Niclosamide induces mitochondria fragmentation and promotes both apoptotic and autophagic cell death

  • Mitochondrial dynamics not only involves mitochondrial morphology but also mitochondrial biogenesis, mitochondrial distribution, and cell death. To identify specific regulators to mitochondria dynamics, we screened a chemical library and identified niclosamide as a potent inducer of mitochondria fission. Niclosamide promoted mitochondrial fragmentation but this was blocked by down-regulation of Drp1. Niclosamide treatment resulted in the disruption of mitochondria membrane potential and reduction of ATP levels. Moreover, niclosamide led to apoptotic cell death by caspase-3 activation. Interestingly, niclosamide also increased autophagic activity. Inhibition of autophagy suppressed niclosamide-induced cell death. Therefore, our findings suggest that niclosamide induces mitochondria fragmentation and may contribute to apoptotic and autophagic cell death.

RFLPs of Mitochondrial DNA in Korean Wild Soybeans

  • Mitochondrial DNA restriction fragment length polymorphisms are convenient markers for identifying cytoplasmic variation among plants. We have collected 212 wild soybeans (Glycine soja Sieb. et Zucc) from all over Korea, and classified mitochondrial genome types based on hybridization patterns in DNA gel-blot analyses using two mitochondrial DNA clones, cox2 and atp6, as probes. Korean wild soybean was classified with eight-mtDNA types, and some of the mtDNAs showed geographical clines among the regions. The diversity index of the mtDNA was much higher in the western and southern regions than in the eastern and northern regions of Korea, respectively. Dissemination and distributive characteristics of wild soybeans in Korea were discussed.

The Complete Mitochondrial Genome of Dendronephthya gigantea (Anthozoa: Octocorallia: Nephtheidae)

  • We sequenced the whole mitochondrial genome of Dendronephthya gigantea (Anthozoa: Octocorallia: Nephteidae), the first mitochondrial genome sequence report in the Family Nephtheidae. The mitochondrial genome of D. gigantea was 18,842 bp in length, and contained 14 protein coding genes (atp6 and 8, cox1-3, cytb, nd1-6 and 4L, and msh1), two ribosomal RNAs, and only one transfer RNA. The gene content and gene order is identical to other octocorals sequenced to date. The portion of the noncoding regions is slightly larger than the other octocorals (5.08% compared to average 3.98%). We expect that the information of gene content, gene order, codon usage, noncoding region and protein coding gene sequence could be used in the further analysis of anthozoan phylogeny.

Major ginsenosides from Panax ginseng promote aerobic cellular respiration and SIRT1-mediated mitochondrial biosynthesis in cardiomyocytes and neurons

  • Background: Aerobic cellular respiration provides chemical energy, adenosine triphosphate (ATP), to maintain multiple cellular functions. Sirtuin 1 (SIRT1) can deacetylate peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α) to promote mitochondrial biosynthesis. Targeting energy metabolism is a potential strategy for the prevention and treatment of various diseases, such as cardiac and neurological disorders. Ginsenosides, one of the major bioactive constituents of Panax ginseng, have been extensively used due to their diverse beneficial effects on healthy subjects and patients with different diseases. However, the underlying molecular mechanisms of total ginsenosides (GS) on energy metabolism remain unclear. Methods: In this study, oxygen consumption rate, ATP production, mitochondrial biosynthesis, glucose metabolism, and SIRT1-PGC-1α pathways in untreated and GS-treated different cells, fly, and mouse models were investigated. Results: GS pretreatment enhanced mitochondrial respiration capacity and ATP production in aerobic respiration-dominated cardiomyocytes and neurons, and promoted tricarboxylic acid metabolism in cardiomyocytes. Moreover, GS clearly enhanced NAD+-dependent SIRT1 activation to increase mitochondrial biosynthesis in cardiomyocytes and neurons, which was completely abrogated by nicotinamide. Importantly, ginsenoside monomers, such as Rg1, Re, Rf, Rb1, Rc, Rh1, Rb2, and Rb3, were found to activate SIRT1 and promote energy metabolism. Conclusion: This study may provide new insights into the extensive application of ginseng for cardiac and neurological protection in healthy subjects and patients.

Mitochondrial activity in illuminated leaves of chlorophyll-deficient mutant rice (OsCHLH) seedlings

  • The rice CHLH gene encodes the $Mg^{2+}$-chelatase H subunit, which is involved in chlorophyll biosynthesis. Growth of the chlorophyll-deficient oschlh mutant is supported by mitochondrial activity. In this study, we investigated the activity of mitochondrial respiration in the illuminated leaves during oschlh seedling development. Growth of mutant plants was enhanced in the presence of 3% sucrose, which may be used by mitochondria to meet cellular energy requirements. ATP content in these mutants was, however, significantly lowered in light conditions. Low cytosolic levels of NADH in illuminated oschlh mutant leaves further indicated the inhibition of mitochondrial metabolism. This down-regulation was particularly evident for oxidative stressresponsive genes in the mutant under light conditions. Hydrogen peroxide levels were higher in oschlh mutant leaves than in wild-type leaves; this increase was largely caused by the impairment of the expression of the antioxidant genes, such as OsAPXl, OsRACl, and OsAOXc in knockout plants. Moreover, treatment of mesophyll protoplasts with ascorbic acid or catalase recovered ATP content in the mutants. Taken together, these results suggest that the light-mediated inhibition of mitochondrial activity leads to stunted growth of CHLH rice seedlings.

Cysteine improves boar sperm quality via glutathione biosynthesis during the liquid storage

  • Objective: Sperm is particularly susceptible to reactive oxygen species (ROS) stress. Glutathione (GSH) is an endogenous antioxidant that regulates sperm redox homeostasis. However, it is not clear whether boar sperm could utilize cysteine for synthesis GSH to protect sperm quality from ROS damage. Therefore, the present study was undertaken to elucidate the mechanism of how cysteine is involved in protecting boar sperm quality during liquid storage. Methods: Sperm motility, membrane integrity, lipid peroxidation, 4-hydroxyIlonenal (4-HNE) modifications, mitochondrial membrane potential, as well as the levels of ROS, GSH, and, ATP were evaluated. Moreover, the enzymes (GCLC: glutamate cysteine ligase; GSS: glutathione synthetase) that are involved in glutathione synthesis from cysteine precursor were detected by western blotting. Results: Compared to the control, addition of 1.25 mM cysteine to the liquid storage significantly increased boar sperm progressive motility, straight-line velocity, curvilinear velocity, beat-cross frequency, membrane integrity, mitochondrial membrane potential, ATP level, acrosome integrity, activities of superoxide dismutase and catalase, and GSH level, while reducing the ROS level, lipid peroxidation and 4-HNE modifications. It was also observed that the GCLC and GSS were expressed in boar sperm. Interestingly, when we used menadione to induce sperm with ROS stress, the menadione associated damages were observed to be reduced by the cysteine supplementation. Moreover, compared to the cysteine treatment, the γ-glutamylcysteine synthetase (γ-GCS) activity, GSH level, mitochondrial membrane potential, ATP level, membrane integrity and progressive motility in boar sperm were decreased by supplementing with an inhibitor of GSH synthesis, buthionine sulfoximine. Conclusion: These data suggest that boar sperm could biosynthesize the GSH from cysteine in vitro. Therefore, during storage, addition of cysteine improves boar sperm quality via enhancing the GSH synthesis to resist ROS stress.

모과추출물의 C2C12 근육세포에서 근분화 및 에너지대사조절인자 발현 증진 효과 연구 (Effects of Chaenomelis Fructus Extract on the regulation of myoblasts differentiation and the expression of biogenetic factors in C2C12 myotubes)

  • Objective : The present study was conducted to investigate the effects of Chaenomelis Fructus (CF) on the regulation of biogenesis in C2C12 mouse skeletal muscle cells. Methods : C2C12 myoblasts were differentiated into myotubes in 2% horse serum-containing medium for 5 days, and then treated with CF extract at different concentrations for 48 hr. The expression of muscle differentiation markers, myogenin and myosin heavy chain (MHC) and mitochondrial biogenesis-regulating factors, peroxisome proliferator-activated receptor gamma coactivator 1alpha (PGC1α), sirtuin1 (Sirt1), nuclear respiratory factor1 (NRF1) and transcription factor A, mitochondrial (TFAM), and the phosphorylation of AMP-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) were determined in C2C12 myotubes by reverse transcriptase (RT)-polymerase chain reaction (RT-PCR) and western blot, respectively. The cellular glucose levels and total ATP contents were measured by cellular glucose uptake and ATP assays, respectively. Results : Treatment with CF extract (0.01, 0.02, and 0.05 mg/㎖) significantly increased the expression of MHC protein in C2C12 myotubes compared with non-treated cells. CF extract significantly increased the expression of PGC1α and TFAM in the myotubes. Also, CF extract significantly increased glucose uptake levels and ATP contents in the myotubes. Conclusion : CF extract can stimulate C2C12 myoblasts differentiation into myotubes and increase energy production through upregulation of the expression of mitochondrial biogenetic factors in C2C12 mouse skeletal muscle cell. This suggests that CF can help to improve skeletal muscle function with stimulation of the energy metabolism.

Energy Status of Neurospora crassa Mutant nap in Relation to Accumulation of Carotenoids

  • N crassa mutant strain nap showed reduced growth rate, decreased electric membrane potential, and elevated intracellular ATP content in comparison to the wild type. Blue light induced a hyperpolarization of the membrane potential in both strains. The analysis of oxidative and phosphorylation activities of mitochondria isolated from the two strains has revealed that nap utilized more efficient oxidative pathways. The higher intracellular ATP content in the nap was presumably due to impaired transport systems of the plasma membrane, and to a lesser extent to the functioning of the fully competent respiratory chain. The excess ATP possibly accounts for carotenoid accumulation in the mutant.

미토콘드리아 기능 이상과 암 (Mitochondrial Dysfunction and Cancer)

  • 미토콘드리아는 세포 에너지 공급을 위한 에너지 대사의 주요 세포 소기관으로 칼슘 조절, 활성 산소(ROS) 생성, 세포 사멸(apoptosis)을 조절하는데도 중요한 역할을 한다. 이러한 미토콘드리아에 발생한 기능 이상은 신경퇴행질환, 루게릭병, 심혈관계 질환, 정신 질환, 당뇨, 암과 같은 다양한 질병과 연관이 있다. 미토콘드리아 기능 이상 관련 질병들은 노화와 관련된 질병이 주를 차지하며, 이 논문에서는 그 중에서도 암에 초점을 맞춰 서술하고자 한다. 미토콘드리아 기능 이상은 발암을 유도하며, 많은 암 종에서 발견된다. 암종에 따라 미토콘드리아 기능 이상을 일으키는 요인들이 다르며, 이러한 변화는 치료 내성, 전이와 같은 암 악성화도 유발한다. 미토콘드리아 기능 이상의 요인으로는 미토콘드리아 수 부족, 주요 물질 제공 불능, ATP 합성 기능 이상 등이 존재하나, 암 발병과 악성화에 영향을 미치는 주요 원인으로 미토콘드리아 DNA (mtDNA)의 감소(depletion)를 들 수 있다. 미토콘드리아 기능 이상은 분자 활성 변화 혹은 발현 변화를 통해 암 악성화를 일으키나, 어떠한 변화가 암 악성화를 야기하는지 구체적으로 알려진 바가 없다. 미토콘드리아 기능 이상과 암의 상관관계는 대부분 미토콘드리아 기능 이상 세포를 이용하여 연구하는데, 그 제작 방법으로는 EtBr에 의한 화학적 방법과 shRNA, Crispr/Cas9과 같은 유전자 수선 (gene editing) 방법 등이 있다. 이러한 기법으로 제작된 미토콘드리아 기능 이상 세포주는 암을 비롯한 미토콘드리아 기능 이상에 의한 다양한 질병 연구에 이용되고 있다.

HT-22 신경세포에서 아밀로이드 베타 펩티드에 의한 미토콘드리아와 세포 손상 기전에서 FUN14 도메인 함유 단백 1의 역할 (FUN14 Domain-Containing Protein 1 Is Involved in Amyloid Beta Peptide-Induced Mitochondrial Dysfunction and Cell Injury in HT-22 Neuronal Cells)

  • Fun14 도메인 함유 단백 1(Fun14 domain-containing protein 1, FUNDC1)은 미토콘드리아 외막에 존재하는 단백질로, 미토콘드리아의 마이토파지(mitophage) 기전 조절에 관여하는 것으로 알려져 있다. 본 연구에서는 해마 뉴런 기원의 HT-22 세포에서 아밀로이드 베타 펩티드(Aβ)에 의한 미토콘드리아와 세포 손상 과정에서 FUNDC1의 개재 가능성과 역할을 조사하였다. HT-22 세포에서 Aβ를 처리하면 처리 시간에 의존적으로 FUNDC1의 발현 감소가 관찰되었다. 또한 MTT 환원능과 세포 내 ATP 농도, 미토콘드리아 막전압의 감소, 반응성 산소종의 생성과 미토콘드리아 Ca2+ 부하의 증가 등 미토콘드리아의 기능적 손상을 나타내는 지표들의 변화와 함께 세포사멸의 증가가 관찰되었다. FUNDC1의 발현을 일시적으로 차단한 세포군에서도 미토콘드리아의 기능적 손상을 나타내는 지표 변화와 세포사멸의 증가가 관찰되었다. 반면에 FUNDC1을 일시적으로 과발현시킨 세포군에서는 Aβ 처리에 의한 미토콘드리아 손상과 세포 사멸이 유의하게 억제되었다. 이와 같은 결과들은 Aβ에 의한 미토콘드리아와 세포 손상 기전에 FUNDC1이 중요하게 관여할 가능성을 시사한다.
