The microsatellites are the short tandem repeats of 1 to 6 bp long monomer sequences that are repeated several times. These short tandem repeats are considered to be generated by the slipped strand mispairing. Based on the unique capability of alternating purine-pyrimidine residues to form Z-DNA, the possible role of the microsatellites in gene regulation has been proposed. The microsatellites are highly polymorphic, follow Mendelian inheritance and are evenly distributed throughout the genomes of eukaryotes. They are easy to isolate and the polymerase chain reaction based typing of the alleles can be readily automated. These properties make them the preferred markers for comparison of the genetic structure of the closely related breeds/populations; very high-resolution genetic mapping and parentage testing etc. The microsatellites have rapidly replaced the restriction fragment length polymorphism (RFLP) and the random amplified polymorphic DNA (RAPD) in most applications in the population genetics studies in most species, including the various farm animals viz. cattle, buffalo, goat, sheep and pigs etc. More and more reports are now available describing the use of microsatellites in pigs ranging from measurement of genetic variation between breeds/populations, developing high resolution genetic maps to identifying and mapping genes of biological and economic importance.
In this manuscript, a review of the diversity of Chinese indigenous goat breeds according to data from body stature and appearance, chromosome group, blood proteins, DNA molecular markers (mitochondria DNA, random amplified polymorphic DNA, microsatellite DNA, major histocompatibility complex) has been introduced. All of these provide efficient tools for the diversity analysis of Chinese indigenous goat breeds and are very important for biodiversity conservation, restoration of declining goat breeds, the priority defining in Chinese indigenous goat breeds' protection and the selection of nature preservation zones. Many Chinese indigenous goat breeds with small population size in the isolated mountains or reservoir areas are verging the potential threat of extinction, effectively lost with the rapid destroying of ecological environment. On the other hand, as a result of the introduction of modern commercial goat breeds and shortage of effective conservation, some populations, such as Small-xiang goat and Tibetan goat decrease rapidly in number of sires. In the interests of the long-term future of the goat breeds in China, conservation of goat breeds' genetic resources should be considered urgently and some conservation measures should be adopted. In addition, the continuing development of molecular biology will further enhance conservation of diversity of Chinese indigenous goat breeds.
Stock enhancement is considered to be a valuable approach for restoring fishery resources. Because no specific official institution in Taiwan is responsible for the production of fry, the released fry are purchased directly from the private sector. However, fishermen from the private industry have not established a genetic background, so the genetic composition for each batch of released fry is unclear. Mangrove red snapper (Lutjanus argentimaculatus), a prominent species released in Taiwan, was collected after its official release. One hundred and two field samples were compared with four batches of hatchery fry (n = 685) by using a microsatellite-based multiplex PCR assay. Four of the field samples (3.9%; 4/102) were revealed to be from a fish farm and most likely from a single batch. This study revealed that wild mangrove red snappers are genetically different from those originating from farms, and their origins can be traced through molecular markers, even without information on breeding stocks.

The genetic diversities and relationships of 10 Chinese indigenous pig breeds and three exotic pig breeds have been evaluated using 26 microsatellites recommended by the Food and Agriculture Organization & the International Society of Animal Genetics (FAO-ISAG). The allele frequencies, genetic heterozygosity (H) and polymorphism information content (PIC) have been calculated. The results showed that genetic diversity of Chinese indigenous pig breeds is higher than that of the introduced pig breeds. The clustering of 10 breeds is generally consistent with their geographical distribution.
Gummy stem blight (GSB), a common and serious disease in cucurbits worldwide, is caused by three genetically distinct species: Stagonosporopsis cucurbitacearum (syn. Didymella bryoniae), S. citrulli, and S. caricae. In Korea, however, the three species of Stagonosporopsis have been barely characterized. In this study, 21 Stagonosporopsis isolates were recovered from watermelon (Citrullus lanatus) and muskmelon (Cucumis melo) leaves and stem showing blight symptoms collected from 43 fields in Korea. Sequence analysis performed with an internal transcribed spacer region was not competent to differentiate the Stagonosporopsis isolates. On the contrary, analysis of β-tubulin (TUB) genes and three microsatellite markers, Db01, Db05, and Db06, successfully differentiated Stagonosporopsis isolates. Further sequence analysis identified two Stagonosporopsis species, S. citrulli and S. caricae, and one previously unknown species of Stagonosporopsis. Representative isolates from three species caused dark water-soaked lesions on the detached watermelon and muskmelon leaves with no significant differences in the aggressiveness. Our results indicate that the S. citrulli, S. caricae, and unknown Stagonosporopsis sp. are all causal agents of GSB for both watermelon and muskmelon. This is the first report of a new species and the population structure of Stagonosporopsis species causing GSB in Korea.

Among the 13 species of otters in the world, only one Eurasian otter (Lutra lutra) is found in South Korea. In the Korean Peninsula, otter pelts were historically valuable and expensive commodities used for international trade, and otters have long been poached as hunting animals. Recent rapid economic development in South Korea has increased habitat fragmentation and loss, creating a continuing threat to the natural environment. Otters live only in the area of rivers and streams as a family group and are territorial (linear habitat). Due to these limited conditions of otter habitat, the population size of otter is lower than that of onshore mammals. According to recent research, DNA analyses using microsatellite markers have shown that only approximately 7-21 otter individuals inhabit river systems for a length of 50-230 km. Korea's urban streams are associated with many threats that hinder otters from inhabiting them. Many areas around the urban streams are surrounded by high concrete riverbanks, and the risk of roadkill is also high. Nevertheless, ecological restoration projects in the urban rivers will contribute greatly to the stable inhabitation of otters. Detailed otter conservation strategies, such as the elimination of threat factors, improvement of habitat environment, and restoration of food resources and shelter, will provide a positive restoration effect on otter and river ecosystems as well.
국내 감자품종들의 품종간 유연관계를 분석하고 품종구분을 위한 DNA 표지인자를 개발하기 위하여 SSR(simple sequence repeats) 분석 및 다중초위성체 마커세트(multiplex-SSR set)를 개발하였다. 기존에 보고된 92개의 SSR 마커를 디자인 하고 이들을 이용하여 국내에서 육성된 24개 감자 품종에 대해 유전적 다양성을 분석하였다. 92개의 SSR 마커 중 PIC(polymorphism information contents) 값이 높은 14개의 SSR 마커를 선발하였고 PIC 값은 SSR 마커별로 0.48에서 0.89로 나타났고, 평균 값은 0.79였다. PSSR-29의 PIC 값은 0.48로 가장 낮은 값을 나타내었으며 PSSR-191에서 0.89로 가장 높은 값을 보였다. 선발된 14개의 SSR 마커를 이용하여 UPGMA 집괴분석 결과 24개의 감자 품종 중 21개의 품종이 2개의 집단으로 구분 할 수 있었으며 I 집단과 II 집단에는 각각 16개, 5개의 품종들이 군집되었으나 3개의 품종은 군집되지 않았다. 선발된 14개의 SSR 마커를 이용한 결과 24개의 품종에서 총 121개의 대립인자가 확인되었으며 각 마커별 대립인자는 3개에서 34개까지 확인되었고 평균 10.8개로 나타났다. 선발된 SSR 마커 중에서 PSSR-17, PSSR-24, PSSR-29 마커를 조합하여 다중초위성체 마커세트(multiplex-SSR set)를 개발하였다. 다중초위성체 마커세트는 한번의 PCR 반응과 PAGE 분석 만으로 본 연구에서 사용된 국내 24개의 감자 품종을 구분할 수 있었며 PIC 값은 0.95로 나타났다.
본 연구는 연산오계(천연기념물 제265호)와 이를 기원으로 하는 5개 지역별 오계 집단의 유전적 특성 및 차별성을 분석하기 위해 25개의 초위성체(MS) 마커를 이용하여 총 9개 집단 243수를 대상으로 유전자형을 분석하였다. 마커별 다형성 분석 결과, 총 153개의 대립유전자가 확인되었으며, $H_{\exp}$와 PIC의 경우 MCW0145에서 각각 0.640, 0.570으로 가장 높았고, $H_{obs}$는 MCW0252에서 0.607로 가장 높은 값을 나타내었다. 반면, LEI0166에서 $H_{\exp}$, $H_{obs}$, PIC가 각각 0.248, 0.204, 0.202로 가장 낮았다. 집단간 유전거리 분석 결과로는 9개 집단중 YSO 집단과 SUO 집단이 가장 가까운(0.073) 반면, LG 집단과 CBO 집단 사이에서 가장 먼(0.937) 것으로 확인되었다. 집단의 실제 구조를 확인하기 위한 집단별 균일도를 분석한 결과, 공시된 9개의 집단은 3개의 집단으로 구분했을 때 최적의 K값(7.96)을 얻을 수 있었으며, 5개의 오계 집단(YSO, ARO, CBO, CNO, SUO) 및 LG 집단과 CN RIR 집단은 각각 1, 2, 3번 군집에 분포하고 있는 것으로 나타났다. 한편, GBO 집단의 경우 1번과 3번 클러스터에 걸쳐서 분포하고 있는 것으로 보아 사육과정에서 타집단과의 교잡이 일어났을 것으로 추정된다. 이러한 결과를 통해 추후 오계 유전자원에 대한 국가 수준의 유전적 특성평가 및 관리의 기초 자료로 유용하게 활용될 것으로 기대된다.
본 연구는 30개의 MS 마커를 이용하여 한국재래염소 3개 집단(당진, 장수, 통영)과 1개 농가집단을 대상으로 집단 내 및 집단간의 유전적 다양성, 계통유전학적 유연관계 분석 및 한국재래염소 3개 집단간의 유전적 균일성을 검증하여 우리 고유유전자원으로서의 가치를 구명하고자 실시하였다. 대립유전자형 분석 결과, 총 277개의 대립유전자형 중 집단-특이 대립유전자형은 102개(36.8%)였으며, 다형성지수인 이형접합도의 관측치($H_O$)는 0.416~0.651, 다형정보량(PIC)은 0.462~0.679로 산출되었다. Nei의 $D_A$유전거리를 토대로 개체별 NJ 계통수를 작성한 결과 집단별로 독립적인 그룹을 형성하였는데, 한국재래염소 3개 집단 간의 유전거리에 비해 한국재래염소 집단과 농가 집단 간의 유전거리는 2배 이상을 보였다. 한국재래염소 집단의 실제적인 분류 및 분류된 군락의 균일도를 STRUCTURE software를 이용하여 분석한 결과, 실제 공시한 집단 수와 동일한 3개의 군락으로 분류가 가능했고, 각 집단에 대한 균일도는 통영(84.1%), 장수(78.1%), 당진(69.9%)의 결과를 나타냈다. 본 연구를 통하여 한국재래염소 집단의 유전적 다양성, 유연관계 및 유전적 균일성을 확인하였다. 본 연구에서 확인된 한국재래염소의 유전적 특성은 우리 고유자원에 대한 과학적인 근거자료이며, 나아가 가축유전자원에 대한 국가수준의 보존, 평가 및 이용에 활용될 수 있을 것으로 사료된다.
1973년 이스라엘 잉어(향어)가 한국에 양식을 위해 도입된 이후 현재까지 품종개량에 대한 연구가 전무한 실정이다. 본 연구는 지속적인 근친교배로 인해 낮아진 국내 이스라엘 잉어의 유전적 다양성을 회복하고, 성장이 빠르고 비늘 개선을 위해 유전적 기반 교잡육종 연구를 수행하였다. 본 연구는 한국의 이스라엘 잉어의 품종개량을 위하여 국내 이스라엘 잉어와 중국의 송푸거울 잉어를 이용하여 4개의 교배구를 설정하여 F1을 생산하였다. 친어의 형태 및 유전학적 거리를 고려하여 교배지침을 설정하였다. 본 연구는 유전적 다양성과 친자분석을 위하여 microsatellite 마커와 유전형 데이터를 활용하였다. 그 결과, 국내 친어의 평균 대립유전자와 기대이형접합율은 8.3과 0.743이며, F1은 13.0과 0.764이었다. 국내 이스라엘 잉어와 중국 송푸거울 잉어의 품종 간 교배를 통하여 국내 이스라엘 잉어보다 F1의 유전적 다양성이 회복되었음을 나타내었다. 한국의 일반 이스라엘 잉어는 17개월에 1.7 kg이었고, 개량된 이스라엘 잉어는 2.2 kg에 도달하였다. 또한, KC(한국×중국) 교배그룹의 비늘수치는 2.52, 친어그룹의 비늘수치는 3.15로 나타나 F1은 친어보다 낮은 비늘수치(0.63)를 나타내었다. 품종개량된 이스라엘 잉어(F1; CK, KC)는 친어그룹 (F0)보다 비늘이 20% 개선되었으며, 일반 이스라엘 잉어에 비해 체중(27%)과 비늘(25%)이 향상되었다. 유전적 데이터를 기반으로 개발된 이스라엘 잉어는 상업성이 좋아 국내 이스라엘 양식업에 크게 기여할 것으로 생각된다.
