With explosively growing PPI databases, the computational approach for a prediction and configuration of PPI network has been a big stream in the bioinformatics area. Recent researches gradually consider physicochemical properties of proteins and support high resolution results with integration of experimental results. With regard to current research trend, it is very close future to complete a PPI network configuration of each organism. However, direct applying the PPI network to real field is complicated problem because PPI network is only a set of co-expressive proteins or gene products, and its network link means simple physical binding rather than in-depth knowledge of biological process. In this paper, we suggest a protein functional flow model which is a directed network based on a protein functions' relation of signaling transduction pathway. The vertex of the suggested model is a molecular function annotated by gene ontology, and the relations among the vertex are considered as edges. Thus, it is easy to trace a specific function's transition, and it can be a constraint to extract a meaningful sub-path from whole PPI network. To evaluate the model, 11 functional flow models of Homo sapiens were built from KEGG, and Cronbach's alpha values were measured (alpha=0.67). Among 1023 functional flows, 765 functional flows showed 0.6 or higher alpha values.
The recent spread of novel and highly variant pathogenic viruses, including the coronavirus (SARS-CoV-2), has increased the demand for diagnostic testing for rapid confirmation. This has resulted in investigating the functional capability of each space, and preparing facility guidelines to secure the safety of medical technologists. During viral evaluations, there is a requirement of negative pressure facilities along with thread separation, during pre-treatment of samples and before nucleic acid amplification. Space composition therefore needs to be planned by considering unidirectional air flow. This classification of safety management facilities is designated as biosafety level 2, and personal protective equipment is placed accordingly. In case of handling dangerous materials, they need to be carried out of the biosafety cabinet, and sterilizers are required for suitable disposal of infectious agents. A common feature of domestic laboratories is maintenance of the sample pre-treatment space at a negative pressure of -2.5 Pa or less, and arranging separate pre-treatment and reagent preparation spaces during the test process. We believe that the data generated in this study is meaningful, and offers an efficient direction and detailed flow for separation of the inspection process and space functions. Moreover, this study introduces construction of the laboratory by applying the safety management standards.

Treatment of Leptomeningeal carcinomatosis (LMC) from solid cancers has not advanced noticeably since the introduction of intra-cerebrospinal fluid (CSF) chemotherapy in the 1970's. The marginal survival benefit and difficulty of intrathecal chemotherapy injection has hindered its wide spread use. Even after the introduction of intraventricular chemotherapy with Ommaya reservoir, frequent development of CSF flow disturbance, manifested as increased intracranial pressure (ICP), made injected drug to be distributed unevenly and thus, the therapy became ineffective. Systemic chemotherapy for LMC has been limited as effective CSF concentration can hardly be achieved except high dose methotrexate (MTX) intravenous administration. However, the introduction of small molecular weight target inhibitors for primary cancer treatment has changed the old concept of 'blood-brain barrier' as the ultimate barrier to systemically administered drugs. Conventional oral administration achieves an effective concentration at the nanomolar level. Furthermore, many studies report that a combined treatment of target inhibitor and intra-CSF chemotherapy significantly prolongs patient survival. Ventriculolumbar perfusion (VLP) chemotherapy has sought to increase drug delivery to the subarachnoid CSF space even in patients with disturbed CSF flow. Recently authors performed phase 1 and 2 clinical trial of VLP chemotherapy with MTX, and 3/4th of patients with increased ICP got controlled ICP and the survival was prolonged. Further trials are required with newly available drugs for CSF chemotherapy. Additionally, new LMC biologic/pharmacodynamic markers for early diagnosis and monitoring of the treatment response are to be identified with the help of advanced molecular biology techniques.
Soybean straw (SS)-based activated carbon was employed as a precursor to prepare carbon molecular sieves (CMSs) via chemical vapor deposition (CVD) technique using methane as carbon source. Prior to the CVD process, SS was activated by 0.5 wt% $ZnCl_2$, followed by a carbonization at $500^{\circ}C$ for 1 h in $N_2$ atmosphere. $N_2$ (77 K) adsorption-desorption and $CO_2$ (273 K) adsorption tests were carried out to analyze the pore structure of the prepared CMSs. The results show that increasing the deposition temperature, time or methane flow rate leads the decrease in $N_2$ adsorption capacity, micropore volume and average pore diameter of CMSs. The adsorption selectivity coefficient of $CO_2/CH_4$ achieves as high as 20.8 over CMSs obtained under the methane flow rate of $30mL\;min^{-1}$ at $800^{\circ}C$ for 70 min. The study demonstrates the prepared CMSs are a candidate adsorbent for $CO_2/CH_4$ separation.
Coalescence process of binary immiscible fluid mixtures having bimodal size distributions, prepared by mixing two pre-sheared samples at different shear rates, ${\gamma}_{pre1}\;and\;{\gamma}_{pre2}$, under shear flow at a final shear rate, ${\gamma}_f$, are examined by transient shear stress measurements and microscopic observations in comparison with the results for simply pre-sheared samples having narrow size distributions (unimodal distribution samples). Component fluids are a silicone oil (PDMS) and a hydrocarbon-formaldehyde resin (Genelite) and their viscosities are 14.1 and 21.0 $pa{\cdot}sec$ at room temperature $(ca.\;20^{\circ}C)$, respectively. The weight ratio of PDMS: Genelite was 7:3. Three cases, $({\gamma}_{pre1}=7.2sec^{-1},\;{\gamma}_{pre2}=12.0sec^{-1}\;and\;{\gamma}_f=2.4sec^{-1}),\;({\gamma}_{pre1}=0.8sec^{-1},\;{\gamma}_{pre2}=4.0sec^{-1}\;and\;{\gamma}_f=2.4sec^{-1}),\;and\;({\gamma}_{pre1}=7.2sec^{-1},\;{\gamma}_{pre2}=12.0^sec^{-1}\;and\;{\gamma}_f=7.2sec^{-1})$ the first case, transient shear stress did not show any significant difference but domains larger than the initial state are observed at short times. In the latter cases, there exist undershoot of shear stress, reflecting existence of deformed large domains, which is confirmed by the direct observation. It is concluded that coalescence between large and small domains more frequently occur than coalescence between the domains with similar size in the bimodal distribution samples.
Four taxa of the genus Phyllostachys were analysed with PCR-based molecular inter-simple sequence repeat (ISSR) determine markers to determine their phylogenetic relationships. Many species of this genus are regarded as economical and ecologically important in the world. With the nine primers screened, ISSR assay generated a total of 64 reproducible bands. Analysis of ISSR from individual plants of genus Phyllostachys resulted in 43 polymorphic bands with 70.49%. When species were grouped by four taxa, within group diversity was 0.092 (Hs), while among group diversify was 0.499 (Gst) on a per locus basis. The estimated gene flow (Nm) between the pairs of species was 0.559. Hence, we can expect weak or low gene flow among species. Total mean genetic diversity ($H_T$) for four taxa was 0.291. The ISSR data allowed us to resolve well-supported clades in the genus Phyllostachys. The four taxa of the genus Phyllostachys analyzed were distinctly related to a monophyletic.

[99mTc]Tc-Hexamethylpropylene amine oxime (HMPAO) is currently used as a regional cerebral blood flow imaging agent for single photon emission computed tomography (SPECT). The HMPAO ligand exists in two isomeric forms: d,l and meso showing different properties in vivo. Later studies indicated that brain uptake patterns of 99mTc-complexes formed from separated enantiomers differed. Separation of enantiomers is difficult by fractional crystallizations method. Usually, the substance is obtained in low chemical yield in a time-consuming procedure. Furthermore, the final product still contains some impurity. So we have developed new efficient route for synthesis of R,R- and S,S-HMPAO enantiomeric compounds in 6-steps. Nucleophilic substitution (SN2) reactions of 2,2-dimethylpropane-1,3-diamine either with S- (1a) or R-methyl2-chloropropanoate (1b) were performed to produce compounds R,R- (2a) or S,S-isomer (2b) derivatives protected with benzylchloroformate (Cbz), respectively. And then Weinreb amide and methylation reaction using Grignard reagent, oxime formation with ketone group and deprotectiion of Cbz group by hydrogenolysis gave S,S- (7a) or R,R-HMPAO (7b), respectively. Entaniomeric compounds were synthesied with high yield and purity without any undesired product. The 7a or 7b kits containing 10 ㎍ SnCl2-2H2O were labeled with 99mTc with high radiolabeling yield (90%).
The research on the flow and heat transfer characteristics of lead bismuth(LBE) is significant for the thermal-hydraulic calculation, safety analysis and practical application of lead-based fast reactors(LFR). In this paper, a new CFD model is proposed to solve the thermal-hydraulic analysis of LBE. The model includes two parts: turbulent model and turbulent Prandtl, which are the important factors for LBE. In order to find the best model, the experiment data and design of 19-pin hexagonal rod bundle with spacer grid, undertaken at the Karlsruhe Liquid Metal Laboratory (KALLA) are used for CFD calculation. Furthermore, the turbulent model includes SST k - 𝜔 and k - 𝜀; the turbulent Prandtl includes Cheng-Tak and constant (Prt =1.5,2.0,2.5,3.0). Among them, the combination between SST k - 𝜔 and Cheng-Tak is more suitable for the experiment. But in the low Pe region, the deviation between the experiment data and CFD result is too much. The reason may be the inlet-effect and when Pe is in a low level, the number of molecular thermal diffusion occupies an absolute advantage, and the buoyancy will enhance. In order to test and verify versatility of the model, the NCCL performed by the Nuclear Thermal-hydraulic Laboratory (Nuthel) of Xi'an Jiao tong University is used for CFD to calculate. This paper provides two verification examples for the new universal model.
Glycomacropeptide(GMP) was purified from cheese whey which is obtaining as a byproduct in cheese producing. Cheese whey was first concentrated 10 times with a ultrafiltration aparratus, and then heated at 95$^{\circ}C$ for 5 min. The concentrated fraction was centrifuged at 20,000$\times$g for 30 min to remove fat layer. The supernatant layer enriched GMP protein was fractionated by ion exchange chromatography on DEAE-Sepharose Fast Flow column. GMP was bound to DEAE resin and eluted with 0.1~0.25 M NaCl when using a linear NaCl gradient from 0 M to 0.5 M. The purified GMP gave a single band of 24 kDa which seems to be trimer molecular weight in SDS-PAGE, and migrated to the same molecular weight with control GMP obtained commercially. Its amino acid composition were consistent with that of standard GMP. About 0.71 g of GMP was recovered from 1 L of cheese whey. These results indicate that glycomacropeptide could be simply purified from cheese whey by using ultrafiltration and DEAE column chromatography.
Purpose: To investigate the influence of exogenous p53 upregulated modulator of apoptosis (PUMA) expression on cell proliferation and apoptosis in human non-small cell lung cancer A549 cells and transplanted tumor cell growth in nude mice. Materials and Methods: A549 cells were divided into the following groups: control, non-carrier (NC), PUMA (transfected with pCEP4-(HA) 2-PUMA plasmid), DDP ($10{\mu}g/mL$ cisplatin treatment) and PUMA+DDP (transfected with pCEP4-(HA)2-PUMA plasmid and $10{\mu}g/mL$ cisplatin treatment). The MTT method was used to detect the cell survival rate. Cell apoptosis rates were measured by flow cytometry, and PUMA, Bax and Bcl-2 protein expression levels were measured by Western blotting. Results: Compared to the control group, the PUMA, DDP and PUMA+DDP groups all had significantly decreased A549 cell proliferation (p<0.01), with the largest reduction in the PUMA+DDP group. Conversely, the apoptosis rates of the three groups were significantly increased (P<0.01), and the PUMA and DDP treatments were synergistic. Moreover, Bax protein levels significantly increased (p<0.01), while Bcl-2 protein levels significantly decreased (p<0.01). Finally, both the volume and the weights of transplanted tumors were significantly reduced (p<0.01), and the inhibition ratio of the PUMA+DDP group was significantly higher than in the single DDP or PUMA groups. Conclusions: Exogenous PUMA effectively inhibited lung cancer A549 cell proliferation and transplanted tumor growth by increasing Bax protein levels and reducing Bcl-2 protein levels.
