미니돼지 및 일반 돼지의 임신기, 분만 및 포유기 중의 모돈의 행동학적 특성 비교 (The Comparison of the Behavior between Miniature Pigs and Conventional Sows during Gestation to Lactation Period)

  • This study was conducted to compare the aspects of behavior between miniature pigs and conventional sows during gestation and lactation periods. Miniature pigs and conventional sows at 3 parity were used in this experiment with 4 pigs per treatment. The behavioral patterns and stereotyped behaviors were observed on day 60 and 90 of gestation, at farrowing, and on day 10 of lactation before and after the feeding. On 60 and 90 days of pregnancy, both treatments showed the tendency of frequent lateral lying rather than other postures. On ventral lying, eating and drinking, miniature pigs spent more time than conventional pigs at day 90 of gestation (P<0.01, P=0.0539, P<0.05, respectively). The occurrence of stereotyped behaviors included bar biting and bar mouth chewing. At 90 day of pregnancy was observed, conventional pigs was higher than miniature pigs (P<0.05). At farrowing, miniature pigs spent more time on ventral lying, standing and walking than conventional sows (P<0.05), however, there were more frequency of drinking and eating. And on day 10 of lactation, there was no significant difference in stereotyped behaviors between miniature pigs and conventional sows. In conclusion, miniature pigs were generally more sensitive than conventional sows although sows were reared in stall during gestation and lactation period and the behavioral pattern during lactating period showed the similarity between miniature pigs and conventional sows for protection of nursing pigs. In consideration of the general reproductive performance of conventional sows, the high sensitivity and stress of miniature pigs did not affect the reproductive performance of miniature pigs.

Intestine Ischemia/reperfusion Induces ER Stress and Apoptosis in Miniature Pigs

  • The miniature pig is a very suitable donor species in xenotransplantation of human organs. Intestine ischemia/reperfusion (I/R) is associated with high morbidity and mortality. Endoplasmic reticulum (ER) stress and apoptosis has been associated with the onset of diverse diseases. Thus, we examined the effect of intestine I/R on the expression of ER stress and apotptosis related molecules. In the present study, I/R induced phosphorylation of protein kinase-like endoplasmic reticulum kinase (PERK), IRE, and ATF-4. I/R also increased the expression of the proapoptotic transcription factor CAAT/enhancer-binding protein homologous protein (CHOP). In addition, I/R decreased the expression of Bcl-2, but increased the expression of Bax, cleaved PARP, and cleaved caspase-3. Moreover, I/R increased splicing form of XBP-1 mRNA and the expression of caspase-6 and caspase-3 mRNA. In conclusion, intestine I/R induced ER stress and apoptosis in miniature pig.

Endocrine Profiles and Blood Chemistry Patterns of Cloned Miniature Pigs in the Post-Puberty Period

  • Although the majority of surviving pigs cloned by somatic cell nuclear transfer (SCNT) appear to be physiologically normal, there is a general lack of detailed hemato-physiologic studies for the period of early adulthood to substantiate this claim. In the present study, we investigated variation in blood chemistry and endocrinological parameters between mesenchymal stem cells (MSCs) derived from cloned and normal age-matched female and male miniature pigs. Cloned females and males showed normal ranges for complete blood count assessments. Biochemical assessments showed that ${\gamma}$-GGT, ALT and cholesterol levels of male and female clones were significantly (P<0.05 or P<0.01, respectively) higher than that of age-matched control miniature pigs. Variations in insulin and IGF-1 were higher in female clones than in male clones and controls. Thus, although female and male cloned miniature pigs may be physiologically similar to normal animals, or at least within normal ranges, a greater degree of physiological and endocrinological variation was found in cloned pigs. The above variation must be taken into account before considering cloned female or male miniature pigs for various biomedical applications.

  • We generated a transgenic male cloned pig which was derived from fibroblast of white Yucatan miniature pig. After 2 weeks of birth, umbilical hernia which was not easily reduced was identified. Considering the usefulness of cloned pig, surgical treatment for umbilical hernia correction was performed and a cloned pig has been maintained healthy. This is the first report and can be useful for the treatments of umbilical hernia of cloned piglets.

Characterization of Insertional Variation of Porcine Endogenous Retroviruses in Six Different Pig Breeds

  • Pigs may need to be exploited as xenotransplantation donors due to the shortage of human organs, tissues and cells. Porcine endogenous retroviruses (PERVs) are a significant obstacle to xenotransplantation because they can infect human cells in vitro and have the potential for transmission of unexpected pathogens to humans. In this research, 101 pigs, including four commercial breeds (23 Berkshire, 13 Duroc, 22 Landrace and 14 Yorkshire pigs), one native breed (19 Korean native pigs) and one miniature breed (10 NIH miniature pigs) were used to investigate insertional variations for 11 PERV loci (three PERV-A, six PERV-B and two PERV-C). Over 60% of the pigs harbored one PERV-A (907F8) integration and five PERV-B (B3-3G, B3-7G, 742H1, 1155D9 and 465D1) integrations. However, two PERV-A loci (A1-6C and 1347C1) and one PERV-B locus (B3-7F) were absent in Duroc pigs. Moreover, two PERV-C loci (C2-6C and C4-2G) only existed in Korean native pigs and NIH miniature pigs. The results suggest that PERV insertional variations differ among pig breeds as well as among individuals within a breed. Also, the results presented here can be used for the selection of animals that do not have specific PERV integration for xenotransplantation research.

Subpopulations of miniature pig mesenchymal stromal cells with different differentiation potentials differ in the expression of octamer-binding transcription factor 4 and sex determining region Y-box 2

  • Objective: Human mesenchymal stromal cells (MSCs) exhibit variable differentiation potential and can be divided accordingly into distinct subpopulations whose ratios vary with donor age. However, it is unknown whether the same is true in pigs. This study investigated MSC subpopulations in miniature pig and compared their characteristics in young (2 to 3 months) and adult (27 to 35 months) pigs. Methods: Osteogenic, chondrogenic, and adipogenic capacity of isolated MSCs was evaluated by von Kossa, Alcian blue, and oil red O staining, respectively. Cell surface antigen expression was determined by flow cytometry. Proliferative capacity was assessed with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. Expression of marker genes was detected by quantitative real-time polymerase chain reaction. Results: Porcine MSCs comprised cells with trilineage and bilineage differentiation potential (tMSCs and bMSCs, respectively) and non-differentiating stromal cells (NDSCs). The tMSC and bMSC fractions were smaller in adult than in young pigs (63.0% vs 71.2% and 11.6% vs 24.0%, respectively, p<0.05); NDSCs showed the opposite trend (25.4% vs 4.8%; p<0.05). Subpopulations showed no differences in morphology, cell surface antigen expression, or proliferative capacity, but octamer-binding transcription factor 4 (OCT4) expression was higher in tMSCs than in bMSCs and NDSCs (p<0.05), whereas sex determining region Y-box 2 (SOX2) expression was higher in tMSCs and bMSCs than in NDSCs (p<0.05). Aging had no effect on these trends. Conclusion: Porcine MSCs comprise distinct subpopulations that differ in their differentiation potential and OCT4 and SOX2 expression. Aging does not affect the characteristics of each subpopulation but alters their ratios.

Effect of Osmolarity of Culture Medium on Imprinting and Apoptotic Gene Expression in Miniature Pig Nuclear Transfer Embryos

  • This study was conducted to investigate the development and gene expression in miniature pig nuclear transfer (mNT) embryos produced under different osmolarity culture conditions. Control group of mNT embryos was cultured in PZM-3 for 6 days. Treatment group of mNT embryos was cultured in modified PZM-3 with NaCl (mPZM-3, 320 mOsmol) for 2 days, and then cultured in PZM-3 (270 mOsmol) for 4 days. Blastocyst formation rate of the treatment group was significantly higher than the control and the apoptosis rate was significantly lower in treatment group. Bax-$\alpha$ and caspase-3 mRNA expression were significantly higher in the control than the treatment group. Also, the majority of imprinting genes were expressed aberrantly in in vitro produced mNT blastocysts compared to in vivo derived blastocyst H19 and Xist mRNA expression were significantly lower in the control than the treatment group or in vivo. IGF2 mRNA expression was significantly higher in the control than the treatment group or in vivo. IGF2r mRNA expression was significantly lower in the control. Methylation profiles of individual DNA strands in H19 upstream T-DMR sequences showed a similar methylation status between treatment group and in vivo. These results indicate that the modification of osmolarity in culture medium at early culture stage could provide more beneficial culture environments for mNT embryos.

미니돼지에서 정상 폐 환기/관류 신티그라피 (Normal Lung Ventilation/Perfusion Scintigraphy in Miniature Pigs)

  • 미니돼지에서 폐 환기/관류 신티그라피를 실시한 결과 환기스캔에서는 기능적 분포는 왼쪽 폐가 44.2%, 오른쪽 폐에서는 56.2%로 나타났으며, 관류스캔에서는 왼쪽 폐가 46.87%, 오른쪽 폐가 54.97% 임을 확인할 수 있었고 사람의 폐용적과 유사한 결과를 보임을 확인할 수 있었다. 또한 다른 방사성가스보다 짧은 시간을 필요로 하며 기계적 환기로도 스캔이 가능한 Technegas로 환기 스캔을 실시하여 미니돼지의 마취시간을 줄 일 수 있었으며 미니돼지에서 더 용이하게 폐기능을 측정할 수 있었다. 따라서 본 연구를 통해 폐 환기/관류 신티그라피가 미니돼지에서 정상 폐기능 및 폐질환과 관련된 실험을 할 때 폐기능을 측정하는 좋은 진단법이 될 수 있으며 미니돼지의 정상 폐기능에 관한 연구가 앞으로 폐질환과 관련된 미니돼지의 실험에 대한 정보를 제공할 수 있으리라 생각된다.
