Physiological Activities of Sesame, Black Sesame, Perilla and Olive Oil Extracts (참기름, 흑참기름, 들기름 및 올리브유 추출물의 생리활성)

  • This study investigated the physiological activities and antimicrobial effects of sesame, black sesame, perilla and olive oil extracts. Total flavonoid contents of sesame, black sesame, perilla and olive oil extracts were 2.7, 1.9, 3.0 and 1.4%, respectively, while total phenol contents were 6.5, 4.5, 4.1 and 10.1%, respectively. The electron donating abilities of sesame oil extract were markedly higher than black sesame, perilla or olive oil extract (p<0.05). The SOD-like activities of black sesame, perilla and olive oil extracts were 67.2%, 90.2% and 46.7%, respectively; in contrast, sesame oil extract did not show SOD-like activity. The order of the nitrite-scavenging abilities of sesame, black sesame, perilla and olive oil extracts was sesame> black sesame> perilla> olive oil extract (p<0.05). Olive oil extract showed strong antimicrobial activity to Bacillus cereus, Micrococcus luteus, Escherichia coli and Salmonella Enteritidis. The black sesame oil extract showed weak antimicrobial activity to Micrococcus luteus and Escherichia coli; conversely, sesame and perilla oil extracts did not show any antimicrobial activity.

A Comparative Study of the Detectable Methods of Residual Oxytetracyeline in Muscle of Flounder (Paralichthys olivaceus) with Simplified Screening Test (넙치 근육중 잔류 옥시테트라싸이클린의 간이스크리닝 검출방법 비교연구)

  • By standardized method, Bacillus subtilis BGA, Bacillus cereus var. mycoides ATCC 11778, and Micrococcus luteus ATCC 9341 were seeded on the muller hinton agar (Difco) plate, and pH was adjusted to 6.0, 7.2, and 8.0. Five agar plates, B. subtilis (pH 6.0), B. cereus (pH 6.0), B. subtilis (pH 7.2), B. subtilis (pH 8.0), and M. luteus (pH 8.0), were employed as test plates of modified EEC 4-plate method. Oxytetracycline (OTC) with a diet was orally administered to flounder, Paralichthys olivaceus, at 100 mg/kg once a day. After oral administration, modified EEC 4-plate method by the three screening test using muscle-direct, extraction-disk and direct-disk methods was conducted for 3 fish at 1, 3, 5, 10, 15, 20, 25 and 30 days. Muscle-direct treatment of B. subtilis (pH 6.0) was found to be dubious positive (${\pm}$) at the 1st day after the administration; thereafter, it was found to be negative to the last day of the experiment. Extraction-disk and direct-disk treatment of B. subtilis (pH 6.0) were found to be negative from the 1st day to the last day after the administration. B. subtilis (pH 7.2), B. subtilis (pH 8.0), and M. luteus (pH 8.0) by the three screening tests, were found to be negative all the way after the administration. On the other hand, B. cereus (pH 6.0) by the three screening tests was clearly found to be positive for the first 15 days after the administration, and then muscle-direct and direct-disk treatment of B. cereus (pH 6.0) were found to be dubious positive at 20th days after the administration. However extraction-disk treatment of B. cereus (pH 6.0) was clearly found to be negative at the same stage; thereafter, the three screening tests of B. cereus (pH 6.0) were found negative to the last of the experiment. These findings showed that to have equal sensitivity to those determination for the residual detection of OTC, and also confirmed that B. cereus was effective test organism for the monitoring of OTC.

Monitoring of Microbial Contamination in Air Purifier for Preventing Cross-contamination (교차오염방지를 위한 공기정화제품에서 미생물 오염도 분석)

  • Many atmospheric pollutants including chemical agents, house dust, and microorganisms cause building-related illnesses through respiration in humans. This study was conducted to analyze the profiles of microbial pollutants in air purifiers used in home, office and playschool. Dominant eleven species of microorganisms were isolated and identified in environmental air and air purifiers. Among them, Staphylococcus sp., Micrococcus sp. and Bacillus sp. are the most dominant species. By phylogenetic analysis of the 16S rRNA gene, the dominant bacteria were identified as Staphylococcus epidermidis, Micrococcus luteus and Bacillus epidermidis, respectively. It has been known that these bacterial species are closely related with food spoilage and human infectious disease. Thus, these results indicate that microbial pathogens related with human illnesses through respiration will be contaminated in air purifiers and also need to develop a method to control those of pathogens for human health.

Isolation and Characterization of Airborne Bacteria and Fungi in Indoor Environment of Elementary Schools (초등학교 실내환경에서 공기 중 세균과 진균의 분리 및 특성)

  • Indoor airborne bacterial and fungal concentrations were examined at classrooms and corridors of 3 elementary schools in Ulsan. Airborne microorganisms were collected with an impaction-type air sampler using plate count agar and dichloran rose bengal chloramphenicol agar. During the semester, concentrations of bacteria ranged $168{\sim}3,887 MPN/m^3$ at classrooms and $168{\sim}6,339 MPN/m^3$ at corridors, while those of fungi ranged $34{\sim}389 MPN/m^3$ at classrooms and $91{\sim}507 MPN/m^3$ at corridors. The bacterial concentrations showed larger variations between situations and schools compared to those of fungi. When airborne bacteria were isolated and identified, 84% were observed as Gram-positive, and Micrococcus spp. was the most abundant group with 61% of tested isolates, followed by genus Staphylococcus with 10%. The Micrococcus spp. isolates, of which 75% were identified as M. luteus, appeared to be from human origins. The protective pigments and substantial cell wall of Micrococcus may provide selective advantage for their survival in the air. We also isolated and identified 15 genera of filamentous fungi. The most common culturable fungi were Cladosporium, Aspergillus and Penicillium, and these 3 genera were 69% of fungal isolates. Genus Stachybotrys, of which S. chartarum is a well known producer of many potent mycotoxins, was also detected from one of the schools. further systematic studies are necessary with an emphasis on species identification and mycotoxin production of isolated fungal genera, including Aspergillus, Penicillium, and Stachybotrys.

Isolation of Leuconostoc mesenteroides subsp. mesenteroides DU-0608 with Antibacterial Activity from Kimchi and Characterization of Its Bacteriocin

  • A bacteriocin-producing strain, DU-0608, was isolated from Kimchi and identified as Leuconostoc mesenteroides subsp. mesenteroides. The bacteriocin from isolate was inhibitory against Listeria monocytogenes, Micrococcus luteus and several strains of lactic acid bacteria. The bacteriocin was inactivated by pepsin, trypsin, $\alpha$-chymotrypsin, protease, $\alpha$-amylase and lipase, but not by catalase or by heating at $100^{\circ}C$ for 60 min. The molecular weight of the bacteriocin was estimated approximately 6 kDa. The inhibitory effect was bactericidal and rapid. Following treatment with isolate bacteriocin, cells of indicator strain (Lactobacillus sake JCM 1157) were damaged at the end regions of the cell wall, whereas the cells treated with nisin were damaged at many places around the cell wall.

  • The scientific analysis of buried fabrics from the 16th century tomb of Kim HeumJo was conducted focusing on the conservation of fabrics, In order to find out the appropriate cleaning solvent and detergent for historical textiles physical chemical and biological analysis was conducted. The following results are obtained from this study : 1, The buried fabrics from the tomb of Kim HeumJo were composed of cotton silk and ramie. Most of fabrics had lost their original colors faded to brown. It was revealed variations in weaves and patterns were very developed at that times. 2. The chemical components of soils are hydrocarbons alkyl alcohols nitrogen compounds aromatic organic acid which is supposed to be from a human body microorganisms and their by-products. 3. Seven kinds of fungi Actinomycetes Corynebacterium spp Micrococcus luteus Bacillus Clostridium were isolated from the fabrics. The most common fungus was Bacillus.

One Pot Synthesis of Novel Cyanopyridones as an Intermediate of Bioactive Pyrido[2,3-d]Pyrimidines

  • Synthesis, structural characterization, and biological activity studies of novel pyrido[2,3-d]pyrimidines (10a-h, 11a-h) are described. Cyclization of cynoacetamides (4, 5) with malonitrile (7) and aldehyde (6a-h) via Hantzsch pyridine synthesis afforded cyanopyridones (8a-h, 9a-h), which on cyclization with formic acid under microwave conditions led to the final product. All the reactions are significantly faster and the isolated yields are remarkably higher in microwave conditions compared to the conventionally heated reactions. The compounds were tested in vitro for their antibacterial activity against Escherichia coli, Pseudomonas aeruginosa, Bacillus subtillus, Staphylococcus aureus, and Micrococcus luteus and antifungal activity against Trichphyton longifusus, Candida albicans, Microsporum canis, Fusarium solani. Compounds 10b, 10e, 11b and 11e exhibited good antibacterial and antifungal activities compared with standards.

  • 육류중에 잔류하는 항생물질 및 항균제를 검출하기 위해 본 실험에서는 3종의 균주 Bacillus subtilis ATCC 6633, Micrococcus luteus ATCC 9341, BAcillus cereus var. mycoides ATCC 11778을 사용하여 실험하였다. 시료의 clean-up은 항생·항균제의 물리화학적 성질을 고려하여 우선 McIlvaine buffer를 가하여 homogenize하고 hexane으로 defatting 시킨 후, chloroform으로 추출한 액과 Sep-Pak C18과 Bakerbond SPE carboxylic acid column에 흡착시킨 후 추출한 액을 각각 시험용액 A, B, C,로 하였다. 각 test solution을 paper disk를 사용하여 함균 배지에 올려놓고 overnight culture 후 inhibition pattern을 통해 여러 종류의 항생·항균제를 계통적으로 검출하였다. 본 실험에서 macrolide계와 tetracycline계 등은 0.1ppm 이하의 detection limit을 보였으며, penicillinrP는 0.001ppm이하의 높은 detection limit을 나타내므로서 시료중에 잔류하는 극미량까지도 검출할 수 있었다. 본 방법은 식육중의 잔류 항생·항균제를 동시에 간단하게 계통적으로 분류하는데 있어 좋은 방법이라고 생각되며 항생·항균제의 체계적인 1차 screening 수단으로서 유용한 방법이라 사료된다.

ANTIBIOTICS RESIDUES IN RAW MILK IN THAILAND

  • One thousand eight hundreds and twenty two samples of raw milk were detected for antibiotic residues using Bacillus subtilis ATTCC 6633, B. stearothermophilus var. calidolactis C 593 and Micrococcus luteus ATCC 9341 as test organisms, were carried out from July 1991 through June 1992. Apparent antibiotic residues were found through out the study period, except in January. The detection rate varied from 0.7% in March and May to 11% in April. One hundred and thirty six (72%) samples of the 187 screening positive samples were considered to contain only the indigenous antimicrobial agents. Of the total, 51 (2.8%) samples were positive for antibiotic residues. Among the tested organisms, B. stearothermophilus var. calidolactis was the most sensitive organism in detection of the antibiotic residues.

  • The cDNA, encoding human lysozyme (HLY) which was isolated from a human placenta cDNA library, has been well characterized (Yoshimura et al., 1988). Based on the communication, we have prepared an artificial HLY gene from chemically synthesized 38-oligomer with high codon usage in Saccharomyces cerevisiae. For directing the synthesis and secretion of HLY in S. cerevisiae, an expression vector, pHKl was constructed by inserting the HLY gene, containing a synthetic HLY secretion signal sequence, between the yeast GAP promoter and PH05 terminator. From a lysoplate assay, we have confirmed an yeast transformant harboring a pHK1 which makes a clearing zone on the overlayed Micrococcus luteus. This result means a chemically synthesized HLY gene which was normally expressed and secreted in yeast.
