Effect of 'DaoCao' on the inflammatory cytokines in Human Jurkat cell and THP-1 cell (나초(糯草) 에탄올추출물이 Human 유래 Jurkat 세포와 THP-1 세포의 알러지 및 염증 사이토카인에 미치는 영향)

  • Objectives : The aim of this study is to investigate anti-inflammatory activity using various extracts of rice straw (DaoCao) extract (RS).Methods : To investigate the anti-inflammatory effect of RS, we examined the effect of RS on cytokines production on THP-1 cell. Cells were cultured in incubator (37℃, CO25%, 0.5% FBS-RPMI, 1X106cells/ml). One hour after,Dermatophagoides pteronissinus(Dp., 10 ug/ml) was treated into cell and at 6 hour after, each different concentrations(0.1, 1 and 10 ug/ml) of RS were treated. The cells were incubated for 16 hours and harvest the supernatant. The levels of IL-4, IL-5, IL-6, IL-8, MCP-1 and TNF-αwere determined using a commercially available ELISA kit.Results : We investigated whether RS has the inhibition of inflammatory response in Jurkat cells and THP-1 cells. RS suppressed secretion of IL-4, IL-5, and TNF-αinduced by house dust mites in Jurkat cells. It showed significant effects for all concentrations. RS suppressed the increased expression of IL-6, IL-8 and MCP-1 after treatment with mite in THP-1 cells. These results suggest that RS may be used as a valuable agent for treating allergic diseases such as atopy due to its anti-inflammatory property.Conclusions : RS showed significant biological activities with anti-inflammatory in the human T cells. These results suggest that RS may be used as a valuable agent for treating allergic diseases such as atopy due to its anti-inflammatory property. In terms of Korean traditional medicine, we expect the results to contribute to building of EBM (Evidence-Based Medicine).

Groundwater Flow Analysis using Numerical model in Small Basin (소규모유역의 수치모헝을 이응한 지하수 유동해석)

  • The applied model for this study area is WINFLOW using mite element method, It is thought that the simulation result by WINFLOW model under the steady flow state reflects well the ground water distribution within the reliability level which shows the error range of 1.1% to 8.0% from the comparison between the computed values and the observed, and analyzed that the constant head distribution is shown along the east-west direction and gentle and stable head gradient along the north-south direction. Ground water of the study area shows stable movement from the south to the stream area, and the particle trace for each location shows relatively linear shape from the upstream to the pumping location while the radius of influence according to the pumping amount shows a significant difference at the down stream area from the pumping location. The simultaneous pumping from P and P1 shows more complicated appearance, not the increase of the radius of influence than pumping from a single well P or P1, and it is analyzed that the particle path takes nearly linear form. It is known that the flow direction of the ground water and the velocity of the flow affect on the magnitude of the radius of influence of the wells from the fact that the more decreasing pattern of the ground water head is observed at the side of the well and the down stream area than the upstream area when the ground water moves from south to north regarding the radius of influence according to the pumping amount. Satisfactory results in analyses of ground water movement are obtained through the significant reduction of the physical uncertainties in the flow system as well as the relatively convenient model application using WINFLOW model which is proposed in this study.

  • Tsutstugamushi disease is a major febrile disease that generally occurs in the fall in Korea with hemorrhagic fever with renal syndrome and leptospirosis. This disease is often accompanied by interstitial pneumonia, acute renal failure and liver failure. The causative agent, namely Orientia tsutsugamushi, is transmitted to humans through the bite of a laval trombiculid mite, which is commonly known as a chigger. A 78 year old man was admitted in October 2004 with intractable fever and a drowsy mentality. Two weeks earlier, he visited a private clinic complaining of a simple skin rash. He was treated with antihistamine and steroid, but his symptoms were aggravated and he was referred to our hospital. His physical examination and laboratory findings showed a septic shock status. The maculopapular rash had spread over his face, chest, abdomen and extremities. Eschar was observed in lower back area but it was too difficult to distinguish it from other skin rashes. His chest X-ray appeared as diffuse nodular patchy consolidations in the bilateral lung parenchyme. He was treated with a mechanical ventilator and doxycycline under th suspicion of Tsutstugamushi disease. However, he suffered multiorgan failure accompanied by acute respiratory distress syndrome, acute renal failure and acute hepatitis. He was treated in the intensive care unit for approximately 12 weeks and his general condition was recovered.

Distribution and Population Density of Rodents and Chigger Mites in Gokseong-gun of Jeollanam-do, Korea (전남 곡성군에서 포획한 들쥐 분포 및 털진드기의 개체군 밀도)

  • The distribution and population density of rodents and chigger mites were investigated between April 2013 and November 2013 in the Gokseong-gun of Jeollanam-do, Korea. A total of 79 rodents were collected using Sherman collapsible traps, and among them, 73 were Apodemus agrarius (92.4%), 4 were Crocidura lasiura (5.0%), and 2 were Microtus fortis (2.5%). Sixty-eight out of the 79 rodents were parasitized by chiggers, showing an infestation rate of 86.0% and a chigger index of 74.5; the chigger index of A. agrarius was 75.4. From the trapped field rodents, 5,063 chigger mites were collected and identified with 6 species of 2 genera. Leptotrombidium scutellare was the dominant species with 3,535 chiggers (69.8%), followed by L. pallidum with 777 chiggers (15.3%). This results showed that the distribution of chigger mites differ in dominant species according to seasons. L. pallidum was the predominant chigger collected in April (34.3%), May (70.0%), and June (55.1%); whereas L. scutellare was the predominant chigger collected in September (75.5%), October (71.8%), and November (74.2%).

Increased Allergen-specific IgE Values and Lymphocyte Proliferation Response to House Dust Mites in Dogs with Atopic Dermatitis (개의 아토피성 피부염에 대한 집먼지 진드기 항원 특이적 IgE와 림프구 증식 반응의 증가)

  • We examined the responses of PBMCs to house dust mite (HDM) allergen in atopic and healthy, non-atopic dogs to identify differences in lymphocyte reactivity that might reflect the immunologic status of atopic dermatitis. Thirteen of 20 (65%) atopic dogs showed a positive lymphocyte proliferative response to HDM allergen. The rate of response was significantly higher in the atopic dogs than that in healthy, non-atopic dogs insensitive to the allergen (P = 0.007). The proliferative responses were positively correlated with the level of HDM-specific IgE in serum (P = 0.035), and were thereby confirmed to reflect the activity of lymphocytes competent to promote IgE production. These results suggest that HDM-specific lymphocytes were present in peripheral blood and played a role in the pathogenesis of canine atopic dermatitis.

Anti-asthmatic Effect of Alismatis Rhizoma and Alisol Acetate B Combination Therapy in a Murine Asthma Model (택사와 alisol B acetate의 병용 투여가 천식 동물 모델에 미치는 영향)

  • Objectives: The aim of the study was to evaluate the anti-asthmatic effect of alismatis rhizoma and alisol acetate B combination therapy in a murine asthma model. Methods: C57BL/6 mice were sensitized to and challenged with a mixture of ragweed, dust mite, and aspergillus to induce an asthma animal model. Alismatis rhizoma extract and alisol acetate B combination therapy was co-administered only in the experimental group. To evaluate the anti-asthmatic effect of the combination therapy, inflammatory cell counts in bronchoalveolar lavage (BAL) fluid were determined, and tissue was examined histologically with hematoxylin and eosin (H & E) and periodic acid-Schiff (PAS) stains, by enzyme-linked immunosorbent assay (ELISA) of IgE, IL-4, and IL-5, and with reverse transcription polymerase chain reaction (RT-PCR) of IL-5, IL-33, MUC5AC. Results: Alismatis rhizoma and alisol acetate B combination therapy reduced the number of inflammatory cells, alleviated histologic features, and down-regulated all the investigated asthma mediators, IgE, IL-4, IL-5, IL-33, and MUC5AC. Conclusions: According to the above results, alismatis rhizoma and alisol acetate B combination therapy may have therapeutic potential for asthma.

Effects of Sun Ginseng on the Crypt Cell Survival in Mice Irradiated with High-energy X-ray (선삼 투여가 고에너지 X-선을 조사한 마우스에서 소장움 재생에 미치는 효과)

  • Six week-old ICR mite which were divided into four groups including NC, RC, RR and RS were injected with sun ginseng (RS), red ginseng (RR) and saline (RC) intraperitonedlly as an amount of 60 mg/g body weight at 1 hour, 12 hours and 36 hours before the irradiation of high-energy X-ray and the mire were sacrificed at three and a half days after the irradiation. The RS group were significant increase in the weight of spleen (p<0.01) and the numbers of jejunal crypt cells (P<0.01), WBC (p<0.05), lymphocytes (p<0.05) and neutrophils (p<0.05) in comparison with the RC group. The RR group were significant increase in the numbers of jejunal crypt cells (p<0.001), WBC (p<0.05) and neutrophils (p<0.05) in comparison with the RC group. The RS group exhibited a more increase in the weights of spleen and thymus and the numbers of jejunal crypt cells and all items of hematological examination than the RR group. The values of ALT (alanine transaminase) and AST (aspartate transaminase) were significantly elevated (p<0.05) by radiation and they were significantly decreased (p<0.05) in the RS group to the values of the NC group. Taken together the above results, sun ginseng demonstrated a jejunal crypt survival effect, the protective effects on hepatocytes and immune and hematopoietic cells in mice irradiated with high-energy X-ray, and those radiation protective effects were a little higher in comparison with red ginseng.

  • Extracts of 43 species of plants were tested for their insecticidal and acaricidal activities against six species of insect pests and one mite species. The methanol extract of Ginkgo biloba leaves selectively was found to have potent insecticidal activity against Nilaparvata lugens (Stal), whereas steam distillate of Thujopsis dolabarata var. hondai sawdust showed potent insecticidal activities with a broad spectrum. The methanol extract of Pinus densiflora leaves and steam distillate of T. dolabrata var. hondai sawdust exhibited potent activities against Tetranychus urticae (Koch).

The Effect of a Cooperative System of Oriental and Western Medicine in the Treatment of Allergic Rhinitis

  • Background and Purpose : Allergic rhinitis is a well-known, relatively controllable chronic disease. Although a number of methods for treating allergic rhinitis have been tried, many patients have not been satisfied with their treatment. Therefore, this study tried to evaluate the effect of a cooperative system of Oriental and Western medicine and to develop a new diagnosis protocol for treatment of allergic rhinitis. Methods : We measured improvement rate and acoustic rhinometry after the allergeninduction test and performed a filter paper test as a nonspecific hypersensitivity test with 60 patients who are allergic to house dust mite. Patients were divided into two groups, one treated with Western medicine only and one treated with both Western and Oriental herbal medicine. For the group with Western medicine only, antihistamine for one week and local steroid medicine for two weeks were prescribed. For the group with combined medicine, Oriental herbal medicine was prescribed according to the patient s constitution, along with Western medicine. After all treatments, the above tests were re-performed and the improvement rate was compared. Results and Conclusion : We observed better results in the group treated with both Western and Oriental herbal medicines, comparing improvement rate and the alteration of total nasal volume through acoustic rhinometry after the allergen induction test. In the filter paper test, there was no significant difference between the two groups. In conclusion, we showed the additive effect of Oriental herbal medicine without any severe side effects compared with treatment with Western medicine only. In this study, we set only two patient groups, but further study is required to create various experimental groups and compare among them. We suggest that it might enhance understanding of the improved effect of Oriental herbal medicine in the therapy of allergic rhinitis.

A mite Element Modeling for the Puncture Drop Test of a Cask with the Failure of Impact Limiter (충격완충체의 효과를 고려할 수 있는 운반용기의 파열낙하시험 유한요소해석 방법)

  • Transport package for radioactive material should be structurally safe under puncture drop condition and its safety should be verified by test and numerical analysis. Most finite element analyses for puncture drop have been performed without modeling the impact limiter since failure is occurred in the materials of the impact limiter. This paper presents a new modeling methodology, where an element is eroded in case that the material's failure criteria are reached at the element's integration point, to investigate the effect of the impact limiter in the puncture process. The effectiveness of the proposed scheme is shown through the puncture drop analysis of hotcell transport cask, which is under design in KAERI. The results show that about 80 percent of the total impact energy is absorbed due to the deformation of impact limiter. Using the present method the puncture drop can be analyzed more accurately, but it would give conservative results compared to the actual test condition.
