Fourteen Unrecorded Species of Agaricales Underw. (Agaricomycetes, Basidiomycota) from the Republic of Korea

  • Agaricales species form pileate-stipitate fruiting bodies and play important roles in maintaining the terrestrial ecosystem as decomposers, symbionts, and pathogens. Approximately 23,000 Agaricales species have been known worldwide, and 937 species have been recorded in the Republic of Korea. However, most of them were identified solely based on morphological characteristics that often led to misidentifications. The specimens collected from 2018 to 2020 in the Republic of Korea were identified based on phylogenetic analysis of the internal transcribed spacer (ITS) sequences. Their identities were confirmed by microscopic characteristics. As a result, 14 Agaricales species were discovered for the first time in the Republic of Korea. They belonged to nine genera: Agaricus, Calocybe, Cortinarius, Hygrocybe, Inocybe, Lepista, Leucoagaricus, Marasmius, and Psathyrella. Detailed macroscopic and microscopic descriptions were provided to help distinguish these species. The morphological and molecular data provided in this study will serve as reliable references for the identification of Agaricales species.

Taxonomy of Botryotrichum luteum sp. nov. based on Morphology and Phylogeny Isolated from Soil in Korea

  • In this study, a fungal strain KNUF-22-025 belonging to the genus Botryotrichum was isolated from the soil in Korea. The cultural and morphological characteristics of this strain differed from those of closely related species. On malt extract agar, strain KNUF-22-025 showed slower growth than most of the related species, except B. domesticum. The conidia size (9.6-21.1×9.9-18.4 ㎛) of strain KNUF-22-025 was larger than those of B. piluliferum, B. domesticum, and B. peruvianum but smaller than those of B. atrogriseum and B. iranicum. Conidiophores in strain KNUF-22-025 (137 ㎛) were longer than those in other closely related species but shorter than those in B. atrogriseum. Multi-locus analysis of molecular markers, such as ITS, 28S ribosomal DNA, RBP2, and TUB2 revealed that strain KNUF-22-025 was distinct from other Botryotrichum species. Thus, this strain is proposed as a novel species based on morphological characteristics along with molecular phylogeny and named Botryotrichum luteum sp. nov.

Characterization of Newly Recorded Talaromyces veerkampii Isolated from Field Soil in Korea based on Morphology and Multigene Sequence Analysis

  • A fungal isolate belonging to the phylum Ascomycota was isolated and identified as Talaromyces veerkampii in 2017 during a survey of fungal diversity in field soils in Korea. This fungal isolate was identified and described based on macro- and micromorphological and molecular characterization. The identification was also based on partial 18S-ITS1-5.8S-ITS2-28S rDNA and calmodulin (CaM)-encoding gene sequencing data. Talaromyces veerkampii has not been previously reported in Korea. Thus, we report here a newly discovered species from soil in Korea along with its morphological and molecular characteristics.

Characteristics of Bio-Piezoelectric Generator Using Edible Collagen Powder (식용 콜라겐 분말을 적용한 바이오 압전 발전기의 특성)

  • Because collagen is inherently piezoelectric, research is being actively conducted to utilize it to harvest energy. In this study, a collagen solution was prepared using edible low-molecular-weight peptide collagen powder, and collagen films were fabricated using a dip coating method. The collagen films prepared by dip coating showed a smooth surface without defects such as pinholes or cracks. Dehydrothermal treatment of the collagen films was performed to induce a stable molecular structure through cross-linking. The collagen film subjected to dehydrothermal treatment at 110 ℃ for 24 h showed a thickness reduction rate of 19 %. Analysis of the collagen films showed that the crystallinity of the collagen film improved by about 7.9 % after dehydrothermal treatment. A collagen film-based piezoelectric nanogenerator showed output characteristics of approximately 13.7 V and 1.4 ㎂ in a pressure test of 120 N. The generator showed a maximum power density of about 2.91 mW/m2 and an output voltage of about 8~19 V during various human body movements such as finger tapping. The collagen film-based piezoelectric generator showed improved output performance with improved crystallinity and piezoelectricity after dehydrothermal treatment.

Morphological and molecular characterization of root-lesion nematode Pratylenchus hippeastri from Korea

  • The root-lesion nematode Pratylenchus spp. is the most important plantparasitic nematode due to its worldwide distribution, wide host ranges, and migratory endoparasitic characteristics. One population of Pratylenchus collected from the giant pussy willow (Salix chaenomeloides Kimura) in the Andong area as part of a nematode survey in Korea was characterized morphologically and by molecular methods. The analysis of morphological measurements and morphometric characteristics, as well as DNA sequencing of the rRNA large subunit (LSU) D2/D3 expansion segments and the internal transcribed spacer (ITS) gene sequence, confirmed the identity of this population as P. hippeastri. This study is the first report of P. hippeastri associated with Salix chaenomeloides in Korea and worldwide. Further studies on distribution and pathogenicity in different P. hippeastri host crops, such as grapevines, strawberries, and apples, are necessary. The taxonomic keys to 16 Pratylenchus species in Korea are provided.

  • In this study, we developed the measuring system based on light addressable potentiometric sensor for the quantitative analysis of penicillin that is very important element in medicine and pharmacy, clinic. It is investigated the response characteristics by enzyme reaction with penicillinase. First, the surface pre-treatment process of the $Si_{3}N_{4}$ was established. The coupling agent was made using self assembled monolayer method and it was confirmed the immobilization process by AFM. Also, as the measuring system, potentiostat, signal processing part etc. was made by Lab VIEW software, it was reduced detecting time as well as simplifying the system. Fabricated device was shown excellent pH response characteristics, 57 mV/ pH in the range of pH $2{\sim}11$. The response characteristics was 60 mV/decade in the range of $0.1{\sim}10{\;}mM$.

Study of Mechanical and Hygroscopic Characteristics of Nanoclay/Epoxy Nanocomposites (나노클레이/에폭시 나노-복합재료의 기계적 및 흡습 특성에 관한 연구)

  • In this study, the moisture related hygroscopic characteristics and mechanical properties of epoxy-clay nanocomposites were investigated by experiments as a function of the weight fraction of nanoclay. The hygroscopic and mechanical properties including the moisture saturation amount, moisture diffusivity, adhesive strength, and tensile properties were obtained by moisture absorption test and various tensile tests, respectively. Also, the molecular dynamics (MD) simulation was devised to study of hygroscopic characteristics of nanocomposites and the results were compared to experimental results as a function of the nanoclay content. It was demonstrated that the proposed MD simulation technique can be successfully used for the prediction of the effects of the nanoclay on the moisture diffusion characteristics.

Identification of Erysiphe izuensis on Rhododendron yedoense f. poukhanense in Korea Based on Morphological and Molecular Characteristics (형태 특징 및 분자 분석에 의한 산철쭉 흰가루병균 Erysiphe izuensis 동정)

  • The Rhododendron powdery mildew fungus Erysiphe izuensis has been recorded throughout Asia, including China, Japan, Korea, and Russia. In Korea, E. izuensis has been identified based on morphological characteristics of the anamorph found on introduced Rhododendron spp. We here describe the first identification of E. izuensis collected from Rhododendron yedoense f. poukhanense in Korea. Morphological characteristics of the teleomorph as well as internal transcribed spacer sequences of the species are provided. Moreover, we describe the characteristics of the primary conidia and conidial surface pattern of E. izuensis for the first time.

  • The importance of wild plant resources along with the development of high biotecyhnology is highlighted for exploitation of new materials which can make the added value. The goal of this study is providing fundamental data bases for developing new materials through the analyses of morphological characteristics, antibiotics and molecular cytogenetics in Baegseon (Dictamnus dasycarpus Turcz.). Baegseon has several characteristics; there are two types of flower color, pink and white, the seed germination starts about February 20th, the maximum flowering season is around May 17th in southern Korea, the growth period is about 60 days. The number of chromosomes are 2n=2x=36, the size of chromosomes in metaphase is $4.2{\sim}8.1{\mu}m$. The amount of 2C nuclear DNA is 1.93pg, and there is no variation of genome size amoung varieties. The extraction juice of baegseon young roots has the excellent antibiotic activity against the mold (Botrytis cinerea).

  • A lot of clones of the genus Dioscorea species have been introduced from some tropical and subtropical regions since 1997. The aim of this study was to determine characteristics of the production process in yams(Dioscorea spp). By utilizing 51clones of water yams(Dioscorea spp), some morphological characteristics were investigated at the field. Intraspecific genetic relationship of 51 variation types of the Yam classified by their external morphological characteristics such as leaf and tuber shape were assessed by DNA using random and specific primers. D. Alata and D. opposita were showed different relationship between yield and the growth of their aboveground parts. This suggests that even in the same species there were differences in yield volume and translocation of assimilation products, depending on the types. D. alata were distinguished from others Dioscorea species at 62% level in AFLP analysis. Also in principal component analysis, D. alata were showed the class from II to V.
