The global nematicides market is expected to continue growing. With an increasing demand for synthetic chemical-free organic foods, botanical nematicides are taking the lead as replacements. Consequently, in the recent years, there have been vigorous efforts towards identification of the active secondary metabolites from various plants. These include mostly glucosinolates and their hydrolysis products such as isothiocyanates; flavonoids, alkaloids, limonoids, quassinoids, saponins, and the more recently probed essential oils, among others. And despite their overwhelming potential, variabilities in quality, efficacy, potency and composition continue to persist, and commercialization of new botanical nematicides is still lagging. Herein, we have reviewed the history of botanical nematicides and regional progresses, the potency of the identified phytochemicals from the key important plant families, and deciphered some of the impediments involved in standardization of the active compounds in addition to the concerns over the safety of the purified compounds to non-target microbial communities.
This paper presents the complete genome sequence of a novel marine actinomycete, Streptomyces sp. MMBL 11-1. The genome of Streptomyces sp. MMBL 11-1 was obtained through next-generation sequencing using the PacBio Sequel system and Illumina platform provided by Macrogen, Korea. The assembled genome consists of five contigs, with a total length of 8,496,900 bp and a G+C content of 71.6%. The genome harbors multiple biosynthetic gene clusters (BGCs) associated with producing microbial natural products (MNPs). The comprehensive genomic information of this type of strain will serve as a valuable resource for identifying other marine actinomycetes strains.
The purpose of this study was to investigate the effects of the addition of various amounts (0, 1, 3, 5%) of Mokdanpi(P. suffruticosa) powder on the shelf life and the physicochemical, sensory characteristics of rice cakes and noodles to examine the possibility of using Mokdanpi as a natural food preservative for food products. The decrease in the moisture content of rice cake and noodle samples during storage was much less in Mokdanpi-added groups than the control group. The color L value increased and a and b values decreased in the rice cake samples, and color L value, b value decreased and a value increased in the noodle samples during storage. The microbial growth was less in Mokdanpi-added groups than the control group. In sensory evaluation, there were no significant differences among the control and 1, 3%-added groups in terms of color, flavor, taste, moistness, cohesiveness and overall quality; however, 5%-added groups received significantly less score than others (p<0.05). As the storage time increased, the sensory scores decreased significantly in the control and 1 %-added groups of both rice cakes and noodles (p<0.05), but no changes were observed in 3, 5%-added samples. The results suggested that Mokdanpi is effective in increasing the shelf life and inhibiting the staling of rice cakes and noodles.
Rhodotorula is a group of pigment-producing yeasts well known for its intracellular biosynthesis of carotenoids such as ${\beta}-carotene$, ${\gamma}-carotene$, torulene and torularhodin. The great potential of carotenoids in applications in food and feed as well as in health products and cosmetics has generated a market value expected to reach over $2.0 billion by 2022. Due to growing public concern over food safety, the demand for natural carotenoids is rising, and this trend significantly encourages the use of microbial fermentation for natural carotenoid production. This review covers the biological properties of carotenoids and the most recent findings on the carotenoid biosynthetic pathway, as well as strategies for the metabolic engineering methods for the enhancement of carotenoid production by Rhodotorula. The practical approaches to improving carotenoid yields, which have been facilitated by advancements in strain work as well as the optimization of media and fermentation conditions, were summarized respectively.
본 연구를 통하여 P. aeruginosa PR3를 이용하여 DOD를 생산하기 위해 저가의 기질로서 필리핀 너트유가 효과적으로 사용될 수 있음을 확인하였으며 배지에 첨가되는 여러 영양인자들의 영향을 조사하여 DOD 생산성을 크게 향상시킬 가능성이 있음도 확인하였다. 따라서 DOD 생산에 이용되는 올레산을 식물성오일로부터 별도의 생산과정을 거쳐 생산하지 않고 식물성오일자체를 직접 기질로 사용함으로서 PR3 균주를 이용하여 고부가가치의 DOD를 효율적으로 생산할 수 있다는 것을 확인하였다.
Oleamide (cis-9-octadecenamide) is endogenous primary amide of fatty acid that is produced in small amounts in animal brains. It is known to induce sleep and to lower temperature by destroying the lipid plasma membrane structure of cells, thereby disclosing gap junction channels. To develop a new biological production method for oleamide, a screening program was conducted to isolate a microorganism producing oleamide. Among 1,500 soil microorganisms tested, KK90378 exhibited a potent positive reaction with Dragendoff`s reagent, used to detect the primary amide of oleamide. KK90378 was identified as a Streptomyces species based on cultural and morpohological characteristics, the presence of diaminopimelic acid in the cell wall, and the sugar patterns for the whole-cell extrat. Streptomyces sp. KK90378 produced oleamide 3 days after culture at $28^{\circ}C$, pH 7.2 A series of purification steps, including hexane extraction, silica gel column, and preparative thin layer chromatographies, were performed for the purification of oleamide. A spectrophotometric analysis using $^1H$, $^13C$-NMR, and GC-MS confirmed that the chemical structure of the purified oleamide was identical to that of authentic oleamide.
조록나무는 제주도 및 일본 혼슈 이남, 중국 동남부, 타이완 등에 분포하는 조록나무과의 상록 교목으로, 항산화 및 tyrosinase, elastase의 억제에 효과적인 것으로 알려져 있지만 NO에 대한 억제 효능은 미미한 것으로 보고되었다. 이에 본 연구는 조록나무 잎 추출물(DL)에 biorenovation 생물 전환 기법을 적용하여 항 염증 활성을 증진 시키고자 수행되었다. 이들의 활성은 LPS로 자극된 RAW264.7 염증 모델에서 평가 되었으며 NO, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) 및 전 염증성 사이토카인에 대한 억제 실험이 수행되었다. 그 결과, biorenovation을 적용한 조록나무 잎 추출물(DLB)는 독성이 없는 농도에서 DL대비 향상된 NO와 prostaglandin E2 억제효능을 나타내었으며, 이들의 합성 효소인 iNOS 및 COX-2의 발현에도 유의한 억제 경향을 나타내었다. 또한 대표적인 전 염증성 사이토 카인인 tumor necrosis factor-α, Interleukin 6, Interleukin-1β 에서도 향상된 억제 효능을 확인 하였다. 이러한 결과를 근거로 우리는 biorenovation을 통해 DL의 항염증 효능이 개선될 수 있으며, DLB가 효과적인 천연 항염증 소재로 적용될 수 있음을 제시한다.
본 연구에서는 황토노각플러스 추출물의 생리활성 평가를 통해 화장품 원료로서의 가능성을 조사하였다. 황토노각 플러스 추출물에 대한 항산화 효과는 DPPH/ABTS 라디컬 소거능 활성 및 FRAP 분석, 세포 생존율은 MTT assay, 항염증 효과는 nitric oxide(NO) 생성억제 및 iNOS 단백질 발현 수준, 항균 효과는 총 3종 균주 Staphylococcus aureus, Staphylococcus epidermidis 및 Escherichia coli에 대한 한천 확산법을 측정하였다. 메탄올추출물(MN)과 열수추출물(WN)의 총폴리페놀 함량은 각각 2.92±0.01 mgGAE/g, 1.67±0.02 mgGAE/g로 나타났다. MN 추출물의 DPPH, ABTS 및 FRAP 값은 모든 농도에서 WN 추출물보다 높게 나타났다. RAW264.7 세포에 대한 두 추출물의 세포 독성은 관찰되지 않았다. 더욱이 1 mg/mL농도에서 MN과 WN 추출물의 NO 생성량은 11.69 μM과 20.4 μM로 측정되었으며, MN 추출물의 iNOS 단백질 발현 수준은 추출물 농도 의존적으로 억제되었다. 또한 MN 추출물은 S. epidermidis 균주에서만 항균효능을 보였다. 이러한 결과들로 황토노각플러스 추출물은 항산화, 항염, 항균효능을 지닌 천연물질 후보로써 가능성이 높다고 판단된다.
Isozyme PAGE에 의한 isozyme polymorphism 및 random amplified polymorphic DNA(RAPD) pattern의 비교를 통하여 한국에서 수집된 63개의 표고 버섯(Lentinus edodes)품종의 유연관계를 조사하여 보았다. NTSYS PC program을 통하여 비교에 사용되어진 3가지 isozyme 즉 esterase, peroxidase, acid phophatase 등은 각각 10, 7, 3개의 isozyme polymorphism type으로 나뉘어질 수 있었고, 이러한 3개의 isozyme type들은 group-average method를 이용하여 최종적으로 6개의 집단으로 구분되어질 수 있었다. 또한 RAPD를 통한 표고 품종간 유연관계 비교는 사용되어진 총 20개의 random primer 들중 3개의 primer(OPA 03, OPA 18, OPA 19)가 품종간 polymorphism을 보였고 사용되어진 전체 random primer의 PCR product들의 집단 분석을 통해 최종적으로 5개의 집단으로 구분할 수 있었다. 사용되어진 두 가지 방법 즉, isozyme polymorphism및 RAPD pattern에 의해 구분되어진 63개 표고 버섯 품종의 subgroup level에서의 절대적인 유사성은 관찰할 수 없었다. Yii-Mattila(1996) 등에 의하면 품종간 비교시 isozyme analyses에서와는 달리 RAPD analyses에서는 많은 수의 strain-specific band들이 얻어질 수 있고 이것이 두 방법간의 유사성을 관찰하지 못하게 하는 원인이라고 보고하였다.
Carotenoid는 isoprenoid 화합물로써 3-15개의 이중 결합이 결합된 긴 polyene 구조를 가지며, 이러한 구조적 특성에 의해 최대 흡수파장대가 결정된다. 카로티노이드는 전형적으로 $C_{40}$ 탄화수소 골격으로 이루어져 있으며, 그 중에는 산소를 포함하고 있는 작용기로 인해 cyclic 또는 acyclic의 크산토필을 형성하기도 한다. 현재 대부분의 세계 시장을 점유하고 있는 합성 카로티노이드의 대안을 찾기 위해 천연물(식물, 미생물, 갑각류 부산물) 유래의 카로티노이드를 생산 및 활용하기 위한 연구가 진행되고 있다. 그럼에도 불구하고, 오직 몇몇 카로티노이드(${\beta}-carotene$, lycopene, astaxanthin, canthaxanthin, lutein)만이 천연물 소재에서 분리 또는 발효되어 상업적으로 이용된다. 카로티노이드가 만성질환 예방 및 발암 억제 작용에 효과가 있음이 밝혀지면서 카로티노이드 시장이 급격히 증가하였다. 사료, 기능성 식품 및 의약품 소재로써의 카로티노이드의 중요성이 증가함에 따라 카로티노이드 생산법에 대한 연구가 진행되었으며, 이러한 관점에서 미생물 및 식물을 이용한 대사공학적 접근법에 의한 카로티노이드 대량생산법을 개발하게 되었다. 본 논문에서는 생산 균주, 대사공학 적용에 따른 대량 생산 방법, 생물학적 작용기전 및 산업적 이용을 중심으로 설명하고자 한다.
