The Effect of 4-Week Health Promotion Summer Camp on the Metabolic Syndrome and Insulin Resistance among Obese Elementary Students (비만 초등학생의 4주 여름 건강증진캠프 참여가 대사증후군 및 인슐린저항성에 미치는 효과)

  • The purpose of the current study was to investigate the effects of 4-week health promotion intervention program consisting of high-intensity aerobic exercise to improve energy consumption and cardiorespiratory capacity, circuit training to strengthen muscular strength and endurance and education for lifestyle changes on the obesity level, physical fitness, insulin resistance and metabolic syndrome among overweight or obese elementary school students. Twenty three overweight or obese children as obese group and 15 normal body weight children as normal group were recruited. The health promotion program consisted of two exercise sessions and one education session, 3days/week in 4-week. Obesity level(body mass index, waist circumference, %body fat), physical fitness(muscular strength, muscular endurance, flexibility, cardiopulmonary fitness), insulin resistance(homeostasis model assessment of insulin resistance) and metabolic syndrome risk factors(blood pressure, fasting glucose, triglycerides, high density lipoprotein cholesterol) were measured. There was not significant decrease in obesity level; however, there were significant improvement in physical fitness, insulin resistance and metabolic syndrome risk factors after program participation among obese children. As a result, through participation in the summer camp consisting mainly of exercise, the improvement of the physical fitness level and the decrease of insulin resistance had an effect on the reduction of the metabolic syndrome frequency.

The Effect of Low Impact Development Techniques on Urban Runoff (저영향개발기법이 도시 유출에 미치는 영향)

  • Due to rapidly increased urbanization, impervious area has been extended and concerns about urban flooding has been increased as well. A lot of effort has been made to restore the urban water circulation. Low Impact Development (LID) technology that consist of retention, infiltration, and evapotranspiration has begun to attract attention to simulate the hydrologic phenomenon before and after development. Many researches on the technique is being actively conducted. In this study, the effect on reducing runoff in urban catchment was analyzed and evaluated by applying LID techniques using SWMM and six scenarios. A SWMM-LID model was built for the Gasan 1 rainwater pumping station basin, and Green Roof and Permeable Pavement were selected as LID techniques to be applied. As a result, the reduction effect of the permeable pavement was larger than green roof. In the future, the results could be used to design a LID facility using the characteristics of the watershed, and other urban water resource factors such as river and groundwater levels that affect each other should be considered, so that the entire system can be considered.

Assessment of testicular steroidogenic enzymes expression in experimental animal model following withdrawal of nandrolone decanoate

  • Anabolic steroids are frequently used to increase the growth rate of meat-producing animals. Exposure to an anabolic-androgenic steroid, nandrolone decanoate (ND), is associated with expressional reduction of testicular steroidogenic enzymes. However, the effect of withdrawal of ND exposure on the expression of these testicular molecules has not been thoroughly explored. The current research investigated expression changes of testicular steroidogenic enzymes in rats at several recovery periods (2, 6, and 12 weeks) after the stop of ND treatment with different doses (2 and 10 mg/kg body weight) for 12 weeks. Body and testis weights were recorded, and transcript levels of molecules were determined by quantitative real-time polymerase chain reaction (PCR). The immunohistochemistry was used to examine the changes of immuno-intensities of molecules. At 6 and 12 weeks of the recovery period, the 10 mg/kg ND-treated rats were lighter than other experimental groups. The interstitial compartment vanished by ND treatment filled up as the recovery period became longer. The expression of steroidogenic acute regulatory protein was returned to the control level at 12 weeks of the recovery period. Expression levels of cytochrome P450 side-chain cleavage and 17a-hydroxylase were increased in 2 mg/kg ND-treated group at 6 weeks of the recovery period, and transcript levels of these molecules in 2 and 10 mg/kg ND-treated groups at 12 weeks of the recovery period were significantly lower than the control. Expression levels of 3β-hydroxysteroid dehydrogenase (HSD) type I and 17β-HSD type 3 in 2 mg/kg ND-treated group were comparable with those of control at 12 weeks of the recovery period, but not in 10 mg/kg ND-treated group. Expression of cytochrome P450 aromatase (Cyp19) was reverted to the control level at 2 weeks of the recovery period. Except for Cyp19, there was a visible increase of immuno-staining intensity of other testicular steroidogenic enzymes in the Leydig cells as the recovery period progressed. This research has demonstrated that the cease of ND administration could restore the expression of testicular steroidogenic enzymes close to the normal level. Nevertheless, a relatively long recovery period, compared to the ND-exposure period would be required to retrieve normal expression levels of testicular steroidogenic enzymes.

A Study on Improvement of Hydrologic Cycle by Selection of LID Technology Application Area -in Oncheon Stream Basin- (LID 기술 적용 지역 선정에 따른 물순환 개선 연구 -온천천 유역을 대상으로-)

  • The frequency by water disaster in urban areas are increasing continuously due to climate change and urbanization. Countermeasures are being conducted to reduce the damage caused by water disasters. An analysis based on permeability, one of the parameters that affect runoff, is needed to predict quantitative runoff in urban watersheds and study runoff reduction. In this study, the SWAT model was simulated for the oncheon stream basin, a representative urban stream in Busan. The permeability map was prepared by calculating the CN values for each hydrologic response unit. Based on the permeability map prepared, EPA SWMM analyzed the effect of LID technology application on the water cycle in the basin for short-term rainfall events. The LID element technology applied to the oncheon stream basin was rooftop greening in the residential complex, and waterproof packaging was installed on the road. The land cover status of the land selected based on the permeability map and the application of LID technology reduced the outflow rate, peak flow rate, and outflow rate and increased the infiltration. Hence, LID technology has a positive effect on the water cycle in an urban basin.

CRISPR/Cas9-mediated knockout of Rag-2 causes systemic lymphopenia with hypoplastic lymphoid organs in FVB mice

  • Recombination activating gene-2 (RAG-2) plays a crucial role in the development of lymphocytes by mediating recombination of T cell receptors and immunoglobulins, and loss of RAG-2 causes severe combined immunodeficiency (SCID) in humans. Rag-2 knockout mice created using homologous recombination in ES cells have served as a valuable immunodeficient platform, but concerns have persisted on the specificity of Rag-2-related phenotypes in these animals due to the limitations associated with the genome engineering method used. To precisely investigate the function of Rag-2, we recently established a new Rag-2 knockout FVB mouse line ($Rag-2^{-/-}$) manifesting lymphopenia by employing a CRISPR/Cas9 system at Center for Mouse Models of Human Disease. In this study, we further characterized their phenotypes focusing on histopathological analysis of lymphoid organs. $Rag-2^{-/-}$ mice showed no abnormality in development compared to their WT littermates for 26 weeks. At necropsy, gross examination revealed significantly smaller spleens and thymuses in $Rag-2^{-/-}$ mice, while histopathological investigation revealed hypoplastic white pulps with intact red pulps in the spleen, severe atrophy of the thymic cortex and disappearance of follicles in lymph nodes. However, no perceivable change was observed in the bone marrow. Moreover, our analyses showed a specific reduction of lymphocytes with a complete loss of mature T cells and B cells in the lymphoid organs, while natural killer cells and splenic megakaryocytes were increased in $Rag-2^{-/-}$ mice. These findings indicate that our $Rag-2^{-/-}$ mice show systemic lymphopenia with the relevant histopathological changes in the lymphoid organs, suggesting them as an improved Rag-2-related immunodeficient model.

CRISPR/Cas9-mediated knockout of CD47 causes hemolytic anemia with splenomegaly in C57BL/6 mice

  • CD47 (integrin-associated protein), a multi-spanning transmembrane protein expressed in all cells including red blood cells (RBCs) and leukocytes, interacts with signal regulatory protein ${\alpha}$ ($SIRP{\alpha}$) on macrophages and thereby inhibits phagocytosis of RBCs. Recently, we generated a novel C57BL/6J CD47 knockout ($CD47^{-/-}$ hereafter) mouse line by employing a CRISPR/Cas9 system at Center for Mouse Models of Human Disease, and here report their hematological phenotypes. On monitoring their birth and development, $CD47^{-/-}$ mice were born viable with a natural male-to-female sex ratio and normally developed from birth through puberty to adulthood without noticeable changes in growth, food/water intake compared to their age and sex-matched wild-type littermates up to 26 weeks. Hematological analysis revealed a mild but significant reduction of RBC counts and hemoglobin in 16 week-old male $CD47^{-/-}$ mice which were aggravated at the age of 26 weeks with increased reticulocyte counts and mean corpuscular volume (MCV), suggesting hemolytic anemia. Interestingly, anemia in female $CD47^{-/-}$ mice became evident at 26 weeks, but splenomegaly was identified in both genders of $CD47^{-/-}$ mice from the age of 16 weeks, consistent with development of hemolytic anemia. Additionally, helper and cytotoxic T cell populations were considerably reduced in the spleen, but not in thymus, of $CD47^{-/-}$ mice, suggesting a crucial role of CD47 in proliferation of T cells. Collectively, these findings indicate that our $CD47^{-/-}$ mice have progressive hemolytic anemia and splenic depletion of mature T cell populations and therefore may be useful as an in vivo model to study the function of CD47.

Institutional Dynamics of In-Work Poverty Determination: Distributive Process of Labor Markets, Households, and the Welfare State Using Korean Welfare Panel Study, 2008-15 (근로빈곤 결정의 제도 동학: 노동시장과 가구, 복지국가 분배 과정 분석)

  • This paper adopts a distributive performance process model of in-work poverty based on labor markets, households, and welfare states and analyzes the 4-11 waves of the Korean Welfare Panel Study during 2008-15. Previous studies on in-work poverty have focused on the definitions and concepts of in-work poverty by analyzing employment and unemployment persistence and repetition dynamics, but rarely paid attention to institutional distributive performance. In this regard, this study preforms a stepwise analysis of labor markets, households, and welfare states as a process of income generation in labor markets, satisfaction of welfare needs and income pooling at households, and deduction of social security contribution and income tax as well as receipt of public transfer income at welfare states. Results of empirical analysis show that in-work poverty had been on increase during 2008-11, followed by a decrease between 2012-15. At labor market stages, full time status had the most prominent impact on in-work poverty process, while status by employment and contract type have generated a huge variation as well. At household stages, household work intensity and number of earners contributed to reduction of in-work poverty, but the relations did not seen to be straightforward. However, welfare state played little role in lifting employees out of in-work poverty. In terms of institutional distributive process, in-work poverty was prevalent in either household-welfare state stage or labor market-household-welfare stage. Non-vulnerable group in terms of in-risk poverty was around 80% of the sample during the period of analysis, the size of which has remained constant.

An Examination of Financial Feasibility and Redistributive Effect of Universal Basic Income (기본소득의 재정적 실현가능성과 재분배효과에 대한 고찰)

  • This article critically reviews the arguments that deny the financial feasbility and effectiveness of universal basic income as an alternative to existing social security systems and makes some suggestions to design effective and efficient basic income schemes. Regarding the financial feasibility of universal basic income, I argue that replacement of the existing regressive tax expenditures with universal basic income without raising tax rates can effectively reduce tax burden or provide income support to a majority of people except the rich. Addition of basic income to the tax base and reduction of the number of beneficiaries of public assistance and the amount of cash payment for them can further help save money. Regarding the redistributive effect, I note that the targeting ability of the existing social security systems is not good and that "the paradox of redistribution" that universal-type programs tend to be more redistributive than selective programs applies to universal basic income as well. I demonstrate significant redistributive effect of a hypothetical revenue-neutral basic income scheme and reviews several empirical studies done in Korea and abroad to show that basic income can be more effective in redistribution than social insurances or public assistance programs. Lastly, I emphasize the need to construct a reliable tax-benefit microsimulation model to help researchers to study redistributive effects of basic income schemes and other taxes and social policies.

An experimental study on the operation mode of rapid flooding protection system in tunnel (축소모형실험을 통한 터널 내 급속침수 차폐자동화 시스템 작동형태에 대한 연구)

  • This study focuses on the verification of a rapid protection automation system using an inflatable structure. The inflatable structure is an automatic rapid protection system against human and material damage when the subsea tunnel is flooded. Especially, it is essential for construction and operation of subsea tunnels. In this study, we have experimentally verified the rapid protection automation system using the inflatable structure designed for this problem. In order to verify this, a model tunnel with a 40: 1 reduction ratio was constructed, and air pressure of 0.1 bar and 0.15 bar was injected to divide the tunnel according to the expansion rate at 10 sec and 20 sec. According to the results of the study, the protection efficiency was better at 0.15 bar than 0.1 bar when the expansion structure was expanded, and the protection efficiency and influent control efficiency were different according to the pneumatic injection time of the inflating structure. As a result of this study, it was found that the higher the internal air pressure of the inflated structure and the faster the inflation of rate, the more effectively the inflated structure was inflated. As a result of this study, it is necessary to further study the wedge type structure which is useful for the storage method of expansion structure, shape and expansion derivative, inhibition of expansion structure during protection and control of inflow water.

Anti-inflammation and hangover relief effects of Schisandra chinensis (SC) and Lycium chinense (LC) water extracts depending on drug processing and fermentation (포제 및 발효 가공에 따른 오미자와 구기자 물 추출물의 항염증 및 숙취해소 효과)

  • Schisandra chinensis (SC) and Lycium chinense (LC) were widely distributed in Asia and the fruit has been used traditionally for medicinal herbs. The processing method was solid-state fermentation using Aspergillus oryzae for 48 h after stir-frying treatment at $220^{\circ}C$ for 12 min. In this study, in vitro the anti-inflammatory effect and in vivo hangover reduction were compared to unprocessed SC and LC water extract. Anti-inflammatory effects have been evaluated in pro-inflammatory mediators which were secreted by lipopolysaccharide (LPS)-induced RAW 264.7 macrophages. Nitric oxide (NO) was determined using Griess reaction. Proinflammatory cytokines such as tumor necrosis factor $(TNF)-{\alpha}$ and interleukin $(IL)-1{\beta}$ were measured by enzyme-linked immunosorbent assays (ELISA). Alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activities were compared to processed SC or LC and mixtures thereof (1:1). In vivo study was compared to hangover relief in alcohol-fed mice. After administering a mixture of SC and LC (300 mg/kg) water extract (1:1), mice were fed 3 g/kg of ethanol. Serum was collected at 1, 3, and 5 h intervals to analyze ethanol and acetaldehyde levels using a colorimetric assay kit. The processed SC and LC water extracts compared to raw materials significantly inhibited LPS-induced NO and inflammatory cytokine production in RAW 264.7 cells. The results of the hangover mouse model are also consistent with anti-inflammatory effects. These results suggest that processed SC and LC extracts may be functional materials for the treatment of inflammation and hangover.
