국내에서 개발된 제초제저항성 벼(밀양 204호, 익산 483호)의 비표적 곤충과 거미에 미치는 생리적 영향 (Physiological Effects of Herbicide-resistant Genetically Modified Rice (Milyang 204 and Iksan 483) Developed in Korea on Non-target Insects and a Spider)

  • 본 연구에서 국내에서 개발된 제초제저항성 GM 벼인 밀양 204호와 익산 483호가 비표적 곤충과 거미에 미치는 영향을 평가하였다. GM 벼와 non-GM 벼에 대한 보리수염진딧물과 벼멸구의 기주선호도에는 차이가 없었으며, GM 벼를 섭식한 벼멸구를 포식한 황산적거미의 체중 변화에도 통계적 유의성이 없었다. 밀양 204호와 그 대조 모본 non-GM 벼에서 서식한 벼멸구를 포식한 황산적거미의 생존율에는 차이가 없었으나, 벼멸구를 통해 익산 483호에 노출된 황산적거미의 생존율은 대조 모본 non-GM 벼에 비해 통계적으로 낮은 생존율을 나타내었다. 또한 익산 483호의 화분을 섭식한 꿀벌 유충은 밀양 204호와 다른 non-GM 벼 화분을 제공하였을 때 보다 현저하게 긴 용기간을 보였다. 농업생태계에서 중요한 포식 천적인 황산적거미와 양봉 산업으로서 중요한 기능을 하는 꿀벌에 GM 벼가 부정적인 영향을 보임에 따라 국내에서 개발된 제초제저항성 GM 벼의 농업 환경 노출 이전에 충분한 추가 연구와 안전성 평가가 선행되어야 할 것으로 사료된다.

  • 국내에서 개발된 GM 쌀인 밀양 204호, 익산 483호의 정성 PCR 분석법의 개발을 위해 쌀의 내재 유전자로써 SAMDC1(S-adenosylmethionine decarboxylase)에 특이적인 primer (OsSAMDC1-5'/3')쌍을 제작하여 쌀을 포함한 20개 작물에 대해 PCR을 수행하여 쌀에 특이적으로 증폭되는 것을 확인하였다. 또한, 밀양 204호에 삽입된 GUS 유전자와 NOS terminator 연결 부위를 증폭시켜 172 bp의 PCR 산물을 얻을 수 있는 primer(Os204-5'/OsNOS-3')와 익산 483호에 삽입된 bar 유전자와 NOS terminator 연결 부위를 증폭시켜 161 bp의 PCR 산물을 얻을 수 있는 primer(Os483-5'/OsNOS-3')을 이용하여 국내 개발된 GM 쌀인 밀양 204호, 익산 483호의 PCR 정성 분석법을 확립하였다.

Evaluation of Crossability, Seed Dormancy and Overwintering Ability in Glufosinate Ammonium-Resistant GM Rice and Their Hybrids with Non-GM and Weedy Rice

  • This study was conducted to investigate the crossability, seed dormancy and overwintering ability of rice plant in GM (glufosinate ammonium-resistant lines. Iksan 483 and Milyang 204) and non-GM (their parents) or red rice (Andongaengmi). Seed-setting rate was not significantly different between GM and non-GM rice varieties. Iksan 483 and Milyang 204 showed the similar level of seed germination rate from 30 to 50 days after heading as compared to non-GM rice varieties. After overwintering in paddy field, seed germination rate of GM and non-GM rice varieties ranged from 14.3 % to 57.6 % in dry soil condition, but there was no germination in wet-soil except red rice. The result in wet-soil condition may help to set up a strategy for reducing the risk of gene flow of transgene via dispersal of seeds of GM plants. The crossability, seed dormancy and seed overwintering of Iksan 483 and Milyang 204, herbicide resistant GM rice varieties, were not significantly different compared to non-GM rice varieties. The results might be helpful to reduce the risk of transgene dispersal from GM crop via seeds and pollens.

  • 밀양 204호는 작물과학원 영남농업연구소 생명공학연구팀이 직파적응 제초제저항성 벼 품종을 육성할 목적으로 '98년 bar 유전자를 동진벼에 아그로박테리움법으로 형질 전환하여 제초제저항성 벼를 양성하였다. '98/'99년 동계에 단간 내도복 양질인 주남벼를 인공교배하여 약배양 등 육종프로그램에 적용하여 우량계통을 선발하고 밀양 204호로 계통명을 부여하였다. GM 벼와 Non-GM 벼의 농업적 특성을 UPOV 및 국립종자관리소의 품종등록 기준에 따른 농업적특성 중 차이가 있었던 것은 엽색도, 지엽의 형태, 간장, 수당립수 등 이었다.

Pollen-Mediated Gene Flow between Glufosinate Ammonium-Tolerant GM and Non-GM Rice

  • To assess the risk of genetically modified (GM) rice on the agricultural ecosystem, agronomic characteristics, pollen longevity and outcrossing rate between GM (Iksan 483 and Milyang 204) and non-GM (their wild types and female parents) varieties were investigated using the bar gene as a tracer marker in paddy field. The agronomic characteristics of two GM rice were similar to their female-parents (non-GM rice) except heading date and 1,000 grain weight of Iksan 483, and they did not show a difference by the introgression of the bar gene as the genetic traits of rice varieties. Pollen viability was more than 90% just after shedding, and it was rapidly decreased below 50% at 5 minutes after shedding both GM and non-GM varieties. The Pollen longevity was lost after 30 minutes of anthesis. When the distance of gene flow from GM to non-GM rice detected to 6 m from the edge of GM rice plant, the maximum distance of pollen dispersal was 4.5m and 3.9m in Iksan 483 and Milyang 204, respectively, and that was increased in order of west, south, east, and north to the dominant wind direction, west-south. Mean outcrossing rate was very low as 0.003 and 0.001% within 1.5 m from the edge of Iksan 483 and Milyang 204, and the GM hybrids by the pollen dispersal did not detected over 4.5 m from the edge of GM rice plant. The results may help to establish the strategy which reduce the risk of pollen-mediated gene flow between GM and non-GM rice.

형질전환 벼 이용 품종 조기개발 체계 확립 (Establishment of a New Breeding Scheme for Rapid Release of Variety Using bar Gene Transformed Rice)

  • 형질전환 식물체를 직접 이용하기에는 후대에 유전적인 고정 및 포장검정이라는 문제가 대두되는데, 이를 해결하기 위하여 본 연구는 바스타저항성 유전자가 도입된 벼와 직파적 응성 계통을 교잡한 후 약배양에 의한 형질전환체 고정 등 기존 육종체계와 연계하여 제초제 저항성 직파적응 계통을 조기에 육성할 수 있는 체계를 확립하고자 수행하였다. 제초제 저항성 직파적응 계통을 조기에 육성하고자 1999년 동계에 교배된 동진벼 Ds15(Ba $r^{R}$)/주남벼 등 5조합으로부터 2000년 $F_1$을 양성하고, 이들을 PCR분석으로 제초제 저항성 유전자 bar도입을 확인한 후 약배양을 실시하여 얻어진 1,508개의 재분화 식물체 중 제초제 저항성 유전자가 도입된 169개체 ( $A_2$)에서 채종하였다. 이들을 2001년 포장에 공시하여 42계통을 선발하고,2002년에 동진벼 Ds15 (Ba $r^{R}$)/주남벼 조합에서 수량 등 주요 농업형질이 우수한 7계통을 생산력 검정시험에 공시한 결과 제초제 저항성이면서 직파적응성 등 기타 농업형질이 우수한 YR23235Acp79를 선발하여 밀양 204호로 계통명을 부여하였다. 결과적으로 형질전환체를 이용하여 교배부터 계통선발까지 3년 만에 목적하는 계통을 조기에 육성할 수 있는 생력육종체계가 확립되었다.

QTL Mapping of Genes Related with Grain Chemical Properties Based on Molecular Map of Rice

  • This study was conducted to investigate the chromosomal locations and effects of quantitative trait loci (QTL) associated with chemical properties of rice (Oryza sativa L.). One hundred sixty four recombinant inbred lines (MGRILs) of $F_{11}$ were derived from the cross between Milyang 23, Tongil type, and Gihobyeo, japonica type. They were evaluated for 7 traits of chemical property in rice. Transgressive segregation was observed for all traits examined. Eight significant QTLs were detected (LOD$\geq$2.0) for five traits, including two QTLs for amylose content, two QTLs for potassium content, one QTL for ratio of magnesium to potassium, one QTL for fat content and two QTLs for ash content. Phenotypic variation explained by each QTL ranged from 7.2% to 14.4%. However, no significant QTL was detected for magnesium and protein contents. In amylose content and ash content M alleles originated from Milyang 23 were responsible for increasing these traits and J alleles originated from Gihobyeo also responsible for increasing these traits. Pleiotropic effects of single QTLs on different traits are observed.

Molecular Analysis of Bacterial Community Structures in Paddy Soils for Environmental Risk Assessment with Two Varieties of Genetically Modified Rice, Iksan 483 and Milyang 204

  • The impacts of planted transgenic rice varieties on bacterial communities in paddy soils were monitored using both cultivation and molecular methods. The rice field plot consisted of eighteen subplots planted with two genetically modified (GM) rice and four non-GM rice plants in three replicates. Analysis with denaturing gradient gel electrophoresis (DGGE) of PCR-amplified 16S rRNA genes revealed that the bacterial community structures were quite similar to each other in a given month, suggesting that there were no significant differences in bacterial communities between GM and non-GM rice soils. The bacterial community structures appeared to be generally stable with the seasons, as shown by a slight variation of microbial population levels and DGGE banding patterns over the year. Comparison analysis of 16S rDNA clone libraries constructed from soil bacterial DNA showed that there were no significant differences between GM and non-GM soil libraries but revealed seasonal differences of phyla distribution between August and December. The composition profile of phospholipid fatty acids (PLFA) between GM and non-GM soils also was not significantly different to each other. When soil DNAs were analyzed with PCR by using primers for the bar gene, which was introduced into GM rice, positive DNA bands were found in October and December soils. However, no bar gene sequence was detected in PCR analysis with DNAs extracted from both cultured and uncultured soil bacterial fractions. The result of this study suggested that, in spite of seasonal variations of bacterial communities and persistence of the bar gene, the bacterial communities of the experimental rice field were not significantly affected by cultivation of GM rice varieties.
