The Effect of Barbaloin on LPS-stimulated Inflammatory Reaction in Mice Peritoneal Macrophages

  • Barbaloin is a major component of Aloe vera, which has been used for a laxative. Also, barbaloin is C-glucoside of aloe emodin anthrone which is founded in Aloe vera. Barbaloin has varieties of pharmacological activity such as inhibitory effects on inflammation, histamine release, cancer and microbial infection. But the effect of barbaloin on lipopolysaccharide (LPS)-stimulated macrophages has not been understood. In this study, we evaluated the effects of barbaloin against LPS-stimulated production of nitric oxide (NO), inflammatory cytokines and MAPKs activation in macrophage. We treated barbaloin (0.1, 1, 10, $100{\mu}M$) in LPS-stimulated mice peritoneal macrophage. Our results showed that barbaloin significantly inhibited production of NO and cytokines of tumor necrosis factor $(TNF)-{\alpha}$, interleukin (IL)-6, interleukin $(IL)-1{\beta}$ in LPS-stimulated peritoneal macrophage. Moreover, barbaloin inhibited the phosphorylation of ERK and JNK in a dose dependent manner. These results indicated that barbaloin could be useful for inflammatory diseases.

This document provides comprehensive guidelines for the proper shutdown and reactivation of UHPLC systems to prevent damage and maintain operational efficiency. • Shutdown: Remove the column and replace it with a union to avoid blockages. Flush the system with a compatible solvent mix, clean mobile phase reservoirs to prevent microbial growth, flush the pump with storage solvent, and clean the autosampler, including the needle and injection port. • Reactivation: Inspect the system for wear or damage, gradually reintroduce mobile phases starting with a weak solvent, reinstall the column securely, and perform system checks on baseline stability, pressure consistency, and detector performance. By adhering to these guidelines, laboratories can ensure the longevity and reliability of their UHPLC systems, maintaining high analytical performance and minimizing downtime. 반응표면분석법을 활용한 Pantoea agglomerans SRCM 119864의 Indole-3-acetic acid 생산 배지 최적화 (Optimization of the Indole-3-Acetic Acid Production Medium of Pantoea agglomerans SRCM 119864 using Response Surface Methodology)

  • 본 연구에서는 토양으로부터 분리한 Indole-3-acetic acid (IAA) 고생산 균주인 Pantoea agglomerans SRCM 119864의 IAA 생산량을 증가시키기 위해 반응표면분석법을 활용하여 배지 조성 최적화를 수행하였다. Plackett-Burman design (PBD)을 이용하여 전구체인 L-tryptophan 이외의 IAA 생산에 영향을 주는 배지 성분 11개의 영향을 조사하였으며, 통계학적 분석을 통해 IAA 최적생산을 위한 배지 인자로 sucrose, tryptone, sodium chloride를 선정하였다. PBD에서 선정된 3가지 인자와 L-tryptophan의 농도 최적화를 수행하기 위해 적은 실험수로도 최적값을 분석할 수 있는 hybrid design을 설계하였다. 실험 모형에서 예측한 P. agglomer- ans SRCM 119864 균주의 IAA 최적 생산을 위한 배지 조성과 농도는 sucrose 13.38 g/l, tryptone 18.34 g/l, sodium chloride 9.71 g/l, L-tryptophan 6.25 g/l로 분석되었으며, 이때의 IAA 생산량은 64.34 ±1.07 mg/l로 예측되었다. ANOVA 분석을 통하여 실험 모형의 통계학적 유의성과 적합성을 검증하였으며, 설계한 최적 조성배지에서 모델 검증실험을 수행하여 IAA 생산량을 측정한 결과, 예측된 IAA 생산량과 매우 유사함을 확인하였으며, 최종 최적화 수행을 통해 IAA 생산량을 기본 배지 대비 44.56% 증가시켰다. 본 연구를 통하여 토양에서 IAA를 고생산하는 균주를 선별하여 배지 조성 최적화를 수행하였으며, 이를 바탕으로 지속가능한 농업 및 작물 생산량 증대를 위한 산업화 기초자료로서 활용될 수 있을 것으로 기대된다.

Protective effects of Bacillus subtilis against Salmonella infection in the microbiome of Hy-Line Brown layers

  • Objective: This study investigated the effects of Bacillus subtilis CSL2 (B. subtilis CSL2) administration before Salmonella challenge on the fecal microbiota and microbial functionality of Hy-line Brown (HLB) laying hens. Methods: Fecal samples were collected from control (CON), Salmonella-infected (SAL) and Salmonella-infected, probiotic-treated (PRO) groups before and after Salmonella challenge for microbiome analysis using 16S rRNA gene pyrosequencing. Results: Infection with Salmonella led to decreased microbial diversity in hen feces; diversity was recovered with Bacillus administration. In addition, Salmonella infection triggered significant alterations in the composition of the fecal microbiota. The abundance of the phylum Firmicutes decreased while that of Proteobacteria, which includes a wide variety of pathogens, increased significantly. Bacillus administration resulted in normal levels of abundance of Firmicutes and Proteobacteria. Analysis of bacterial genera showed that Salmonella challenge decreased the population of Lactobacillus, the most abundant genus, and increased populations of Pseudomonas and Flavobacterium genera by a factor of 3 to 5. On the other hand, Bacillus administration caused the abundance of the Lactobacillus genus to recover to control levels and decreased the population of Pseudomonas significantly. Further analysis of operational taxonomic units revealed a high abundance of genes associated with two-component systems and secretion systems in the SAL group, whereas the PRO group had more genes associated with ribosomes. Conclusion: The results of this study indicate that B. subtilis CSL2 administration can modulate the microbiota in HLB laying hens, potentially acting as a probiotic to protect against Salmonella Gallinarum infection.

신경회로망을 이용한 순환식 돈분폐수 처리시스템의 모니터링

  • 본 연구에서는 순환식 돈분 폐수 처리 시스템에서의 미생물 분포에 따른 폐수 처리 효과를 모델링하기 위해 신경회로망과 PCA를 이용하는 새로운 방법을 제안하였다. PCA 분석 결과를 바탕으로 신경회로망의 최적 입력 조건을 찾고, 실측 데이터를 이용하여, 폐수 처리 시스템의 각 탱크를 별도로 학습함으로써 비교적 적은 수의 데이터에도 불구하고 정확한 모델링 결과를 얻었다. 제안한 시스템은 폐수 처리 시스템의 효과적인모니터링 시스템으로 사용할 수 있으며, 향후 실제 돈분 처리 시스템에서 원하는 기준의 방류수를 얻기 위한 최적의 입력조건 (미생물밀도 등)을 결정하는데 있어서 에뮬레이터로 사용될 수 있을 것으로 기대된다.

Bacterial Communities of Biofilms Sampled from Seepage Groundwater Contaminated with Petroleum Oil

  • The diesel-degrading activities of biofilms sampled from petroleum-contaminated groundwaters in urban subway drainage systems were examined in liquid cultures, and the microbial populations of the biofilms were characterized by denaturing gel gradient electrophoresis (DGGE) and 16S rDNA sequence analysis. Biofilm samples derived from two sites (19 K and 20 K) at subway Station N and Station I could degrade around $80\%$ of applied diesel within 20 and 40 days, respectively, at $15^{\circ}C$, and these results were strongly correlated with the growth patterns of the biofilms. The closest phylogenetic neighbor of a dominant component in the 19 K biofilm was Thiothrix fructosivorans strain Q ($100\%$ similarity). Four dominant strains in the 20 K biofilm were closely related to Thiothrix fructosivorans strain Q ($100\%$ similarity), Thiothrix sp. CC-5 ($100\%$ similarity), Sphaerotilus sp. IF14 ($99\%$ similarity), and Cytophaga-Flexibacter-Bacterioides (CFB) group bacterium RW262 ($98\%$ similarity). Three dominant members in the Station I biofilms were very similar to uncultured Cytophagales clone CRE-PA82 ($91\%$ similarity), Pseudomonas sp. WDL5 ($97\%$ similarity), and uncultured CFB group bacterium LCK-64 ($94\%$ similarity). The microbial components of the biofilms differed depending on the sampling site. This is the first report on the isolation of clones highly similar to Thiothrix fructosivorans and Thiothrix sp. from biofilms in petroleum-polluted groundwaters, and the first evidence that these organisms may play major roles in petroleum degradation and/or biofilm-development.

Enzymatic Transformation of Ginsenoside Rb1 by Lactobacillus pentosus Strain 6105 from Kimchi

  • Ginsenoside (ginseng saponin), the principal component of ginseng, is responsible for the pharmacological and biological activities of ginseng. We isolated lactic acid bacteria from Kimchi using esculin agar, to produce ${\beta}$-glucosidase. We focused on the bio-transformation of ginsenoside. Phylogenetic analysis was performed by comparing the 16S rRNA sequences. We identified the strain as Lactobacillus (strain 6105). In order to determine the optimal conditions for enzyme activity, the crude enzyme was incubated with 1 mM ginsenoside Rb1 to catalyse the reaction. A carbon substrate, such as cellobiose, lactose, and sucrose, resulted in the highest yields of ${\beta}$-glucosidase activity. Biotransformations of ginsenoside Rb1 were analyzed using TLC and HPLC. Our results confirmed that the microbial enzyme of strain 6105 significantly transformed ginsenoside as follows: Rb1${\rightarrow}$gypenoside XVII, Rd${\rightarrow}$F2 into compound K. Our results indicate that this is the best possible way to obtain specific ginsenosides using microbial enzymes from 6105 culture.

MICROBIAL COLONIZATION AND DIGESTION OF FEED MATERIALS IN CATTLE AND BUFFALOES I. GUINEA GRASS

  • An experiment was conducted to determine whether there were any apparent differences in the microbial population, colonization pattern and digestion of guinea grass in situ, between cattle and swamp buffalo. Percentage losses in dry matter (DM), nitrogen (N) and neutral detergent fibre (NDF) of guinea grass were significantly (p<0.01) higher when incubated in the rumen of buffalo than in cattle. Buffalo also showed significantly (p<0.05) faster degradation rates than cattle for each grass component (DM, N, DNF). Light microscopy and SEM examination of the incubated grass materials showed that there were no apparent differences in the pattern of bacterial and fungal invasion and colonization of the grass materials between cattle and buffalo. Attachment of bacteria and fungal zoospores on the grass fragments occurred at 15 min after rumen incubation. After 3 h of rumen incubation, dense population of bacteria was observed in the thin-walled mesophyll and parenchyma tissues, whereas root-like fungal rhizoids were observed in both thin-walled and thick-walled cells. By 6 h, eroded zones were apparent in the thin-walled tissues and in thick-walled tissues with profuse rhizoids. After 24. 48 and 72 h of rumen incubation, most thin-walled tissues were degraded leaving mostly the thick-walled tissues. The predominant bacteria were the curved rods resembling Butyrivibrio sp., the thick rods resembling Fibrobacter sp., the diplococcoids resumbling Ruminococcus sp. And spirochetes. Fungi were predominantly those with spherical or oval sporangia. Fusiform sporangia with acuminate apices which resembled Ruminomyces sp. Were of lesser occurrence. Few protozoa were found on the grass fragments at all incubation times.

Helicobacter pylori의 생육에 영향을 미치는 성분과 식품첨가물에 관한 연구 (Studies on Food Components and Food Additives Affecting the Growth Patterns of Helicobacter pylori)

  • The purpose of this study was to choose prinicipal food components contained in diet foods and food additives used for manufacturing processed foods and elucidate their in vivo effects on the growth pattern of Helicobacter pylori. To do this the antibacterial effects of various sources of carbon nitrogen and mineral as an effect agent on Helicobacter pylori were first assessed based upon bacterial growth degree. results show that the source of carbon tested had different effects on bacterial growth of Helicobacter pylori. It was revealed that a promotional effect of monosaccharides resulted in enhanced growth of Helicobacter pylori compared with disaccharides and polysacchrides, in particular glucose was observed to be most effective in growth of Helicobacter pylori among monosaccharides teste whereas mannose to hinder the growth of Helicobacter pylori. Polyols such as sorbitol mannitol maltitol and xylitol was however observed to show no promotion or suppression effect on growth of Helicobacter pylori. Apart from these the sources of amino acid and inorganic nitrogen were chosen and tested to assess the promotion or suppression effect of nitrogen sources on growth of helicobacter pylori. It was found that amino acid such as lysine showed its promotion effect on the growth of Helicobacter pylori while arginine (NH4)2SO4 and NH4Cl showed no effect on its growth. Ammoia and urea were however observed to have a positive effect on the growth of Helicobacter pylori. Among these effect agents lysine and methionine were revealed to show the most positive effect on growth of Helicobacter pylori. Minerals such as MgSO4 KH2PO4 and MgCl2 appered to exert their positive growth effects whereas CaCl2 and CaSo4 had a little effect. In addition FeSO4 FeCl2 and FeCl3 brought suppression on the growth of helicobacter pylori. In studies of the growth of Helicobacter pylori by food additives ascorbic acid showed extreme suppression on its growth,. Sodium nitrate and sodium chloride were also found to be of negative effect on the growth of Helicobacter pylori in rder of degree whereas tocopherol had nothing to do with microbial growth.

Differentiation of Beef and Fish Meals in Animal Feeds Using Chemometric Analytic Models

  • Purpose: The research presented in this paper applied the chemometric analysis to the near-infrared spectral data from line-scanned hyperspectral images of beef and fish meals in animal feeds. The chemometric statistical models were developed to distinguish beef meals from fish ones. Methods: The meal samples of 40 fish meals and 15 beef meals were line-scanned to obtain hyperspectral images. The spectral data were retrieved from each of 3600 pixels in the Region of Interest (ROI) of every sample image. The wavebands spanning 969 nm to 1551 nm (across 176 spectral bands) were selected for chemometric analysis. The partial least squares regression (PLSR) and the principal component analysis (PCA) methods of the chemometric analysis were applied to the model development. The purpose of the models was to correctly classify as many beef pixels as possible while misclassified fish pixels in an acceptable amount. Results: The results showed that the success classification rates were 97.9% for beef samples and 99.4% for fish samples by the PLSR model, and 85.1% for beef samples and 88.2% for fish samples by the PCA model. Conclusion: The chemometric analysis-based PLSR and PCA models for the hyperspectral image analysis could differentiate beef meals from fish ones in animal feeds.
