Changes of Proteolytic Activity and Amino Acid Composition of the Tissue Extract from Sea Cucumber Entrails during Fermentation with Salt (해삼내장(內臟)젓갈 숙성중(熟成中) 단백질분해효소(蛋白質分解酵素)의 활성(活性)과 아미노산(酸) 조성(組成)의 변화(變化))

  • This study was undertaken to ascertain food and nutritional evaluating data on the processing of fermented sea cucumber (Stichopus japonicus) entrails. In the experiment, the crude proteolytic enzyme from the entrails tissue of raw and fermented sea cucumber during the days of ripening was extracted. The optimal activity condition for the crude enzyme and the compositional changes of amino acid of the protein and free amino acid in the raw and fermented sample were also investigated. 1. Less than three kinds of proteolytic enzymes that each enzyme has optimal activity condition at pH 3.1 $50^{\circ}C$(A-enzyme), pH 5.7 $50^{\circ}C$(B-enzyme) and pH 7.7 $45^{\circ}C$(C-enzyme), respectively were believed to be exist in the entrails tissue of sea cucumber. 2. A-enzyme and C-enzyme were strongly inhibited with the increase of the salt concentration, and B-enzyme was activated at the 1% salt concentration and was inhibited above the 5% salt concentration. 3. The result of the effect of several salt ions on the proteolytic activity showed that A-enzyme was slightly inhibited in the presence of all salt ions added, B-enzyme was activated in the presence of the all salt ions except $Cu^{2+}$ and C-enzyme was activated in the presence of $Ca^{2+}$ and $Mn^{2+}$, and inhibited by $Cu^{2+}$, $Co^{2+}$ and $Mg^{2+}$. 4. When the effects of the ripening days on the proteolytic activity of the crude enzymes were analysed, the activity of the A-enzyme was slightly weakened with the lapse of the fermentation days, whereas the B-enzyme was not influenced by the fermentation days. 5. In the analysis of amino acid composition of the protein of the samples, the 8 days fermented sea cucumber entrails showed the diminution of all kinds of amino acid. Apparently diminished amino acids were arginine, alanine, glutamic acid, glycine, serine, valine, threonine and lysine etc., and methionine, histidine and isoleucine were slightly decreased. 6. In the analysis of free amino acid composition of the 8 days fermented sample, glutamic acid, aspartic acid, leucine and lysine were rich, while histidine, methionine, proline and tyrosine were poor. The most of free amino acids were increased during the fermentation procedure and especially in lysine, histidine, threonine, glutamic acid, methionine, valine and leucine.

CRANIOFACIAL STRUCTURE AND ARCH DIMENSION OF ADULT CLASS III MALOCCLUSION (성인 III급 부정교합자의 악안면골격구조 및 치열궁형태에 관한 연구)

  • This study was conducted to discern differences of craniofacial, dentoalveolar structure and model measurements between sex and between class n openbite group and non-openbite group. The sample consisted of 49 adult patients with class Il malocclusion. 24 linear measurements, 22 angular measurements and 12 ratios were selected in lateral cephalometry. Also, arch width, length, anterior crowding, average molar relation were measured or calculated in diagnostic model. The data were evaluated by t-test and multiple discriminant analysis. The results were as follows, 1. Most linear measurements, with the exception of MnBL and AUDH, were significantly larger in male(p<0.05). but, intermaxillary relations and spatial position of maxilla and mandible relative to cranial base were not different for both sex. 2. With the exception of upper and lower anterior crowding, lower arch width, upper arch length, AMR, male exhibited significantly larger measurements in model analysis (p<0.05). 3. Size differences of maxilla and mandible between openbite and non-openbite group were not significant(p>0.05). but openbite group showed significantly increased genial angle(p<0.05), FH-CoGo(p<0.01), FH-NA(p<0.01) and FH-NB, FH-NPog (p<0.05). 4. ALFH and PUDH were larger(p<0.05) in openbite group. this result served as compensation for the spatial position of mandible relative to cranial base. AUPUDH (p<0.001) and ALPLDH(p<0.05) were lower in openbite group. upper anterior crowding was the only measurement which showed difference between openbite and non-openbite group(p<0.05). 5. For the purpose of classifying adult class n openbite and non-openbite group, multiple discriminant analysis was done genial angle, ALPLDH, AUPUDH, FH-NA were included in multiple discriminant equation. 39 cases($92.86\%$) were correctly classified when applied to the sample used in this study.

The Evaluation of Feed Value and Growth Characteristics of Sasa quelpaertenisis Nakai by Horse Grazing in the Woodland of Jeju (말 방목에 의한 제주조릿대의 생육특성 및 사료 가치 평가)

  • This study was conducted to evaluate the feed value and growth characteristics of Sasa quelpaertensis Nakai by horse grazing in the woodland of Jeju. Crude protein (CP), NDF, ADF and minerals contents at different growth stages of Sasa quelpaertensis were analyzed. The apparent digestibilities of DM, CP, NDF and ADF were determined by total collection method using 5 Jeju horses. The yield and growth characteristics of S. quelpaertensis Nakai by horse grazing were investigated. CP contents in the currentand the previous-year-sprouted leaves of S. quelpaertenisis were $16.6{\pm}1.0%$ and $12.2{\pm}1.1%$, respectively. The digestibility of DM, CP, NDF and ADF in the current-year-sprouted leaves of S. quelpaertenisis were $47.2{\pm}1.2$, $67.8{\pm}1.0$, $47.0{\pm}1.3$ and $47.1{\pm}1.1%$, respectively. The current-year-sprouted S. quelpaertenisis tended to have high nutritional values and digestibilities compared to those of the previous-year-sprouted. The dry matter yield, plant length, leaf width and density of S. quelpaertensis Nakai were clearly decreased by horse grazing. These results show that considering the feed value and growth characteristics, the natural S. quelpaertenisis Nakai can be used as a feed source for horses.

Vertical Distribution of the Heavy Metal in Paddy Soils of Below Part at Guundong Mine in Milyang, Korea (구운동 폐광산 하류 논토양의 토심별 중금속함량)

  • This study was conducted to investigate form of pollution brought by residual of mine tailing in agricultural land, and get basic information need for environment restoration. Guundong mine was completely restored region by implementation the soil pollution prevention plan. The districts is soils in Guundong mine vicinity the Mahul-ri, Muan-myeon, Miryang city, Gyeongsangnam-do. The nature of soil studied is the Shinra series andesite and mineral deposits which contain brimstone and heavy metals such as gold, silver, copper, lead, and zinc. The residual mine tailing and around agricultural land of heavy metals analyzed with 0.1N HCI solubility. The chemical properties of surface soil in upper part around mining area were pH 4.3-4.4, organic matter 19-21 g $kg^{-1}$, available $P_2O_5$ 85 mg $kg^{-1}$, exchangeable Ca 0.21-0.25 $cmol_c\;kg^{-1}$, exchangeable Mg 0.04 $cmol_c\;kg^{-1}$. The pH, exchangeable Ca, and Mg were increased with soil depth. The contents of 0.1N HCl extractable Cu, Cd, Pb, Cr, and Ni in soil (siteI) which influenced by outflow water from mine tailing were 97, 0.6, 197, 0.28 및 0.12 mg $kg^{-1}$, respectively. The vertical distribution of heavy metals in soil varied considerably among the metals kind. In case of siteI, The content of Cu, Pb, and Cr in soil was highest at surface soil. However, the content of Cd, Zn, Ni, and Mn was high at middle part of soil profile.

Characteristics of Commercial Sigumjang meju (시판 전통 시금장 메주의 품질특성조사)

  • It is very distinctive that sigumjang meju was produced with smoking after molding. Every shapes of traditional sigumjang meju gathered from Kyeongsangbukdo were doughnut types. Fermentation time were $60{\sim}90$ days. Aerobic bacteria were $6.8{\times}10^7\;cfu/g$, and anaerobic bacteria, yeasts, molds count $3.2{\times}10^6\;cfu/g,\;1.0{\times}10^6\;cfu/g\;and\;4.0{\times}10^6\;cfu/g$, respectively. Analysis showed: moisture content, $10.5{\pm}2.6%$; fat content, $2.9{\pm}1.1%$; protein content, $8.3{\pm}0.7%$; ash content, $3.8{\pm}0.7%$; pH, $6.0{\pm}0.5$; L-value, $54.5{\pm}4.7$; a-value, $+3.3{\pm}0.7$; and b-value, $+10.7{\pm}2.0$. K content was the most in quantity among the minerals in sigumjang meju. Maltose was most abundant component among the tested sugars followed by mannitol. Total content of amino acid was $1524.9{\pm}1295.3\;mg%$. Proline was the most abundant component among the amino acids. The ratio of essential amino acid was $30.0{\pm}12.9%$. The principal fatty acids were 18:2, 18:1 and 16:0. Saturated fatty acid and mono unsaturated fatty acid were $23.2{\pm}1.9%\;and\;23.5{\pm}5.2%$, respectively. The PUFA/SFA was $2.3{\pm,}0.3$ which higher than 1.

The Study on the Amount and Major Compositions of Excreta from Swine (돼지의 분뇨 배설량 및 분뇨 성분 조사)

  • This work was carried out to investigate the quantity of excreta and its composition in crossbred pigs (Yorkshire ${\times}$ Landrace ${\times}$ Duroc) at different stages of growth. Twelve young piglets (average BW weight of $19.0{\pm}0.33kg$) were used in this study. Pigs were divided into four phases during growing time and two phases during finishing time. The average excreta production for growing pig was 3.46 kg/head/day (feces: 1.07 kg, urine: 2.39 kg). The average moisture contents of feces and urine were 70.54% and 97.39%, respectively. Contents of Calcium, Magnesium, Copper, Plumbum, and Arsenic were 1.00%, 0.26%, 10.47 mg/kg, 2.43 mg/kg, and 1.02 mg/kg, respectively. The concentration of the water pollutants like Biochemical Oxygen Demand ($BOD_5$), Chemical Oxygen Demand (COD), Suspended Solid (SS), Total Nitrogen (TN) and Total Phosphorus (TP), excreted from pig were 96335, 61073, 207466, 8104 and 4209 mg/L in feces and 7364, 7149, 2715, 10110 and 613 mg/L in urine at the end of test, respectively. The daily loading amount of water pollutants ($BOD_5$, COD, SS, TN, and TP, respectively) in pig excreta were 102.1, 61.8, 221.6, 8.7, and 3.9 g/head/day in feces, and 19.3, 16.7, 8.0, 22.2, and 1.3 g/head/day in urine, respectively. The Nitrogen, $P_2O_5$, and $K_2O$ contents in the excreta of pigs were 0.96, 0.83 and 0.42% in feces, and 0.80, 0.09 and 0.53% in urine, respectively. Finally, this work was suggested to give basic information to swine farms.

Effects of Allium Vegetable Intake on Levels of Plasma Glucose, Lipid and Minerals in Streptozotocin Induced Diabetic Rats (Allium속 식용식물의 섭취가 Streptozotocin 유발 당뇨 흰쥐의 혈당, 지질 및 무기질 수준에 미치는 영향)

  • The effects of Allium vegetables on blood glucose levels and lipid metabolism in streptozotocin (S12) induced diabetic rats were investigated. Diabetes mellitus was induced by S1'2 injection (45 mg/kg 5.w.) into the tail vein. Sprague-Dawley rats weighing $220\;{\pm}\;10\;g$ were randomly assigned to 7 groups: normal, S1'2-control and five Allium groups (Allium cepa, Allium fistulosum, Allium sativum, Allium tuberosum and Allium victorialisL Normal and S12control groups were fed an AIN-93 diet and five Allium groups were fed a modified diet containing. 10% Allium powder each for 4 weeks. Body weight, diet intake, food efficiency ratio (FER) and organ weights- were monitored. Activities of aspartate aminotransferase (AST) & alanine aminotransferase (ALT) were observed: Plasma lev~ls of glucose, free fatty acid, triglyceride and HDL-cholesterol were analyzed. Levels of glycogen, cholesterol and triglyceride in liver were determined. Levels of malondialdehyde (MDA) in liver, lung, kidney, and pancreas were assayed. The hepatic contents of chromium (Cr) , iron (Fe), zinc (Zn) and manganese (Mu) were measured. The Allium sativum group had weight gain and suppressed a hypertrophy of the kidney significantly. The activity of ALT was significantly lowered in the diabetic groups except Allium sativum group compared to STZ-control group. The Allium sativum and Allium tuberosum groups showed the hypoglycemic effects at 4 weeks. There were no significant differences between the control and all the other diabetic groups in the plasma levels of cholesterol, HDL-cholesterol, triglycerides and free fatty acids. Most of the Allium groups except Allium fistulosum were observed significantly lowered level of MDA in the lung compared to STZ-control group. The diabetic rats fed the Allium cepa and Allium sativum have shown significantly lowered hepatic Zn contents. The results suggested that the intake of the Allium vegetables may be effective in the antihyperglycemia by lowering blood glucose levels.

Initial Charge/Discharge of $LiCoO_2$ Composite Cathode with Various Content of Conductive Material for the Lithium ion Battery (리튬이온전지용 $LiCoO_2$정극의 도전재료에 따른 초기 충방전 특성)

  • Initial electrochemical characteristics of $LiCoO_2$ electrode for lithium ion battery with various content of super s black as conductive material were evaluated through the charge/discharge with the potential range of 4.3V to 2.0V versus $Li^+/Li^+$. The rate of C/4 and C/2 by the 3 electrode test cell composed with an electrolytic solution of 1 mol/l $LiPF_6/EC+DEC(1:3\;by\; weight)$. Lithium was used as reference electrode. High impedance charge behavior was observed at early stage of charge. In the case of $3\%w/w$ of super s black as conductive material, the specific resistance of the high impedance releasing was $3.82\;{\Omega}\;{\cdot}\;g-LiCoCo_2$ at the current density of $0.5 mA/cm^2$, which corresponds 7 times of the specific resistance of electrode $(0.728 g-LiCoO_2)$. At second charge, the specific resistance of the high impedance releasing was 63 mn · g-Lico02, which corresponds 12eio of the specific resistance of electrode and only $1.7\%$ of that of the first charge. The first charge and discharge specific capacities at C/4 rate were 160-161 and $153\~155mAh/g-LiCoO_2$, respectively, to lead $95.4\~96.4\%$ of coulombic efficiencies and ca. $6 mAh/g-LiCoO_2$ of initial irreversible specific capacity. Specific resistance at the end of charge and rest showed low value at content of super s black between 2 and $7\%w/w$, which agreed with characteristics of irreversible specific capacity. Capacity densities were reduced by the increasing the content of conductive material. They were 447 and 431mAh/ml when 2 and $2.9\%w/w$ of super s black were used, respectively, at the rate of C/4.

Analysis of Growth Response by Non - destructive, Continuous Measurement of Fresh Weight in Leaf Lettuce 1. Effect of Nutrient Solution and Light Condition on the Growth of Leaf Lettuce (비파괴 연속 생체중 측정장치의 개발 및 이에 의한 상추의 생장반응 분석 l. 양액의 이온 농도 및 명ㆍ암 처리가 생장에 미치는 영향)

  • These studies were carried out to develop a system for non -destructive and continuous measurement of fresh weight and to analyse the growth response of leaf lettuce under the different nutrient solution and light condition with this system. The developed measurement system was consisted of four load cells and a microcomputer. The output from the system was highly positive correlation with the plant fresh weight above the surface of the hydroponic solution. The top fresh weight of plant could be measured within the error $\pm$ 1.0g in the range of 0 - 2000g. The top fresh weight of leaf lettuce increased 44 times at 18th day after transferring to the nutrient solution, and the maximum growth rate was observed at 13th day after transferring. The growth rate was 10.7- 29.6% per day during 18 days. Optimum concentration of the nutrient solution for the growth of lettuce was 1.4 - 2.2 mS/cm of EC level. When the light condition was changed from dark to light, the fresh weight was temporarily decreased, but the fresh weight increased under the opposite condition. Top fresh weight of leaf lettuce in the darkness normally increased within 12 hours after darkness treatment, and then slowly increased until 78 hours under continuous dark condition. After that times, the fresh weight began to decrease.

Clinicopathological Analysis of Glomerulonephritis in Children (소아 사구체신염의 임상 및 병리학적 분석)

  • Purpose: To evaluate the prevalence and clinical manifestations of various glomerulonephritis (GN) in children, a clinicopathological anlysis of 310 biopsied cases were performed. Method: We conducted retrospective study with review of histopathologic findings and clinical manifestations of the 310 cases diagnosed as glomerulonephritis by percutaneous renal biopsy which were done between January 1986 and December 1996 at department of pediatrics, Pusan Paik hospital. Results: 1) Male to female ratio was 1.54:1 and the range of age was from 13 months to 15 years 10 months. 2) Among these, 217 (70.0%) patients were belong to primary GN and 93 (30.0%) patients were belong to secondary GN. As a whole, the most common pathologic diagnosis was minimal change lesion (MC, 32.6%), which was followed by IgA nephropathy (IgAN, 15.8%), $Henoch-Sch\"{o}nlein$ purpura nephritis (HSPN, 13.5%), Poststreptococcal glomerulonephritis (PSAGN, 8.1%). 3) Clinical manifestations of patients were asymptomatic urinary abnormality (43.2%), nephrotic syndrome (41.0%), acute glomerulonephritis (14.2%), chronic glomerulonephritis (1.0%), rapidly progressive glomerulonephritis (0.6%). 4) In primary GN, the most common pathologic diagnosis was MC (46.5%), IgAN (22.6%), thin glomerular basement membrane (GBM) disease (7.8%), membranoproliferative glomerulonephritis (MPGN, 5.5%), mesangial proliferative glomerulonephritis (MesPGN,4.6%), focal segmental glomerulosclerosis (FSGS, 4.6%), membranous nephropathy (MN, 0.9%), sclerosing glomerulonephritis (SCGN, 0.9%), crescentic glomerulonephritis (CreGN, 0.5%) and non-specific glomerulonephritis (NonspGN, 6.0%). 5) Major causes of secondary GN were HSPN (45.2%), PSAGN (26.9%), hepatitis B associated glomerulonephritis (HBGN, 17.2%), lupus nephritis (LN, 6.5%), Alport syndrome (2.2%), hemolytic uremic syndrome (1.0%), fibrillary glomerulonephritis (1.0%) in descending order. Conclusions: There are some differences of the results of clinicopathological stuidies of glomerulonephritis in children because of its different indications of renal biopsy, pathologic classification of renal disease and methods of analysis among investigators. In order to establish more reliable data of incidence and classification of childhood glomerulonephritis in Korea, multicenter cooperative study were necessary.
