Involvement of the CXC Chemokines Mig and IP-10 in Response to M. bovis BCG in Mice

  • The non-ELR-containing CXC chemokines Mig and IP-10 have been shown to function as chemotactic cytokines for activated T lymphocytes. In this study, we examined the potential involvement of Mig and IP-10 in antimycobacterial response of mice immunized or infected with M. bovis BCG. The accumulation of Mig and IP-10 mRNA in resident peritoneal monocytes ($RPM{\Phi}$) was slightly reduced by stimulation with vBCG, and the degree was greater for 24 hr culture even though IFN-${\gamma}$ was added. Expression of Mig, IP-10, and IFN-${\gamma}$ in 24 hr delayed-type hypersensitivity (DTH) response was stronger in vBCG-immune mice than in the non-immune. The increase of DTH measured by foot-pad thickness appears to be clearly related to the levels of chemokines Mig and IP10 messages and those of IFN-${\gamma}$ and IL-12. Stimulation with vBCG for 2 days decreased or completely dropped the levels of Mig message in non-immune or immune splenocytes, respectively, whereas IP-10 message was slightly decreased in 2 days culture. Moreover, messages for IL-12 (p40) showed similar kinetics for Mig. The levels of Mig and IP-10 mRNA during the course of infection with BCG were not readily changed in lungs, livers, and spleens from BCG-infected mice. Although there was no obvious changes of Mig and IP-10 messages in the target organs during infection process, we found that the infection progressed over the first 3 wk before being contained by the emerging immune response suggested from detectable amount of IFN-${\gamma}$ mRNA around this time. In view of selectivity of chemokines Mig and IP-10 for activated T cells, these data suggest that chemokine Mig and IP-10, especially in collaboration with IL-12 and IFN-${\gamma}$, may playa role as T cell recruiters in immune response against mycobacterial infection.

극성가변 AC 펄스 MIG용접기를 이용한 아크 브레이징 (The Arc Brazing by Variable Polarity AC Pulse MIG Welding Machine)

  • MIG brazing is used for many parts without melting base metal because of high productivity. Pulsed MIG brazing can be used to further reduce heat input and to improve the process stability. However, a significant amount of zinc in galvanized sheet steel is burned off in the area of brazes. Therefore, the brazing method to reduce the heat input is needed. In the brazing for galvanized sheet steel, variable polarity AC pulse MIG arc brazing can be applied to more decrease the heat input by setting EN-ratio adequately. In this research, we studied for the variable polarity AC pulse MIG arc brazing to decrease the heat input by using ERCuSi-A wire. As the result of increasing EN-ratio, melting ratio of base metal and burning off of zinc were reduced in galvanized sheet steel.

Lipopolysaccharide Synergizes with Interferon-${\gamma}$ to Induce Expression of Mig mRNA in Mouse Peritoneal Macrophages

  • Lipopolysaccharide (LPS) is responsible for the tissue injury that occurs following the invasion of multicelluar organisms by Gram-negative microbes. The effect of LPS on IFN-$\gamma$-induced chemokine Mig gene expression in mouse peritoneal macrophages was investigated. Very little Mig mRNA was detectable upon exposure to LPS without IFN-$\gamma$. Although LPS alone is only minimally effective, LPS plus IFN-$\gamma$ synergized to produce a high level of Mig mRNA in the peritoneal macrophages. This synergy was not dependent on a new protein synthesis, and was not controlled at the level of the gene transcription. Futhermore, LPS did not increase IFN-$\gamma$-induced Mig mRNA stability. Accordingly, it is suggested the LPS may synergize the expression of IFN-$\gamma$-induced Mig mRNA through a process that depends on a pretranscriptional level or concurrent Mig mRNA translation.

Synergistic Effect of Lipopolysaccharide and Interferon-$\beta$ on the Expression of Chemokine Mig mRNA

  • Expression of monokine induced by IFN-$\gamma$(Mig) mRNA is well-known to strictly depend on Interferon-$\gamma$(IFN-$\gamma$). Lipopolysaccharide (LPS) alone Is weakly effective on Mig mRNA expression in mouse Peritoneal macrophages. This study was undertaken to investigate the synergistic effect of LPS and IFN-$\beta$ on chemokine Mig gene expression in mouse peritoneal macrophages. Although IFN-$\beta$ alone was minimally effective, LPS plus IFN-$\beta$ synergized to produce a high level of Mig mRNh. The synergistic effect of LPS and IFN-$\beta$ (LPS/IFN-$\beta$) on Mig mRNA expression was strain-specific. The most effective synergistic effect of LPS/IFN-$\beta$ on the mRNh expression was found in simultaneous stimulation of LPS/IFN-$\beta$. This synergy was modulated at the level of the gene transcription and was not dependent on a new protein synthesis. Synergistic effect of LPS/IFN-$\beta$ also required the activation of $NF-_KB$. Accordingly, these data suggest that LPS/IFN-$\beta$ synergizes the expression of Mig mRNA through a process that depends on a pretranscriptional level and/or coincident Mig mRNA transcription.

  • 최근 에너지 절감과 환경개선 문제가 대두되면서 아크용접에서도 깨끗하고 불량률이 낮은 아크용접기기의 개발을 본격적으로 추진하고 있다. 본 연구에서는 알루미늄 등과 같은 비철금속에 주로 적용되는 MIG 아크용접에서의 용접성능을 개선하고자 한다. 기존의 직류펄스 MIG 아크용접은 박판 금속에 용접을 하는 경우 아크전압이 낮아 단락이 빈번하게 발생하고 용접 불량을 일으킨다. 이를 개선하기 위하여 중첩교류 펄스를 적용한 MIG 아크용접방법을 제안하고 중첩교류펄스의 설계와 실험을 통하여 그 타당성을 검증하였다.

  • Friction stir welding results in low distortion and high joint strength compared with other welding procedures, and is able to join all aluminium alloys that are not considered as virtually weldable with classical liquid state techniques. The comparative study on high cycle fatigue properties between A16005-T6 friction stir welds and MIG weld joints have been performed and fracture mechanisms for the fatigue specimens were investigated. Although mechanical properties are lower than the corresponding base material, FSW joints of A16005-T6 become higher at tensile and fatigue strength in comparison with the traditional fusion weld(MIG). The fracture surfaces of FSW and MIG fatigue specimens cleary show different aspects of the fracture morphology. MIG weldments were characterized by voids and cleavage(brittle fracture) but FSW specimens showed the presence of ductile fracture surface.

MIG 헤드 가상 갭에 의한 재생 전압 스펙트럼 Bump의 컴퓨터 시뮬레이션 (Computer simulation of Playback spectrum bumps due to pseudo gaps of MIG head)

  • 본 연구에서는 MIG 헤드의 재생 전압 스펙트럼에 나타나는 Bump들의 발생 원인을 설명하고, 이론적으로 예상된 주파수에서 Bump들의 진폭을 컴퓨터로 시뮬레이션 하였다. MIG 헤드의 센더스트와 페라이트 사이의 가상 갭(pseudo gap)이 없는 경우와 양쪽 모두 $0.02\;\mu\textrm{m},\;0.2\;\mu\textrm{m}$, 인 가상 갭을 갖는 3가지 종류의 MIG 헤드들을 설정하고, 금속 분말(Metal Powder) 테입을 사용한 것으로 가정하였다. 시뮬레이션 결과, 이론적으로 예상한 주파수에서 Bump들이 발생하였으며, Bump 진폭 또한 예상치와 거의 일치하였다.

Plasma-MIG 하이브리드 용접에서 용적 이행모드 현상 모니터링에 대한 연구 (Study on the Welding Mode Transition Phenomena in Monitoring Plasma-MIG Hybrid Welding)

  • Recently in the welding field, the establishment of unmanned and automated systems are rapidly developing. Accurate interpretation of the welding phenomenon is applied a number of monitoring systems. In this paper, butt welding (6t) type I using Plasma-MIG welding was carried out. And we evaluated characteristics of the Al-5083 aluminium alloy in Plasma-MIG hybrid welding. Process variables including the plasma current, MIG voltage, wire feeding rate and the welding speed were used. Butt welding was conducted 1 pass. Argon gas was used as the protective gas that results from the experiment were able to achieve full penetration. In addition to monitoring the welding process occurring during MIG welding current, welding votage and Plasma current, voltage were collected in real time, the photodiode and CCD cameras observing the phenomenon that the welding is in progress were measured using a quantity of light.

Comparison of steels via SMAW and MIG welding methods under industrial loads

  • In this study, the deflection and deformation behaviours of IPN80 steel beam and column were investigated under the different industrial loads. Single-sided welds were applied to IPN80 steel beams using shielded metal arc (SMAW) and metal inert gas welding (MIG) method in the form of T-type. After that, the performance of SMAW and MIG welded joints were identified using beam bending test under 500 and 3000 N loads. SMAW and MIG methods were compared with each other to understand the deflection and deformation behaviours of the welded steel structures. Lower deformation and deflection were obtained in MIG welded steel beams. The results show that, steel beams welded MIG method has higher load capacity than SMAW welded ones. MIG welding method is more reliable than the SMAW method for the combining performance and load capacity.

철도차량 AI 하부구조의 MIG 용접 조건에 따른 기계적 특성에 관한 연구 (Mechanical Properties of Metal Inert Gas Welding Conditions of Railway-Vehicle Aluminum Under Frame)

  • In this study, the mechanical properties of railway-vehicle aluminum under frame was investigated based on the metal inert gas (MIG) welding conditions. An aluminum-alloy (6005A-T6) extruded material used in the lower panel of a railway vehicle was connected through MIG welding to determine the mechanical properties of MIG welds. Argon shielding gas and filler materials, such as ER5356 and ER4043, were used as consumable welding materials. For the welding conditions of the test specimen, welding frequencies of 2.5 and 4.5 Hz were applied using the SynchroPuls function, and the root faces were 1.0 and 1.5 mm. The mechanical properties of the MIG welds were determined through tensile, bending, and fatigue tests.
