Effects of atopic dermatitis induced materials on the expression of cytokine genes in human monocytes and mast cells (인간의 단핵구와 비만세포에서 다양한 아토피 유발물질이 사이토카인 유전자의 발현에 미치는 영향)

  • Objectives : On an experimental basis, the effects of atopic dermatitis induced materials on the expression of cytokine genes in human monocytes (THP-1, U937) and mast cells were studied. This study was carried out to be considered a fundamental knowledge in the research on the good of oriental medicine. Methods : After culturing THP-1, U937, and HMC-1, with the three different concentrations of LPS ($1\;{\mu}g/ml$), DPE ($10\;{\mu}g/ml$), and DNCB ($1\;{\mu}g/ml$), atopic dermatitis induced materials were treated in the culture medium. To investigate cytokine genes expression patterns, with lysis buffer and separation reagent, total RNA was extracted from THP-1, U937, and HMC-1 at intervals of 0, 12, 24, and 48 hours. Both cytokine mRNA expression patterns by atopic dermatitis induced materials and change of cytokine genes expression patterns in relation to atopy by selenium were analyzed with RT-PCR. Also IL-4 and INF-$\gamma$, which were secreted in the HMC-1, were analyzed using ELISA method. Results : 1. After treating THP-1 and U937 with LPS, DPE, and DNCB, there was no significant change in cytokine genes themselves, but various cytokines (IL-4, IL-6, IL-8, IL-13, IFN-$\gamma$, IFN-a, MCP-1, B2-MG) were expressed. 2. In the case of HMC-1, the expressions of IL-6 and IL-8 were significantly increased in the analysis of mRNA expression by dust mite allergens in DPE. 3. As a result of ELISA method, it is certain that IL-4 and IFN-$\gamma$ protein were secreted in the HMC-1 by DPE. 4. Selenium, an essential trace element, decreased the IL-10 and IL-13 expression in the HMC-1 by DPE. Conclusion : The results suggest that it is necessary to choose proper atopic dermatitis induced materials and suitable cultured cells in establishment of in vitro model of atopic dermatitis.

  • Collection of data for this study was conducted by 271 questionnaire responded by the consumers of purchasing anti-allergy bedding products, and analysed statistically using SPSS. Research results through the questionnaire, it was determined to be the cause of allergic disease consumers most house dust mites. More than 40, laundry convenience is more important. Sewing and laundry management respondents of high school or less. Properties for the anti-allergic bedding satisfaction in women than in men, laundry, storage convenience, to prevent dust skimp on the effects of satisfaction was convenient. 30-40s for light weight than satisfaction, high durability and anti-allergic effect on satisfaction was higher than 20 respondents more than 50 respondents. Than 20 respondents more than 40 respondents also appeared to be more about the durability that meets. Anti-allergic bedding was soft to the touch, warmth, dust Blow out prevention effects, anti-allergic performance satisfaction of the properties has a positive effect on repurchase intent. It were also identified as important anti-allergic bedding inclined to the performance of a soft feel and anti-allergic recommended.

Toxicity of Pesticides to Minute Pirate Bug, Orius strigicollis Poppius (Hemiptera: Anthocoridae), a Predator of Thrips (총채벌레의 천적인 으뜸애꽃노린재에 대한 농약 독성)

  • Toxicities of 51 pesticides (25 insecticides, 11, acaricides, 11 fungicides and 4 adjuvants) commonly used to control greenhouse insect, mite, and disease pests were evaluated to minute pirate bug, Orius strigicollis nymphs and adults at the recommended concentration. Among 25 insecticides tested, fipronil, lufenuron, acetamiprid+fipronil, $\alpha$-cypermethrin+flufenoxuron and buprofezin + amitraz showed low toxicity to O. strigicollis. Among acaricides, acequinocyl, bifenazate, chlorfenapyr, etoxazole, fenpyroximate, flufenoxuron, milbemectin, spirodiclofen and tebufenpyrad showed low toxicity to O. strigicollis. All fungicides and adjuvants tested were very low toxicity. It may be suggested from these results that five insecticides, nine acaricides, eleven fungicides and four adjuvants could be incorporated into the integrated thrips management system with O. strigicollis in greenhouses.

Observation on the Fauna of Arthropods form Apple Orchards in Winter in Kyongbuk Province (경북지방 사과원의 동계 절지동물(Arthropoda)상)

  • Arthropods of 3 Classes, 19 Orders and 58 Families were collected by the enticing band from the trunk of apple trees in winter in Kyongbuk province. The two spotted spider mite, Tetranychu.v urricae Koch, was dominant species with 53.0%, followed by Eriosomtr lanigerutn Hausmann. 16.7%. Oribatida 13.3% and Collembola 9.3%. Of those, herbivores were majority, followed by some decomposers and few natural enemies. Arthropods of 2 Classes, 12 Orders and 17 Families were observed from the fallen leaves. T. urric,ae 63.1% was dominant, followed by Collembola 13.797~ and other Arthropods. While 2 Classes, 9 Orders and 18 Families were investigated from the soil of apple orchards. They were collembola 37.9%. Oribatida 34.476, T. urticae Ih.l% and others in few numbers. Decomposers were majority, followed by herbivores and few natural enemies.

  • In this report, an experiment has been conducted to test the quantitative changes of nucleic acids in the nuclei of the epidermal cell of the galls, caused by Eriophyes kuko Kishida on the leaf of Lycium chinense Mill by means o( microspectrophotometric techniques, the two-wave-length methods. And the sizes of the epidermal cells and nuclei have been measured. The experimental results were summarized as follows: 1) It has been found that as the gall grows, the diameter of the epidermal cells and their nuclei increased and they were larger than those of tile healthy ones. 2) Microspectrophotometric measurement of nuclei by the 'Two-wave-length method' after staining with Feulgen reagent showed no changes in DNA content in the early stage of the gall. As the gall matured, however, DNA content of the gall increased more than that of the healthy leaf. 3) RNA-measurement of nuclei stained with Azur-B in DNase treated epidermal cells of the gall revealed that temporary increase in RNA content occurred in early to middle stages after the gall formation. As the gall matured, however, RNA content of the gall decreased more as against that of the healthy leaf.

  • We investigated the inhibitory effect of an oral administration of a Sophora flavescens Aiton ethanol extract (SFE) on the development of atopic dermatitis (AD) by using NC/Nga model mice. The induction of atopic dermatitis-like lesion was conducted by the removal of the back hairs and topical application of a mite antigen (Dermatophagoides farinae, Df) on to the back skin twice a week for 8 weeks. SFE was orally administered at a different doses (100-400 mg/kg). Atopic dermatitis-like skin lesions were evaluated by dermatitis scores, skin histology and immunological parameters (serum levels of IgE, TARC/CCL17, MDC/CCL22, and CTACK/CCL27). Oral administration of SFE significantly inhibited the clinical sign of Df-induced atopic dermatitis, including dermatitis score and leukocyte infiltration. Moreover, SFE suppressed significantly the serum IgE and Th2 chemokine (TARC/CCL17, MDC/CCL22, and CTACK/CCL27) levels in a concentration dependent manner. These results suggest that oral administration of SFE could reduce significantly the clinical signs and Th2 chemokines in Df-induced atopic dermatitis model mice. Therefore, SFE may be effective substances for the management of AD in human.

  • This study investigated the soil oribatid mite community at planatations of the Korean white pine, planted in different years. The soil samples $(10\times10\times5cm)$ were taken monthly from Jun 1988 to July 1989, and soil microarthropods in th sample were extracted using the Tullgren funnel for 72 hors. The diversity indices decreased at the older plantation sites with the maximum point in January and the minimum in September. The richness indices showed the maximum point in June and the minimum in may. The dominance indices increased at the older plantation and showed the maximum point in April, May, the minimum in January. The indices of aggregation of Oribatid mites species showed concentrated distribution in the average 1.9. The older plantation, the more indices increase. The seasonal variation of th indices tended to be greatest in April, August and at lowest in January. The richness indices was positively correlated with shannon-wiener, evenness indices. The dominance, aggregation indices was negatively correlated with shannon, evenness, richness indices.

Insecticidal and Acaricidal Activities of African Plant Extracts against the Brown Planthopper and Two-Spotted Spider Mite (아프리카산 식물체 추출물의 벼멸구 및 점박이응애에 대한 살충 및 살비활성)

  • Total 31 samples from 21 African plant species in 13 families were tested for their insecticidal and acaricidal activities against Nilaparvata lugens (Stal) and Tetranychus urticae (Koch) adults through topical application an leaf-dipping methods, respectively. The insecticidal and acaricidal activities were both plant parts and species dependent. The methanol extracts from whole plants of Casia occidental is and Cassia tora (Caesalpinaceae), an stem of Prosopis chinensis (Mimosaceae) revealed potent insecticidal activity against N. lugens. Potnet acaricidal activity against T. urticae was obtained from the methanol extracts from whole plants of Celosia trigyna (Amaranthaceae) and Combretum micronthum (Combretaceeae), leaves of Combretm glutinotum, and leaves and fruits of prosopis chinensis.

Monitoring of Acaricide Resistance in Field-Collected Populations of Tetranychus urticae (Acari: Tetranychidae) in Korea (점박이응애 야외개체군의 살비제 저항성 모니터링)

  • Eight field-collected populations of the two-spotted spider mites, Tetranychus urticae (Koch) from apple orchards of different geographical areas were tested for resistance to seven acaricides by leaf disk method in comparison with a susceptible strain. Marked regional variations of susceptibility were observed. Only low to moderate resistance to azocyclotin, fenpropathrin, propargite, and abamectin was obtained. However, high resistance to dicofol, fenpyroxymate, and pyridaen by eight field-collected populations was produced. Resistance to dicofol and fenpyroxymate was widespread. All of the strains tested were susceptible to one or more o the acaricides used. These results indicate that careful selection of the chemical used against any population of the two-spotted spider mite might result in satisfactory control.

The Leaves of Broussonetia kazinoki Siebold Inhibit Atopic Dermatitis-Like Response on Mite Allergen-Treated Nc/Nga Mice

  • Broussonetia kazinoki Siebold. (B. kazinoki) has long been used in the manufacture of paper in Asian countries. Although B. kazinoki leaves (BK) have been employed in dermatological therapy, use of BK has not been tested in patients with atopic dermatitis (AD). Using Nc/Nga mice, which are genetically predisposed to develop AD-like skin lesions, we confirmed the efficacy of BK in AD treatment. BK extract was applied topically to Dermatophagoides farinae-induced AD-like lesions in Nc/Nga mice, and the effects were assessed both clinically and by measuring skin thickness on the back and ears. We measured the effects of BK extract on plasma levels of IgE and IL-4. We also measured the ability of BK extract to inhibit the secretion of hTARC in HaCaT cells after stimulation by TNF-${\alpha}$ and IFN-${\gamma}$. We found that BK extract significantly reduced ear and dorsal skin thickness and the clinical signs of AD, as well as significantly down-regulating the plasma levels of IgE and IL-4 (p<0.01 for each comparison). Moreover, $500{\mu}g/mL$ of BK extract inhibited hTARC secretion in HaCaT cells by activated TNF-${\alpha}$/IFN-${\gamma}$ by about 87%. These findings suggest that topical application of BK extract has excellent potential in the treatment of AD.
