Characterizing LipR from Pseudomonas sp. R0-14 and Applying in Enrichment of Polyunsaturated Fatty Acids from Algal Oil

  • In this study, Pseudomonas R0-14, which was isolated from Arctic soil samples, showed a clear halo when grown on M9 medium agarose plates containing olive oil-rhodamine B as substrate, suggesting that it expressed putative lipase(s). A putative lipase gene, lipR, was cloned from R0-14 by genome walking and Touchdown PCR. lipR encodes a 562-amino-acid polypeptide showing a typical α/β hydrolase structure with a catalytic triad consisting of Ser153-Asp202-His260 and one α-helical lid (residues 103-113). A phylogenetic analysis revealed that LipR belongs to the lipase subfamily I.3. LipR was successfully expressed in Escherichia coli, purified, and biochemically characterized. Recombinant LipR exhibited its maximum activity towards p-nitrophenyl butyrate at pH 8.5 and 60℃ with a Km of 0.37 mM and a kcat of 6.42 s-1. It retained over 90% of its original activity after incubation at 50℃ for 12 h. In addition, LipR was activated by Ca2+, Mg2+, Ba2+, and Sr2+, while strongly inhibited by Cu2+, Zn2+, Mn2+, and ethylenediaminetetraacetic acid. Moreover, it showed a certain tolerance to organic solvents, including acetonitrile, isopropanol, acetone, methanol, and tert-butanol. When algal oil was hydrolyzed by LipR for 24 h, there was an enrichment of n-3 long-chain polyunsaturated fatty acids, including eicosapentaenoic acid (1.22%, 1.65-fold), docosapentaenoic acid (21.24%, 2.04-fold), and docosahexaenoic acid (36.98%, 1.33-fold), and even a certain amount of diacylglycerols was also produced. As a result, LipR has great prospect in industrial applications, especially in food and/or cosmetics applications.

호박류의 조리방법에 따른 무기질 성분의 변화 (Effect of Cooking Methods on Elemental Composition of Pumpkin (Cucurbitaceae spp.))

  • 본 연구는 애호박, 쥬키니, 단호박 및 늙은호박 조리방법에 따른 무기성분 변화를 확인하고자 하였다. 조리방법은 데치기, 전자레인지 및 찌기를 이용하였으며, 조리시간은 3분, 5분 및 10분으로 설정하였다. 설정된 시간에 따라 3가지 조리방법을 이용하여 조리한 후 건조하였다. 건조된 시료는 분쇄 후 microwave법을 이용하여 분해하였으며, 다량 무기성분은 ICP-OES로, 미량 무기성분은 ICP-MS를 이용하여 분석하였다. 4종의 호박에 함유된 다량 무기성분은 칼륨, 인, 칼슘, 마그네슘, 황, 철, 아연, 나트륨 순으로 확인되었으며, 미량 무기성분은 망간, 구리, 루비듐, 바륨, 니켈, 크롬, 갈륨, 코발트, 리튬, 바나듐, 셀레늄 순으로 검출되었다. 4종의 호박에 함유된 무기성분은 데치기, 전자레인지 조리 및 찌기에서 모두 감소하였으며, 데치는 조리에서 가장 많은 감소를 보였다. 조리시간에 따른 무기성분의 함량 변화는 조리시간이 증가할수록 낮은 함량을 보였으며, 5분과 10분에서 큰 차이를 확인하였다. 미량 무기성분의 함량은 조리방법과 조리시간에 따른 큰 변화를 확인할 수 없었다. 호박의 종류에 따른 무기성분의 조성은 비슷하였으며, 무기성분의 함량은 약간의 차이를 나타내었다. 애호박과 늙은호박의 무기성분은 찌는 조리법에서 높은 잔존율을 보였으며, 쥬키니와 단호박은 전자레인지 조리 시 가장 높은 잔존율을 나타내었다. 결과적으로 조리방법과 시간은 호박의 무기성분 잔존량에 영향을 주는 것으로 확인되었다.

화력발전소의 혼합연료 사용에 따른 중금속 배출량 변화 연구 (A Study on the Changes in Heavy Metal Emissions when Using Mixed Fuel in a Thermal Power Plant)

  • Objectives: The aim of this research is to explore the total heavy metals from a coal-fired power plant burning bituminous coal with wood pellets due to the implementation of the Renewable Portfolio Standard policy (RPS, 10% of electricity from renewable energy resources by 2023). Methods: The research was carried out by collecting archival data and using the USEPA's AP-42 & EMEP/EEA compilation of emission factors for use in calculating emissions. The Monte Carlo method was also applied for carrying out the calculations of measurement uncertainty. Results: In this paper, the results are listed as follows. Sb was measured at 110 kg (2015) and calculated as 165 kg (2019) and 201 kg (2023). Cr was measured at 1,597 kg (2015) and calculated as 1,687 kg (2019) and 1,728 kg (2023). Cu was measured at 2,888 kg (2015) and calculated as 3,133 kg (2019) and 3,264 kg (2023). Pb was measured at 2,580 kg (2015) and calculated as 2,831 kg (2019) and 2,969 kg (2023). Mn was measured at 3,011 kg (2015) and calculated as 15,034 kg (2019) and 23,014 kg (2023). Hg was measured at 510 kg (2015) and calculated as 513 kg (2019) and 537 kg (2023). Ni was measured at 1,720 kg (2015) and calculated as 1,895 kg (2019) and 1,991 kg (2023). Zn was measured at 7,054 kg (2015) and calculated as 9,938 kg (2019) and 11,778 kg (2023). Se was measured at 7,988 kg (2015) and calculated as 7,663 kg (2019) and 7,351 kg (2023). Conclusion: This shows that most heavy metals would increase steadily from 2015 to 2023. However, Se would decrease by 7.9%. This analysis was conducted with EMEP/EEA's emission factors due to the limited emission factors in South Korea. Co-firewood pellets in coal-fired power plants cause the emission of heavy metals. For this reason, emission factors at air pollution control facilities would be presented and the replacement of wood pellets would be needed.

삼광광상의 모암변질과 원소분산 (Element Dispersion and Wallrock Alteration from Samgwang Deposit)

  • 삼광광상은 선캠브리아기 경기육괴의 화강편마암내에 발달된 단열대(NE, NW)를 따라 충진한 8개의 괴상맥으로 구성된 중열수 석영맥광상이다. 이 광상의 광화작용은 여러번의 단열작용에 의해 형성된 두시기의 석영+방해석시기(광화I시기)와 방해석시기(광화II시기)로 구성된다. 광화I시기의 열수작용에 의한 변질작용은 견운모화, 녹니석화, 탄산염화, 황철석화, 규화, 및 점토화작용등이 관찰되며 견운모대는 석영맥과 접촉한 부분에서 녹니석대는 석영맥으로부터 멀어짐에 따라 관찰된다. 견운모대의 모암변질광물은 대부분이 견운모 및 석영이며 일부 일라이트, 탄산염광물, 녹니석으로 구성된다. 녹니석대의 모암변질광물은 주로 녹니석, 석영과 소량 견운모, 탄산염광물 및 녹염석으로 구성된다. 견운모의 Fe/(Fe+Mg) 값은 0.45${\sim}$0.50(0.48$\pm$0.02)이며 백운모-펜자이트족에 해당되고 녹니석의 Fe/(Fe+Mg) 값은 0.74${\sim}$0.81(0.77$\pm$0.03)이고 대부분 브런스비자이트에 해당된다. 견운모와 녹니석에 대한 $Al_{IV}$-FE/(FE+Mg)의 다이어그램은 변질시 같은 광종의 견운모와 녹니석의 형성온도를 나타내는 지시자로써 유용하다. 이것은 계산된 녹니석 단종의 활동도가 $a3(Fe_5Al_2Si_3O_{10}(OH)_6$=0.0275${\sim}$0.0413, $a2(Mg_5Al_2Si_3O_{10}(OH)_6$=1.18E-10${\sim}$7.79E-7, $a1(Mg_6Si_4O_{10}(OH)_6$=4.92E-10${\sim}$9.29E-7로서 삼광광상의 녹니석은 iron-rich 녹니석으로 비교적 고온 (T>450$^{\circ}C$에서 모암과 평형상태에서 온도가 감소함에 따라 형성되었음을 알 수 있다. 모암변질시 ${\alpha}Na^+$, ${\alpha}K^+$, ${\alpha}Ca^{2+}$${\alpha}Mg^{2+}$는 각각 ${\alpha}Na^+$=0.0476($400^{\circ}C$), 0.0863($350^{\circ}C$), ${\alpha}K^+$=0.0154($400^{\circ}C$), 0.0231($350^{\circ}C$), ${\alpha}Ca^{2+}$=2.42E-11($400^{\circ}C$), 7.07E-10($350^{\circ}C$), ${\alpha}Mg^{2+}$=1.59E-12($400^{\circ}C$), 1.77E-11($350^{\circ}C$)이며 열수용액의 pH는 5.4${\sim}$6.4($400^{\circ}C$), 5.3${\sim}$5.7($350^{\circ}C$)로서 모암변질시 열수용액는 약산성이었음을 알 수 있다. 모암변질시 이득원소(부화원소)는 $TiO_2$, $Fe_2O_3(T)$,CaO, MnO, MgO, As, Ag, Cu, Zn, Ni, Co, W, V, Br, Cs, Rb, Sc, Bi, Nb, Sb, Se, Sn 및 Lu 등이며 특히 대부분의 광상에서 Ag, As, Zn, Sc, Sb, S,$CO_2$ 등의 원소가 현저하게 증가하므로 중열수 및 천열수 금-은광상의 탐사에 지시원소로서 활용될 수 있을 것이다.

광환경 조절이 인삼의 생육과 진세노사이드 함량에 미치는 영향 (Effect of Controlled Light Environment on the Growth and Ginsenoside Content of Panax ginseng C. A. Meyer)

  • Background: The photosynthetic efficiency cool-season, semi-shade ginseng is normal at low morning temperatures, but drops at high afternoon temperatures. Therefore, optimal plant performance would be ensured if it were possible to control daily light transmission rates (LTR). Methods and Results: Plants were grown in a controlled light environment that replicated 11 AM conditions and comparatively analyzed against plant grown under normal conditions. Growth in the controlled light environment resulted in a 2.81 fold increase in photosynthetic efficiency with no change in chlorophyll content, although LTR were high due to low morning temperatures. Increased aerial plant growth was observed in the ginseng plants adapted to the controlled light environment, which in turn influenced root weight. An 81% increase in fresh root weight (33.3 g per plant on average) was observed in 4-year-old ginseng plants grown in controlled light environment compared to the plants grown following conventional practices (18.4 g per plant on average). With regard to the inorganic composition of leaves of 4-year-old ginseng plants grown in controlled light environment, an increased in Fe content was observed, while Mn and Zn content decreased, and total ginsenoside content of roots increased 2.37 fold. Conclusions: Growth of ginseng under a favorable light environment, such as the condition which exist naturally at 11 AM and are suitable for the plant's photosynthetic activity creates the possibility of large scale production, excellent-quality ginseng.

선박 엔진용 Fuel Pump Block 소재(CK35)의 피로파손 원인규명 (A Cause Analysis of Fatigue Failure of Fuel Pump Block Material(CK35) for Marine Engine)

  • DIN CK35 (JIS S35CK) steels have been used as a material in fuel pump blocks for marine engines. Failures in the inner surface of a drilling hole, due to the initiation of fatigue cracks have been frequently reported. However, the mechanism initiating these cracks and growths has not been clearly diagnosed yet. This study was conducted using a scraped fuel pump block, containing an initiated fatigue crack in the inner surface of a drilling hole. Initially, the cracks and fractured surfaces inside the block were investigated using an optical microscope and a SEM (Scanning Electron Microscope). In addition, microstructure observation, fatigue life test and fatigue crack growth test were performed using a specimen, which was taken from the same block. Results from these tests are summarized as follows; (1) The early crack in the block was supposed to occur inside the inner surface of the drilling hole. (2) The fatigue endurance of this material was about 330 Mpa. (3) The early crack was generated in the cavitations created by the breakdown of a big inclusion, or separation between the big inclusion and the base metal, in which the fundamental ingredients of the inclusion were C, 5, and Mn. (4) In order to prevent these types of failures, the suppression of inclusions inflow by improving the casting process, formation of fine inclusions by applying a heat treatment process, and coating of the surface of the drilling hole were required.

Otolith microchemistry reveals the migration patterns of the flathead grey mullet Mugil cephalus (Pisces: Mugilidae) in Korean waters

  • Background: The flathead grey mullet Mugil cephalus has the widest distribution among mugilid species. Recent studies based on mitochondrial DNA sequences showed that the species comprises at least 14 different groups, three of which occur in the northwest Pacific. We analyzed the otolith microchemistry of M. cephalus at several locations in Korea to improve understanding of migration pattern and population origin. Results: We collected 123 sagittal otoliths from seven locations and determined their concentrations of eight elements (7Li, 24Mg, 55Mn, 57Fe, 60Ni, 63Cu, 88Sr, and 138Ba) using laser ablation inductively coupled plasma mass spectrometry. Mean otolith elemental ratios differed significantly among the locations. The Sr:Ca, Fe:Ca, and Ba:Ca ratios were significantly higher than others, and useful chemical signatures for investigating the habitat use of M. cephalus populations. We identified five diverse and complicated migration patterns using the otolith data that we collected: estuarine resident (type I), freshwater migrant (type II), estuarine migrant (type III), seawater resident (type IV), and seawater migrant (type V). A canonical discriminant analysis plot revealed separation of two groups (type II in the Yellow Sea vs. other types in remaining locations). Two locations on Jeju Island, despite their close proximity, had fish with quite different migration patterns, corroborating previous molecular studies that distinguished two groups of fishes. Conclusion: We successfully showed that the migration patterns of the Korean mullet varied by location. Only fish from the western sector of Jeju had a unique migration pattern, which is likely confined population in this area. Among the eight otolith elements measured, the Sr:Ca ratio was found to be the best indicator of migration pattern and population origin.

Comparing the Dyeing Properties of Synthetic Mordants and Hwangsu Spring

  • To evaluate the Hwangsu spring as natural mordants effect on dyeing. The natural mordant effect of Hwangsu spring was analyzed by pH, temperature, trace elements and organic matter. Hwangsu spring having pH 2.42 and $14.4^{\circ}C$ was harvested at the end of September, 2011, in Yeongcheon(Gyeongbuk Province, Korea) and left on a $0^{\circ}C$ and $25^{\circ}C$ before use. The Result of ICP analysis, it contain bned Fe(414.9 ppm), Al(88.9 ppm), Mn(4.9 ppm). Dyeing and post-mordant procedure; Fabrics(cotton, silk, rayon) were dyed with natural colorants(Sopbora Japonica L., Caesalpinia sappan L., Allium cepa L.) for 20 min at $80^{\circ}C$. Hwangsu spring, aluminium sulfate $14-18H_2O(Al_2(SO_4)_3{\cdot}14-18H_2O)$, iron sulfate peptahydrate($FeSO_4{\cdot}7H_2O$), copper sulfate pentahydrate($CuSO_4{\cdot}5H_2O$) were used by post mordants. But in the case of Caesalpinia sappan L. was dyed after pre-mordanting with Al(II). The fabrics were dyed with each mordant solutions at $25^{\circ}C$ for 10min. The colorless also was measured by color-differnce meter. Comparison with a synthetic mordants, the K/S values of cotton fabrics dyed with Hwangsu sping were increased.

Enhanced pectinase and β-glucosidase enzyme production by a Bacillus subtilis strain under blue light-emitting diodes

  • Bacillus subtilis B22, a chemotrophic and aerobic bacterial strain was isolated from homemade kimchi, identified by 16S rRNA gene sequencing. B22 was primarily screened by biochemical, carbon source utilization tests. B22 was used to produce pectinase and ${\beta}$-glucosidase by submerged fermentation under different light sources. B22 was incubated in pectin media and basal media (pH 7.0) under blue, green, red and white light-emitting diodes (LEDs), fluorescent white light, and in darkness at $37^{\circ}C$, orbital shaker 150 rpm for 24 hours. Fermentation under blue LEDs maximized pectinase production ($71.59{\pm}1.6U/mL$ at 24 h) and ${\beta}$-glucosidase production ($56.31{\pm}1.6U/mL$ at 24 h). Further, the production of enzyme increased to pectinase ($156{\pm}1.28U/mL$) and ${\beta}$-glucosidase ($172{\pm}1.28U/mL$) with 3% glucose as a carbon source. Activity and stability of the partially purified enzymes were higher at pH 6.0 to 8.0 and $25-55^{\circ}C$. The effect on the metal ions $Na^+$ and $K^+$ and (moderateactivity) $Mn^{2+}$ and $Ni^{2+}$ increased activity, while $Hg^{2+}$, $Cu^{2+}$, $Fe^{2+}$, and $Fe^{2+}$ inhibited activity. EDTA, phenylmethylsulfonyl fluoride and 5,5-dithiobis (2-nitrobenzoicacid) reduced activity, while tetrafluoroethylene and 1,10-phenanthroline inhibited activity. The amylase was highly tolerant of the surfactants TritonX-100, Tween-20, Tween-80 and compatible with organic solvents methanol, ethanol, isoamylalcohol, isopropanol, t-butylalcohol and the oxidizing agents hydrogen peroxide, sodium perborate and sodium hypochlorite, although potassium iodide and ammonium persulfate reduced activity. These properties suggest utility of pectinase and ${\beta}$-glucosidase produced by B. subtilis B22 under blue LED-mediated fermentation for industrial applications.

Micrococcus luteus에서 정제한 Isocitrate Lyase의 특성 (Characterization of Isocitrate Lyase from Micrococcus luteus)

  • Micrococcus luteus 가 생산하는 isocitrate lyase 의 효소단백질을 ammonium sulfate precipitation, DEAE-cellulose, DEAE-sephacel, 1차 Sephadex G-200, 2 차 Sephadex G-200 column chromatography 과정을 통하여 분리 정제하였으며 정제된 효소의 분자량과 효소학적 특성을 조사하였다. 정제된 isocitrate lyase 는 상기 정제 과정을 통하여 10.2%의 회수율과 38.8 의 정제도를 나타내었으며, 전기영동시 단일밴드를 얻을 수 있었고, SDS-PAGE 에 의해 측정된 분자량을 약 60,000이며 1개의 subunit 로 나타났다. 본 효소의 최적 pH 는 7.5이었으며, 최적 온도는 $40^{\circ}C$ 부근이었고 $45^{\circ}C$까지는 안정하였다. 2가 금속염의 첨가시$ Mg^{2+}$ 를 제외한 다른 금속이온들은 효소활성을 저해하는 것으로 나타났으며 $Mg^{2+}$의 농도는 5 mM 에서 최대활성을 나타내었다. 또한 기질인 DL-isocitrate 의 $K_{m}$ 치는 0.95 mM 로 나타났으며, thiol 화합물의 첨가시 cysteine 은 1mM, glutathione과 $\beta$-mercaptoethanol 은 5 mM 의 농도에서 효소활성이 최대에 달하였다.
