Improvement in microbial stability of photosynthetic bacteria via optimized cell immobilization and lyophilization: Application to the treatment of shrimp aquaculture water

  • Photosynthetic bacteria (PSB) play an important role in water purification, and their application is beneficial for sustainable aquaculture. However, maintaining the microbial stability of PSB from subculturing to preservation is a challenging task. Since improvement in the microbial stability of PSB is a crucial parameter, optimized conditions for cell immobilization and lyophilization were investigated. In PSB immobilization, 0.1-M CaCl2 was found to be the most effective divalent metal ion solution in terms of cost-effectiveness, resulting in beads with a 4-mm diameter and high loading (1.91×109 CFU/mL) of viable cells. Maintenance of cell viability, external appearance, and color of PSB beads was best in 3.5% NaCl during storage. In lyophilization, the addition of skim milk (9%) and dextrose (2%) as cryoprotective additives allowed the highest cell viability. Over an 18-week shrimp breeding period, when optimally manufactured beads and lyophilized powder of PSB were applied to shrimp aquaculture water, NH4+, NO3-, and NO2- were more effectively removed by 55%, 100%, and 100%, respectively, compared to controls. Thus, microbial stability of PSB through optimized cell immobilization and lyophilization was successfully enhanced, enabling a wide application.

  • BACKGROUND: Towards achievement of sustainable agriculture, using microbial inoculants may present promising alternatives without adverse environmental effects; however, there are challenging issues that should be addressed in terms of effectiveness and ecology. Viability and stability of the bacterial inoculants would be one of the major issues in effectiveness of microbial pesticide uses, and the changes within the indigenous microbial communities by the inoculants would be an important factor influencing soil ecology. Here we investigated the stability of the introduced bacterial strains in the soils planted with barley and its effect on the diversity shifts of the rhizosphere soil bacteria. METHODS AND RESULTS: Two different types of bacterial strains of Bacillus thuringiensis and Shewanella oneidensis MR-1 were inoculated to the soils planted with barley. To monitor the stability of the inoculated bacterial strains, genes specific to the strains (XRE and mtrA) were quantified by qPCR. In addition, bacterial community analyses were performed using v3-v4 regions of 16S rRNA gene sequences from the barley rhizosphere soils, which were analyzed using Illumina MiSeq system and Mothur. Alpha- and beta-diversity analyses indicated that the inoculated rhizosphere soils were grouped apart from the uninoculated soil, and plant growth also may have affected the soil bacterial diversity. CONCLUSION: Regardless of the survival of the introduced non-native microbes, non-indigenous bacteria may influence the soil microbial community and diversity.

  • To examine stability and a separate quantitative method of a mixed preparation of lactic acid bacteria, a capsule containing Lactobacillus acidophilus, Lactobacillus bulgaricus and Streptococcus thermophilus was suspended and diluted in sterile water. After the diluted suspension was spread on three media of tryptone glucose extract agar, MRS agar and MRS-sucrose agar, their colonies appeared and were counted. The viable counts exceeded the minimum number of the three bacteria and showed that the mixed preparation was stable at least for 18 months. The results also showed that a separate quantitation of viable cells of the each strain was feasible.

Enzyme and Microbial Activities in Paddy Soil Amended Continuously with Different Fertilizer Systems

  • Soil enzyme and microbial activities are affected by fertilizer and compost applications and can be used as sensitive indicators of ecological stability. Microbial population and soil enzymes viz., dehydrogenase, urease, acid phosphatase and aryl-sulphatase were determined in the long-term fertilizer and compost applied paddy soil. Soil samples were collected from the four treatments (control, compost, NPK and compost+NPK). Long-term NPK+compost application significantly increased activities of urease, dehydrogenase and acid phosphatase than all other treatments. The compost application enhanced activities of urease, dehydrogenase and acid phosphatase than the NPK application. However, arylsulfatase activity was not significantly different between compost and fertilizer application. The highest microbial population was recorded in the NPK+compost treatment. The compost application also resulted in higher microbial population than the NPK application. The above results indicate that ecological stability could be maintained by application of compost alone or with NPK.

미생물 첨가와 사일로 밀도가 총체벼 사일리지의 영양소 함량, 발효특성 및 호기적 안전성에 미치는 영향 (Effects of Microbial Additives and Silo Density on Chemical Compositions, Fermentation Indices, and Aerobic Stability of Whole Crop Rice Silage)

  • 본 연구에서는 미생물 첨가제와 사일로 밀도가 총체벼 곤포 사일리지의 영양소 함량, 발효특성 및 호기적 안전성에 미치는 영향을 규명하고자 수행하였다. 미생물 첨가와 사일로 밀도는 사일리지의 영양소 함량에 영향을 미치지 않았다. 하지만, INO 시험구에서 lactate 함량, lactate와 acetate 비율 및 LAB는 감소하였고, propionate 함량, yeast 및 호기적 안전성은 증가하였다. 사일로 밀도는 pH와 lactate 함량에 영향을 미쳤으나, 처리 구간 차이는 매우 적었다. 이상의 결과에서, L. brevis 5M2와 L. buchneri 6M1을 포함한 혼합형 LAB 첨가제 사용은 총체벼 곤포 사일리지의 호기적 안전성 개선에 유리하며, 사일로 밀도는 베일 압력이 430~770 kgf/cm2일 때 정상적인 사일리지 발효가 일어나는 것으로 확인되었다.

Immobilization and Stability of Lipase from Mucor racemosus NRRL 3631

  • The lipase from Mucor racemosus NRRL 3631 was partially purified by fractional precipitation using 60% ammonium sulfate, which resulted in a 8.33-fold purification. The partially purified lipase was then immobilized using different immobilization techniques: physical adsorption, ionic binding, and entrapment. Entrapment in a 4% agar proved to be the most suitable technique (82% yield), as the immobilized lipase was more stable at acidic and alkaline pHs than the free enzyme, plus 100% of the original activity was retained owing to the thermal stability of the immobilized enzyme after heat treatment for 60 min at $45^{\circ}C$. The calculated half-lives (472.5, 433.12, and 268.5 min at 50, 55, and $60^{\circ}C$, respectively) and the activation energy (9.85 kcal/mol) for the immobilized enzyme were higher than those for the free enzyme. Under the selected conditions, the immobilized enzyme had a higher $K_m$ (11.11 mM) and lower $V_{max}$ (105.26 U/mg protein) when compared with the free enzyme (8.33 mM and 125.0 U/mg protein, respectively). The operational stability of the biocatalyst was tested for both the hydrolysis of triglycerides and esterification of fatty acids with glycerol. After 4 cycles, the immobilized lipase retained approximately 50% and 80% of its original activity in the hydrolysis and esterification reactions, respectively.

Stability of pUC-Derived Plasmids with a Fluorescence Marker in Pectobacterium carotovorum subsp. carotovorum and subsp. betavasculorum

  • The stability of three different kinds of pUC-derived plasmids, pDsRed, pZsYellow, and pGFPuv, was investigated in Pectobacterium strains to utilize those plasmids as tracers. All three plasmids pDsRed, pZsYellow and pGFPuv showed their specific colors in Pectobacterium strains. Especially, the plasmid pDsRed conferred bright pink colonies on the Pectobacterium strains. When the bacteria lost the plasmid pDsRed, the colonies turned white, suggesting that the plasmid could be a good marker system for Pectobacterium strains on different environmental conditions. The effect of the antibiotic pressure on the stability of the plasmid was different depending on the host bacteria. P. carotovorum subsp. betavasculorum was more sensitive to the antibiotic pressure than P. carotovorum subsp. carotovorum Pcc21. However, temperature change significantly affected plasmid stability on both Pectobacterium strains. Almost all strains lost the plasmids with the shift in temperature from $28^{\circ}C$ to $37^{\circ}C$. Presence of the plasmids did not affect bacterial pathogenicity on their own host plants. Among three plasmids, pZsYellow was not useful as a marker because the yellow fluorescent proteins from pZs Yellow were interfered with the yellow natural fluorescence of the plant tissues induced by the defense system. Since the red color of DsRed can be seen with naked eyes, plasmid pDsRed was applicable as a marker. However, the color change was slow so that additional manipulation to increase the expression speed was necessary. Plasmid pGFPuv could serve as a perfect marker without any problem, tracing the reproduction and spread of the plant pathogens perfectly.

바이오디젤의 미생물 안정성 조사 (Microbiological Stability Test of Biodisel)

  • 동ㆍ식물성 기름으로부터 산이나 알카리 촉매를 사용하여 제조한 친환경 에너지인 바이오디젤의 미생물학적 안정성을 테스트하기 위하여 온도와 저장기간에 따른 미생물에 의한 변질정도를 측정하였다. 측정방법으로 역상 현미경 관찰과, 3M petrifilm분석법으로 생균수를 측정하였고, 균체 단백질의 양을 측정하기 위하여 Bradford protein assay법을 사용하였다. 바이오디젤과 석유디젤, 그리고 각각의 시료를 혼합한 시료를 서로 다른 온도조건(25℃와 35℃)에서 혐기적인 조건으로 90일동안 장기간 저장했을 때 바이오디젤, 석유디젤 그리고 혼합한 시료에서 미생물의 생육은 관찰되지 않았다.

  • 전분질의 에탄을 직접발효에 유용한 Saccharomyces diastaticus의 glucoamylase 생산성 향상을 위하여 glucoamylase 유전자 STA1을 함유한 재조합 plasmid pYES 18을 갖는 S. diastaticus의 형질전환 균주에서 성장속도가 plasmid의 안정성에 미치는 영향과 plasmid 안정성에 따른 glucoamylase 생산성을 조사하였다. 최소 선택배지에서는 plasmid가 일정한 수준에서 안정하게 유지되었으나, 균체의 증식과 glucoamylase 생산성은 매우 미약하였다. Starch가 포함된 완전배지에서는 glucose를 첨가해 줌으로써 생육속도를 촉진시킬 수 있었으며 생육속도의 촉진에 의해 plasmid 안정성이 증가되었고 이에 따라 glucoamylase 생산성이 향상됨을 알았다.

근린약국에서 산제로 조제된 아테놀올정의 안정성 (Stability of Atenolol Tablet After Dispensing to Powder form at Community Pharmacies)

  • Prescription filling in powder form is performed in community pharmacy practice to adjust dose for children and patients who cannot swallow whole tablet. However, there are few reports regarding the stability of the active ingredient and possible microbial growth after the medication is dispensed to powder form. This study examined the stability of atenolol, an antihypertensive agent, and microbial growth in the unit dose pouches dispensed at twenty-one community pharmacies located in Taegu area. Randomly chosen first unit dose pouch contained 77.4% of the prescribed dose of the drug and there were only four community pharmacies that dispensed the drug within 10% deviation from the dose prescribed by physician. Surprisingly, there were three community pharmacies that dispensed the drug with greater than 40% deviation, which may pose a major concern regarding the efficacy and safety of the drug prescribed for the treatment of hypertension. Atenolol content during a month did not indicate significant change, showing 5.4%, 4.3%, and 3.3% of decrease in 50%, 80%, and 90% relative humidity conditions, respectively. Microbiological examination during a month showed less than 0.5 microorganism in high power field (hpf) in all the relative humidity conditions tested. Based on this study, pharmacy practice in community pharmacy needs to be rigorously regulated to ensure that the dose of the prescribed drug is properly incorporated into the unit dose pouch dispensed as powder form.
