Effect of 1-methylcyclopropene Treatment on Extension of Freshness and Storage Potential of Fresh Ginseng (1-methylcyclopropene 처리의 수삼의 신선도 유지 및 저장성 연장 효과)

  • Fresh ginseng has a limited storage life due to the quality change caused by microbial spoilage as well as physiological deterioration. The present study investigated the effects of 1-methylcyclopropene (1-MCP) treatment, an inhibitor of ethylene action, on the microbial growth and quality maintenance of fresh ginseng during storage. Harvested fresh ginsengs were treated with $1{\mu}L{\cdot}L^{-1}$ 1-MCP for 20 hours at $4^{\circ}C$ and then stored at room temperature (RT) for 18 days or low temperature ($4^{\circ}C$) for 160 days. After 18 days of storage at RT, the percentage weight loss in 1-MCP treated fresh ginseng (8.3%) is lower than that of control (10.1%). During long-term storage at $4^{\circ}C$, weight losses were increased slightly until 120 days without difference between non-treated and 1-MCP ginsengs. In contrast, after 120 days of storage at $4^{\circ}C$, higher increase in weight loss was observed in non-treated ginsengs than in 1-MCP treated ginsengs. Respiration rate and ethylene production of fresh ginseng were reduced by 1-MCP treatments at RT. The 1-MCP treatment also resulted in lower microbial population compared to those of non-treated ginsengs at RT. However, in ginsengs stored at $4^{\circ}C$ for short-term (45 days), no differences were noted in weight loss and microbial population between 1-MCP treated and non-treated ginsengs. Major ginsenosides was not changed by 1-MCP treatment during the 7 days of storage at RT. Results suggest that 1-MCP treatment can be used to maintain the freshness of ginseng at room temperature for short term storage and at low temperature for long term storage. 1-MCP treatment could be applied on fresh ginseng to avoid deleterious effect of exogenous ethylene during storage and shipping.

Comparative analysis of the microbial communities in raw milk produced in different regions of Korea

  • Objective: The control of psychrotrophic bacteria causing milk spoilage and illness due to toxic compounds is an important issue in the dairy industry. In South Korea, Gangwon-do province is one of the coldest terrains in which eighty percent of the area is mountainous regions, and mainly plays an important role in the agriculture and dairy industries. The purposes of this study were to analyze the indigenous microbiota of raw milk in Gangwon-do and accurately investigate a putative microbial group causing deterioration in milk quality. Methods: We collected raw milk from the bulk tank of 18 dairy farms in the Hoengseong and Pyeongchang regions of Gangwon-do. Milk components were analyzed and the number of viable bacteria was confirmed. The V3 and V4 regions of 16S rRNA gene were amplified and sequenced on an Illumina Miseq platform. Sequences were then assigned to operational taxonomic units, followed by the selection of representative sequences using the QIIME software package. Results: The milk samples from Pyeongchang were higher in fat, protein, lactose, total solid, and solid non-fat, and bacterial cell counts were observed only for the Hoengseong samples. The phylum Proteobacteria was detected most frequently in both the Hoengseong and Pyeongchang samples, followed by the phyla Firmicutes and Actinobacteria. Notably, Corynebacterium, Pediococcus, Macrococcus, and Acinetobacter were significantly different from two regions. Conclusion: Although the predominant phylum in raw milk is same, the abundances of major genera in milk samples were different between Hoengseong and Pyeongchang. We assumed that these differences are caused by regional dissimilar farming environments such as soil, forage, and dairy farming equipment so that the quality of milk raw milk from Pyeongchang is higher than that of Hoengseong. These results could provide the crucial information for identifying the microbiota in raw milk of South Korea.

The Effects of Sugar Content on Growth of Zygosaccharomyces bailii in Rice-syrup (쌀엿 저장시 주요 오염원인 Zygosaccharomyces bailii의 성장에 당도가 미치는 영향)

  • Spoilage yeast was isolated from spoiled rice-syrup and identified by Microbial Identification system (MIDI) as Zygosaccharomyces bailii based on composition of fatty acids exhibiting carbon skeleton of 16:0, 16.1 $({\Delta}cis\;7)$, 18:2 $({\Delta}cis\;9)$, 18:1 $({\Delta}cis\;9)$, and 18:1. Increase in sugar content $(1^{\circ}brix)$ resulted in decreased moisture concentration (1%), and water activity decreased from 0.72 to 0,63 when sugar concentration increased from 77 to $83^{\circ}brix$. Yeast showed very slow growth during three weeks post-inoculation in rice-syrup at 17 and $79^{\circ}brix$, and maximal level of Z. bailii was found at 6 and 10 weeks storage at 15 and $25^{\circ}C$, respectively. At over $81^{\circ}brix$ its growth slowly decreased at beginning of storage, indicating rice-syrup could be preserved safely at above $81^{\circ}brix$ sugar content without Z. bailii contamination.

Effects of sodium diacetate or microbial inoculants on aerobic stability of wilted rye silage

  • Objective: The primary goal was to identify the effectiveness of chemical or biological additives in delaying the deterioration of early-harvested wilted rye silage after exposure to air. Methods: Rye harvested as a whole plant at the early heading stage was wilted for 24 h. The wilted forage was divided into treatments including sodium diacetate (SDA) at 3 (SDA3) and 6 g/kg (SDA6), Lactobacillus plantarum (LP), L. buchneri (LB), or their equal mixture (LP+LB) at 1×106 colony-forming unit/g fresh matter. Results: After 60 d of conservation in 20-L silos, lactic acid was greater in LP and LP+LB silages than other treatments (102 vs 90.2 g/kg dry matter [DM]). Acetic acid was greatest in SDA6 (32.0 g/kg DM) followed by LB (26.1 g/kg DM) and was lowest in LP treatment (4.73 g/kg DM). Silage pH was lower with microbial inoculation and the lowest and highest values were observed in LP and untreated silages, respectively. After 60 d, neutral detergent fiber concentration was lowest in SDA6 silages, resulting in the greatest in vitro DM digestibility (846 g/kg DM). Aerobic stability was longest in SDA6 (176 h) followed by LB treatment (134 h). Instability after aerobiosis was greatest in LP silages (68 h), about 8 h less than untreated silages. After aerobic exposure, yeast and mold numbers were lowest in SDA6 silages, resulting in DM loss minimization. Exhaustion of acetic acid and lactic acid after aerobic exposure was lowest with SDA6 but greatest with untreated and LP silages. Conclusion: Treatment of early-cut wilted rye forage with SDA at 6 g/kg resulted in silages with higher feeding value and fermentation quality, and substantially delayed deterioration after aerobic exposure, potentially qualifying SDA at this load for promotion of silage quality and delaying aerobic spoilage of early-harvested (low DM) rye forage.

  • Certain parts of 190 kinds of medicinal herbs and 171 kinds of original materials of food were extracted by methanol. The extracts were tested their microbial inhibition activities against several food spoilage microorganisms, Micrococcus luteus, Bacillus subtilis, Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Escherichia coli, Saccharomyces cerevisiae, Candida albicans, Penicillium citrinum, Aspergillus flavus and Aspergillus niger. The methanol extracts of Cornus officinalis, Evodia officinalis, Glycyrrhiza glabra, Salvia miltiorrhiza. Schizandrae fructus, Coptidis rhizoma, aroma hop and bitter hop were shown inhibitory effect on certain species of gram(+) bacteria. Aroma hop and bitter hop were shown inhibitory effect on certain species of gram(-) bacteria. The methanol extract of Salvia miltiorrhiza exhibited a strong antibacterial activities. It was purified by solvent fractionation, silicagel column chromatography, prep. TLC, prep. HPLC. The purified active substance was identified as cryptotanshinone by EIMS, $1^H-NMR,\;{13}^C-NMR$ and DEPT. Cryptotanshinone showed a strong antibacterial activity against gram positive bacteria $(MIC\;:\;3.91{\sim}62.50\;{\mu}g/mL)$. Especially, this compound was the most strong activity against Bacillus subtilis $(MIC\;:\;3.91\;{\mu}g/mL)$.

Antifungal Activity of Fistulosides, Steroidal Saponins, from Allium fistulosum L (대파(Allium fistulosum L)로부터 fistulosides의 분리와 분리 물질의 항진균 활성)

  • Allium fistulosum L. (Welsh onion) is a perennial herb that is widely cultivated throughout the world, especially in China, Japan and Korea. Although, various activities were reported, little is known about antimicrobial activity of A. fistulosum L.. In this study, strong antimicrobial substances, fistuloside A, B, and C were isolated from the edible parts of A. fistulosum L. and their antimicrobial activity was evaluated with pathogenic- or food-spoilage microorganism based on disk-diffusion assay, minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) determination. Fistuloside A and fistuloside C showed strong antifungal activity and anti-Proteus activity, while fistuloside B is effective to only fungi. The fistuloside C showed a prominent antifungal activity with $3.1{\sim}6.2{\mu}g/ml$ of MIC and MFC. Our results indicated that fistuloside C has a prominent antifungal activity and support the use of A. fistulosum to treat microbial infection.

The Distribution of Microflora in the Viscera of Sardine, Sardinops melanosticta by the storage Temperature (저장온도에 따른 정어리 내장 미생물상의 변화에 대하여)

  • As the putrefaction of fish is greatly relied on the microorganisms inhabited in the viscera of them, we investigated the microfloral changes in the viscera of sardine, Sardinops melanosticta, which has been caught a lot in adjacent sea of Korea but showed rapid spoilage, after storages with various temperature. The following results were obtained. Viable cell counts at $25^{\circ}C$ of the viscera of sardine were $1.6\times10^5/g$ at the fresh sample, $1.5\times10^5/g$ at the frozen sample, $2.9\times10^8/g$ at the spoiled samples. The most predominant microbial genera from the fresh sardine were Moraxella spp.($31.4\%$) and Pseudomonas spp.($28.6\%$), but Enterobacteriaceae($83.1\%$) was in spoiled sample. While Moraxella spp.($46.2\%$) and Flavobacterium-Cytophaga($21.0\%$) were predominant in the frozen sample and Enterobacteriacear($69.6\%$) was in the thawed-spoiled sample. The rates of proteolytic enzyme producing bacteria were $20\%$ in the fresh sample, $22\%$ in the frozen sample but the rates were increased to $52\%,\;29\%$ in the spoiled sample and the thawed-spoiled sample respectively.

Radurization of the Microorganisms Contaminated in Beef (우육에 오염된 미생물의 감마선 살균)

  • The effects of gamma irradiation (1, 3 and 5 kGy) and packaging methods (air and vacuum) on the growth of microorganisms contaminated in beef was investigated during storage at different temperatures (-20, 4 and $25^{\circ}C$). The initial microbial population of beef was $8.0{\sim}10^2\;CFU/g$ in total aerobic bacteria, $2.0{\times}10^2\;CFU/g$ in total lactic acid bacteria, $8.0{\times}10^1\;CFU/g$ in molds, $6.0{\times}10^2\;CFU/g$ in Pseudomonas sp. and $7.0{\times}10^2\;CFU/g$ in coliforms, respectively. Gamma irradiation at 5 kGy completely eliminated pathogenic bacteria in beef. Gamma irradiation at such dose and subsequent storage at less than $4^{\circ}C$ could ensure hygienic quality prolong the microbiological shelf-life resulting from the reduction of spoilage microorganisms. The different packaging methods of beef caused negligible changes in the growth of microorganisms during storage.

  • In order to examine the antimicrobial effect on dairy processing facilities and products, Prunus mume extinct (PME) was applied to the pilot plant system of dairy industry and yogurt, PME showed thermal and pH stability in the wide spectrum of temperature ($40{\sim}150^{\circ}C$) and pH ($4{\sim}10$) and remarkable antimicrobial activities against dairy spoilage microorganisms. As the result of aseptic treatment of dairy processing facilities with PME microbial colony including coliform bacteria was not detected canpore to those detected in the control. In the level of PME concentration which inhibit the growth of putrefactive microorganisms we could produce yogurt with good scores of sensory evaluation.

Improved Preservation and Shelf-life Extension of Fish Cake by Modified Atmosphere Packaging (변형기체포장에 의한 어묵의 저장성 향상 및 유통기한 연장)

  • Modified atmosphere packaging (MAP) of vacuum, 60% $N_2/40%$ $CO_2$, 100% $N_2$ and 100% $CO_2$ were examined to preserve fried fish cakes and extend their shelf life. When product packages in air were stored at 5, 10, and $15^{\circ}C$ with measurement in quality attributes, aerobic bacterial growth was found to be primary quality factor to determine the shelf life of the product. Huang's model was adopted to obtain the parameters for use in shelf life determination for a variety of packaging and storage conditions. Temperature dependence expressed in Arrhenius equation showed activation energies of 139.4 and 41.8 kJ/mol for inverse of lag time and growth rate of the microbial spoilage, respectively. MAP of 100% $CO_2$ could double the product shelf life at $10^{\circ}C$ compared to air package. High $CO_2$ concentration MAP at low temperature was found to be effective to prolong the shelf life.
