Microbiological Water Quality of Water Purifiers at Elementary Schools in Gunsan Area (군산지역 초등학교 정수기 물의 미생물학적 수질)

  • In this research, we investigated the actual conditions of water purification systems at ten elementary schools located in Gunsan, Korea from July to December, 2007. The results were as follows; The population densities of heterotrophic bacteria in water purifiers ranged from 0 to $1.2{\pm}0.2{\times}10^4$ CFU/ml and those of tap water were in the range from 0 to $1.9{\pm}0.3{\times}10^4$ CFU/ml during investigation periods. Ninety percentage of purified water samples in July and September, 87.2% in October and November, and 93.7% in December turned out not to be suitable for drinking. The seasonal variation of the population densities of heterotrophic bacteria from purified waters was not notable. The total coliform, Salmonella and Shigella were not detected in purified water and tap water during investigation periods. Forty-five species of bacteria were isolated from water purifiers. The identified bacterial genera were Sphingomonas, Methylobacterium, Caulobacter, Novosphingobium, Bosea, Brevundimonas, Aminobacter, Ralstonia, Mitsuaria, Variovorax, Acidovorax, Massilia, Pseudomonas, Acinetobacter, Aeromonas, Bacillus, Staphylococcus, Brevibacillus, Microbacterium, Lapillicoccus, Micrococcus, Arthrobacter, Janibacter, Flavobacterium, Chryseobacterium, and Hymenobacter: Among the isolates, opportunistic pathogens such as Pseudomonas fluorescens, Staphylococcus epidermidis, Flavobacterium johnsoniae, and Acinetobacter johnsonii were also found.

Characteristics of Fermented Omija (Schizandra chinensis Baillon) Sugar Treatment Extracts by Lactobacillus sp. (Lactobacillus sp. 균주를 이용한 오미자 당침액의 발효특성)

  • This study was carried out to investigate the characteristics of FOST (fermented omija sugar treatment extracts) using Lactobacillus brevis HLJ59. Antioxidant activities of FOST were evaluated through viable cell number of L. brevis HLJ59, DPPH radical scavenging activity, reducing power and SOD-like activity, compared to non-FOST(non-fermented omija sugar treatment extracts). Also it was to evaluate Angiotensin Converting Enzyme (ACE) and Urease inhibitory activity of FOST. The viable cell number of L. brevis was about $2.05{\pm}0.21{\times}10^8$, $6.31{\pm}0.56{\times}10^{11}$, and $8.14{\pm}0.14{\times}10^9$ at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. DPPH radical scavenging activity of FOST was about 60.3%, 71.8%, and 44.5% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The reducing power of FOST was about 0.92, 1.19, and 0.73 (OD at 700 nm) at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, and SOD-like activity of FOST was about 50.4%, 53.7%, and 33.4% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. ACE and Urease inhibitory activity by FOST was about 47.4%, 78.2%, 56.4% and 58.1%, 83.4%, 63.2% at ${\times}2$, ${\times}5$, and ${\times}10$ dilution, respectively. The results indicated that the fermentation of omija sugar treatment extracts using Lactobacillus brevis HLJ59 increased the antioxidant activities campared to the non-fermented omija sugar treatment extracts.

Antioxidant Activities and Monacolin K Production on Solid-State Fermentation of Diverse Yam by Aspergillus Species Strain (Aspergillus 속 균주를 이용한 마 품종별 고체발효시 Monacolin K 생산과 항산화 활성)

  • This study was conducted to investigate the characteristics between non-fermented chinese yam and rice (non-FCYR) and fermented chinese yam and rice (FCYR). 1.1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, total polyphenol, flavonoid contents and reducing power were investigated to evaluate the anti-oxidation activities of FCYR. Also, productivity of monacolin K, major medicinal ingredient of FCYR, was investigated, using Aspergillus terreus KCCM 12225. In case of non-FCYR, DPPH radical scavenging activities for three kinds of yam (Dioscorea japonica, Dioscorea batatas, Dioscorea opposita) and rice were estimated about 51.8, 66.4, 42.2 and 7.5%, while in case of FCYR, those for three kinds of yam and rice were estimated to increased about 64.7, 74.7, 52.8, and 32.3%, respectively. Total polyphenol contents of non-FCYR were estimated to 196.9, 265.7, 160.1, and 91.6 mg/kg, while total polyphenol contents of FCYR were estimated to increase about 530.7, 708.3, 427.2, and 265.9 mg/kg, respectively. Total flavonoid contents of FCYR increased to 2.8-3.3 times higher than those of non-FCYR. Reducing power of non-FCYR was about 0.97, 1.28, 0.64, and 0.17 (OD at 700 nm), while, that of FCYR increased about 1.75, 2.38, 1.24, and 0.46. Remakable increase in monacolin K productivity of FCYR was observed. Monacolin K productivity of FCYR was estimated to 467.1, 514.8, 339.2, and 272.5 mg/kg, respectively. In this study, fermented chinese yam (Dioscorea batatas) was estimated to be effective biological activity material.

  • This study was carried out to investigate the characteristics of fermented chinese yam and rice (Dioscorea japonica, Dioscorea batatas, Dioscorea opposita, Rice, FCYR) using Monascus sp. MK805. The extracts from FCYR were measured to examine pigments, antioxidant activities were investigated through DPPH radical scavenging activity, total polyphenol and flavonoid contents, reducing power. Also it was investigated monacolin K productivity by FCYR. Pigments productivity (yellow, orange and red) were 26.2, 13.9, 17.3 at Dioscorea japonica, 41.9, 22.6, 53.2 at Dioscorea batatas, 12.5, 7.5, 9.7 at Dioscorea opposita and 10.1, 7.7, 10.2 at rice, respectively. DPPH radical scavenging activity of FCYR was about 69.7, 79.6, 57.8, and 42.3%, total polyphenol contents of FCYR was about 480.6, 658.7, 379.3, and 212.9 mg/kg, total flavonoid contents of FCYR was about 342.5, 448.4, 235.2, and 168.7 mg/kg, reducing power of FCYR was about 1.57, 2.14, 1.14, and 0.35 (OD at 700 nm), respectively. And then monacolin K productivity of FCYR was about 453.8, 509.5, 332.2, and 263.2 mg/kg, respectively.

Availability of Marine bacteria (Erythrobacter sp. $S{\pi}-1$) for enrichment of livefood in the slime flounder larvae, Microstomus achne (찰가자미 자어에 있어서 먹이생물의 영양강화을 위한 해양세균 (Erythrobacter sp. $S{\pi}-1$)의 이용)

  • This study was performed to compare the effect of enrichement of rotifer and Artemia fed marine bacteria (Erythrobacter sp. S$\pi$-1) with those fed the different diets-(Super Selco, $\omega$-yeast and marine Chlorella) on the slime flounder larvae, Microstomus achne. Because the ratio of docosahexaenoic acid (DHA) to fatty acids in rotifer (dry weight $\%$) fed on Super Selco and ESP-SR including Erythrobacter sp. S$\pi$-1 was higher than that in rotifer fed marine Chlorella, the growth of the slime larvae fed the former showed better than the latter. And the ratio of DHA and highly unsaturated fatty acid (n-3 HUFA) to fatty acids in Artemia (dry weight $\%$) enriched by ESP-A including Erythrobacter sp. S$\pi$-1 were also higher than those in Artemia enriched by others or Artemia without enrichment. Larvae fed on Artemia enriched by ESP-A showed better growth and survival rate than those fed on Artemia enriched by others or Artemia without enrichment. With regard to dietary value of Erythrobacter sp. S$\pi$-1, its use could improve the quality of the live foods for the slime flounder larvae.

Influence of Soil Flooding on Control of Pink Root Disease in Onion Crop (양파 연작지의 분홍색뿌리썩음병 방제를 위한 담수처리 효과)

  • Pink root disease of onion, a known worldwide constraint upon onion production, significantly reduces crop levels in the main Korea cultivation area. In order to examine the effect of flooding on incidence of pink root disease caused by Pyrenochaeta terrestris and on onion growth and on populations of soil fungi, field experiments were conducted during two seasons, 2003/04 and 2004/05 at Onion Research Institute. Populations of soil fungi from fields were assayed on selective media. Flooding treatment was effective in reducing populations of P. terrestris, Pythium spp., and Rhizoctonia sp. in soil; fungal populations in soils flooded for 90 days were reduced to 1/2 to 1/3 of those in non-flooded soils. In nursery bed, protective activities of soils flooded for over 60 days were 93.5 to 99.2% and their pink root incidences were less than 5%, which were 1/11 to 1/18 of that in control. Increased yield of onion bulbs was associated with control of soil borne pathogenic fungi by flooding treatments. As flooding period was prolonged, bulb grade showed the tendency to increase. Soil flooding for over 60 days resulted in effective disease control, facilitated accurate planning of plant population in the field, extension of the growing-season and consequently higher yields of better quality.

Bonding Quality of Adhesives Formulated with Okara Hydrolyzates and Phenol-formaldehyde Resins for Bonding Fancy Veneer onto High-density Fiberboard (두부비지 가수분해물과 페놀수지로 조제한 마루판 화장용 접착제의 접착성능)

  • In our study, the potential of okara as an ingredient of new bio-based adhesives was investigated for the production of fancy-veneered flooring boards. Okara was hydrolyzed by 1% sulfuric acid solution (AC) and 1% sodium hydroxide solution (AK). Phenol formaldehyde (PF) prepolymers were prepared as a cross-linker of okara hydrolyzates. Then, okara-based adhesive resins were formulated with 35% AC, 35% AK and 30% PF prepolymer on solid content basis. The adhesive resins were applied on high-density fiberboards (HDF) with the spread rate of $300g/m^2$. After that, oak fancy veneers are covered on the HDF, and then pressed with the pressure of $7kg/m^2$ at $120^{\circ}C$. The experimental variables were three mole ratios of formaldehyde to phenol (1.8, 2.1, 2.4), three assembly time (0, 10, 20 min), and two press time (90 sec, 120 sec), respectively. The fancy-veneered high-density fiberboards were tested by dry tensile strength, glueline failure by wetting and formaldehyde emission. Tensile strength of the boards exceeded the requirement of KS standard. The formaldehyde emissions were approached at the E0 level specified in KS standard. Based on these results, okara can be used as an ingredient of environmentally friendly adhesive resin systems for the production of flooring boards.

The Effects of Gamma Irradiation on the Microbiological, Physicochemical and Sensory Quality of Peach (Prunus persica L. Batsch cv Dangeumdo) (감마선 조사에 의한 복숭아의 미생물학적, 이화학적 품질 및 관능적 품질 변화)

  • The effect of gamma irradiation ($0.5{\sim}2$ kGy) on the microbiological, physicochemical and sensory properties of peaches was investigated during 6 day storage at $20{\pm}3^{\circ}C$. Total aerobic bacteria, yeasts and molds significantly decreased with increasing dose level. In Hunter's color values, L-values decreased and a- and b-values increased with increment of irradiation dose level. Hardness significantly decreased with increment of irradiation dose level whereas pH, soluble solid, and total polyphenol contents increased with increment of irradiation dose level. DPPH (1,1-diphenyl-2-picrylhydrazyl) radical-scavenging activity of irradiated peach was higher than that of control and its activity increased with increment of irradiation dose level. Vitamin C content was not affected by irradiation. In sensory test, overall acceptabilities of irradiated samples were higher than those of control. These results suggest that gamma irradiation on peach was effective for microbiological safety while improving the antioxidant activity, but not good on color and texture of peach.

Identification of the Volatile Components in Korean Ordinary Kochujang (한국재래식(韓國在來式) 고추장의 향기성분동정(香氣成分同定))

  • The present paper was attempted to obtain the basic date concerning a reasonable preparing method and optimum fermentation conditions of Kochujang (Red pepper paste). To establish the standard quality of Kochujang, the chemical compositions and the volatile components of Kochujang was discussed. The native Kochujang collected from 80 households contained 40.51% of moisture, 6.00% of salts, 3.25 % of crude fat, 10.30 % of crude ash, pH 4.79, 9.28ml of tillable acidity. 19.60% of reducing sugar, 179.51 mg/100g of amino nitrogen and 4.43% of total nitrogen. The volatiles of Kochujang were extracted by a steam-distillation under the reduced pressure and determined by gas chromatography-mass spectrometry (GC-MS). Total volatile components identified in the native Kochujang were 46 components, i. e., 30 components in neutral fraction, 8 components in phenolic fraction and 8 components in acidic fraction, respectively. But no components were detected basic fraction.

Molecular epidemiological study of measles virus throughout an imported epidemic outbreak in Gyeonggi-do in 2014 (2014년 경기지역에서 발생한 해외유입 홍역 바이러스의 분자역학적 특성 분석)

  • Measles virus is a highly contagious, exanthematic virus, preventable by the use of an effective live-attenuated vaccine. However, measles virus remains endemic in many area of the world causing nearly 200,000 deaths per year and still a major cause of child mortality, mostly in developing countries. In March 2014, Republic of Korea was certified as a 'national measles elimination' by the WHO as a result of a high-quality case-based surveillance system and population immunity, which was achieved by a high vaccination rate (>95.0% since 1996). But, since the beginning of 2014, the Gyeonggi province has experienced a resurgence of measles cases. In this study, we investigated the characteristics of measles viruses isolated from confirmed measles in Gyeonggi province during January 1, 2014 ~ July 31, 2014, 60 isolates were obtained from 72 confirmed measles specimens. Genotypic distributions and genetic diversities of isolated measles virus were analyzed by sequencing of nucleoprotein (N) gene. 58 (96.7%) imported cases were identified. The predominant genotype was B3, which reflects the circulating measles virus in adjacent countries. The sequences of nucleoprotein (N) gene of isolated MeV were showed that the strains characterized showed the highest degree of identity (99%) with the Philippine related strains in 2013-2014. Therefore, infected traveler returning from the Philippines transmitted secondary infection in Korea.
