In current animal production in Japan, a large surplus of nitrogen and phosphorus is given to animals as their feed which are mostly imported from outside of our own country. Today, an excess of nitrogen and phosphorus from animal manure has been spread out of the area of animal production and the surroundings. These components have become the major reason for eutrophication of ground, surface and inland water. Nutritional studies for the reduction of nitrogen and phosphorus from animal waste has been done by many researchers. The reduction of excess protein in animal feed and the supplementation of deficient essential amino acids to feed have a possibility to increase the biological value of feed and to reduce nitrogen excretion, especially, via urine. The use of phytase activity to degrade phytate and to release utilizable inorganic phosphorus make it possible to cut an excess supply of feed additive inorganic phosphorus and to reduce phosphorus excretion from animal waste.
Hexavalent chromium in aqueous solutions was successfully removed via sorption and reduction in the presence of waste leaves. Cr(VI) removal followed a first-order reaction, and removal rates were proportional to the amount of waste leaves used in the tests. Most of Cr(VI) were removed via sorption in early stages of the tests, but the reduction reaction played a significant role in Cr(VI) removal later. Solution pHs were continuously decreased due to the microbial activity, which was induced from the microorganisms attached on waste leaves. The decreased solution pHs further enhanced the sorption and reduction of Cr(VI). To characterize the microorganisms found in the tests, a denaturing gradient gel electrophoresis (DGGE) method was used. The majority of microorganisms were composed of Bacillus sp. which can reduce Cr(VI). Thus, waste leaves can be effective reactive media for the treatment of Cr(VI) in the subsurface.
To understand the degradation processes of organic matter related to sulfate reduction by Sulfate Reduction Bacteria (SRB) in the tidal flat sediments of Hwang-do and Sogeun-ri, Tae-an Peninsula in Chungnam-do, biogeochemical characteristics were analyzed and highlighted using specific microbial biomarkers. The organic geochemical parameters (TOC, ${\delta}^{13}C_{org}$, C/N ratio, long-chain-n-alkane) indicate that most of the organic matter has been derived from marine phytoplankton and bacteria in the fine-grained sediment of Sogeun-ri, although terrestrial plant components have occasionally been incorporated to a significant degree in the coarse-grained sediment of Hwang-do. The concentration of sulfate in pore water is a constant tendency with regard to depth profile, while methane concentration appears to be slightly different with regard to depth profile at the two sites. Especially, the sum of bacteria fatty acid (a-C15:0 + i-C15:0 + C16:1w5) confirms that the these concentrations in Sogeun-ri are related to the degradation of Benzene, Toluene, Ethylbenzene and Xylene (BTEX) compounds from the crude oil retained in the sediments as a result of the Hebei Spirit oil-spill accident in 2007. The methane-related microbial communities as shown by lipid biomarkers (crocetane, PMI) are larger in some sedimentary sections of Hwang-do than in the Sogeunri tidal flat. These findings suggest that methane production by microbiological processes is clearly governed by SRB activity along the vertical succession in organic-enriched tidal flats.
Objective: Aim of this study was to evaluate the influence of freeze-dried acid whey addition and the use a game meat (fallow deer) on a microbial content and the biogenic amines formation in dry fermented sausages. Methods: The experiment involved dry fermented sausages made in two variants from beef and from fallow deer. Each variant was divided into five groups: control (with a curing mixture), reference (with a sea salt), sample with a liquid acid whey and two samples with the addition of reconstituted freeze-dried acid whey in different concentrations. Changes in lactic acid bacteria (LAB), Enterobacteriaceae content and biogenic amines content were determined. Results: The microbial content changes suggest that addition of acid whey slightly affected LAB content in comparison with the control and reference sample, but the addition of freezedried acid whey resulted in a reduction of Enterobacteriaceae content in the sausages from fallow deer or a similar level in the beef sausages compared with the control and reference sample. Both changes in LAB and Enterobacteriaceae content were more evident in case of sausages made from fallow deer. Addition of acid whey (liquid and a higher amount of freezedried) and use of fallow deer meat to produce the sausages resulted in a significant reduction of total biogenic amines content. Conclusion: The addition of acid whey (liquid and higher amount of freeze-dried) resulted in a significant reduction of total biogenic amines content in dry fermented sausages made from fallow deer meat.
Microbial enhanced oil recovery (MEOR) is being used more widely, and the biological contributions involved in MEOR need to be identified and quantified for the improvement of field applications. Owing to the excellent interfacial activity and the wide distribution of producing strains in oil reservoirs, lipopeptides have proved to be an essential part of the complex mechanisms in MEOR. In this study, crude lipopeptides were produced by a strain isolated from an indigenous community in an oil reservoir. It was found that crude lipopeptides can effectively reduce the IFT (interfacial tension) to 10-1~10-2 mN/m under high salinity without forming stable emulsions, and the wettability of natural sandstone can be enhanced (Amott index, from 0.36 to 0.48). The results of core flooding experiments indicate that an additional 5.2% of original oil in place can be recovered with a 9.5% reduction of injection pressure. After the shut-in period, the wettability of the core, the reduction of injection pressure, and the oil recovery can be improved to 0.63, 16.2% and 9.6%, respectively. In the microscopic flooding experiments, the crude oil in membrane, cluster, and throat states contribute nearly 90% in total of the additional oil recovery, and the recovery of membranestate oil was significantly enhanced by 93.3% after shut in. Based on the results in macro and pore scale, the IFT reduction and the wettability alteration are considered primary contributors to oil recovery, while the latter was more dominant after one shut-in period.
The characteristics of As and heavy metals depend on the oxidation/reduction condition of the soil environment. The most heavy metals are immobilized by the reduction condition whereas As, Fe and Mn become more soluble. Therefore this study estimated the stabilization efficiency of the agricultural paddy soil in the vicinity of the abandoned mine using a flooded column test including analysis of the soil solution, contaminants fractionation and rice grain. Limestone and steelmaking slag were used as amendments for stabilization of the contaminated soil. In an analysis of the soil solution, the mobile characteristics of Fe and Mn, which were used as electron acceptors of the microorganisms, were controlled by increasing the pH by adding alkali amendments. This means that the contaminants combined with Fe and Mn can be stable under flooded reduction condition. However, the concentrations of cationic heavy metals (Cd, Pb, and Zn) were also decreased without amendments because the carbonates produced from microbial respiration increased the pH of the soil solution. In the amended soil, the specific sorbed fraction of As and carbonates fraction of heavy metals were increased when compared to the control soil at the end of the column test. Especially in heavy metals, the increase of carbonates fraction seems to be influenced by alkali amendments rather than microbial respiration. Because of the stabilization effect in the flooded paddy soil, the contents of As and Zn in rice grain from amended soil were lower than that of the control soil. But additional research is needed because of the relatively higher Pb content identified in the rice grain from the amended.
Biotechnological treatment of sulfate- and metal-ions-containing acidic wastewaters from mining and metallurgical activities utilizes sulfate-reducing bacteria to produce sulfide that can subsequently precipitate metal ions. Reducing sulfate at a low pH has several advantages above neutrophilic sulfate reduction. This study describes the effect of sulfide removal on the reactor performance and microbial community in a high-rate sulfidogenic gas-lift bioreactor fed with hydrogen at a controlled internal pH of 5. Under sulfide removal conditions, 99% of the sulfate was converted at a hydraulic retention time of 24 h, reaching a volumetric activity as high as 51 mmol sulfate/l/d. Under nonsulfide removal conditions, <25% of the sulfate was converted at a hydraulic retention time of 24 h reaching volumetric activities of <13 mmol sulfate/l/d. The absence of sulfide removal at a hydraulic retention time of 24 h resulted in an average $H_2S$ concentration of 18.2 mM (584 mg S/I). The incomplete sulfate removal was probably due to sulfide inhibition. Molecular phylogenetic analysis identified 11 separate 16S rRNA bands under sulfide stripping conditions, whereas under nonsulfide removal conditions only 4 separate 16S rRNA bands were found. This shows that a less diverse population was found in the presence of a high sulfide concentration.
Sealing of leakage in waterfront or water-retaining structures is one of the major issues in geotechnical engineering practices. With demands for biological methods as sustainable ground improvement techniques, bioclogging, defined as the reduction in hydraulic conductivity of soils caused by microbial activities, has been considered as an alternative to the chemical grout techniques for its economic advantages and eco-friendliness of microbial by-products. This study investigated the feasibility of bioaugmentation and biostimulation methods to induce fermentation-based bioclogging effect in coarse sands. In the bioaugmentation experiments, effects of various parameters and conditions, including grain size, pH, and biogenic gas generation, on hydraulic conductivity reduction were examined through a series of column experiments while Leuconostoc mesenteroides, which produce an insoluble biopolymer called dextran, was used as the model bacteria. The column test results demonstrate that the accumulation of bacterial biopolymer can readily reduce the hydraulic conductivity by three-to-four orders of magnitudes or by 99.9-99.99% in well-controlled environments. In the biostimulation experiments, two inoculums of indigenous soil bacteria sampled from waterfront embankments were prepared and their bioclogging efficiency was examined. With one inoculum containing species capable of fermentation and biopolymer production, the hydraulic conductivity reduction by two orders of magnitude was achieved, however, no clogging was found with the other inoculum. This implies that presence of indigenous species capable of biopolymer production and their population, if any, play a key role in causing bioclogging, because of competition with other indigenous bacteria. The presented results provide fundamental insights into the bacterial biopolymer formation mechanism, its effect on soil permeability, and potential of engineering bacterial clogging in subsurface.

The consumption of king oyster mushrooms has steadily increased owing to their unique flavor and delicate texture. This study extended the storage period of king oyster mushrooms (Pleurotus eryngii ) via gamma irradiation. Irradiated samples (0, 0.5, 1, 2, and 3 kGy) were stored at 4℃ with 80% relative humidity for up to 28 days, and the experiments were conducted every 7 day. Microbiological analysis revealed a dose-dependent reduction in total aerobic bacteria, Pseudomonas spp., and yeasts and molds, and gamma irradiation above 2 kGy effectively controlled microbial contamination for up to 28 days. At the 28th day of storage, the irradiated king oyster mushrooms exhibited delayed browning through the reduction of tyrosinase activity. Moreover, firmness reduction (%) was 80.59±1.89% for the non-irradiated group and 42.80±1.28, 34.57±1.13, 31.05±3.24, and 39.73±0.94% for the irradiated group (0.5, 1, 2, and 3 kGy), respectively. These results were supported by the scanning electron microscopy photos, which showed smaller pores in the irradiated group than in the non-irradiated group. This study demonstrated that 2 kGy of gamma irradiation effectively reduces microbial contamination and delays the browning and softening of king oyster mushrooms for up to 28 days.
Today, application of additives to replace cement in order to improve concrete mixes is widely promoted. Micro-silica is among the best pozzolanic additives which can desirably contribute to the concrete characteristics provided it is used properly. In this paper, the effects of AP2RC and P1RB micro-silica gels on strength characteristics of normal concrete are investigated. Obtained results indicated that the application of these additives not only provided proper workability during construction, but also led to increased tensile, compressive and flexural strength values for the concrete during early ages as well as ultimate ones with the resulting reduction in the porosity lowering permeability of the micro-silica concrete. Furthermore, evaluation of microbial contamination of the mentioned gels showed the resultant contamination level to be within the permitted range.
