생물학적 처리공정에서 응집제 사용에 따른 미생물 활성도 영향 (Effect of Microbial Activity by Using the Coagulants in the Biological Treatment Process)

  • Alum은 단분자성 알루미늄이 주종이었고, PAC는 고분자성 알루미늄을 함유하고 있었다. Alum과 PAC 모두 응집제 주입에 따른 총인 제거는 향상되었다. 응집제 사용은 미생물 활성도에 영향을 미치는 것으로 나타났다. Alum에 비해 PAC가 미생물 활성도에 대한 영향을 적게 미치는 것으로 나타났다. 그리고 슬러지 반송에 따른 미생물 활성도와 개체수에 대한 영향은 미미한 것으로 나타났다.

Population Structure of Fusarium graminearum from Maize and Rice in 2009 in Korea

  • We performed diagnostic PCR assays and a phylogenetic analysis using partial sequences of TEF1 (translation elongation factor-1) to determine the trichothecene chemotypes and genetic diversity of F. graminearum isolates from maize and rice samples collected in 2009 in Korea. PCR using a species-specific primer set revealed a total of 324 isolates belonging to the putative F. graminearum species complex. PCR with trichothecene chemotypespecific primers revealed that the nivalenol (NIV) chemotype was predominant among the fungal isolates from rice (95%) in all provinces examined. In contrast, the predominant chemotype among the corn isolates varied according to region. The deoxynivalenol (DON) chemotype was found more frequently (66%) than the NIV chemotype in Gangwon Province, whereas the NIV chemotype (70%) was predominant in Chungbuk Province. Phylogenetic analysis showed that all DON isolates examined were clustered into lineage 7, while the NIV isolates resided within lineage 6 (F. asiaticum). Compared with previous studies, the lineage 6 isolates in rice have been predominantly maintained in southern provinces, while the dominance of lineage 7 in maize has been evident in Gangwon at a slightly reduced level.

Symbiobacterium toebii Sp. nov., Commensal Thermophile Isolated from Korean Compost

  • A thermophilic nonspore-forming rod isolated from hay compost in Korea was subjected to a taxonomic study. The microorganism, designated as $SC-1^T$, was identified as a nitrate-reducing and nonmotile bacterium. Although the strain was negatively Gram-stained, a KOH test showed that the strain $SC-1^T$ belonged to a Gram-positive species. Growth was observed between 45 and $70^{\circ}C$. The optimal growth temperature and pH were $60^{\circ}C$ and pH 7.5, respectively. The G+C content of the genomic DNA was 65 mol% and the major quinone types were MK-6 and MK-7. A phylogenetic analysis based on 16S rDNA sequences revealed that the strain $SC-1^T$ was most closely related to Symbiobacterium thermophilum. However, the level of DNA-DNA relatedness between strain $SC-1^T$ and the type strain for Symbiobacterium thermophilum was approximately 30%. Accordingly, on the basis of the phenotypic traits and molecular systematic data, the strain $SC-1^T$ would appear to represent a new species within the genus Symbiobacterium. The type strain for the new species is named $SC-1^T$ ($=KCTC\;0307BP^T;\;DSM15906^T$).

Estimation of Dominant Bacterial Species in a Bench-Scale Shipboard Sewage Treatment Plant

  • Recently, an innovative method for wastewater treatment and nutrient removal was developed by combining the sequence batch reactor and membrane bioreactor to overcome pollution caused by shipboard sewage. This system is a modified form of the activated sludge process and involves repeated cycles of mixing and aeration. In the present study, the bacterial diversity and dominant microbial community in this wastewater treatment system were studied using the MACROGEN next generation sequencing technique. A high diversity of bacteria was observed in anaerobic and aerobic bioreactors, with approximately 486 species. Microbial diversity and the presence of beneficial species are crucial for an effective biological shipboard wastewater treatment system. The Arcobacter genus was dominant in the anaerobic tank, which mainly contained Arcobacter lanthieri (8.24%), followed by Acinetobacter jahnsonii (5.81%). However, the dominant bacterial species in the aerobic bioreactor were Terrimonas lutea (7.24%) and Rubrivivax gelatinosus (4.95%).

Evaluation of SERS Nanoparticles to Detect Bacillus cereus and Bacillus thuringiensis

  • Purpose: This research evaluated five types of nanoparticles to develop a surface-enhanced Raman spectroscopy (SERS) method for the rapid detection of two Bacillus species (Bacillus cereus and Bacillus thuringiensis) that are commonly found on fresh produce, which can cause food poisoning. Methods: Bacterial concentrations were adjusted to a constant turbidity, and a total of $30{\mu}L$ of each Bacillus cell suspension was prepared for each nanoparticle. A point-scan Raman system with laser light source of wavelength 785 nm was used to obtain SERS data. Results: There was no qualitative difference in the SERS data of B. cereus and B. thuringiensis for any of the five nanoparticles. Three gold nanoparticles, stabilized in either citrate buffer or ethanol, showed subtle differences in Raman intensities of two Bacillus species at $877.7cm^{-1}$. Conclusions: Among the three types of nanoparticles, the gold nanoparticles stabilized in citrate buffer showed the lowest standard deviation, followed by gold nanoparticles stabilized in ethanol. This result supports the potential application of gold nanoparticles for SERS-based detection of B. cereus and B. thuringiensis.

후자린산(Fusaric acid) 생성 Fusarium 종의 신속 검출 PCR (Rapid Detection Method for Fusaric Acid-producing Species of Fusarium by PCR)

  • 후자린산은 Fusarium이 생성하는 독소로서 다른 Fusarium 독소와 함께 독성을 증진시킬 수 있다. 후자린산 독소를 특이적으로 검출하기 위해 후자린산의 생합성유전자 중 전사인자인 FUB10을 증폭하는 프라이머를 제작하였다. Fub10-f와 Fub10-r 프라이머쌍으로 PCR을 수행했을 때, F. oxysporum, F. proliferatum, F. verticillioides, F. anthophilum, F. bulbicola, F. circinatum, F. fujikuroi, F. redolens, F. sacchari, F. subglutinans, F. thapsinum에서 약 550 bp의 단일밴드가 증폭되었으며 이들은 모두 후자린산을 생성하는 것으로 알려졌다. 반면 트라이코쎄신을 생성하는 종에서는 FUB10 특이 밴드가 증폭되지 않았다. 후자린산은 푸모니신을 생성하는 종에서 함께 생성될 수 있기 때문에 FUB10 프라이머와 푸모니신 생합성유전자인 FUM1 프라이머를 이용한 multiplex PCR을 수행하였다. 그 결과 푸모니신 생성종인 F. proliferatum과 F. verticillioides에서 밴드가 모두 증폭되었으며 이는 두 가지 독소를 생성할 수 있는 종에서 동시 검출이 가능함을 시사하였다.

Description of 17 unrecorded bacterial species isolated from freshwater showing antibiotic resistance in Korea

  • As part of the research program "2018 Rapid screening and identification of freshwater microorganisms using MALDI-TOF/MS library" freshwater samples were collected from a branch of the Nakdong River. Almost 300 antibiotic-resistant bacterial strains were isolated from freshwater samples and subsequently identified by 16S rRNA gene sequencing. Seventeen strains among the isolates shared high 16S rRNA gene sequence similarity (>99.0%) with known species that were not previously recorded in Korea, and each of the isolates also formed a robust phylogenetic clade with the closest species. These species were phylogenetically diverse, belonging to four phyla, seven classes, 10 orders, and 13 genera. At the genus and class level, the previously unrecorded species belonged to Rhodovarius, Xanthobacter, and Shinella of the class Alphaproteobacteria; Ottowia, Simplicispira, and Zoogloea of Betaproteobacteria; Pseudomonas, Acinetobacter, and Shewanella of Gammaproteobacteria; Arcobacter of Epsilonproteobacteria; Sphingobacterium of Sphingobacteriia; Trichococcus of Bacilli; and Leucobacter of Actinobacteria. The previously unrecorded species were further characterized by examining their gram-staining, colony and cell morphology, biochemical properties, and phylogenetic position.

A report of 20 unrecorded bacterial species in Korea, isolated from soils of coastal areas in 2022

  • To obtain unrecorded bacterial species in Korea, various soils of coastal areas were collected from the Republic of Korea in 2022. After plating the samples on marine agar and incubating aerobically and anaerobically, approximately 1,700 bacterial strains were isolated and identified using 16S rRNA gene sequences. A total of 20 strains showed ≥98.7% 16S rRNA gene sequence similarity with validly published bacterial species but not reported in Korea, indicating they are unrecorded bacterial species in Korea. The unrecorded bacterial strains belonged to four phyla, six classes, 15 orders, 16 families, and 19 genera which were assigned to Blastomonas and Sphingomonas of the class Alphaproteobacteria; Pseudidiomarina, Kushneria, Salinicola, and Salinisphaera of the class Gammaproteobacteria; Evansella, Virgibacillus, and Paenibacillus of the class Bacilli; Cyclobacterium of the class Cytophagia; Pedobacter of the class Sphingobacteriia; and Demequina, Ornithinimicrobium, Blastococcus, Jatrophihabitans, Kineococcus, Glaciihabitans, Aeromicrobium and Streptomyces of the class Actinomycetes. The details of the 20 unreported species, including Gram reaction, morphology, biochemical characteristics, and phylogenetic position are also provided in the description of the strains.

낙동강 담수 미세조류 군집 분포 변화 (Distribution Changes of Freshwater Microalgae Community in the Nakdonggang River, Korea)

  • Distribution changes in microalgae communities were studied in the Nakdonggang River at two sampling stations (St.1 Gyeongcheongyo Bridge (GB) and St.2 Daedong Wharf (DW)) at monthly intervals from January 2021 to November 2021. A total of 83 taxa included 82 species, 1 forma, belonging to 49 genera, 32 families, 21 orders, and 8 classes. The most important groups were Bacillariophyta and Chlorophyta. The number of species ranged from 5 to 24 in GB, and from 9 to 21 taxa in DW. The contribution of Bacillariophyta to the total species richness was the highest during all survey periods, and Chlorophyta yielded the next highest value in the study area. The dominant taxa were Aulacoseira ambigua, A. ambigua f. japonica, and Ulnaria acus in this study. Cluster analysis and non-metric multidimensional scaling (nMDS) analysis based on Bray- Curtis similarity identified 4 major groups, which corresponded to microalgae assemblages and their characteristic species. Correlation was analyzed through the CCA analysis. It was found that there was a correlation between the microalgae and environmental factors. It was revealed that the divided groups were distinguished because of the differences by the survey period. Therefore, seasonal change was judged as a major factor affecting the distribution of microalgae communities.

Production of L-DOPA by Thermostable Tyrosine Phenol-lyase of a Thermophilic Symbiobacterium Species Overexpressed in Recombinant Escherichia coli

  • A thermostable tyrosine phenol-lyase gene of a thermophilic Symbiobacterium species was cloned and overexpressed in Escherichia coli in order to produce the biocatalyst for the synthesis of 3, 4-dihy-droxyphenyl-L-alanine (L-DOPA). The substrates used for the synthetic reaction were pyrocatechol, so-dium pyruvate, and ammonium chloride. The enzyme was stable up to $60^{\circ}C$, and the optimal temperature for the synthesis of L-DOPA was $37^{\circ}C$ . The optimal pH of the reaction was about 8.3. Enzyme activity was highly dependent on the amount of ammonium chloride and the optimal concentration was estimated to be 0.6 M. In the case of pyrocatechol, an inactivation of enzyme activity was observed at con-centrations higher than 0.1 M. Enzyme activity was increased by the presence of ethanol. Under op-timized conditions, L-DOPA production was carried out adding pyrocatechol and sodium pyruvate to the reaction solution intermittently to avoid substrate depletion during the reaction. The concentration of L-DOPA reached 29.8 g/l after 6 h, but the concentration didn t increase further because of the formation of byproducts by a non-enzymatic reaction between L-DOPA and pyruvate.
