAK cells from AA donors than GG donors (P<0.05). These data suggest that either IL-2-330(rs2069762)T/G GG donors or IL-2R rs2104286 A/G AA donors are excellent candidates for allogeneic LAK cell immunotherapy.

Effect of Chemical Composition, Molecular Weight, Carbon Black Concentration and Temperature on the Flow Behavior of Styrene/Butyl Methacrylate Homo- and Co-polymer Particles (화학적 구성, 분자량, 카본블랙의 농도 및 온도에 따른 스티렌/부틸메타크릴레이트 단일중합체 및 공중합체 입자의 유동성)

  • We measured shear viscosity of polymeric particles, prepared by suspension polymerization with hydrophobic silica as a stabilizer at $75^{\circ}C$, by a capillary rheometer. Shear viscosity displayed a non-Newtonian behavior with an increase in weight average molecular weight. Measurement of shear viscosity at 170 and $190^{\circ}C$ with copoly(styrene/butyl methacrylate) (co-PSB) particles by varying the ratio between styrene (St) and butyl methacrylate (BMA) showed that shear viscosity was a function of molecular weight, temperature and compositional ratio. When the ratio was 7/3, 5/5 and 3/7, shear viscosity slightly reduced with an increase in BMA concentration despite similar weight average molecular weights. We found that shear viscosity of copolymers with BMA concentrations exceeding 70% displayed a sharp reduction at high shear rates. It is speculated that increased PBMA chain length contributes to enhanced flowability of copolymers. When carbon black was incorporated into co-PSB, shear viscosity progressively increased with increasing carbon black concentration. The increase in shear viscosity, however, was less pronounced compared to the cases of molecular weight increase.

Flow Behavior of Polystyrene and Poly(butyl methacrylate) Composite Particles Filled with Varying Concentrations of Carbon Black (다양한 농도의 카본블랙을 함유하는 폴리스티렌 및 폴리뷰틸메타크릴레이트 복합체 입자의 유동성)

  • We measured shear viscosity of polystyrene (PS) and poly(butyl methacrylate) (PBMA) particles, with a capillary rheometer, prepared by suspension polymerization with 1.0 wt% hydrophobic silica as a stabilizer by varying the initiator concentration at $75^{\circ}C$. PS particles with weight average molecular weight of 66,500 g/mol displayed a Newtonian behaior at low shear rates at $190^{\circ}C$. With increasing molecular weight, PS particles showed shear thinning over the entire range of shear rates. For PBMA particles, steady shear measurement was carried out at $170^{\circ}C$. PBMA particles with weight average molecular weight of 156,700 g/mol showed a Newtonian behaior only at low shear rates. PBMA particles also showed shear thinning with an increase in molecular weight and its pattern similar to that of PS. When carbon black was incorporated into PS and PBMA polymers, steady shear measurement was conducted at $170^{\circ}C$. An increase in carbon black concentration in PS and PBMA composite particles exhibited a progressive increase in shear viscosity. The increase in shear viscosity, however, was less pronounced compared to an increase as a function of molecular weight. Preparing PS composites containing carbon black by internal mixing resulted in an increase in shear viscosity. Its increase, however, was found to be less than that shown in PS composite particles. We speculate that this is caused by an enhanced dispersion of carbon black particles with an internal mixer. Yield behavior was not observed in any of the samples we selected in this experiment.

Purification and Characterization of 5,10-Methenyltetrahydrofolate Synthetase from Chicken Liver (닭의 간 유래의 5,10-Methenyltetrahydrofolate Synthetase의 정제 및 특성)

  • 5,10-Methenyltetrahydrofolate synthetase from chicken liver was purified through 30-70% ammonium sulfate fractionation, Q Sepharose Fast Flow anion exchange and Source 15Phe hydrophobic interaction chromatography. Specific activities of cell extract, ammonium sulfate, Q Sepharose Fast Flow and Source 15Phe were 0.0085, 0.031, 0.80 and 1.27 U/mg, respectively. Purification fold activities of cell extract, ammonium sulfate, Q Sepharose Fast Flow and Source 15Phe were 1, 3.7, 94.1 and 149.4, respectively. HPLC gel permeation chromatography and SDS-polyacrylamide electrophoresis experiments indicated that the enzyme is a monomeric protein with a molecular weight of 22.8 kDa. Km for 5-methyl THF and Mg-ATP were $7.1\;{\mu}M$ and $63\;{\mu}M$, respectively. Optimum temperature and pH were $30^{\circ}C$ and 6.0, respectively. The data for metal ion specificity and stoichiometry showed that the maximum activity was obtained with a 1:l. ratio of $Mg^{2+}$. The ATP and Km values increased in the order of MgATP, MgCTP, MgUTP and MgGTP, and the maximum activities also decreased in the same order, indicating MgATP as the most efficient substrate. The enzyme was chemically modified only by tetranitrometane and 1-ethyl-3-(3-dimethyl aminopropyl)-carbodiimide, indicating that tyrosine and carboxylate are present in the active site.

  • Textile printing prints around twenty bilion linear meters of textile each year. Rotary and flat bed screen printing requires pre and post treatments, leading to the loss of dyes and the environmental problems due to effluents. Digital ink jet printing can offer a solution to the existing problems, especially the environmental problems, in addition to its flexibility. Pigments are used as a dispersion inks in the digital inkjet textile printing. Molecular dynamic simulation like Stokesian dynamic simulation was employed to simulate the behavior of pigments and velocity distribution under the pressure driven flow in the printer nozzle. The simulation shows that the particle distribution in the flow are uniform if particle volume fraction is low, the ratio of nozzle and particle diameter is large, and the dimensionless average suspension velocity is low.

Critical Role of Glu175 on Stability and Folding of Bacterial Luciferase: Stopped-flow Fluorescence Study

  • Bacterial luciferase is a heterodimeric enzyme, which catalyzes the light emission reaction, utilizing reduced FMN (FMNH2), a long chain aliphatic aldehyde and $O_2$, to produce green-blue light. This enzyme can be readily classed as slow or fast decay based on their rate of luminescence decay in a single turnover. Mutation of Glu175 in $\alpha$ subunit to Gly converted slow decay Xenorhabdus Luminescence luciferase to fast decay one. The following studies revealed that changing the luciferase flexibility and lake of Glu-flavin interactions are responsible for the unusual kinetic properties of mutant enzyme. Optical and thermodynamics studies have caused a decrease in free energy and anisotropy of mutant enzyme. Moreover, the role of Glu175 in transition state of folding pathway by use of stopped-flow fluorescence technique has been studied which suggesting that Glu175 is not involved in transition state of folding and appears as surface residue of the nucleus or as a member of one of a few alternative folding nuclei. These results suggest that mutation of Glu175 to Gly extended the structure of Xenorhabdus Luminescence luciferase, locally.

Prediction of Burnt Gas Properties for Kerosene Fuel-rich Preburner (케로신 연료과잉 예연소기의 연소가스 물성치 예측)

  • A Fuel-rich preburner using kerosene fuel is operated in a non-equilibrium condition and a prediction of burnt-gas properties is not easy from a chemical equilibrium analysis. A premixed counter-flow flame analysis was conducted for the prediction of burnt-gas properties. JP10 was selected for a representative kerosene fuel and a non-equilibrium combustion analysis was accomplished in supercritical condition using UC San Diego reaction mechanism. The premixed counter-flow flame was assumed for stationary and stable flame, and the temperature result in present study was overestimated rather than the experimental results from Huzel. From the difference of the temperature result, other properties, heat capacity, specific heat ratio and molecular weight had some differences against the experimental results. Moreover, the present results was more similar to the experimental results than those of the equilibrium analysis.

Associations Between Mammography and Ultrasound Imaging Features and Molecular Characteristics of Triple-negative Breast Cancer

  • Background: The triple-negative breast cancer (TNBC) is an aggressive cancer characterized by the absence of estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor 2 (HER2). Preoperative mammography and ultrasound features of TNBC may potentially suggest characteristics of the disease and assist in treatment decisions. Materials and Methods: The study covered 153 patients with TNBC from May 2011 to May 2012 who were confirmed by postoperative pathology results in our hospital. We compared the radiological findings among the patients and sought to determine the significant iconographic features. The biomarkers p53 and Ki-67 are regarded as significant factors in TNBC. They were therefore used to divide the TNBC into four groups for assessment of relationships with TNBC imaging features. Results: On mammography, most TNBCs exhibit obscure (44.3%) masses. On ultrasound, the majority of masses (95.4%) were predominantly indistinct (50.7%), irregular (76.0%) or featuring posterior echo enhancement/shadowing. Color Doppler flow imaging (CDFI) emphasized hypervascular (32.9%) masses. Differences in CDFI by ultrasound among the four groups were statistically significant (p=0.009). There were obvious differences in the percentages of spiculated margin (p=0.049) and intensive posterior echo (p=0.006) with spotty flow imaging by ultrasound between the Ki-67 (+) p53 (+) and other groups. Conclusions: A combination of mammography and ultrasound revealed the imaging characteristics of TNBC included an obscure mass with less attenuated posterior echoes and some vascularity. A worse prognosis was associated with spiculated margin and intensive posterior echoes with spotty flow imaging.

Preparation and Electrochemical Applications of Pore-filled Ion-exchange Membranes with Well-adjusted Cross-linking Degrees: Part I. All Vanadium Redox Flow Battery (가교도가 조절된 세공충진 이온교환막의 제조 및 전기화학적 응용: Part I. 전 바나듐 레독스 흐름전지)

  • In this study, we have developed pore-filled ion-exchange membranes (PFIEMs) filled with ionomer in a thin polyethylene porous film (thickness = $25{\mu}m$) and investigated the charge-discharge characteristics of the all vanadium redox flow battery (VRFB) employing them. Especially, the degree of crosslinking and free volume of the PFIEMs were appropriately controlled to produce ion-exchange membranes exhibiting both the low membrane resistance and low vanadium permeability by mixing crosslinking agents having different molecular size. As a result, the prepared PFIEMs exhibited excellent electrochemical properties which are comparable to those of the commercial membranes. Also, it was confirmed through the experiments of vanadium ion permeability and VRFB performance evaluation that the PFIEMs showed low vanadium ion permeability and high charge-discharge efficiency in comparison with the commercial membrane despite their thin film thickness.
